Background: A major challenge in implementing personalized medicine in pediatrics is identifying appropriate drug dosages for children. The majority of drug dosing studies have been based on adult populations, often with modification of the dosing for children based on size and weight. However, the growth and development experienced by children between birth and adulthood represents a dynamically changing biological system, with implications for effective drug dosing, efficacy as well as potential drug toxicity. The purpose of this study was to apply a metabolomics approach to gain preliminary insights into the ontogeny of liver function from newborn to adolescent.
Methods: Metabolites …

BACKGROUND: Mis-implementation-defined as failure to successfully implement and continue evidence-based programs-is widespread in public health practice. Yet the causes of this phenomenon are poorly understood.
METHODS: We develop an agent-based computational model to explore how complexity hinders effective implementation. The model is adapted from the evolutionary biology literature and incorporates three distinct complexities faced in public health practice: dimensionality, ruggedness, and context-specificity. Agents in the model attempt to solve problems using one of three approaches-Plan-Do-Study-Act (PDSA), evidence-based interventions (EBIs), and evidence-based decision-making (EBDM).
RESULTS: The model demonstrates that the most effective approach to implementation and quality improvement depends on the …

Extracellular vesicles (EVs) are secretory lipid membranes with the ability to regulate cellular functions by exchanging biological components between different cells. Resident skin cells such as keratinocytes, fibroblasts, melanocytes, and inflammatory cells can secrete different types of EVs depending on their biological state. These vesicles can influence the physiological properties and pathological processes of skin, such as pigmentation, cutaneous immunity, and wound healing. Since keratinocytes constitute the majority of skin cells, secreted EVs from these cells may alter the pathophysiological behavior of other skin cells. This paper reviews the contents of keratinocyte-derived EVs and their impact on fibroblasts, melanocytes, and …

It has been shown that carbinolamides are intermediates in the enzyme-catalyzed synthesis of -amidated peptides. Although carbinolamides are important intermediates in the production of these hormones and have roles in other biological venues, little attention has been given to their mechanisms of reaction in water, and, thus, it is difficult to speculate how their reaction would be enzymatically catalyzed. The purpose of the work presented here was to synthesize structural analogs of the biological intermediates and study their mechanism of breakdown in water. -Hydroxyhippuric acid, a substrate for the enzymatic system, provides an easily modified template for the study of …

Allograft rejection after renal transplantation remains a challenge to overcome. Interleukin (IL)-21, a cytokine with pleiotropic effects, maintains immune homeostasis post-transplantation. Here, we report higher levels of IL-21 in kidney transplant recipients with non-rejection (NR) than in recipients with T cell-mediated rejection (TCMR, P < 0.001) and antibody-mediated rejection (ABMR, P = 0.005). We observed a negative correlation between IL-21 and creatinine (Cr) levels (P = 0.016). The receiving operating characteristic (ROC) curve showed a promising diagnostic value of IL-21 to identify acute rejection with an area under the curve (AUC) of 0.822 (P < 0.001). In contrast, exogenous administration of IL-21 accelerated acute rejection in a comparative translational kidney transplant (KT) mouse model. Reduced IL-21 levels in the peripheral blood were observed in KT mice after IL-21 injection. Further analysis revealed that increased IL-21 levels in the spleen induced proliferation of CD4+ T cells and CD19+ B cells after IL-21 treatment. Our findings suggest a critical function of IL-21 in kidney transplantation and the potential involvement of the IL-21/IL-21R pathway in acute rejection management.

Background and Objectives: Among the multiple factors posited to drive the health inequities that black men experience, the fundamental role of stress in the production of poor health is a key component. Allostatic load (AL) is considered to be a byproduct of stressors related to cumulative disadvantage. Exposure to chronic stress is associated with poorer mental health including depressive symptoms. Few studies have investigated how AL contributes to depressive symptoms among black men. The purpose of the cross-sectional study was to examine the association between AL and depressive symptoms among middle- to old age black men. Research Design and Methods: …

To reduce the danger of powerful super-intelligent AIs, we might make the first such AIs oracles that can only send and receive messages. This paper proposes a possibly practical means of using machine learning to create two classes of narrow AI oracles that would provide chess advice: those aligned with the player's interest, and those that want the player to lose and give deceptively bad advice. The player would be uncertain which type of oracle it was interacting with. As the oracles would be vastly more intelligent than the player in the domain of chess, experience with these oracles might …

BACKGROUND: Breast cancer treatment is based on estrogen receptors (ERs), progesterone receptors (PRs), and human epidermal growth factor receptor 2 (HER2). At the time of metastasis, receptor status can be discordant from that at initial diagnosis. The purpose of this study was to determine the incidence of discordance and its effect on survival and subsequent treatment in patients with breast cancer brain metastases (BCBM).
METHODS: A retrospective database of 316 patients who underwent craniotomy for BCBM between 2006 and 2017 was created. Discordance was considered present if the ER, PR, or HER2 status differed between the primary tumor and the …

Sonoporation-Mediated Loading Of Trehalose In Cells For Cryopreservation., Charles W. Shaffer Iv, David F. Grimm, Michael A. Menze, Jonathan A. Kopechek
Undergraduate Research Events
Trehalose, a non-reducing disaccharide, is present in many microorganisms and metazoans. In these organisms, trehalose acts as a stress protectant and helps preserve lipid membranes of cells during states of desiccation and freezing. Trehalose is required on both sides of the cell membrane to achieve a significant cryoprotective effect. Specific loading methods for trehalose are required since this sugar is impermeant to mammalian cells. Trehalose loading in mammalian cells has been achieved by fluid-phase endocytosis and genetic modification for the expression of trehalose transporters, however cryoprotective outcomes are unable to compete with established methods of cryopreservation for mammalian cells. Sonoporation …

Diabetic retinopathy is one of the leading causes of visual disability and blindness worldwide. It is estimated that 4.8% out of 37 million cases of blindness were due to diabetic retinopathy, globally. It affects patients suffering from prolonged diabetes, which probably results in permanent blindness. The earliest symptoms surfaced when the patients have vision problems. Therefore, regular eyes examination and early intervention normally controls this disease. Many studies for early intervention and prevention of diabetic retinopathy uses various predictive models. The booming of database and digital storage technology creates an abundance of health records. Thus, data mining techniques helps uncover …

The complexities of modern biomedicine are rapidly increasing. Thus, modeling and simulation have become increasingly important as a strategy to understand and predict the trajectory of pathophysiology, disease genesis, and disease spread in support of clinical and policy decisions. In such cases, inappropriate or ill-placed trust in the model and simulation outcomes may result in negative outcomes, and hence illustrate the need to formalize the execution and communication of modeling and simulation practices. Although verification and validation have been generally accepted as significant components of a model's credibility, they cannot be assumed to equate to a holistic credible practice, which …

Тaxonomic Position Of Orthochirus Olivaceus (Karsch, 1881), The Type Species Of The Genus Orthochirus Karsch, 1892 (Scorpiones: Buthidae), František Kovařík, Victor Fet, Manal Siyam
Euscorpius
We address the taxonomic position of Orthodactylus olivaceus Karsch, 1881, the type species of the genus Orthochirus Karsch, 1892. For over a century, this taxon was considered a synonym of Orthochirus scrobiculosus (Grube, 1873); however, the latter was recently limited to Central Asia (Turkmenistan) (Kovařík et al., 2020). Analysis of Karsch’s holotype confirmed that Orthochirus olivaceus (Karsch, 1881), is a valid species. Its type locality published as “Sicily” is clearly incorrect. It is a senior synonym of Butheolus aristidis Simon, 1882, syn. n., described from Sudan (Nubia). We confirm its distribution in northern Sudan and southern Egypt.

Buprestidae, Cerambycidae, And Siricidae Collected In Baited Funnel Traps On Drummond Island, Chippewa County, Michigan, Robert Haack
The Great Lakes Entomologist
Trapping of bark- and wood-infesting insects in 2015 and 2016 at multiple locations on Drummond Island in northern Lake Huron, using baited multi-funnel traps, yielded 4 species of Buprestidae, 24 Cerambycidae, and 4 Siricidae. In 2015, all funnel traps were baited with the plant volatiles α-pinene, ethanol, and cis-3-hexenol, and were either black or green in color, and placed either at heights similar to the lower canopy of nearby trees (4-5 m) or at 1-2 m above groundline. In 2016, all traps were green, hung at 1-2 m above groundline, and baited with the cerambycid pheromones fuscumol acetate, monochamol, and …

Ailanthus Altissima Aqueous Extract Deters Spodoptera Frugiperda Oviposition, Ryan L. Wagner, Jordan A. Card
The Great Lakes Entomologist
Ailanthus altissima (Mill.) Swingle (Simaroubaceae), the tree of heaven, is a highly invasive tree species containing phytochemicals with a range of biological activities. Exposure to novel chemistry from invasive plant species may result in changes to established Lepidopteran behaviors, including feeding or oviposition. However, the impact of A. altissima chemistry on insect behavior has not been extensively explored. Therefore, A. altissima extract was tested for oviposition deterrence effects against Spodoptera frugiperda (Smith) (Noctuidae). Oviposition substrates were treated with water-soluble extractions of Zea mays (L), A. altissima, or a combination of both. Moths were then placed in chambers containing different …

The white-mold gall midge, Karshomyia caulicola Coquillett, was documented in association with soybean, Glycine max (L.) Merr., infected with the fungus Sclerotinia sclerotiorum (Lib.) de Bary. This mycetophagous cecidomyiid appears widespread in the northern soybean producing region, with confirmed detections from Minnesota, North Dakota and Québec. Though likely not a pest of soybean plants, the presence of K. caulicola in soybean fields may complicate identification, population assessment and decision making for soybean gall midge, Resseliella maxima Gagné, which is a recently described pest of soybean. Here, we provide an overview of the known biology and distribution of K. caulicola and …

Hunting Billbug Sphenophorus Venatus (Coleoptera: Dryophthoridae) Adult Feeding And Attraction To Warm- And Cool-Season Turfgrasses, Alexandra Grace Duffy, Douglas S. Richmond
The Great Lakes Entomologist
The hunting billbug Sphenophorus venatus Say (Coleoptera: Curculionidae: Dryophothorinae) is a generalist stem-boring pest on warm- and cool-season grasses. The objectives of this work were to (1) investigate adult feeding preference for four common turfgrass host species and (2) explore whether adults are attracted to the volatile odors emitted by these grasses. In laboratory feeding assays, S. venatus adults preferred zoysiagrass Zoysia japonica Steud ‘Meyer’ over all other species tested: Bermudagrass Cynodon dactylon ‘Patriot’, Kentucky bluegrass Poa pratensis ‘Barron’, and creeping bentgrass Agrostis stolonifera ‘Penncross’. In y-tube olfactometry assays, only males were attracted to bermudagrass. Our findings suggest that although …

First Records Of Megachile Apicalis (Hymenoptera: Megachilidae) For Illinois Found In Heavily Urbanized Areas Within The City Of Chicago, Andrea M. Gruver, Paul J. Caradonna
The Great Lakes Entomologist
We provide the first record of the non-native Megachile apicalis Spinola (Hymenoptera: Megachilidae) in Illinois. Thirty Megachile apicalis specimens were collected in the summer of 2018 in a highly urban area of Chicago, IL, USA. Our findings suggest that the range of Megachile apicalis appears to be rapidly expanding across North America, inward from both the East and Western Coasts. Megachile apicalis in Chicago has a broad activity period, can take advantage of abundant non-native floral food resources, and has competitive nesting behavior, all of which may facilitate its successful establishment in disturbed urban environments, as well as its continued …

Reinstatement Of Andrena Vernalis Mitchell (Hymenoptera: Andrenidae) From Synonymy With A. Ziziae Robertson, Zachary M. Portman, Ian G. Lane, Gabriella L. Pardee, Daniel P. Cariveau
The Great Lakes Entomologist
Andrena (Micrandrena) ziziae Robertson, 1891 (Andrenidae) is a well-known species found in a variety of habitats in the eastern and central United States and adjacent southern Canada. Andrena (Micrandrena) vernalis Mitchell, 1960 was described from five female specimens in the eastern United States and was synonymized with A. ziziae by Ribble in 1968. Recently collected specimens from throughout Minnesota have revealed that A. ziziae sensu Ribble is actually two species, one of which matches A. vernalis. Here, we reinstate A. vernalis as a valid species and describe the previously unknown male. We provide diagnostic characters …
