Department of Medical Oncology Faculty Papers
t(8;21)(q22;q22) is present in ~5–10% of patients with de novo acute myeloid leukemia (AML) and is associated with a better overall prognosis. Variants of the t(8;21) have been described in the literature, however, their clinical and prognostic significance has not been well-characterized. Molecular profiling of these cases has not previously been reported but may be useful in better defining the prognosis of this subset of patients. We present two cases of variant t(8;21) AML including clinical, cytogenetic, and molecular data.
Polymer-Grafted Nanoparticle Membranes With Unusual Gas Separation Properties, Sanat K. Kumar
Polymer-Grafted Nanoparticle Membranes With Unusual Gas Separation Properties, Sanat K. Kumar
Amgen Seminar Series in Chemical Engineering
Polymer membranes are employed in several critical sustainability applications involving the separation of gas mixtures based on size differences. In spite of their widespread use, important performance challenges remain outstanding - the need to dramatically affect the transport of a desired mixture component and improving mechanical resilience relative to the current state-of-the-art. Here, we develop novel membranes based on polymer-grafted nanoparticles (GNPs) which possess controllable, spatially inhomogeneous gas transport behavior. We show that smaller gases are transported more uniformly than larger solutes in the polymer layer of pure GNPs; these larger gases preferentially move through the interstices between the NPs. …

Introduction: In the USA, nearly one in three people will experience herpes zoster (HZ) in their lifetime. Underserved communities may be at even higher risk due to several factors, including access to healthcare, education, and co-morbid conditions. The purpose of this study was to investigate current knowledge, attitudes, beliefs and practices (KABP) relative to HZ and HZ vaccines in a large urban city.
Methods: A cross-sectional KABP survey was conducted via in-person interview among 381 participants aged ≥ 50 years in Detroit, MI, USA, from June to August 2018. Survey results were stratified into two groups [< 60 and ≥ 60 years of age (YO)] for comparison.

Oral herpes is a highly prevalent infection caused by herpes simplex virus 1 (HSV-1). After an initial infection of the oral cavity, HSV-1 remains latent in sensory neurons of the trigeminal ganglia. Episodic reactivation of the virus leads to the formation of mucocutaneous lesions (cold sores), but asymptomatic reactivation accompanied by viral shedding is more frequent and allows virus spread to new hosts. HSV-1 DNA has been detected in many oral tissues. In particular, HSV-1 can be found in periodontal lesions and several studies associated its presence with more severe periodontitis pathologies. Since gingival fibroblasts may become exposed to salivary …

Guiding analytes to the sensing area is an indispensable step in a sensing system. Most of the sensing systems apply a passive sensing method, which waits for the analytes to diffuse towards the sensor. However, passive sensing methods limit the detection of analytes to a picomolar range on micro/nanosensors for a practical time scale. Therefore, active sensing methods need to be used to improve the detection limit in which the analytes are forced to concentrate on the sensors. In this article, we have demonstrated the manufacturing of nanogap-rich structures for active chemical sensing. Nanogap-rich structures are manufactured from metallic nanoparticles …
Plant Cell-Derived Growth Factors For Ex Vivo Mass Production Of Red Blood Cells, Xiaoting Wang
Plant Cell-Derived Growth Factors For Ex Vivo Mass Production Of Red Blood Cells, Xiaoting Wang
Student Theses and Dissertations
Generation of red blood cells from hematopoietic stem cells (HSCs) used for blood transfusion represents one of the focus in regenerative medicine. Erythropoietic growth factors (eGFs), including stem cell factor (SCF), erythropoietin (EPO) and interleukin 3 (IL-3) are vital for proliferation and differentiation of HSCs into mature red blood cells (RBCs) ex vivo. However, massive production of RBCs requires significant quantity and high quality of eGFs, making manufacturing at large scale cost prohibitive. Plant cell culture is proposed to be a promising bioproduction platform for growth factors based on the advantages of, fast growing, no risk of human pathogen, and …

In aquatic settings, animals directly affect ecosystem functions through excretion of dissolved nutrients. However, the comparative role of egestion as an animal-mediated nutrient flux remains understudied. We conducted a literature survey and meta-analysis to directly compare nitrogen (N), phosphorus (P), and N:P of egestion compared to excretion rates and ratios across freshwater animals. Synthesizing 215 datasets across 47 animal species (all primary consumers or omnivores), we show that the total N and P egestion rates exceed inorganic N and P excretion rates but not total N and P excretion rates, and that proportions of P egested compared to excreted depend …

Optimizing Hydroxyproline-O-Glycan Engineering In Tobacco Transient Protein Expression: Fish Il-22 And Enhanced Green Fluorescent Protein (Egfp), Cristofer Calvo
Student Theses and Dissertations
Despite the advantages of plant-produced recombinant proteins, the most important bottleneck that limits the commercialization is the low protein yields. Plants have a unique type of O-glycosylation that has potential to enhance the stability and solubility of recombinant proteins. Specifically, target gene sequences are fused with a sequence to code for hydroxyproline-O-glycosylated peptide (HypGP) tags. We targeted the expression of a very unstable rainbow trout interleukin 22 (rtIL-22) and a readily-monitored enhanced green fluorescent protein (EGFP). The HypGP-tagged rIL-22 accumulation in planta was significantly enhanced over the untagged protein and showed increase stability. Changes in gene expression of the marker …

Osteolytic bone lesions and hypercalcemia are common, serious complications in adult T cell leukemia/lymphoma (ATL), an aggressive T cell malignancy associated with human T cell leukemia virus type 1 (HTLV-1) infection. The HTLV-1 viral oncogene HBZ has been implicated in ATL tumorigenesis and bone loss. In this study, we evaluated the role of HBZ on ATL-associated bone destruction using HTLV-1 infection and disease progression mouse models. Humanized mice infected with HTLV-1 developed lymphoproliferative disease and continuous, progressive osteolytic bone lesions. HTLV-1 lacking HBZ displayed only modest delays to lymphoproliferative disease but significantly decreased disease-associated bone loss compared with HTLV-1-infected mice. …

Aberrant activation of the NF-κB transcription factors underlies chemoresistance in various cancer types, including colorectal cancer (CRC). Targeting the activating mechanisms, particularly with inhibitors to the upstream IκB kinase (IKK) complex, is a promising strategy to augment the effect of chemotherapy. However, clinical success has been limited, largely because of low specificity and toxicities of tested compounds. In solid cancers, the IKKs are driven predominantly by the Toll-like receptor (TLR)/IL-1 receptor family members, which signal through the IL-1 receptor-associated kinases (IRAKs), with isoform 4 (IRAK4) being the most critical. The pathogenic role and therapeutic value of IRAK4 in CRC have …

Reperfusion of patients after myocardial infarction (heart attack) triggers cardiac inflammation that leads to infarct expansion and heart failure (HF). We previously showed that the circadian mechanism is a critical regulator of reperfusion injury. However, whether pharmacological targeting using circadian medicine limits reperfusion injury and protects against HF is unknown. Here, we show that short-term targeting of the circadian driver REV-ERB with SR9009 benefits long-term cardiac repair post-myocardial ischemia reperfusion in mice. Gain and loss of function studies demonstrate specificity of targeting REV-ERB in mice. Treatment for just one day abates the cardiac NLRP3 inflammasome, decreasing immunocyte recruitment, and thereby …

Bone is an important organ for supports the body that stores reserve of calcium, phosphorus, and other minerals. In fracture conditions where bleeding, soft tissue edema, nerve damage, and blood vessels around the bone damage happen, they can cause the mobilization of these minerals in the surrounding tissue. One of the efforts made in the treatment of these fractures is reconnection, in which it works by filling of bone defect with a matrix and administration of anti-infection. Biomaterial filling in defective bone is thought to accelerate the healing process of bone fracture and prevent osteomyelitis. For this reason, this study …
