Aim: To identify naturally occurring variants of IAPP capable of inhibiting the aggregation of human IAPP and protecting living cells from the toxic effects of human IAPP. Background: The loss of insulin-producing β-cells and the overall progression of type 2 diabetes appears to be linked to the formation of toxic human IAPP (hIAPP, Islet Amyloid Polypeptide, amylin) amyloid in the pancreas. Inhibiting the initial aggregation of hIAPP has the potential to slow, if not stop entirely, the loss of β-cells and halt the progression of the disease. Objective: To identify and characterize naturally occurring variants of IAPP capable of inhibiting …

Going Beneath The Surface: The Ecology Of Metalimnetic Cyanobacteria Layers In Three New Hampshire Lakes, Sabina Perkins Aug 2019

Going Beneath The Surface: The Ecology Of Metalimnetic Cyanobacteria Layers In Three New Hampshire Lakes, Sabina Perkins

Master's Theses and Capstones

Cyanobacteria blooms take different forms: surface scums, subsurface epilimnetic blooms, benthic mats, and in some lakes, metalimnetic layers. There is limited field research on the seasonal behavior of metalimnetic layers and the contribution of layer-produced cyanotoxins to the overall toxin profile of New Hampshire lakes. Three lakes in New Hampshire were monitored monthly pre-summer stratification through post-fall mixis in 2018, tracking the formation and persistence of deep-water cyanobacteria layers and the physical/chemical/light environments where they were found. Cyanobacteria abundance and dominance were quantified through cell counts using an Imaging Flow Cytobot (IFCB) and with phycocyanin fluorescence estimates. Persistent, nearly monospecific …

The Effects Of Climate Change On The Heart Rates & Growth Of Sea Slugs In The Gulf Of Maine, Jennifer Leigh Gibson

Master's Theses and Capstones

In the next 80 years, sea surface temperatures are expected to increase by 1.5o to 2oC and ocean pH is expected to drop by 0.06 to 0.32 units, with exacerbated effects seen in coastal waters. Temperature increase has already forced organisms to shift their range polewards and ocean acidification has negatively affected calcifying organisms. Interactive effects, only more recently studied, vary depending on phylum and life cycle stage. This study examined both the upper thermal tolerance and interactive effect of temperature and acidification on the heart rate of five cold-water species of nudibranchs (Aeolidia papillosa, Cuthona gymnota, Dendronotus frondosus, Flabellina …

Chronic periodontitis (ChP) is a prevalent inflammatory disease affecting 46% of the US population. ChP produces a profound local inflammatory response to dysbiotic oral microbiota that leads to destruction of alveolar bone and tooth loss. ChP is also associated with systemic illnesses, including cardiovascular diseases, malignancies, and adverse pregnancy outcomes. However, the mechanisms underlying these adverse health outcomes are poorly understood. In this prospective cohort study, we used a highly multiplex mass cytometry immunoassay to perform an in-depth analysis of the systemic consequences of ChP in patients before (n = 28) and after (n = 16) periodontal treatment. A high-dimensional …

Using Geographic Information System Technology To Identify Environmental Education Field Sites, Whitney Morgan, Rebecca Barlow

Journal of Extension

Natural resources Extension educators use outdoor environmental education sites to increase public knowledge of environmental issues, but locating appropriate sites can be time-consuming. Geographic information system (GIS) software and geospatial data can help Extension professionals choose such sites more efficiently. We describe our use of GIS technology to select a natural resources field day site in Tuskegee National Forest in Alabama. We used site characteristics such as forest type, road access, and slope to narrow potential sites from 94 possibilities to the two best options. Sources of free and low-cost geospatial data also are discussed.

Regional Variation In Grass, Sedge, And Cereal Cultivation During The Viking Age In Skagafjörður, North Iceland, Melissa M. Ritchey Aug 2019

Regional Variation In Grass, Sedge, And Cereal Cultivation During The Viking Age In Skagafjörður, North Iceland, Melissa M. Ritchey

Graduate Masters Theses

In Viking Age and Medieval Iceland, livestock forage was a critical resource in the Norse agropastoral economy. Cereal cultivation, typically an important part of the Norse economy, may have been more limited in marginal sub-Arctic Iceland. An analysis of macrobotanical seed assemblages from archaeological excavations at 42 Viking Age and Medieval farmsteads in the Skagafjörður region of North Iceland suggests both broad trends and substantial variation over time and space in agropastoral production practices. This study finds that the main components of livestock forage (grass, sedge, and perhaps cereal) are highly variable between regions and over time. Interestingly, barley (Hordeum …

Comparative Spending Of Medicaid Dollars On Child Participants Of Kentucky’S Sobriety Treatment And Recovery Teams Program Versus A Matched Comparison Group., Matthew Thomas Walton

Electronic Theses and Dissertations

Child protective services agencies have long observed the complicating role that parental substance use and addiction plays in cases of child maltreatment. Families who struggle with these problems present unique challenges for child welfare professionals. These families are typically more difficult to engage, more likely to have children removed from the home, and have poorer outcomes when compared to other families. These poorer outcomes often include health problems. Addiction has well-known effects on health, and the specific manifestations of these problems for parents have been documented for years in child protection casework. However, what has been less investigated are the …

Numerous studies have examined the association between pre-admission and/or during nursing program variables and the outcome on the National Council Licensure Examination for Registered Nurses (NCLEX-RN®; National Council of State Boards of Nursing [NCSBN]; 2015) but very few studies have examined post-graduation variables. This study examined post-graduation variables associated with the outcome on the NCLEX-RN® examination. The purpose of this exploratory field study was twofold. The first aspect was to identify the benchmark score on a post-graduation test (the Kaplan RN Readiness Exam; Sanders, 2014) correlated with passing the NCLEX-RN® examination. The second aspect was to explore factors that the …

The Gsk-3Β-Fbxl21 Axis Regulates Tcap Via Ubiquitin-Mediated Proteasomal Pathway In The Cytoplasm, Jiah Yang Aug 2019

The Gsk-3Β-Fbxl21 Axis Regulates Tcap Via Ubiquitin-Mediated Proteasomal Pathway In The Cytoplasm, Jiah Yang

Dissertations and Theses (Open Access)

Protein turnover is one of the most essential mechanisms controlling circadian rhythms. F-Box and Leucine Rich Repeat Protein21 (FBXL21) is a circadian E3 ligase which shows oscillatory mRNA transcripts and protein levels. It was previously found to perform subcellular compartment-specific E3 ligase activities targeting the core clock proteins CRYPTOCHROME(CRY)1/2. Here we identified a new sarcomeric target substrate, Telethonin(TCAP), which also shows circadian oscillation in its mRNA transcript and protein expression and, importantly, interaction with FBXL21 in an anti-phasic manner. Via computational and pharmacological tests, we identified Glycogen Synthase Kinase-3β(GSK-3β) as a regulator of FBXL21. Biochemical and molecular characterizations demonstrated that …

The Role Of Secretory Phospholipase A2 Group Iia In Obesity And Metabolism, Michael S. Kuefner

Theses and Dissertations (ETD)

Secretory phospholipase A2 group IIA (PLA2G2A) is a member of a family of secretory phospholipases previously implicated in inflammation, atherogenesis, and antibacterial actions. These enzymes hydrolyze glycerophospholipids at the sn-2 position releasing lysophospholipids and fatty acids. Though studies have shown PLA2G2A is pro-inflammatory and promotes atherosclerosis, no research has analyzed the role of this enzyme in obesity and metabolism. Studies in the past 5-10 years utilizing various knock- out or over-expression mouse models have analyzed the role of different secretory phospholipase A2s (sPLA2) in metabolic diseases. From these studies, it is known that at least seven of the 11 sPLA2 …

An Integrated Model Of Eating Disorder Risk And Protective Factors: Implications For Research And Treatment, Leslie Alison Davis-Waddle

Electronic Theses and Dissertations

Body dissatisfaction and disordered eating behaviors are significant public health concerns, especially among college-age women. Despite extensive research, the complex nature of disordered eating and its etiology has prevented a satisfactory predictive model from being developed and validated and has therefore hindered the development of effective prevention and intervention strategies. This study aimed to integrate four of the most common etiological theories into a model to predict disordered eating behaviors. Bivariate correlations and moderation analyses were conducted to determine the strength of relationships between variables and to determine whether a protective factor (i.e., self-compassion) moderates the effects of risk factors …
