Role Of Glycolytic Metabolism In Glioblastoma Multiforme, Shreya Udawant Dec 2020

Role Of Glycolytic Metabolism In Glioblastoma Multiforme, Shreya Udawant

Theses and Dissertations

PI3K/AKT/mTOR signaling pathway is activated in 90% of all Glioblastoma Multiforme (GBM) tumors. We used a dual targeting approach to interfere with the phosphoinositide 3-kinase- mammalian target of rapamycin (PI3K-mTOR) pathway by treating U87MG GBM cells with NVP-BEZ235 (PI3K and mTOR a dual inhibitor) and, their downstream effects on molecular regulation on glycolytic genes were investigated using RNA-sequencing and gene set enrichment analysis (GSEA). RNA-seq identified 7803 differentially regulated genes in response to NVP-BEZ235. GSEA identified two glycolysis-related gene sets had significant differences (P < 0.05) in gene expression between control and NVP-BEZ235 treated samples. The GSEA core enrichment reported 17 and 35 genes were significantly downregulated (P adj < 0.05) in response to NVP-BEZ235 treatment in two different glycolysis related gene sets. We validated the expression of genes using qRT-PCR in U87MG cells as well as other GBM cell lines. Our results suggest novel insights on hallmark metabolic reprogramming associated with the PI3K-mTOR inhibition and provides advances in developing molecular targeted therapies.

Differentiating Between Invasive And Native Populations Of Bighead And Silver Carp Using Ms-Aflp, Erika Renee Sarvela Dec 2020

Differentiating Between Invasive And Native Populations Of Bighead And Silver Carp Using Ms-Aflp, Erika Renee Sarvela

Theses

When a species is introduced outside their native range, the genetic diversity of the introduced population is generally decreased due to the founder effect, and the fitness of individuals in the introduced population may decrease due to inbreeding depression. Invasive species are a paradox to this paradigm because while the initial population size of an invasive species may be small in their non-native range, the individuals are able to survive, eat, and reproduce so successfully, that they have deleterious effects on native species. One mechanism that invasive species use to overcome a lack of genetic diversity and adapt to their …

Identification Of Micrornas Related To Type 2 Diabetic Osteopath, Mouza Mohammed Saeed Alfashti Alaleeli Dec 2020

Identification Of Micrornas Related To Type 2 Diabetic Osteopath, Mouza Mohammed Saeed Alfashti Alaleeli

Theses

T2DM is linked to an increase in the fracture rate as compared to the non-diabetic population even with normal or raised Bone Mineral Density (BMD). Previous studies have addressed the question of how T2DM induces osteoporosis; the exact underlying mechanism is still elusive. Bones undergo continuous remodelling throughout life. Bone remodelling implicates the coupling of osteoclastic bone resorption and osteoblastic bone formation. This process is controlled by numerous genetic, hormonal, and neurogenic mechanisms. Osteoporosis is a result of bone loss that occurs by uncoupled remodelling. The existing evidence indicates that short non-coding RNAs known as microRNAs (miRNAs), are the key …

Systems Genetics And Systems Biology Analysis Of Paraquat Effects In Bxd Recombinant Inbred Mice, Carolina Del Valle Torres Rojas

Theses and Dissertations (ETD)

Paraquat (PQ) is a chemical herbicide that is used in many countries including the United States. It is also highly acutely toxic to humans and has been used as a means of suicide. As PQ is applied mainly in agricultural settings, it moves to soil and well water. Chronic low dose exposure via drinking water may have adverse effects on humans, including increased risk for sporadic Parkinson’s disease (sPD). The etiology of sPD is unclear and the most accepted hypothesis states it is the result of the interaction between environmental factors and genetic susceptibility. Increasing evidence led us to infer …

Early Diagnosis Of Alzheimer’S Disease In The Primary Care Setting, Raymond R. Romano Dec 2020

Early Diagnosis Of Alzheimer’S Disease In The Primary Care Setting, Raymond R. Romano

Theses and Dissertations (ETD)

The burden of Alzheimer’s disease (AD) affects not just the individual but also families, providers, and society. Early recognition and diagnosis of AD may reduce cost by reducing interaction with the health care system, earlier initiation of treatment, and prolonging time to long- term care. Primary care providers, the first contact for diagnosis of patients with AD, are not fulfilling the potential of early diagnosis for a variety of reasons. Biomarkers of AD emerge on average 15 to 20 years before clinical diagnosis, yet currently established biomarkers are not easily available in the primary care setting. A growing body of …
