Thermal-stress events associated with climate change cause coral bleaching and mortality that threatens coral reefs globally. Yet coral bleaching and coral cover patterns vary spatially and temporally, and few studies have identified bright spots where corals have the potential to survive through climate change. This study synthesizes over 10,000 field observations globally of coral bleaching and coral cover from 1997to 2017 and uses a suite of environmental and temperature metrics to analyze coral bleaching and coral cover patterns. 3d Printed Microporous Collagen Constructs Using The Freeze-Fresh Methodology, Thais Sousa Senzano

Scaffold microporosity is known to play a critical role in governing cellular response including cell migration, proliferation, and tissue-specific differentiation. While fabrication approaches such as solvent leaching and freeze-casting are commonly used for the generation of microporous biomaterial scaffolds, these methods provide little control over scaffold geometry and creation of complex tissue structures. Extrusion-based 3D printing, an additive manufacturing method, is a highly versatile layer-by-layer printing approach that allows for the fabrication of easily customizable 3D scaffolds with complex architecture using a vast selection of polymeric bioinks. These 3D printed constructs are porous at the macroscale, but achieving microporosity (i.e., < 100 μm) in printed constructs is challenging due to the sub-optimal resolution of the extrusion-based printing method. A recent study using alginate inks introduced a new fabrication technique termed Freeze-FRESH(FF)that combines extrusion printing and freeze-casting approaches to generate 3D constructs with hierarchical microporosity. However, the porosity of the resultant alginate constructs was comparable despite changing the freezing temperature. In the current study, the FF method was modified to print collagen constructs with greater control of microporous architecture. Highly concentrated collagen type I ink was used to 3D print collagen constructs using the freeform reversible embedding of suspended hydrogels (FRESH)approach. Modification of the FF technique entailed melting of the FRESH bath post printing via incubation at 37 °C followed by freezing and lyophilization to allow for collagen gelation and better heat transport during the freezing process. The effect of freezing temperature on micropore size, swelling degree, degradation, and mechanical properties of printed constructs was assessed. Finite element (FE) models were generated to predict the mechanical properties of microporous scaffolds. In addition, the effect of microporosity on Saos-2 cell morphology, proliferation, infiltration, and ALP activity was evaluated. Results from the study showed that freezing temperature effectively modulated micropore size and that constructs with larger micropore size were more stable. Microporosity had no effect on swelling ratio yet caused a decrease in mechanical properties of collagen constructs; FE models confirmed experimental results. Cell metabolic activity and infiltration was enhanced in microporous constructs with larger pore size, yet there was no effect on cell morphology, and ALP activity. In conclusion, the modified FF technique can be effectively used to fabricate hierarchically porous 3D collagen constructs.

School principals address problems that extend way beyond the classroom. This qualitative study is about dispositions of emotional intelligence in school principals. The participants interviewed were novice school principals or principal coaches. The results showed that principals tend to address the needs of others more than their own. Strategies are shared on how to improve emotional intelligence. Additionally, the study provides recommendations that school districts can implement to ensure the success of school principals.

On March 11, 2020, Miami-Dade County had its first diagnosed case of COVID-19 and quickly became the top hotspot of COVID-19 cases in Florida. Hospitals were challenged to find effective treatments to combat the virus. A collaborative effort between Doctors Hospital and Boca Raton Regional Hospital established a robust convalescent plasma program for critically ill patients diagnosed with COVID-19 within Baptist Health South Florida.

Background: In this study, we investigated if a combination of the nutraceutical supplement, EvenFlo and folic acid will be superior to the standard stand-alone use of folic acid.

Methods: We conducted a randomized double-blind, active-controlled, clinical trial. A total of 70 subjects with SCD ages 5-12 years were enrolled into the study with 35 in the intervention group and 35 in the control group; 61 completed the trial (32 from the intervention group and 29 from the control group).

In this paper, we report a simple and inexpensive paper-based microfluidic device for detecting nitrate in water. This device incorporates two recent developments in paper-based technology suitable for nitrate detection and has an optimized microfluidic design. The first technical advancement employed is an innovative fibrous composite material made up of cotton fibers and zinc microparticles that can be incorporated in paper-based devices and results in better nitrate reduction. The second is a detection zone with an immobilized reagent that allows the passage of a larger sample volume. The importance of microbiota in the health and diseases of farm animals has been well-documented for diverse animal species. However, studies on microbiotas in turkey and turkey farms are relatively limited as compared to other farm animal species. In this study, we performed a comprehensive survey of the litter microbiotas in 5 commercial turkey farms in the Northwest Arkansas (H, M, V, K, and R farms) including one farm with positive incidence of cellulitis (R farm). Altogether 246 boot swabs were used for 16S rRNA gene profiling of bacterial communities. At phylum level, 11 major bacterial phyla (≥0.01%) were recovered. …

This research study evaluates the outcomes of pastoral counseling on mental health within a predominantly African American congregation in the southeastern United States. The research study is intended to build a further understanding of the pastor’s role in counseling individuals with mental health conditions, given individuals seek counseling as a treatment option. To evaluate those perceptions, the researcher analyzed data collected from a survey. The outcomes from this qualitative data study suggest that pastoral counseling alone is ineffective given several factors such as individual failure to seek counseling, no suggested mental-health related issues within the sampled population, and ethnicity barriers. …

Over the past thirty years, research has resulted in increased understanding of these experiences and the unique trajectory that values-based, as opposed to fear-based, traumas can cause. Researchers posit that teachers’ excessive absences may be caused by a lack of commitment or dissatisfaction with their jobs due to school environments and relationships, and a lack of commitment can ultimately influence student achievement. This correlational study sought to determine any relationship between teachers’ absenteeism and job satisfaction as mitigated by perceptions of favoritism. Self-induced moral injuries caused by compulsive sexual behavior can result in experiences of sexual shame. Shame is a pervasive experience of unworthiness, and it can become a significant obstacle to life and relationship satisfaction because it leads to negative cognitions and isolating behaviors. Self-induced moral injuries occur when a person's wrongdoing challenges their worldview and spiritual beliefs. This study’s purpose was to better understand how narcissistic admiration and rivalry relate to relationship satisfaction and to investigate the satisfaction found in homogamous versus heterogamous narcissistic relationships. The study explored relationship satisfaction of married participants (N = 764), as moderated by their partner’s perceived level and subtype of narcissism. It also examined direct correlations between participant narcissistic admiration and rivalry and relationship satisfaction, as well as correlations between participant and perceived partner narcissism scores. Lanthanide elements have been recently recognized as “new life metals” yet much remains unknown regarding lanthanide acquisition and homeostasis. In Methylorubrum extorquens AM1, the periplasmic lanthanide-dependent methanol dehydrogenase XoxF1 produces formaldehyde, which is lethal if allowed to accumulate. This property enabled a transposon mutagenesis study and growth studies to confirm novel gene products required for XoxF1 function. The identified genes encode an MxaD homolog, an ABC-type transporter, an aminopeptidase, a putative homospermidine synthase, and two genes of unknown function annotated as orf6 and orf7. Viral pneumonia remains a global health threat despite worldwide vaccination and therapeutic programs. The influenza pandemic of 1918 and SARS-CoV2 pandemic of 2019-2020 are cautionary reminders demanding the need for novel treatment strategies. Moreover, in addition to causing acute disease, respiratory virus infections are often complicated by chronic lung pathologies, such as asthma induction, progression, and exacerbation. Hyperpolarized Magnetic Resonance Imaging (HP MRI) is an emerging modality that enables non-invasive interrogation of cells and tissues with unprecedented biochemical detail. This technology provides rapid imaging measurements of the activity of a small quantity of molecules with a strongly polarized nuclear magnetic moment. This polarization is created in a polarizer separate from the imaging magnet, and decays continuously towards a non-detectable thermal equilibrium once the imaging agent is removed from the polarizer and administered by intravenous injection. Specialized imaging strategies are therefore needed to extract as much information as possible from the HP signal during its limited lifetime.

Intratumoral genetic heterogeneity describes the molecular differences among subclones within a tumor and is a major barrier to effective therapy in many solid and liquid cancers, including chronic lymphocytic leukemia (CLL). Rare, treatment-resistant subclones can expand to compose relapsed disease during tumor evolution. Examination of malignant evolution in the context of specific treatment provides insight into the molecular lesions that mediate therapeutic response and resistance. Pancreatic ductal adenocarcinoma (PDAC) is a highly aggressive malignancy with a very poor patient prognosis (5-year survival of ≤ 7%). While transcriptional profiling has aided in the classification of this disease into at least two broader subtypes, this alone has so far been insufficient to inform on more nuanced patterns of oncogenic dependency. We hypothesized that a more comprehensive and granular characterization of PDAC disease diversity is required to establish relevant context for targeted therapy. Trypanosoma brucei is a parasitic eukaryote and is the causal agent of the disease known as African sleeping sickness, transmitted via the bite of a tsetse fly. If left untreated, the parasite ultimately crosses the blood-brain eventually leading to death. Currently there are 5 approved drugs used to treat it, each with toxic side effects and effective at specific disease stages. A more effective and less toxic drug candidate is highly sought after. The essential enzyme, dUTPase, is an excellent drug target in the parasite while utilizing S. cerevisiae as a model system.

Cell stress and DNA damage activate the tumor suppressor p53, triggering transcriptional activation of a myriad of target genes. The molecular, morphological, and physiological consequences of this activation remain poorly understood in vivo. We activated a p53 transcriptional program in mice by deletion of Mdm2, a gene which encodes the major p53 inhibitor. By overlaying tissue-specific RNA-sequencing data from pancreas, small intestine, ovary, kidney, and heart with existing p53 ChIP-sequencing, we identified a large repertoire of tissue-specific p53 genes and a common p53 transcriptional signature of seven genes which included Mdm2 but not p21.
