Covariate-Adaptive And Response-Adaptive Clinical Trial Designs, Lu Wang Aug 2020

Covariate-Adaptive And Response-Adaptive Clinical Trial Designs, Lu Wang

Dissertations and Theses (Open Access)

Most of the research on Response-adaptive randomization(RAR) designs has been focused on clinical trials with a single endpoint. However modern clinical trials are often complex, with multiple competing objectives and multiple endpoints. We overcome the obstacles introduced by the large number of unknown parameters and the possible correlations between the multiple endpoints. We obtained the optimal allocation proportions for the following two optimization problems: (1) maximizing the power of the test of homogeneity with a xed sample size, and (2) minimizing the expected weighted number of failures with a fixed power. Further, we implemented these optimal allocations through response-adaptive randomization …

Chapter 1

Information disclosure can help overcome problems associated with asymmetric information. For example, Lewis (2011) argues that sellers with high-quality products will reveal more detailed information about their products, allowing them to charge higher prices. Using data from eBay marketplace, I empirically study the effect of information disclosure in mitigating asymmetric information. I find that auctions generally sell for a higher price when sellers disclose more information, and this effect appears stronger when the quality level of the product is more uncertain. However, when other quality assurance mechanisms are available, sellers may not rely on information disclosure purely as …

The Influence Of Cerebellar Transcranial Direct Current Stimulation On Motor Function In Parkinson’S Disease, Lidio Lima De Albuquerque

UNLV Theses, Dissertations, Professional Papers, and Capstones

Parkinson's disease (PD) is the most common movement disorder and the second most common neurodegenerative disorder. PD is characterized by dopaminergic cell loss in the substantia nigra pars compacta, which leads to a reduction in dopamine in the striatum. These physiological mechanisms lead to a number of motor impairments such as bradykinesia, rigidity, tremor, and postural instability that severely limit the ability of individuals with PD to perform many essential daily living activities. Although current pharmacological, surgical, and physical exercise treatment approaches are valuable they are either only mildly effective, expensive, or associated with a variety of side effects. Therefore, …

A Symbiosis Between A Dark Septate Fungus, An Arbuscular Mycorrhiza, And Two Plants Native To The Sagebrush Steppe, Craig Lane Carpenter

Boise State University Theses and Dissertations

Plant roots form symbioses with various fungi, including arbuscular mycorrhizae (AMFs) and dark septate endophytes (DSEs). The symbiosis between plants and AMFs has been extensively studied and is generally considered to be mutualistic. Much less is known about the symbiosis between plants and DSE. In sagebrush habitats, DSEs are common, but their effects on the vegetation are unclear. As a first step to study these effects, I isolated and cultured a DSE from the roots of the shrub Artemisia tridentata. Based on partial sequences of five genes and phylogenetic analyses, the isolated fungus was a non-described species within the …

Making The Case For A Right To A Healthy Environment For The Protection Of Vulnerable Communities: A Case Of Coal-Ash Disaster In Puerto Rico, Sarah Dávila-Ruhaak

Michigan Journal of Environmental & Administrative Law

The connection between the environment and human rights is not a surprising one. The enjoyment of human rights depends on a person’s ability to live free from interference and to have his or her rights protected. The interdependence of human rights and the protection of the environment is manifested in the full and effective enjoyment of the right to a healthy environment. This article argues that in order to protect vulnerable persons and communities facing environmental harm, a human rights framework—specifically the right to a healthy environment—must be applied. A human rights approach complements environmental justice work, recognizing that individuals …

Hyperammonaemia in children can lead to grave consequences in the form of cerebral oedema, severe neurological impairment and even death. In infants and children, common causes of hyperammonaemia include urea cycle disorders or organic acidaemias. Few studies have assessed the role of extracorporeal therapies in the management of hyperammonaemia in neonates and children. Moreover, consensus guidelines are lacking for the use of non-kidney replacement therapy (NKRT) and kidney replacement therapies (KRTs, including peritoneal dialysis, continuous KRT, haemodialysis and hybrid therapy) to manage hyperammonaemia in neonates and children. Prompt treatment with KRT and/or NKRT, the choice of which depends on the …

Capturing Functional Epigenomes For Insight Into Metabolic Diseases., Fiona Allum, Elin Grundberg

Manuscripts, Articles, Book Chapters and Other Papers

BACKGROUND: Metabolic diseases such as obesity are known to be driven by both environmental and genetic factors. Although genome-wide association studies of common variants and their impact on complex traits have provided some biological insight into disease etiology, identified genetic variants have been found to contribute only a small proportion to disease heritability, and to map mainly to non-coding regions of the genome. To link variants to function, association studies of cellular traits, such as epigenetic marks, in disease-relevant tissues are commonly applied.

SCOPE OF THE REVIEW: We review large-scale efforts to generate genome-wide maps of coordinated epigenetic marks and …

Premise AVP1 (H+-pyrophosphatase) and MIOX4 (myo-inositol oxygenase) are genes that, when overexpressed individually, enhance the growth and abiotic stress tolerance of Arabidopsis thaliana plants. We propose that pyramiding AVP1 and MIOX4 genes will further improve stress tolerance under water-limited and salt-stress conditions. Methods MIOX4 and AVP1 reciprocal crosses were developed and phenomic approaches used to investigate the possible synergy between these genes. Results Under normal and stress conditions, the crosses had higher foliar ascorbate content than the wild-type and parental lines. Under water-limited conditions, the crosses also displayed an enhanced growth rate and biomass compared with the control. The observed …

Epithelial Responses To Porphyromonas Gingivalis In The Community Context., Zackary Ray Fitzsimonds Aug 2020

Epithelial Responses To Porphyromonas Gingivalis In The Community Context., Zackary Ray Fitzsimonds

Electronic Theses and Dissertations

Porphyromonas gingivalis, a keystone pathogen for periodontal disease, initiates a complex molecular dialogue with gingival epithelial cells, leading ultimately to disruption of host signaling pathways. Dysbiotic host responses are also thought to contribute to the initiation and progression of oral squamous cell carcinomas. Recently, the homeostatic commensal Streptococcus gordonii has been shown to antagonize P. gingivalis-induced epithelial cell signaling events in host cells, such as proliferation and migration. The aim of this study was to characterize pathways that P. gingivalis targets to disturb host signaling, with a specific focus on pathways that S. gordonii can restore to homeostatic …
