The Vimentin Cytoskeleton: When Polymer Physics Meets Cell Biology, Alison E. Patteson, Robert J. Carroll, Daniel V. Iwamoto, Paul A. Janmey
Physics - All Scholarship
The proper functions of tissues depend on the ability of cells to withstand stress and maintain shape. Central to this process is the cytoskeleton, comprised of three polymeric networks: F-actin, microtubules, and intermediate filaments (IFs). IF proteins are among the most abundant cytoskeletal proteins in cells; yet they remain some of the least understood. Their structure and function deviate from those of their cytoskeletal partners, F-actin and microtubules. IF networks show a unique combination of extensibility, flexibility and toughness that confers mechanical resilience to the cell. Vimentin is an IF protein expressed in mesenchymal cells. This review highlights exciting new …

In early October 2020, the Trump administration’s Environmental Protection Agency sent a letter to Oklahoma governor Kevin Stitt. The letter granted Stitt environmental regulatory control over all of the tribal lands in the state. Among other things, this gives Stitt the power to determine whether hazardous waste can be dumped on tribal lands, the ability to make decisions regarding whether and where fracking can take place, and the ability to determine if and where large-scale industrial animal agriculture, with all its attendant pollution, can operate in tribal jurisdictions.

Dynamic contrast-enhanced (DCE) MRI provides both morphological and functional information regarding breast tumor response to neoadjuvant chemotherapy (NAC). The purpose of this retrospective study is to test if prediction models combining multiple MRI features outperform models with single features. Four features were quantitatively calculated in each MRI exam: functional tumor volume, longest diameter, sphericity, and contralateral background parenchymal enhancement. Logistic regression analysis was used to study the relationship between MRI variables and pathologic complete response (pCR). Predictive performance was estimated using the area under the receiver operating characteristic curve (AUC). The full cohort was stratified by hormone receptor (HR) and …

Lyme Disease In Kentucky: Prevalence, Awareness, And Diagnosis In A Conventionally Low-Endemic Area, Rachel L. Carden Ots
Honors Theses
Lyme Disease is the fastest-growing vector-borne illness in the United States – including in low-endemic states, such as Kentucky; given Kentucky’s low Lyme disease incidence in prior decades, it has since experienced an increase of cases despite a lack of discussion on the condition and its implications on patient health and wellbeing. To measure current levels of knowledge and awareness of Lyme disease in Kentucky residents, a survey was distributed to Microbiology and Honors Program students at Eastern Kentucky University, and its results were discussed. While Lyme disease cases do continue to rise, the general public of Kentucky lacks in …

Iopamidol (an iodinated X-ray contrast medium) and bromide are precursors in the formation of halogenated disinfection byproducts (DBPs). The interactions of these precursors are vital to elucidate the formation of halogenated DBPs during chloramination. This work investigated the formation of total organic halogen and select individual DBPs in two laboratory-chloraminated source waters containing iopamidol and bromide. Experiments were carried out in batch reactors containing Barberton source water (BSW) and Cleveland BSW (CSW), spiked with iopamidol (5 mu M), bromide (15 mu M), and 100 mu M monochloramine. Total organic iodine concentrations were approximately equal regardless of source water since they …

Introduction: The progression and clinical course of head and neck squamous cell carcinoma (HNSCC) relies on complex interactions between cancer and stromal cells in the tumor microenvironment (TME). Among the most abundant of these stromal cells are cancer-associated fibroblasts (CAFs). While their contribution to tumor progression is widely acknowledged, and various CAF-targeted treatments are under development, the relationship between CAF density and the clinicopathologic course of HNSCC has not been clearly defined. Here we examine the published evidence investigating the relationship of cancer-associated fibroblasts to local recurrence and indicators of prognostic significance in HNSCC.
Methods: We conducted a meta-analysis of …

Tackle Epithelial-Mesenchymal Transition With Epigenetic Drugs In Cancer, Bo Dong, Zhaoping Qiu, Yadi Wu
Pharmacology and Nutritional Sciences Faculty Publications
Epithelial-mesenchymal Transition (EMT) is a de-differentiation process in which epithelial cells lose their epithelial properties to acquire mesenchymal features. EMT is essential for embryogenesis and wound healing but is aberrantly activated in pathological conditions like fibrosis and cancer. Tumor-associated EMT contributes to cancer cell initiation, invasion, metastasis, drug resistance and recurrence. This dynamic and reversible event is governed by EMT-transcription factors (EMT-TFs) with epigenetic complexes. In this review, we discuss recent advances regarding the mechanisms that modulate EMT in the context of epigenetic regulation, with emphasis on epigenetic drugs, such as DNA demethylating reagents, inhibitors of histone modifiers and non-coding …

Desirability Of Outcome Ranking For The Management Of Antimicrobial Therapy (Door Mat): A Framework For Assessing Antibiotic Selection Strategies In The Presence Of Drug Resistance, Brigid M. Wilson, Robin L.P. Jump, Federico Perez, Robert A. Bonomo
Faculty Scholarship
The complexities of antibiotic resistance mean that successful stewardship must consider both the effectiveness of a given antibiotic and the spectrum of that therapy to minimize imposing further selective pressure. To meet this challenge, we propose the Desirability of Outcome Ranking approach for the Management of Antimicrobial Therapy (DOOR MAT), a flexible quantitative framework that evaluates the desirability of antibiotic selection. Herein, we describe the steps required to implement DOOR MAT and present examples to illustrate how the desirability of treatment selection can be evaluated using resistance information. While treatments and the scoring of treatment selections must be adapted to …

(1) Background: The atopic march is defined by the increased prevalence of allergic diseases after atopic dermatitis onset. In fact, atopic dermatitis is believed to play an important role in allergen sensitization via the damaged skin barrier, leading to allergic diseases such as allergic asthma and allergic rhinitis. The eosinophil, a pro-inflammatory cell that contributes to epithelial damage, is one of the various cells recruited in the inflammatory reactions characterizing these diseases. Few studies were conducted on the transcriptome of this cell type and even less on their specific microRNA (miRNA) profile, which could modulate pathogenesis of allergic diseases and …

In a previous study, air sampling using vortex air samplers combined with species-specific amplification of pathogen DNA was carried out over two years in four or five locations in the Salinas Valley of California. The resulting time series data for the abundance of pathogen DNA trapped per day displayed complex dynamics with features of both deterministic (chaotic) and stochastic uncertainty. Methods of nonlinear time series analysis developed for the reconstruction of low dimensional attractors provided new insights into the complexity of pathogen abundance data. In particular, the analyses suggested that the length of time series data that it is practical …

This study assesses the spatial and temporal characteristics of nighttime surface urban heat island (SUHI) effects over Greater Cairo: the largest metropolitan area in Africa. This study employed nighttime land surface temperature (LST) data at 1 km resolution from the Moderate Resolution Imaging Spectroradiometer (MODIS) Aqua sensor for the period 2003–2019. We presented a new spatial anomaly algorithm, which allowed to define SUHI using the most anomalous hotspot and cold spot of LST for each time step over Greater Cairo between 2003 and 2019. Results demonstrate that although there is a significant increase in the spatial extent of SUHI over …
