Global DNA hypomethylation promotes genomic instability through the accumulation of DNA damage and resulted in an increasing rate of mutation. To stabilize the genome, cells possess an epigenetic mechanism to reduce DNA tension that causes DNA damages, named Replication-Independent Endogenous DNA Double-Strand Breaks (RIND-EDSBs). Due to their biological roles, this type of physiologic EDSBs act as youth-associated genomic-stabilizing DNA gaps (Youth-DNA-GAPs). They are localized within methylated genome and maintained by non-histone HMGB1 and SIRT1 proteins. Reduction of RIND-EDSBs was found in hypomethylated genome, including cancer and aging. High Efficiency Video Coding (HEVC) has dramatically enhanced the coding efficiency compared to the previous video coding standard, H.264/AVC. However, the existing rate control updates its parameters according to a fixed initialization, which can cause error prediction of bit allocation to each coding tree unit (CTU) in frames. In this work, a learning-based mapping between rate control parameters and video contents is proposed to achieve an accurate target bit rate and good video quality. This study is divided into two phases. The first phase is about the investigation of cell viability and damage of white blood cells after sorting in both a spiral microchannel and contraction and expansion array (CEA) at appropriate flow rates for cell separation. Three techniques such as Trypan Blue staining, Scanning Electron Microscopy (SEM) and Wright-Giemsa staining are employed in the experiments. Durian (Durio zibethinus L.), an economically important fruit of Southeast Asian countries, expresses unique flavor at the edible ripe stage, resulting from metabolic changes during the ripening process. Nevertheless, the study in such modulations is still limited. In the first manuscript, we profiled metabolites from unripe and ripe pulps of durian cv. Monthong using capillary electrophoresis- (CE) and gas chromatography- (GC) time-of-flight mass spectrometry (TOF/MS) and obtained the total 94 ripening-related metabolites from a combination of the two datasets, of which we focused on flavor-related metabolites. Nowadays, biomass conversion has become a more attractive process to produce fuels and various high-value chemicals from biological materials. From many kinds of catalysts, the metal-supported gamma-alumina (γ-Al2O3) is a widely used catalyst for various transformation processes, especially in the aqueous phase reaction. Typically, coke formation on γ-Al2O3 causes catalyst deactivation in the thermal reaction process. Manganese dioxide (MnO2) is the effective electrocatalysts used as a cathode for Zn-Air batteries (ZABs) accounted for the acceleration of the oxygen reduction reaction (ORR). There are various studies on the ORR activity of MnO2 rather than deactivation mechanisms. In this work, the deactivation mechanisms of α-MnO2 and β-MnO2 was investigated by density functional theory and computational hydrogen electrode. It was found that, the phase transformation from the α-MnO2 (211) to β-MnO2 (110) shows that the catalyst deactivation is due to the higher overpotential of β-MnO2 (110) making electrocatalyst efficiency low. This thesis investigates biodiesel production from palm oil by transesterification using extruded CaO catalysts. The preparation parameters of extruded CaO catalysts were investigated including 1) binder type including activated alumina, diatomite and kaolin, 2) calcination temperature at 700, 800 and 900°C, 3) mass ratio of CaO to the binder at 1: 1, 3: 1 and 6: 1. การศึกษานี้แบ่งออกเป็นสองวัตถุประสงค์หลัก วัตถุประสงค์แรกเป็นการศึกษาผลกระทบของมุมของหลุมจุลภาครูปร่างทรงพีระมิดฐานสี่เหลี่ยมข้าวหลามตัดทั้ง 3 รูปร่างคือ มุมแหลม (66 องศา) มุมด้านเท่า (90 องศา) และมุมป้าน (106 องศา) ต่อการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อสำหรับการเลี้ยงที่ใช้การไหลแบบต่อเนื่อง โดยแต่ละหลุมจุลภาควางเรียงชิดกันเพื่อลดพื้นที่ว่างไม่ให้เซลล์ไปเกาะหรือเกิดการสูญเสียเซลล์ไป ในบริเวณก้นหลุมจุลภาคมีทรงกระบอกรัศมี 450 ไมโครเมตร ติดตั้งอยู่ ซึ่งเป็นตำแหน่งที่จะเกิดการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อขึ้น และทำให้ความลึกรวมทั้งหมดของหลุมจุลภาคเท่ากับ 550 ไมโครเมตร ในการทดลองได้ใช้อัตราการไหลเท่ากับ 10 ไมโครลิตรต่อชั่วโมง ทั้งจากผลการคำนวณ และผลการทดลองพบว่า ในกรณีหลุมจุลภาคที่มีมุม 66 องศา ของไหลที่ไหลผ่านหลุมมีการเบี่ยงเบนทิศทางเข้าสู่กึ่งกลางหลุมมากกว่าหลุมแบบอื่น และน่าจะทำให้เกิดการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อของเซลล์ลำบาก เพราะถูกรบกวนจากของไหลค่อนข้างสูง ในกรณีหลุมจุลภาคที่มีมุม 90 องศา ของไหลมีการเบี่ยงเบนทิศทางเข้าสู่กึ่งกลางลดลง ทำให้มีการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อได้ดีขึ้น อย่างไรก็ตาม เมื่อหลุมจุลภาคมีมุม 106 องศา ของไหลมีการเบี่ยงเบนทิศทางเข้าหากึ่งกลางหลุมน้อยลงมาก ทำให้เซลล์บางส่วนไม่ถูกพัดพามารวมกันที่บริเวณก้นหลุมทรงกระบอก โดยเซลล์บางส่วนติดอยู่บนบริเวณผนังด้านข้าง ทำให้การสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อลดต่ำลง ดังนั้นผลการศึกษาจึงบ่งชี้ว่าหลุมจุลภาคทรงพีระมิดฐานสี่เหลี่ยมข้าวหลามตัดมุม 90 องศา มีความเหมาะสมที่สุดในการสร้างกลุ่มเซลล์คล้ายเนื้อเยื่อ นอกจากนั้น ผลการทดลองยังแสดงให้เห็นด้วยว่าอัตราการรอดชีวิตของเซลล์มีแนวโน้มดีกว่าหลุมจุลภาคแบบอื่นอีกด้วย วัตถุประสงค์ที่สองคือการสร้างระบบของไหลจุลภาคที่ใช้ต้นทุนต่ำ และรวดเร็ว โดยการสร้างแม่พิมพ์ด้วยการพิมพ์สามมิติด้วยเรซิ่น สำหรับการพิมพ์สามมิติจะทำให้สร้างแม่พิมพ์ที่มีรูปร่างที่ซับซ้อนมากได้ แต่ขนาดที่เล็กสุดยังจำกัดอยู่ และความแข็งแรงทางกล และความทนทานต่อความร้อนไม่ดีนัก อย่างไรก็ตาม วิธีการที่พัฒนาที่ขึ้นสามารถใช้งานได้ดีในการหล่อสารพอลิไดเมทิลซิโลเซนสำหรับระบบของไหลจุลภาค ซึ่งใช้งานอย่างมากในงานวิจัยทางด้านนี้

งานวิจัยนี้ศึกษาการนำเม็ดพลาสติกชีวภาพมาใช้เป็นแหล่งสารอินทรีย์คาร์บอนและตัวกลางในถังปฏิกรณ์ชีวภาพแอน็อกซิกชนิดเบดเคลื่อนที่ โดยเม็ดพลาสติกชีวภาพ 3 ชนิดที่ใช้ในการทดลอง ได้แก่ PLA, PHBV และ PBS แบ่งการทดลองออกเป็น 3 ช่วง การทดลองช่วงแรกศึกษาสมบัติทางกายภาพของเม็ดพลาสติกชีวภาพ พบว่าพลาสติกชีวภาพชนิด PBS มีสมบัติความชอบน้ำ ขณะที่ PLA และ PHBV ไม่ชอบน้ำ ผลการละลายน้ำของเม็ดพลาสติกชีวภาพพบว่า PBS ตรวจพบสารอินทรีย์คาร์บอนละลายในน้ำประปาและน้ำปราศจากไอออน เพิ่มขึ้นสูงกว่า PLA และ PHBV ตามลำดับ เมื่อทดสอบการดูดซับไนเตรทของเม็ดพลาสติกชีวภาพ พบว่าที่ค่า พีเอชในช่วงที่มากกว่า 4 พื้นผิวของเม็ดพลาสติกชีวภาพทั้ง 3 ชนิดจะแสดงประจุเป็นลบ จึงไม่พบการดูดซับหรือดูดติดผิวของไนเตรทบนตัวกลาง การทดลองช่วงที่สองศึกษาประสิทธิภาพการกำจัดไนเตรทในถังปฏิกรณ์ชีวภาพแอน็อกซิกชนิดเบดเคลื่อนที่เมื่อทำการเดินระบบแบบทีละเท พบว่าเม็ดพลาสติกชีวภาพชนิด PHBV และ PBS มีประสิทธิภาพการกำจัดไนเตรทมากกว่าร้อยละ 90 ในขณะที่ PLA กำจัดไนเตรทได้เพียงร้อยละ 48 เมื่อทำการศึกษาค่าจลนพลศาสตร์การกำจัดไนเตรท พบว่ากระบวนการดีไนตริฟิเคชันที่เกิดขึ้นเป็นปฏิกิริยาอันดับศูนย์ ความเข้มข้นไนเตรทคงเหลือของการทดลองแบบทีละเทเข้าใกล้ศูนย์ที่ระยะเวลา 12 และ 15 ชม. ตามลำดับ เมื่อใช้เม็ดพลาสติกชีวภาพชนิด PHBV และ PBS เป็นแหล่งสารอินทรีย์คาร์บอน การทดลองช่วงสุดท้ายเป็นการเดินระบบแบบต่อเนื่องในถังปฏิกรณ์ชีวภาพแอน็อกซิกชนิดเบดเคลื่อนที่ ผลการทดลองพบว่าถังปฏิกรณ์ที่บรรจุเม็ดพลาสติกชีวภาพ มีประสิทธิภาพการกำจัดไนเตรทที่สภาวะคงตัวเท่ากับร้อยละ 12.87±4.92, 8.11±5.31, 12.33±8.37 และ 84.83±4.58 มีระยะเวลากักน้ำ 12, 16, 20 และ 24 ชม. ตามลำดับ โดยจลนพลศาสตร์ของการกำจัดไนเตรทจัดเป็นปฏิกิริยาอันดับหนึ่ง เนื่องจากมีปริมาณของสารอินทรีย์คาร์บอนในระบบอย่างจำกัด ทำให้ปฏิกิริยาดีไนตริฟิเคชันเกิดได้ช้าลง ขณะที่เมื่อใช้เม็ดพลาสติกชีวภาพ PBS เป็นแหล่งอินทรีย์คาร์บอน ถังปฏิกรณ์ที่บรรจุตัวกลางดังกล่าวมีประสิทธิภาพการกำจัดไนเตรทร้อยละ 99.47±0.52, 99.34±1.83, 100 และ 98.91±0.95 ที่ระยะเวลากักน้ำ 12, 8, 4 และ 2 ชม. ตามลำดับ เนื่องจากในระบบมีสารอินทรีย์คาร์บอนเพียงพอต่อการเกิดกระบวนการดีไนตริฟิเคชันจึงทำให้ปฏิกิริยาเกิดขึ้นได้อย่างสมบูรณ์

Colorectal cancer (CRC) is genetically and transcriptomically heterogeneous disease. Molecular subtyping of colorectal cancer using consensus molecular subtype (CMS) system demonstrated the potential predictive value for tumor progression and treatment response. However, the CMS system was developed from data of whole tissues containing both cancer and non-tumor transcripts components for classification which does not directly represent intrinsic heterogeneity of cancer cells. In this study genetic profiles of CRC organoids were investigated first, and the results indicate chromosomal instability (CIN) and microsatellite instability (MSI) as pathogenic pathways of CRC. Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) are severe cutaneous adverse drug reactions with high mortality rates. Sequelae such as blindness remained even after recovery. Patients with SJS/TEN should be accurately diagnosed and received appropriate treatment as soon as possible. Therefore, factors for severity prediction are necessary. This study aimed to clarify clinical parameters and biological markers that can predict acute severe ocular complications (SOCs) in SJS/TEN. House dust mite (HDM) represents one of the most important allergenic sources worldwide. HDM allergens trigger in atopic patients potent Th2-biased inflammatory responses leading to the development of allergic rhinitis/asthma and atopic dermatitis. The HDM allergic response is mediated mainly by the allergen-specific Th2 cells and IgE. Due to its several mechanisms of action, curcumin (CUR) has been employed as adjuvant therapy in treating psoriasis. Limited oral bioavailability was the problem for the development of CUR for therapeutic purposes. Therefore, many approaches, including the prodrug strategy, have been devised to address Cur's drawbacks. CUR was conjugated with mycophenolic acid (MPA) as a new prodrug in this research. Characterization of MPA-CUR structure was done by FT-IR, 1H-NMR, 13C-NMR, and MS. MPA-CUR showed water solubility at 0.73 μM and log P at 2.33, respectively. Mercury ion (Hg2+) is one of the most challenging problems due to its high toxicity at low concentration and bioaccumulative effects affecting human health. Herein, we propose a strategy to exploit the fluorescence quenching of carbon quantum dots (CQDs) combined with surface-enhanced Raman scattering (SERS) technique to detect Hg2+. CQDs exhibit strong fluorescence emission interfering SERS measurement. However, with the presence of Hg2+, fluorescence from CQDs can be quenched, and the SERS spectrum can be collected. Halophilic Bacilllus subtilis P5-6 contains bacteriocin encoding genes (subtilin and subtilosin). It has been reported as a potential strain developed to be a protective culture that could reduce viable count of Staphylococcus aureus and prolong the freshness when applied in cheese. The aim of this research was to evaluate bacteriocin generation property of P5-6 and to optimize the bacteriocin production methods for application as food bio-preservative in protective culture form. In this work, a simple method for urinary oxalate determination was developed using testing paper modified with reddish-brown Fe(III)-curcumin complex. Oxalate in sample interacted with Fe(III) ions on the surface of the testing paper to form a Fe(III)-oxalate complex in solution, resulting in a decrease of the amount of Fe(III)-curcumin complex on the paper surface. The amount of the remaining Fe(III)-curcumin complex on the testing paper was inversely proportional to the oxalate level in the sample solution. A change of the testing paper color from reddish-brown to yellow could be observed and the color intensity was determined by ImageJ program. …

Nitrogen-doped carbon dots (NCDs) were synthesized from amino acids via the hydrothermal method under an acidic condition. L-histidine, L-phenylalanine, L-tyrosine, L-tryptophan, and glycine were used as a precursor which NCDs synthesized from those precursors were labeled as His-NCDs, Phe-NCDs, Tyr-NCDs, Trp-NCDs, and Gly-NCDs, respectively. Spectroscopic and microscopic characterizations were operated to investigate optical properties, chemical structures, and morphologies of NCDs. The results show that all NCDs show a spherical shape with an average diameter of 4.11±1.20 nm. Paper-based analytical device was fabricated by a polymer screen printing and wax printing methods. The various types of device were designed for simultaneous heavy metal ions detection in synthetic unknown and real water samples. The colorimetric method was applied to detect the heavy metal ions using 1-(2-pyridylazo)-2-naphthol, bathophenanthroline, 1, 5-diphenylcarbazide, bathocuproine, dimethylglyoxime, dithizone, and 4-(2-pyridylazo) resorcinol as complexing agents. However, complexing agents require very highly selectivity. This research was conducted to study the antioxidant traits that contained the total phenolic content (TPC), total flavonoid content (TFC), and the antioxidant capacity (AC), by ABTS assay in the seed of 174 Thai rice cultivars, comprising 152 white, 10 red, and 12 purple pericarp cultivars. The hierarchical cluster analysis was evaluated, and the rice cultivars were classified into four clusters based on the antioxidant. Cluster I comprised of 72 white pericarp cultivars and had the lowest TPC, TFC, and AC. Cluster II contained 80 white cultivars and showed higher TFC values than cluster I. The goals of this research have three main points: (i) to develop a rapid method for inorganic phosphorus quantification, (ii) to investigate spectral reflectance indices representing phosphorus deficiency in rice, and (iii) to determine SNPs associated with phosphorus deficiency tolerance in Thai rice via genome-wide association study (GWAS). The punching method was developed to enhance the high throughput performance of the conventional Pi extraction and molybdate blue assay. Pi content can be extracted using equally small leaf areas without leaf grinding, balancing, and tedious transferring steps. วัณโรค เป็นโรคติดต่อจากสัตว์สู่คนหรือจากคนสู่สัตว์ที่เป็นปัญหาด้านสาธารณสุขที่สำคัญระดับโลก รวมถึงในประเทศไทย เนื่องจากเชื้อ Mycobacterium tuberculosis (M. tb) complex (MTBC) ซึ่งเป็นเชื้อสาเหตุของวัณโรค สามารถพบการติดเชื้อได้ในสัตว์หลายกลุ่ม ทั้งสัตว์เท้ากีบ สัตว์ตระกูลลิง สัตว์เลี้ยงลูกด้วยนมรวมถึงช้าง การวินิจฉัยวัณโรคในช้างทำได้ยาก เนื่องจากช้างเป็นสัตว์ที่มีขนาดใหญ่ จึงไม่สามารถวินิจฉัยได้ด้วยวิธีเดียวกับมนุษย์ เช่น การเอกซเรย์ปอด และการทดสอบทางผิวหนัง นอกจากนี้ วิธีมาตรฐานโดยการเพาะเลี้ยงเชื้อจากน้ำล้างงวงช้างมีความไวต่ำ ใช้เวลานาน และตรวจได้เฉพาะระยะที่แสดงอาการเท่านั้น ดังนั้น การตรวจวินิจฉัยที่แม่นยำ ครอบคลุมทุกระยะการติดเชื้อ M. tb จึงเป็นสิ่งจำเป็น นำมาสู่การประยุกต์ใช้เทคนิค Interferon gamma release assay (IGRA) สำหรับเป็นวิธีทางเลือกในการวินิจฉัยวัณโรค ซึ่งเป็นวิธีตรวจหาอินเตอร์เฟียรอนแกมมา (IFN) ที่มีการหลั่งออกมาจากทีลิมโฟไซต์ชนิด memory หลังถูกกระตุ้นด้วยแอนติเจนจำเพาะของ M.tb งานวิจัยนี้จึงมีวัตถุประสงค์ที่จะพัฒนาวิธี IGRA ให้ใช้งานได้ง่ายกว่าที่มีอยู่เดิม โดยการคัดกรองเพปไทด์สังเคราะห์ของโปรตีน early secreted antigenic target 6 kDa (ESAT6) และ culture filtrate protein 10 kDa (CFP10) ที่สามารถกระตุ้นการหลั่ง eIFN ของเซลล์เม็ดเลือดขาวของช้าง และพัฒนาวิธีการตรวจโดยการกระตุ้นแบบ whole blood culture โดยเริ่มทำการตรวจการติดเชื้อของช้างด้วยวิธี IGRA เดิม (IGRA โดยใช้เซลล์เม็ดเลือดขาว PBMCs) ในตัวอย่างเลือดจากช้าง 20 เชือก โดยมีโปรตีนทั้งสายเป็นแอนติเจน พบว่าช้างมีการติดเชื้อ MTBC 50% ของตัวอย่างทั้งหมด ไม่ติดเชื้อ 30% และ 20% มีการติดเชื้อกลุ่ม Non tuberculous mycobacterium (NTM) จากนั้นทำการคัดกรองเพปไทด์สังเคราะห์ของโปรตีน ESAT6 และ CFP10 ในช้างทั้ง 20 เชือก พบว่าเพปไทด์สังเคราะห์ของโปรตีน …
