Pharmacy Faculty Articles and Research

Adenylyl cyclases (ACs) catalyze the conversion of ATP to the ubiquitous second messenger cAMP. Mammals possess nine isoforms of transmembrane ACs, dubbed AC1-9, that serve as major effector enzymes of G protein-coupled receptors. The transmembrane ACs display varying expression patterns across tissues, giving potential for them having a wide array of physiologic roles. Cells express multiple AC isoforms, implying that ACs have redundant functions. Furthermore, all transmembrane ACs are activated by Gαs so it was long assumed that all ACs are activated by Gαs-coupled GPCRs. AC isoforms partition to different microdomains of the plasma membrane and form …

The runoff or leaching of nitrogen fertilizers from monoculture turfgrass lawns contri-butes to water pollution, and such lawns are susceptible to insect pests and provide few resources for pollinators. One approach to creating more sustainable lawns is to incorporate white clover (Trifolium repens L.), a nitrogen-fixing legume, into grass seed mixtures or existing turfgrass swards. “Dutch” white clover (DWC), a ubiquitous landrace, forms non-uniform clumps when intermixed with turfgrasses, thus it is often considered to be a lawn weed. Recently, several dwarf varieties of white clover have been selected for their small leaf size and low growth habit, allowing …

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has necessitated the rapid development of antibody-based therapies and vaccines as countermeasures. Here, we use cryoelectron microscopy (cryo-EM) to characterize two protective anti-SARS-CoV-2 murine monoclonal antibodies (mAbs) in complex with the spike protein, revealing similarities between epitopes targeted by human and murine B cells. The more neutralizing mAb, 2B04, binds the receptor-binding motif (RBM) of the receptor-binding domain (RBD) and competes with angiotensin-converting enzyme 2 (ACE2). By contrast, 2H04 binds adjacent to the RBM and does not compete for ACE2 binding. Naturally occurring sequence variants of SARS-CoV-2 and corresponding neutralization escape …

The host cell serine protease TMPRSS2 is an attractive therapeutic target for COVID-19 drug discovery. This protease activates the Spike protein of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and of other coronaviruses and is essential for viral spread in the lung. Utilizing rational structure-based drug design (SBDD) coupled to substrate specificity screening of TMPRSS2, we have discovered covalent small-molecule ketobenzothiazole (kbt) TMPRSS2 inhibitors which are structurally distinct from and have significantly improved activity over the existing known inhibitors Camostat and Nafamostat. Lead compound MM3122 (4) has an IC

Nontuberculous mycobacteria, including those in the Mycobacterium avium complex (MAC), constitute an increasingly urgent threat to global public health. Ubiquitous in soil and water worldwide, MAC members cause a diverse array of infections in humans and animals that are often multidrug resistant, intractable, and deadly. MAC lung disease is of particular concern and is now more prevalent than tuberculosis in many countries, including the United States. Although the clinical importance of these microorganisms continues to expand, our understanding of their genomic diversity is limited, hampering basic and translational studies alike. Here, we leveraged a unique collection of genomes to characterize …

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein is the main target for neutralizing antibodies. These antibodies can be elicited through immunization or passively transferred as therapeutics in the form of convalescent-phase sera or monoclonal antibodies (MAbs). Potently neutralizing antibodies are expected to confer protection; however, it is unclear whether weakly neutralizing antibodies contribute to protection. Also, their mechanism of action

Envelope protein-targeted vaccines for flaviviruses are limited by concerns of antibody-dependent enhancement (ADE) of infections. Nonstructural protein 1 (NS1) provides an alternative vaccine target that avoids this risk since this protein is absent from the virion. Beyond its intracellular role in virus replication, extracellular forms of NS1 function in immune modulation and are recognized by host-derived antibodies. The rational design of NS1-based vaccines requires an extensive understanding of the antigenic sites on NS1, especially those targeted by protective antibodies. Here, we isolated human monoclonal antibodies (MAbs) from individuals previously naturally infected with WNV, mapped their epitopes using structure-guided mutagenesis, and …

Branding Kidfluencers: Regulating Content And Advertising On Youtube, Gavin Feller, Benjamin Burroughs

Hank Greenspun School of Journalism and Media Studies Faculty Research

This paper analyzes emerging shifts in YouTube, advertising, and children’s digital media industries through a case study of Pocket Watch, a digital-first production and distribution studio built exclusively for YouTube child stars. Our analysis reveals the company’s strategic use of legacy media industry power, networks, and expertise to transform YouTube stars into global brands through the creation of toy, clothing, and lifestyle product lines across several industries. We further argue that Pocket Watch’s newly formed advertising division, Clock Work, exploits its child partners through problematic native advertising and host selling practices. The strategies implemented by Pocket Watch and other similar …

Understanding Religious Tolerance In Yongchang, China, Liming Gao

Honors Program: Senior Projects (Public)

The formation of China is a process of national integration and a fusion of different beliefs. However, under Chairman Mao (1949-1976) and specifically during the Cultural Revolution (1966-1976), people were reeducated to focus on Communism and expel remnants of traditional Chinese culture including the various religions. Although, after the Cultural Revolution, China reinstated its policy of religious freedom, there were still strict laws against religion. Despite such circumstances, Chinese people still practice their religious beliefs. The Yongchang area, located in Gansu Province in the northwest of China is a typical region of Chinese culture. At the same time, compared to …

Synthesis Of Thresholds Of Ocean Acidification Impacts On Decapods, John I. Spicer, Nina Bednaršek Oct 2021

Synthesis Of Thresholds Of Ocean Acidification Impacts On Decapods, John I. Spicer, Nina Bednaršek

School of Biological and Marine Sciences

Assessing decapod sensitivity to regional-scale ocean acidification (OA) conditions is limited because of a fragmented understanding of the thresholds at which they exhibit biological response. To address this need, we undertook a three-step data synthesis: first, we compiled a dataset composed of 27,000 datapoints from 55 studies of decapod responses to OA. Second, we used statistical threshold analyses to identify OA thresholds using pH as a proxy for 13 response pathways from physiology to behavior, growth, development and survival. Third, we worked with the panel of experts to review these thresholds, considering the contributing datasets based on quality of the …

Mongolian Herders’ Evaluation Of Rangeland Ecosystems Services, Values, And Changes Over The Past Decade, T. Ulambayar, B. Yunden, N. Davaasuren, S. Balt, D. Davaajav, B. Zambuu

IGC Proceedings (1977-2023)

Mongolia's rangeland is one of the largest remaining contiguous ecosystems encompassing 2.6% of the global grasslands, and almost three-fourths of the country's territory provides essential ecosystem services (ESS) for over 3 million Mongolians and 71 million livestock. The well-being of 171,605 pastoral households directly depends on the rangelands receiving provisional services in the forms of nutrition, material use and energy, regulatory services, and cultural services. This study explored herders' perceptions of these ESS, their evaluation for ESS values, and observations of ESS change for the last decade. The study found that Mongolian herders have more benefits from provisional ESS (on …

Conceptualizing Pastoral Development Based On Carbon Sequestration: The Case Of Yabelo District In The Southern Ethiopian Rangelands, D. L. Coppock

IGC Proceedings (1977-2023)

Major challenges for rangeland stewardship in the developing world include how to mitigate the spread of pastoral poverty and environmental degradation. Arresting such trends requires a scale of investment, policy incentives, and institutional commitments not previously observed in pastoral development. Indeed, such a rangeland revolution requires several global events to set the stage, namely: (1) Creation of markets for diverse ecosystem services; (2) recognition that improved rangeland stewardship is vital to mitigate climate change; and (3) distribution of green climate funds in support of local projects. New approaches for pastoral development projects are also needed. Previous projects have largely focused …

Wildlife Conservation And The Role Of The Indigenous Communities Living Around Conservation Areas, N. Parmisa, K. A. Galvin Oct 2021

Wildlife Conservation And The Role Of The Indigenous Communities Living Around Conservation Areas, N. Parmisa, K. A. Galvin

IGC Proceedings (1977-2023)

The Indigenous Kenyan Maasai community has coexisted with the wildlife surrounding it for decades from Nairobi National Park, Maasai Mara and Amboseli. These parks border Maasai lands. Although the northern, eastern, and western perimeters of the Nairobi National Park are fenced, the southern part is not. It is at this point that the Maasai community’s land meets the park. This area also acts as a wildlife dispersal area where wildlife can freely migrate to other parks, including Maasai Mara and Amboseli. The park is only 117sq kms and its vitality depends on the plains to the south where the Maasai …

Shifting Skins: Becoming Multiple During Emergency Online Teaching, Bianca Licata, Catherine Cheng Stahl

Occasional Paper Series

In this essay, we reflect on the emergence of our (new) teacher identities from the phenomenal space created within online learning, following the COVID-19 pandemic of 2020. Thrust from classrooms into in-between spaces mediated by digital technologies, the capricious co-inhabited new learning space functioned as a becoming-other space of identity-play, surfacing from centrifugal intra-actions among human, non-human, and inorganic entities and energies—what we have named a thinning space (authors, forthcoming). It called for becoming shapeshifters together through resisting crystallized roles and (re)claiming a multiplicity of vulnerable thin skins. We draw from the possibilities of existing virtual gaming spaces to …

Telling Tales For Justice And Equity: Storytelling As Public Nepantla Pedagogy, Ayesha Rabadi-Raol

Occasional Paper Series

As the COVID-19 pandemic led to schools moving to online platforms, I launched Tell-a-Tale, a livestreamed, biweekly read-aloud program for children. I designed and implemented each episode to include diverse children’s literature, followed by an artistic response, and finally a discussion about issues of equity and justice. Applying public pedagogy as a theoretical construct, I used this platform to create a space of “public intellectualism and social activism” (Sandlin, O’Malley, Burdick, 2011, p. 338). In this paper I will describe how I used “the pandemic as a portal” (Roy, 2020) to make space for historically marginalized stories and voices take …

In this study, the structural, thermal, and morphological properties of biocomposite films composed of wool keratin mixed with cellulose and regenerated with ionic liquids and various coagulation agents were characterized and explored. These blended films exhibit different physical and thermal properties based on the polymer ratio and coagulation agent type in the fabrication process. Thus, understanding their structure and molecular interaction will enable an understanding of how the crystallinity of cellulose can be modified in order to understand the formation of protein secondary structures. The thermal, morphological, and physiochemical properties of the biocomposites were investigated by Fourier transform infrared (FTIR) …

Bovine viral diarrhea virus’s (BVDV) entry into bovine cells involves attachment of virions to cellular receptors, internalization, and pH-dependent fusion with endosomal membranes. The primary host receptor for BVDV is CD46; however, the complete set of host factors required for virus entry is unknown. The Madin-Darby bovine kidney (MDBK) cell line is susceptible to BVDV infection, while a derivative cell line (CRIB) is resistant at the level of virus entry. We performed complete genome sequencing of each to identify genomic variation underlying the resistant phenotype with the aim of identifying host factors essential for BVDV entry. Three large compound deletions …
