Human cytomegalovirus (HCMV) is an enveloped, single segment, double-stranded DNA virus. HCMV infection causes disease in immunocompromised (HIV patients, transplant recipients) and immunodeficient (fetuses, neonates) populations. Current treatments are effective but are either limited in use or can lead to organ damage and/or antiviral resistance, and no vaccines are available. Additional antiviral targets are needed. HCMV pUL103 is a potential antiviral target. pUL103 is a conserved herpesvirus protein present in the tegument, layer of proteins and RNA between the envelope and capsid of HCMV virions. pUL103 helps reorganize cellular secretory machinery (Golgi, endosomes) to form the cytoplasmic virion assembly compartment …

Cardiolipin (CL) is the signature phospholipid of mitochondria. CL and its remodeling exert critical roles in biological processes both inside and outside of mitochondria. CL abnormalities have been associated with various mitochondrial disorders and aging. Understanding the role of CL in mitochondrial physiology and cellular metabolism could provide valuable insights into cell biology and human health. Several metabolic alterations have been reported in CL-deficient cells, including accumulated lactate, decreased PDH activity, and decreased TCA cycle function. This dissertation connected these findings by showing abnormal NAD+ metabolism in various models lacking CL. Importantly, it shows that NAD+ supplementation improves mitochondrial function …

The traditional drug discovery process is extremely slow and costly. More than 90% of drugs fail to pass beyond the early stage of development and toxicity tests, and many of the drugs that go through early phases of the clinical trials fail because of adverse reactions, side effects, or lack of efficiency. In spite of unprecedented investments in research and development (R&D), the number of new FDA-approved drugs remains low, reflecting the limitations of the current R&D model.

While unisexual flowers have evolved repeatedly throughout angiosperm families, the actual identity of sex determining genes has been elusive, and their regulation within populations remains largely undefined. Additionally, sex liability is often observed in unisexual plants and has been correlated to external and internal cues, suggesting that the genes responsible for unisexual morphology are not necessarily segregating but rather differentially regulated. Understanding these processes will be of significant theoretical and agronomical importance.

B cells are a unique subset of immune cells that, in response to antigen, diversify their antibody repertoire to generate progressively higher affinity antibodies of different isotypes through the processes of affinity maturation/somatic hypermutation (SHM) and class switch recombination (CSR). One of the major sites in which this diversification occurs is in T cell dependent microanatomical structures known as germinal centers (GC). Here, we find that GC B cells express the protein, autoimmune regulator (Aire) in a CD40 dependent manner. In these cells, Aire interacts with activation induced cytidine deaminase (AID), the protein responsible for initiating the processes of diversification …

These would help human health, animal welfare, and conservation of at-risk wildlife all at the same time. One of the most common complications associated with childbirth is pregnancy-related depression. Postpartum Depression (PPD) affects a new mother’s ability to bond with and take care of their baby. When left untreated PPD not only impacts the mother’s health but can also cause behavioral, eating, and sleeping issues for the baby (CDC, 2020). New mothers experiencing PPD have reported feeling stressed, worried, and anxious about their children. New mothers also reported having doubts about their ability to be a good mother. Bilin lyases are enzymes which ligate linear tetrapyrrole chromophores to specific cysteine residues on light harvesting proteins present in cyanobacteria and red algae. The lyases responsible for chromophorylating the light harvesting protein phycoerythrin (PE) have not been fully characterized. In this study, we explore the role of CpeT, a putative bilin lyase, in the biosynthesis of PE in the cyanobacterium Fremyella diplosiphon. Recombinant protein studies show that CpeT alone can bind phycoerythrobilin (PEB), but CpeZ, a chaperone-like protein, is needed in order to correctly and efficiently attach PEB to the beta-subunit of PE. A triflic acid (TfOH) mediated sequential cyclization of ortho-alkynylarylesters and ammonium acetate (NH4OAc) was reported. The reaction took place via a Brønsted acid-mediated intramolecular cyclization of ortho-alkynylarylesters followed by an ammonium acetate participated substitution reaction, forming isoquinolin-1-ones as the major products. Different from most of the known synthetic methods of isoquinolin-1-ones, no metal catalyst was required in the reported reaction. The regioisomers – isoindolin-1-ones were obtained together with isoquinolin-1-ones in a few cases. The intermediate compounds – isochromen-1-ones and isobenzofuran-1-ones were also isolated. The 32nd Annual Barrels meeting was hosted at the Northwestern University Feinberg School of Medicine in Chicago, Illinois on October 17th and 18th, 2019. The annual meeting brings together researchers who utilize the rodent whisker-to-barrel system as a means to understand cortical function and development. This year’s meeting focused on social behaviors, development and cerebellar functions within the barrel system and beyond.

Purpose of review: Impaired reward processing and amotivation are well documented in schizophrenia. We aim to review the current state of neuroimaging and behavioral research addressing components of motivational deficits in this complex and impairing syndrome. Evidence will be integrated to inform the ongoing development of effective strategies for behavioral rehabilitation. Recent findings: While striatal dopamine and aberrant reward prediction errors have been connected to amotivation in schizophrenia, they are not sufficiently full explanations of reward processing impairments. Frontal dysfunction and associated cognitive control deficits also have evidenced involvement in atypical reward prediction, learning, and valuation. Ongoing work supports the …

Joan Robinson and Paul Samuelson found little to agree upon in a correspondence which began in 1946, shortly after the death of Keynes, and ended a year prior to Robinson’s death in 1983. One way to read the correspondence is to keep in mind that Keynesian uncertainty was central to Robinson’s understanding of how capitalist economies function. Samuelson, never impressed by Keynes’s handling of uncertainty, understood capital theory—if not capitalism—in terms of dynamic programming, with its perfect foresight entailments. This is evident throughout his letters to Robinson, although rarely acknowledged in a straightforward way, particularly during the period from 1971 …

This study examined the perspectives of a national sample of pre-doctoral internship directors regarding psychological testing and assessment practices at the internship level. Study participants were from member programs of the Association of Psychology Postdoctoral and Internship Centers (APPIC). The study's goals were to better understand the current state of affairs regarding the role of assessment in internship programs and to develop findings that may be useful to academic training programs and graduate students in psychology. The goal of this dissertation was to create an intervention resource guide containing recommendations that can be utilized by the South Los Angeles Trauma Recovery Center (SLATRC) to implement a brief early somatic intervention for victims of violent crime in acute hospital settings in Southeast Los Angeles. It was decided that an intervention based upon the Trauma Resiliency Model/Community Resiliency Model (TRM/CRM) would best fit the needs of the population. Family homelessness, particularly of mothers and their children, continues to remain an unfortunate epidemic throughout the United States. Personal, relational, and systemic adversity and trauma leave this population at high risk for instability, physical and mental health challenges, and impaired wellbeing. However, despite these difficulties, research strongly suggests that positive functioning can remain in families experiencing homelessness and that many go on to leave shelters and maintain stability, particularly when mothers have effective strategies to manage stress in contexts of adversity. Recent publications report that adults known as Generation Z, between the ages of 18 to 23, increasingly rely upon social media to gain knowledge of social issues. Given social media's embeddedness in Generation Z's life, this study sought to understand if or how social media has influenced and possibly empowered Generation Z to act on social issues, particularly global climate change. This research focused on global leadership and change and competitive athletics. Servant leadership, including the five servant leadership variables; altruistic calling, emotional healing, wisdom, persuasive mapping, and organizational stewardship, as well coach athlete relationships, including the three coach-athlete relationship variables; closeness, commitment, and complementarity, were used to analyze athlete perceptions. This study then found correlations of the associated factors to outcome, at team and individual levels. Sharks are a taxon of significant conservation concern and associated public interest. The scientific community largely supports management policies focusing on sustainable fisheries exploitation of sharks, but many concerned members of the public and some environmental advocates believe that sustainable shark fisheries cannot and do not exist and therefore support total bans on all shark fisheries and/or trade in shark products. The purpose of this research study was to determine the interaction effects national board certified teachers (NBCTs) and teachers with advanced degrees (ADs); while considering school accountability levels, had on student achievement. This study examined whether significant differences existed in student achievement between the eight identified groups of teachers and how the factors of NBCT status, AD status, and accountability contributed. The study examined student achievement in grades three through eight on the end-of-the-year state assessments for the 2017-18 school year. Differential Semantics is a theoretical accounting of the semantic complexity found in natural language, particularly that of the academic and literary registers. It addresses natural language semantics in terms of its contribution to the characterization and expression of creative thought, beginning the perception and conceptualization of objective reality, folloby the metacognitive development of idiosemantic connotations in reference to those conceptualizations, and finally, the intuitive process of implication and inference that facilitates the abstraction and communication of thought.
Fabrication Of Taste Masked Tablets Via Fused Deposition Modeling 3d Printing Paired With Hot-Melt Extrusion Techniques, Honghe Wang

The objective of this work was to develop taste-masked donut-shaped tablet formulations utilizing fused deposition modeling three-dimensional (3D) printing paired with hot-melt extrusion (HME) techniques. Caffeine citrate (CC) was used as the model drug for its bitter taste, and a three-point bend test was performed to assess the printability of filaments. The stiffness constant was calculated to represent the printability by fitting the breaking distances and stress data into Hooke's law. Formulation F6 and F7 filaments exhibited the desired hardness with a “k” value of 48.30 ± 3.52 g/mm3 and 45.47 ± 3.51 g/mm3, respectively, and were successfully printed. The …
