Innate immune signaling through the NLRP3 inflammasome is activated by multiple diabetes-related stressors, but whether targeting the inflammasome is beneficial for diabetes is still unclear. Nucleoside reverse-transcriptase inhibitors (NRTI), drugs approved to treat HIV-1 and hepatitis B infections, also block inflammasome activation. Here, we show, by analyzing five health insurance databases, that the adjusted risk of incident diabetes is 33% lower in patients with NRTI exposure among 128,861 patients with HIV-1 or hepatitis B (adjusted hazard ratio for NRTI exposure, 0.673; 95% confidence interval, 0.638 to 0.710; P < 0.0001; 95% prediction interval, 0.618 to 0.734). Meanwhile, an NRTI, lamivudine, improves insulin sensitivity and reduces inflammasome activation in diabetic and insulin resistance-induced human cells, as well as in mice fed with high-fat chow; mechanistically, inflammasome-activating short interspersed nuclear element (SINE) transcripts are elevated, whereas SINE-catabolizing DICER1 is reduced, in diabetic cells and mice. These data suggest the possibility of repurposing an approved class of drugs for prevention of diabetes.

Preharvest mycotoxin contamination of field-grown crops is influenced not only by the host genotype, but also by inoculum load, insect pressure and their confounding interactions with seasonal weather. In two different field trials, we observed a preference in the natural infestation of corn earworm (CEW; Helicoverpa zea Boddie) to specific maize (Zea mays L.) genotypes and investigated this observation. The field trials involved four maize lines with contrasting levels of resistance to Aspergillus flavus. The resistant lines had 7 to 14-fold greater infested ears than the susceptible lines. Seed aflatoxin B1 (AF) levels, in mock- and A. flavus-inoculated ears were …

The extensive literature on population and development yielded few policy-relevant results before the discovery of the demographic dividend. This dividend refers to a rise in per capita income that results from an increase in workers per capita as a population’s fertility declines. This paper describes the role of the demographic dividend in economic development in developing countries and summarizes policy options for strengthening the dividend. The first section reviews the demographic transition with an emphasis on its later phases when declining fertility and a changing population age structure produce the dividend. Next, the demographic drivers of the dividend and its …

The posterior alpha (α) rhythm, seen in human electroencephalogram (EEG), is posited to originate from cycling inhibitory/excitatory states of visual relay cells in the thalamus. These cycling states are thought to lead to oscillating visual sensitivity levels termed the “neuronic shutter effect.” If true, perceptual performance should be predictable by observed α phase (of cycling inhibitory/excitatory states) relative to the timeline of afferentiation onto the visual cortex. Here, we tested this hypothesis by presenting contrast changes at near perceptual threshold intensity through closed eyelids to 20 participants (balanced for gender) during times of spontaneous α oscillations. To more accurately and …

The skeleton is well-innervated, but only recently have the functions of this complex network in bone started to become known. Although our knowledge of skeletal sensory and sympathetic innervation is incomplete, including the specific locations and subtypes of nerves in bone, we are now able to reconcile early studies utilizing denervation models with recent work dissecting the molecular signaling between bone and nerve. In total, sensory innervation functions in bone much as it does elsewhere in the body-to sense and respond to stimuli, including mechanical loading. Similarly, sympathetic nerves regulate autonomic functions related to bone, including homeostatic remodeling and vascular …

Expression Of Mir-720 Is Correlated With Dnmt3 In Oral Squamous Cell Carcinomas, Andrew Graves, Sukhdeep Sandhu, Karl Kingsley
Dental Medicine Faculty Research
Background: Many factors are involved with cellular differentiation and pluripotency, such as long non-coding and microRNAs (miRNA). miR-720 has been demonstrated to affect development, differentiation and pluripotency in dental pulp stem cells but has also been linked to many cancers (renal, prostate, skin, colorectal, breast, cervical and esophageal) through shared pathways including DNA methyltransferase family members (DNMT)3a and DNMT3b, which are highly upregulated in many oral cancers. However, little is known about the expression (or function, if expressed) of miR-720 in oral cancers. Based upon this lack of information, the primary objective of this project was to evaluate the expression …

Key points
- Nrf2 is a master regulator of endogenous cellular defences, governing the expression of more than 200 cytoprotective proteins, including a panel of antioxidant enzymes.
- Nrf2 plays an important role in redox haemostasis of skeletal muscle in response to the increased generation of reactive oxygen species during contraction.
- Employing skeletal muscle-specific transgenic mouse models with unbiased-omic approaches, we uncovered new target proteins, downstream pathways and molecular networks of Nrf2 in skeletal muscle following Nrf2 or Keap1 deletion.
- Based on the findings, we proposed a two-way model to understand Nrf2 function: a tonic effect through a Keap1-independent mechanism under basal …

Action selection (AS), or selection of an action from a set of alternatives, is an important movement preparation process that engages a frontal-parietal network. The addition of AS demands to arm training after stroke could be used to engage this motor planning process and the neural network that supports it. The purpose of this case series is to describe the feasibility and outcomes associated with task-oriented arm training aimed at engaging the AS behavioral process and the related neural network in three individuals with chronic stroke. Three participants with mild to moderate motor deficits completed 13 to 15 sessions of …

Transposable Element Control Disrupted By Meiotic Drive In A Stalk-Eyed Fly Genome, Josephine A. Reinhardt
Transposable Element Control Disrupted By Meiotic Drive In A Stalk-Eyed Fly Genome, Josephine A. Reinhardt
Biology
Some stalk-eyed flies in the genus Teleopsis carry selfish genetic elements that induce sex ratio (SR) meiotic drive and impact the fitness of male and female carriers. Here, we produce a chromosome-level genome assembly of the stalk-eyed fly, T. dalmanni, to elucidate the pattern of genomic divergence associated with the presence of drive elements. We find evidence for multiple nested inversions along the sex ratio haplotype and widespread differentiation and divergence between the inversion types along the entire X chromosome. In addition, the genome contains tens of thousands of transposable element (TE) insertions and hundreds of transcriptionally active TE …

Radiation therapy is one of the most prevalent procedures for cancer treatment, but the risks of malignancies induced by peripheral beam in healthy tissues surrounding the target is high. Therefore, being able to accurately measure the exposure dose is a critical aspect of patient care. Here a radiation detector based on an organic field‐effect transistor (RAD‐OFET) is introduced, an in vivo dosimeter that can be placed directly on a patient's skin to validate in real time the dose being delivered and ensure that for nearby regions an acceptable level of low dose is being received. This device reduces the errors …

Child (Un)Awareness Of Parental Incarceration As A Risk Factor: Evidence From South Korea, Youngki Woo, Melissa A. Kowalski
Criminal Justice Faculty Publications
A large body of research has been devoted to the relationship between parental incarceration and adverse outcomes for children, but such studies often compare children of incarcerated parents to those whose parents have never been imprisoned. Research is lacking regarding the effects of parental incarceration on children aware of their parent’s imprisonment compared to those who are unaware of their parent’s incarceration. In the current study we use propensity score weighting with a sample of 219 incarcerated Korean parents to examine differences in developmental outcomes between children cognizant of their parent’s incarceration and those who are unaware of parental imprisonment. …

There is increasing concern globally about the enormity of the threats posed by antimicrobial resistance (AMR) to human, animal, plant and environmental health. A proliferation of international, national and institutional reports on the problems posed by AMR and the need for antibiotic stewardship have galvanised attention on the global stage. However, the AMR community increasingly laments a lack of action, often identified as an ‘implementation gap’. At a policy level, the design of internationally salient solutions that are able to address AMR’s interconnected biological and social (historical, political, economic and cultural) dimensions is not straightforward. This multidisciplinary paper responds by …

Advanced fibrosis and portal hypertension influence short-term mortality. Lipocalin 2 (LCN2) regulates infection response and increases in liver injury. We explored the role of intrahepatic LCN2 in human alcoholic hepatitis (AH) with advanced fibrosis and portal hypertension and in experimental mouse fibrosis. We found hepatic LCN2 expression and serum LCN2 level markedly increased and correlated with disease severity and portal hypertension in patients with AH. In control human livers, LCN2 expressed exclusively in mononuclear cells, while its expression was markedly induced in AH livers, not only in mononuclear cells but also notably in hepatocytes. Lcn2−/− mice were protected from liver …

Molecular motions are fundamental to the existence of life, and NMR spectroscopy remains one of the most useful and powerful methods to measure their rates and molecular characteristics. Multiple experimental methods are available for measuring the NMR relaxation properties and these can require different methods for extracting model parameters. We present here a new software application, RING NMR Dynamics, that is designed to support analysis of multiple relaxation types. The initial release of RING NMR Dynamics supports the analysis of exponential decay experiments such as T1 and T2, as well as CEST and R2 and R …

Background: Badly broken primary teeth that require pulp therapy present a high-risk of biomechanical failure due to the loss of tooth substance resulting from pre-existing decay and endodontic therapy itself. The introduction of Biological dentine posts offered an excellent alternative for restoring such teeth. The ideal post material should have physical and mechanical properties that are similar to those of dentine. Accordingly there is no such material other than dentine itself. Another concern in restoring endodontically treated primary teeth is the need to find a material that can resorb in a similar way to natural tooth structure as a part …

Statement of the problem: Headgear is most commonly used to correct anteroposterior discrepancies. Headgear can also be used to make more space for teeth to come in. In this instance the headgear is attached to the molars, via molar headgear bands and tubes, and helps to draw these molars backwards in the mouth, opening up space for the front teeth to be moved back using braces and bands. Stress on PDL of molars teeth and soreness of teeth when chewing, or when the teeth touch, is typical. Adults usually feel the soreness 12e24 h later, but younger patients tend to …

Identification Of Stiffness Variations In Supporting Substances Of A Human Canine Tooth With A Bracket-Beam-Piezoelectric Sensor And Its Electromechanical Impedance, Hector A. Tinoco
Future Dental Journal
In this paper, an experimental method is described to identify the stiffness variations produced by drillings done in different supporting substances of a human canine tooth. To measure the supporting substances parameters through of a canine, a sensor-actuator systemwas developed. The sensor-actuator device was composed of a stainless steel bracket bonded to a steel wire attached to two piezoelectric transducers, with a concentrated mass attached to the end of the wire. To excite the device, high frequency voltage (between 5 and 10 KHz) was applied through the piezo-transducers, which affects the tooth by means of the vibration of the wire. …
