Functional recovery in the end target muscle is a determinant of outcome after peripheral nerve injury. The neuromuscular junction (NMJ) provides the interface between nerve and muscle and includes non-myelinating terminal Schwann cells (tSCs). After nerve injury, tSCs extend cytoplasmic processes between NMJs to guide axon growth and NMJ reinnervation. The mechanisms related to NMJ reinnervation are not known. We used multiple mouse models to investigate the mechanisms of NMJ reinnervation in both sexes, specifically whether macrophage-derived vascular endothelial growth factor-A (Vegf-A) is crucial to establishing NMJ reinnervation at the end target muscle. The isolation, purification, and clinical deployment of antibiotics is one of the major drivers of decrease in morbidity and mortality from infectious bacteria in the 20th century. The rapid, ubiquitous deployment of antibiotics encouraged swift development and distribution of antibiotic resistance. New, novel techniques, technologies, and ultimately therapeutic antimicrobial compounds will be required to counter the rise of antibiotic resistant microbes. Historically, mimicking naturally occurring compounds has been the most fruitful method for discovering new antibiotics; unsurprisingly, many recent efforts have focused on expanding the cultivation and detection of previously unknown microbes and compounds, respectively. Serving as a visual representation of a different time and place, barns and agricultural architecture across America have both historical and cultural values that are imitable. Evoking a strong sense of nostalgia and simplicity, barns are an often-overlooked architectural typology worth preserving. Native to Western New York, historical Wells style barns have served as an iconic agricultural symbol since the late 1800s. As the means and methods of agriculture change with technological development, more and more of these exemplary structures are becoming neglected and obsolete. A Rapid Spectrophotometric Assay For Quantifying Seed Coat Saponins In Quinoa, Sydney Diver Szabo

Background and objectives: The commonly used afrosimetric foam-height method for quantifying saponins in quinoa is rapid but imprecise. A rapid UV/Vis method, utilizing the Liebermann-Burchard (LB) color reaction, was compared to the foam method across a range of saponin levels in washed and unwashed seed. Findings: A 6 min UV/Vis method provided greater precision and accuracy than the afrosimetric method at the lower saponin levels found in washed quinoa. The afrosimetric method did not differentiate saponin levels below 0.6 mg/mL but allows for useful relative comparisons of saponin content in unwashed quinoa where the foam height is large. Retellings of classic fairy tales have become increasingly popular in the past decade, but little research has been done on the novelizations written for a young adult (YA) audience. Critical multicultural analysis determining the effect of race, gender, disability, and more has been completed for both original fairy tale retellings and fairy tale retellings for children, but scholars have neglected popular YA novelizations. Anime tourism has emerged as an important form of tourism resulted from both online and offline human activities and interactions. However, investigations into anime tourism motivations remain limited. Using Chinese anime tourists as study subjects and employing a mixed methods approach, this study develops a five-dimension anime tourism motivation scale with sufficient reliability and validity. The five motivational factors are anime authenticity seeking, novelty, escape/relaxation, socialisation, and anime cultural exploration. The results help understand unique anime tourist behaviours, expand the current knowledge of media-induced tourism, and extend contemporary cultural consumption literature. People from across the Caylloma Province of Peru are adapting to social-ecological changes, such as decreasing water supplies and rapidly retreating glaciers, in different and unequal ways. In this study, we show how this inequality is intersectional; identities like gender and age compound and interact with systems of power to shape how people adapt to these changes at the individual, household, and district scales. We draw from 130 semi-structured interviews with agricultural actors and participant observations in district and regional meetings across four districts, to demonstrate how intersectionality shapes adaptation to social-ecological change. Epidemiological studies indicate that elevated alkaline phosphatase activity is associated with increased cardiovascular disease risk. Other epidemiological data demonstrate that mothers giving multiple childbirths (multipara) are also at increased risk of developing late-onset cardiovascular disease. We hypothesized that these two associations stem from a common cause, the insufficient plasma level of the ectopic mineralization inhibitor inorganic pyrophosphate, which is a substrate of alkaline phosphatase. As alkaline phosphatase activity is elevated in pregnancy, we hypothesized that pyrophosphate concentrations decrease gestationally, potentially leading to increased maternal vascular calcification and cardiovascular disease risk in multipara. Metabolic dysfunction underlies several chronic diseases, many of which are exacerbated by obesity. Dietary interventions can reverse metabolic declines and slow aging, although compliance issues remain paramount. 17α-estradiol treatment improves metabolic parameters and slows aging in male mice. The mechanisms by which 17α-estradiol elicits these benefits remain unresolved. Herein, we show that 17α-estradiol elicits similar genomic binding and transcriptional activation through estrogen receptor α (ERα) to that of 17β-estradiol. In addition, we show that the ablation of ERα completely attenuates the beneficial metabolic effects of 17α-E2 in male mice. Our findings suggest that 17α-E2 may act through the liver and …

Myostatin (MSTN) is a transforming growth factor-β (TGF-β) family member that normally acts to limit muscle growth. The function of MSTN is partially redundant with that of another TGF-β family member, activin A. MSTN and activin A are capable of signaling through a complex of type II and type I receptors. Here, we investigated the roles of two type II receptors (ACVR2 and ACVR2B) and two type I receptors (ALK4 and ALK5) in the regulation of muscle mass by these ligands by genetically targeting these receptors either alone or in combination specifically in myofibers in mice.
