Age-Dependent Effects Of Eedq On Cocaine-Induced Locomotor Activity And D2 Receptor Supersensitivity, Angie Teran Jul 2019

Age-Dependent Effects Of Eedq On Cocaine-Induced Locomotor Activity And D2 Receptor Supersensitivity, Angie Teran

Electronic Theses, Projects, and Dissertations

The neurochemical changes occurring between the preweanling period and adolescence could be crucial for understanding the role development plays in the manifestation of psychotic behaviors. N-ethoxycarbonyl-2-ethoxy-1,2-dihydroquinoline (EEDQ) fully attenuates the DA agonist-induced behaviors of adult rats, while potentiating the DA agonist-induced locomotor activity of preweanling rats. My specific hypotheses were as follows: (1) Systemically administered EEDQ would block the cocaine-induced locomotor activity of adult rats. (2) Systemically administered EEDQ would potentiate the cocaine-induced locomotion of preweanling rats. (3) EEDQ would increase the Emax values (a measure of D2 receptor sensitivity) of preweanling rats, but not adolescent or adult rats. …

The proteins derived from the foliated shell layer of the oyster, Crassostrea virginica, are unusually acidic and highly phosphorylated. Here we report the identification of a gene encoding a member of this class of phosphoproteins that we collectively refer to as folian. Using an in silico approach, a virtual probe was constructed from an N-terminal sequence (DEADAGD) determined for a 48 kDa folian phosphoprotein and used to screen an oyster EST databank. A sequence that matched the N-terminus of the 48 kDa protein was found and used to identify the full-length gene from a C. virginica BAC library. The …

Uncoordinated Maturation Of Developing And Regenerating Postnatal Mammalian Vestibular Hair Cells, Tian Wang, Mamiko Niwa, Zahra N. Sayyid, Davood K. Hosseini, Nicole Pham, Sherri M. Jones, Anthony J. Ricci, Alan G. Cheng

Department of Special Education and Communication Disorders: Faculty Publications

Sensory hair cells are mechanoreceptors required for hearing and balance functions. From embryonic development, hair cells acquire apical stereociliary bundles for mechanosensation, basolateral ion channels that shape receptor potential, and synaptic contacts for conveying information centrally. These key maturation steps are sequential and presumed coupled; however, whether hair cells emerging postnatally mature similarly is unknown. Here, we show that in vivo postnatally generated and regenerated hair cells in the utricle, a vestibular organ detecting linear acceleration, acquired some mature somatic features but hair bundles appeared nonfunctional and short. The utricle consists of two hair cell subtypes with distinct morphological, electrophysiological …

Feminist Ecclesiology: A Trinitarian Framework For Transforming The Church's Institutional And Spiritual Life, Stacy Geere Jul 2019

Feminist Ecclesiology: A Trinitarian Framework For Transforming The Church's Institutional And Spiritual Life, Stacy Geere

LMU Theses and Dissertations

In light of women’s marginal status in church governance and ministry through most of recorded history, feminist trinitarian ecclesiology is needed to transform the church’s institutional and spiritual life. While Vatican II provided the paradigm shift and promising anthropology essential for an egalitarian church, feminist ecclesiology prompts a radical transformation of its hierarchical and patriarchal structures and practices so that it may truly embody the Trinity. Trinitarian life provides practical and radical consequences for Christian life, and provides a model of church marked by relationships of equality, mutuality, unity and reciprocity. It also provides a strong ecclesiological argument for reform …

Ocean velocity defines ocean circulation, yet the available observations of subsurface velocity are under-utilized by society. The first step to address these concerns is to improve visibility of and access to existing measurements, which include acoustic sampling from ships, subsurface float drifts, and measurements from autonomous vehicles. While multiple programs provide data publicly, the present difficulty in finding, understanding, and using these data hinder broader use by managers, the public, and other scientists. Creating links from centralized national archives to project specific websites is an easy but important way to improve data discoverability and access. A further step is to …

1. Marine sponges are host to large, diverse communities of microorganisms. These microbiomes are distinct among sponge species and from seawater bacterial communities, indicating a key role of host identity in shaping its resident microbial community. However, the factors governing intraspecific microbiome variability are underexplored, and may shed light on the evolutionary and ecological relationships between host and microbiome.

2. Here, we examined the influence of genetic variation and geographic location on the composition of the Ircinia campana microbiome.

3. We developed new microsatellite markers to genotype I. campana from two locations in the Florida Keys, USA, and characterised their …

Exploring Construct Validity And Measurement Invariance Of The Cyberbullying Experiences Survey, Kendall Moore Jul 2019

Exploring Construct Validity And Measurement Invariance Of The Cyberbullying Experiences Survey, Kendall Moore

Theses and Dissertations

Given recent calls for advancing valid instrumentation in the field of cyberaggression, the present study evaluated construct validity and measurement invariance for the Cyberbullying Experiences Survey (CES) in a high school and college student sample. A series of confirmatory factor analyses (CFA), reliability analyses, and a nomological net evaluation were conducted to address these aims. The data did not provide support for the hypothesized four-factor model for cyberaggression or cybervictimization (i.e., unwanted contact, malice, deception, and public humiliation). Upon implementing suggested and theoretically supported modification indices, support for a four-factor solution for both cyberaggression and cybervictimization was provided.

To subsequently …

The Role Of The Gut Microbiome In Tumor Development And Response To 5-Fluorouracil In The Apc^Min/+ Mouse, Kristen Michele Hogan Jul 2019

The Role Of The Gut Microbiome In Tumor Development And Response To 5-Fluorouracil In The Apc^Min/+ Mouse, Kristen Michele Hogan

Theses and Dissertations

Colorectal cancer (CRC) is the third most frequent cancer and the second leading cause of cancer related deaths. While CRC is initiated by mutations in critical tumor suppressor genes, its development and progression to malignancy is aided by host derived cells that are recruited to the tumor microenvironment (TME) and by cancer-associated microbiota that constitute the gut microbiome. Components of the gut microbiota have been shown to alter immune cell infiltration into tumors and influence response to therapy, disease stage, and patient survival. In previous studies, we found that ApcMin/+ mice, a model of early colon carcinogenesis, had reduced tumor …

Role Of Epigenome And Microbiome In Cannabinoid And Aryl Hydrocarbon Receptor-Mediated Regulation Of Inflammatory And Autoimmune Diseases, Zinah Zamil Al-Ghezi

Theses and Dissertations

Inflammation is considered to be the underlying cause of a majority of clinical disorders. Thus, studies aimed at understanding the signaling pathways that trigger inflammation could have significant translational impact. In the current study, we investigated the effect of activation of cannabinoid (CB) and Aryl hydrocarbon hydroxylase (AhR) receptors on the inflammatory response. To that end, we used cannabinoids such as Δ (9)-Tetrahydrocannabinol (THC) and cannabidiol (CBD), and a well-established AhR agonist namely 2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD). These were tested against two inflammatory disease models: 1) Experimental autoimmune encephalomyelitis (EAE), an animal model for human multiple sclerosis (MS), and 2) Exposure to …

Online course offerings are becoming more abundant in post-secondary education; such is the case with science courses at Tidal Wave Technical College. Online courses bring many benefits to students including flexibility, convenience, and the ability to learn anywhere, anytime. However, despite these benefits many challenges are also associated with online course offerings including student attrition. Using a non-major biology course as the test model this research sought to trial one possible way to alleviate the problems associated with online learning by incorporating a synchronous online orientation. Data on student attrition, success, and perception was gathered to gain insight into the …

Effects Of Human Dopamine Transporter (Dat) Mutations And Novel Allosteric Modulatory Compounds In Disrupting Hiv-1 Tat-Dat Protein Interaction, Pamela Marie P. Quizon Jul 2019

Effects Of Human Dopamine Transporter (Dat) Mutations And Novel Allosteric Modulatory Compounds In Disrupting Hiv-1 Tat-Dat Protein Interaction, Pamela Marie P. Quizon

Theses and Dissertations

More than 37 million people are living with HIV worldwide. Despite the widespread use of antiretroviral therapy (ART), up to 70% of HIV-positive individuals suffer from cognitive and behavioral deficits collectively known as HIV-associated neurocognitive disorders (HAND). HIV-mediated damage to the dopaminergic system is a mediating factor in HAND. Dopamine (DA) transporter (DAT)-mediated reuptake is essential for maintaining DA homeostasis. Because most ART cannot efficiently cross the blood-brain barrier (BBB), the brain serves as a viral reservoir that facilitates the spread of infection to microglia and astrocytes. Infected cells shed viral proteins such as Tat, which plays a critical role …

Longitudinal Prediction Of Delinquent Behavior In Early Adolescence, Kathleen Watson Macdonell Jul 2019

Longitudinal Prediction Of Delinquent Behavior In Early Adolescence, Kathleen Watson Macdonell

Theses and Dissertations

Childhood behavioral problems have consistently been reported as an antecedent for later problem behavior, including delinquency in adolescence and into adulthood. Parenting behaviors are an important influence on the potentially negative behavioral trajectory from childhood behavior problems to delinquency in early adolescence. The current study sought to provide further understanding of the relationship between teacher-reported third grade childhood behavior problems and two outcomes: ninth grade teacher-reported and ninth grade self-reported delinquency. Additionally, the moderating impact of parental monitoring and parental school involvement, assessed in fifth grade, was explored. Using a longitudinal sample of 556 participants, hypotheses were assessed separately for …

Leveraging Geotagged Social Media To Monitor Spatial Behavior During Population Movements Triggered By Hurricanes, Yago Martín González Jul 2019

Leveraging Geotagged Social Media To Monitor Spatial Behavior During Population Movements Triggered By Hurricanes, Yago Martín González

Theses and Dissertations

In a world of increased mobility and interconnectedness, the study of spatial behavior becomes more relevant than ever. However, multiple researchers have highlighted that the understanding of these dynamic processes has reached a bottleneck derived from the rigidity of traditional spatial behavior inquiry methods and the unavailability of trustworthy and relevant information. These difficulties are even more prominent during emergencies and disasters as these events often create scenarios where spatial behavior does not follow regular and logical patterns and where conventional mobility datasets are often skewed or not existent. Thus, many scholars working within the spatial behavior sub-discipline are pursuing …

Investigating The Neural Correlates Of Nicotine Withdrawal Phenotypes In Mice: Involvement Of Creb-Dependent Nrg3-Erbb4 Signaling In Mediating Anxiety-Like Behavior, Miranda Fisher Jul 2019

Investigating The Neural Correlates Of Nicotine Withdrawal Phenotypes In Mice: Involvement Of Creb-Dependent Nrg3-Erbb4 Signaling In Mediating Anxiety-Like Behavior, Miranda Fisher

Theses and Dissertations

Addiction to nicotine and the ability to quit smoking are influenced by genetic factors. Therefore, it is important to understand how genes and drugs of abuse mechanistically impact each other. One well-characterized protein responsible for regulating both response to drugs and gene expression is the transcription factor cAMP response element-binding protein (CREB). Work from our lab indicates that hippocampal specific alterations in CREB signaling and synaptic plasticity underlie certain nicotine withdrawal (WD) phenotypes in a region-specific manner. We found that CREB deletion in the ventral hippocampus (VH), a region known for regulation of mood and emotion, results in amelioration of …

Disinfection By-Products: Method Optimization For Quantification, Unknown Analysis, And Calculated Toxicity, Amy A. Cuthbertson Jul 2019

Disinfection By-Products: Method Optimization For Quantification, Unknown Analysis, And Calculated Toxicity, Amy A. Cuthbertson

Theses and Dissertations

Disinfected drinking water contains hundreds of disinfection by-products (DBPs) that are formed by the reaction of disinfectants with natural and anthropogenic organic matter, bromide, and iodide. Understanding what these DBPs are is important because millions of people worldwide consume drinking water every day, and human epidemiologic studies have reported cancer, miscarriage, and birth defects from consuming such waters. While more than 700 DBPs are reported in the literature, very few studies quantify complete classes of chlorinated, brominated, and iodinated DBPs. The following document contains five chapters in the format designated for specific scientific journals on this subject. Chapter 1 describes …

Glia Responsivity In Nicotine Dependence, Adewale Oluwamuyiwa Adeluyi Jul 2019

Glia Responsivity In Nicotine Dependence, Adewale Oluwamuyiwa Adeluyi

Theses and Dissertations

Neurons are the primary research target in the exploration and development of novel smoking cessation pharmacotherapies over the years. These research efforts have led to the discovery and development of three FDA-approved smoking cessation aids – nicotine replacement therapy, varenicline, and bupropion, each of which have quantifiable success as smoking cessation aids. However, relapse is still very common and smoking cessation is successful in less than 5% of quit attempts, making paramount the development of novel therapeutics. One untapped therapeutic target are glia, particularly, astrocytes and microglia, which are active participants in the neuroplastic events underlying drug addiction.

First, we …

Association Between Snps In The Vitamin D Binding Protein, Vitamin D Status, And Aggressive Prostate Cancer By Race, Diana Diaz Jul 2019

Association Between Snps In The Vitamin D Binding Protein, Vitamin D Status, And Aggressive Prostate Cancer By Race, Diana Diaz

Theses and Dissertations

Prostate cancer incidence and mortality have been decreasing in recent years, but it remains the most commonly diagnosed cancer among men. Racial disparities exist in prostate cancer with African Americans (AAs) having notably higher rates of incidence and mortality, lower survival rates, and are more likely to be diagnosed with a highergrade cancer compared to European Americans (EAs). Genome-wide association studies have revealed several single nucleotide polymorphisms (SNPs) within the vitamin Drelated GC gene that may be associated with prostate cancer aggressiveness. In addition to SNPs, previous research has suggested that low levels of 25-hydroxyvitamin D (25(OH)D) may be related …

Statistical Analysis Of Interval-Censored Data Subject To Additional Complications, Qiang Zheng Jul 2019

Statistical Analysis Of Interval-Censored Data Subject To Additional Complications, Qiang Zheng

Theses and Dissertations

Survival analysis is an important branch of statistics that studies time to event data (or survival data), in which the response variable is time to a certain event of interest. The most prominent feature of survival data is that the response is not exactly observed due to limits of the study design or nature of the event of interest. Interval-censored data are a common type of survival data and occur frequently in real life studies where subjects are examined at periodical follow ups. The response time is usually not observed, but the status of the event of interest is known …

Role Of Epigenome And Microbiome In Endocannabinoid-Mediated Regulation Of Inflammation During Diet-Induced Obesity, Kathryn Miranda

Theses and Dissertations

Obesity is an inflammatory disease involving accumulation of adipose tissue, gut microbiota dysbiosis, and over-activation of the endocannabinoid system (ECS). Mechanisms involved in ECS regulation of obesity-induced inflammation are not well understood. Presented here are three chapters involving microRNA (miR) regulation of inflammatory adipose tissue macrophages (ATM) during high-fat diet (HFD)-induced obesity and cannabinoid receptor-mediated regulation of gut microbiota dysbiosis.

ATMs are innate immune cells that drive chronic low-grade inflammation during obesity. Polarization between pro-(M1) and anti-(M2-like) inflammatory phenotypes influence insulin sensitivity and energy expenditure; however, the mechanisms involved are unclear. In study one, we characterized miRs involved in ATM …

Co-Occurrence And Non-Overlap Among Behavioral And Health-Related Problems In Preschool Children, Chandni Patel

Theses and Dissertations

Integrating mental health and healthcare services increases the reach of healthcare while reducing costs and stigma associated with seeking mental health services. To provide such services for preschool aged children we must first understand how behaviors related to healthy lifestyle and mental/behavioral health overlap. This study examined the relationship between behavior problems, sleep duration, screen time, and eating patterns and assessed the frequency of co-occurring problems in these areas. Because of the importance of parenting across these domains, the association between parent disciplinary strategies and the co-occurrence of behaviors in the different problem areas was assessed. MANOVAs revealed that there …

Mechanistic Studies Of Atypical Reactions And Uncommon Substrates Native To Bacterial Cytochrome P450 Enzymes OletJe And Nikq, Courtney Elizabeth Wise Jul 2019

Mechanistic Studies Of Atypical Reactions And Uncommon Substrates Native To Bacterial Cytochrome P450 Enzymes OletJe And Nikq, Courtney Elizabeth Wise

Theses and Dissertations

Cytochrome P450 (CYP) enzymes typically perform hydroxylation chemistry on small molecule substrates using atmospheric oxygen and electrons from redox partner proteins. This reactivity is dependent on a thiolate-ligated heme cofactor that also allows for spectroscopic detection of ligation, oxidation, and environmental changes to the iron metal. The onset of the genomics era has resulted in the discovery of CYPs that deviate from this paradigm in terms of reaction performed, cosubstrate requirements, and substrate-scope. This work focuses on two of these unusual P450s – OleT and NikQ. OleT performs an unconventional H2O2-dependent decarboxylation reaction on a standard small molecule fatty acid …

A Mixed Methods Psychometric Validation Of The Microaggression Experiences Among University Women Of Color Scale, Kinjal Pandya

Theses and Dissertations

The purpose of this mixed methods study was to develop a measure of gender and racial microaggressions among university women of color (WOC). Microaggressions are conceptualized as verbal, nonverbal, and environmental slights that can be intentional or unintentional, but communicate derogatory messages towards a group of marginalized people. Despite many qualitative, legal, and narrative reports of microaggressions against university WOC, there are no validated measures specific to the experiences of WOC in academic settings. This study attempts to fill that gap by creating and testing the factor structure of a measure of microaggressions experienced by university WOC. Phase I of …

A Phenomenological Look At What Motivates And Challenges Teachers To Use Outdoor Learning Activities, Kristin Elizabeth Peney

Theses and Dissertations

The specific problem of practice on which this study is focused is the lack of opportunities for students to engage in outdoor learning experiences (OLEs) and one contributing factor to this problem, the hesitance teachers demonstrate towards engaging their students and themselves in OLEs (Rickinson et al., 2004). The purpose of this study was to gain a deeper and more thorough understanding of the beliefs about outdoor learning experiences (OLEs) held by teachers in my context. An investigative action research design using the phenomenological approach was selected for this study as teacher beliefs are complex, and are best understood when …

Inhibition Of Return In Visual Search: Disentangling Overlapping Contributions With Event-Related Potentials, Allison Michelle Pierce Jul 2019

Inhibition Of Return In Visual Search: Disentangling Overlapping Contributions With Event-Related Potentials, Allison Michelle Pierce

Theses and Dissertations

Inhibition of return (IOR) refers to the finding that responses to previously attended locations are slower than those to previously unattended locations. Despite over 30 years of research on IOR, there is still no consensus in the field regarding what the underlying mechanism of this effect is. Although IOR is traditionally studied within spatial cueing paradigms, this effect is thought to reflect a mechanism that facilitates efficient visual search. The following studies explored the hypothesis that multiple processes contribute to the IOR effect in visual search and examined whether these are the same processes that result in IOR in cueing …
