Taxonomy, Bioactivities, And Secondary Metabolites Of Endophytic Actinomycetes Isolated From Medicinal Plants, Nattaporn Klykleung

Fifty endophytic actinomycetes isolated from six medicinal plants in Thailand were identified as Micromonospora (34 strains), Streptomyces (8 strains), Microbispora (3 strains), Nonomuraea (2 strains), Plantactinospora (2 strains), and Amycolatopsis (1 strain) based on phenotypic characteristics and 16S rRNA gene sequencing. Fifteen strains exhibited antimicrobial activity against Gram-positive bacteria. Strain CR1-09T, proposed as Microbispora cathalanthi sp. nov. was active against Staphylococcus aureus ATCC 6538 and Kocuria rhizophila ATCC 9341. Streptomyces sp. 3MP-10 and 3MP-14, novel species were active against S. aureus ATCC 6538, K. rhizophila ATCC 9341, and Bacillus subtilis ATCC 6633. Oxyresveratrol (OXY) is known to exert antioxidant and anti-inflammatory properties but has low oral bioavailability in rats and low permeability in the Caco-2 cell monolayer model. Three ester prodrugs with different acyl chain lengths (oxyresveratrol tetraacetate (OXY-TAc), oxyresveratrol tetrapropionate (OXY-TPr), and oxyresveratrol tetrabutyrate (OXY-TBu) were synthesized to improve the permeability of OXY. Transport of three OXY ester prodrugs across Caco-2 monolayers was higher than that of OXY, and OXY-TAc exhibited the highest permeation. Molecular pharming represents the innovative development of plants as alternative production platforms for recombinant proteins. Human vascular endothelial growth factor (VEGF) is one of the growth factors that has displayed eminent roles in wound healing. This study aimed to develop optimized expression conditions and rapid production of recombinant VEGF in Nicotiana benthamiana. Cordycepin, an adenosine analogue, has displayed numerous pharmacological and therapeutic implications including enhanced apoptosis and inhibition of proliferation of cancer cells, antioxidant property and improving sexual activity. However, oral administration of cordycepin has limited practical use due to its poor bioavailability in the intestine. Here, we developed a novel hybrid nanoparticle system for effective delivery of cordycepin in the gastrointestinal tract by using the bile salt-based liposome/niosome hybrid system (designated as biloniosome). Doxorubicin (DOX) is a chemotherapeutic agent that suffers from its severe adverse effects due to non-selective cytotoxicity. This study aims to design novel beta-thiopropanamide linkers (-S-CH2-C(R)H-CONH-) for DOX conjugation to promote cancer cell selectivity and cytotoxicity. Two series of DOX conjugates with various beta-thiopropanamide linkers were synthesized and evaluated for their cytotoxic selectivity. The first series contains 7 DOX analogues with different beta-thiopropanamide linkers. The purpose of this study was to develop pickering nanoemulsions (NEs) containing amphotericin B (AmB) for fungal keratitis therapy. The phase-solubility profiles of AmB in various aqueous cyclodextrin (CD) solutions were determined. The AmB solubilization in oils and surfactants was also examined. Two CDs i.e., gCD and 2-hydroxypropyl-g-CD (HPgCD), medium-chain triglycerides (MCT oil) and phosphatidylcholine (lecithin) were chosen because of their high solubility increment of AmB. Solution-state by 1H-NMR studies demonstrated the formation of MCT oil/αCD, AmB/gCD and AmB/HPgCD complexes. This study aimed to express recombinant protein human Dentin Matrix Protein 1 (hDMP1) in Escherichia coli and Nicotiana benthamiana and investigated the osteogenic differentiation effects in human Periodontal Ligament Stem Cells (hPDLSCs). The gene encoding hDMP1 was ligated into the pET22b and pBY2R2e expression vectors. Recombinant hDMP1 protein has been productively expressed in E. coli and N. benthamiana. E. coli-produced hDMP1 resulted in 60 kDa and 100, and the optimal condition to produce highly expression was demonstrated at 0.5 mM IPTG and incubation at 37oC for 6 hr. Mansonones are naphthoquinone-containing compounds extracted from the heartwood of Mansonia gagei. Several studies have reported that mansonones have various pharmacological activities such as antibacterial, antifungal, antioxidant, antiestrogenic, antiadipogenic and anticancer effects. Macrophages play major roles to produce several inflammatory mediators in many chronic inflammatory diseases. Current anti-inflammatory drugs target these mediators to alleviate inflammation symptoms. Searching for new anti-inflammatory agents is always needed due to problems from currently used anti-inflammatory drugs. This study aimed to investigate the anti-inflammatory potential of three main compounds, arborinine, N-methylatalaphylline and S-deoxydihydroglyparvin (DDGP), isolated from Glycosmis parva leaves and branches on lipopolysaccharide (LPS)-activated macrophage RAW 264.7 cells. Only DDGP demonstrated a potent inhibitor of LPS-activated macrophages. It inhibited expression of inducible nitric oxide synthase and decreased nitric oxide production with IC50 3.47 ± 0.1 µM. Phytochemical investigations of Cissus javana DC., Dendrobium christyanum Rchb. f., Gastrochilus bellinus Rchb. f. Kuntze and Huberantha jenkinsii (Hook. f. & Thomson) Chaowasku led to the isolation of twenty-seven compounds, six of which were new naturally occurring compounds. From Cissus javana, three known compounds were isolated, including bergenin, stigmast-4-en-3-one, and β-sitosterol. From Dendrobium christyanum, thirteen known compounds were identified, namely, n-eicosyl trans-ferulate, n-docosyl 4-hydroxy-trans-cinnamate, 4,5-dihydroxy-2-methoxy-9,10-dihydrophenanthrene, moscatilin, aloifol I, gigantol, batatasin III, dendrosinen B, coniferyl aldehyde, methyl haematommate, atraric acid, vanillin and diorcinolic acid. Rhinacanthin-C (RN-C) is a major bioactive naphthoquinone ester found in Rhinacanthus nasutus Kurz (Acanthaceae). This compound has potential therapeutic value as an anticancer agent. Rhinacanthin-C was reported to enhance doxorubicin cytotoxicity in breast cancer cells through direct inhibition of P-gp activity. Vital characteristics that potentiate tumor growth are cancer stem cell (CSC) and enhance cell proliferation. Background: The Patient-Reported Outcomes Measurement Information System-29 (PROMIS-29) questionnaire assesses 7 health related quality of life domains. However, research to evaluate the responsiveness and minimal clinically important differences (MCIDs) of the PROMIS-29 scores in patients with chronic low back pain (cLBP) is limited. Purpose: To evaluate responsiveness and estimate the MCIDs for PROMIS-29 scales in patients with cLBP. Method: One hundred and eighty-three participants with cLBP took part in the study. Atopic dermatitis (AD) is an inflammatory skin disease characterized by chronic and recurrent eczematous rash. AD occurs in both children and adults, but children are more affected. An important aggravating factor is caused by Staphylococcus aureus (S. aureus) infection. การสกัดคอลลาเจนจากเศษของครีบปลาหมึกกล้วย (Loligo Duvauceli) ซึ่งเป็นของเสียในอุตสาหกรรมอาหารทะเล, ศิริพงศ์ โสมสิริวัลย์

หมึกกล้วย (Indian squid) สายพันธุ์ Loligo duvauceli เป็นสัตว์ทะเลทางเศรษฐกิจที่สำคัญของประเทศไทย พบว่าครีบหมึกจัดว่าเป็นของเสียในกระบวนการผลิตจากหมึกกล้วยคิดเป็น 12% และมีราคาถูก โดยครีบหมึกกล้วยแห้งประกอบด้วยความชื้น 4.62% โปรตีน 77.08% ไขมัน 15.60% และ เถ้า 3.03% ในงานวิจัยนี้ได้สกัดคอลลาเจนจากครีบหมึกกล้วยบดโดยใช้สารละลายโซเดียมไฮดรอกไซด์ความเข้มข้น 0.5 โมลต่อลิตรเพื่อกำจัดไขมันและโปรตีนอื่นๆ จากนั้นนำไปสกัดต่อด้วยกรดแอซิติกความเข้มข้น 0.5 โมลต่อลิตรที่อุณหภูมิ 4 องศาเซลเซียส แยกส่วนของแข็งและสารละลายออกจากกันด้วยการกรอง และนำสารละลายกรดที่ได้ไปทำแห้งด้วยวิธีแช่เยือกแข็ง โดยของแข็งที่ได้จะเรียกว่า คอลลาเจนที่ละลายในกรด (ASC) ส่วน คอลลาเจนที่ละลายในเอนไซม์เปปซิน (APSC) สกัดได้โดยใช้สารละลายเปปซินปริมาณ 1% โดยน้ำหนักต่อปริมาตรในกรดแอซิติก 0.5 โมลต่อลิตร ในขั้นตอนการสกัดพบว่าผลได้ของคอลลาเจน ASC เท่ากับ 0.69 ± 0.25% และสามารถสกัดคอลลาเจน APSC ได้ 3.02 ± 00.16% เมื่อสกัดที่ 48 ชั่วโมงและ 1.42 ± 0.41% เมื่อสกัดที่ 24 ชั่วโมง ตรวจสอบน้ำหนักโมเลกุลของคอลลาเจนด้วยเทคนิค SDS-Page เทียบกับคอลลาเจนชนิดที่ 1 จากหนังหมูพบว่าคอลลาเจนที่สกัดได้จากครีบหมึกเป็นคอลลาเจนชนิด SQ ([α1]2[α2]1) ซึ่งมีลักษณะคล้ายกับคอลลาเจนชนิดที่ 1 โดย ASC ประกอบด้วยสาย α1 (202.39 kDa), α2 (159.90 kDa) และ β (281.50 kDa) และ APSC ประกอบด้วยสาย α1 (184.19 kDa), α2 (145.52 kDa) และ β (281.50 kDa) แสดงถึงส่วนเทโลเปปไทด์ถูกเอนไซม์เปปซินย่อยส่วนเทโลเปปไทด์ ฟูเรียร์ทรานส์ฟอร์มอินฟราเรดสเปกตรัมของ ASC และ …

น้ำมันตัดเป็นน้ำมันที่ใช้ในการลดแรงเสียดทานและระบายความร้อนของชิ้นงานในกระบวนการผลิต น้ำมันตัดที่ผ่านการใช้งานแล้วจะมีค่าซีโอดี ความขุ่น และของแข็งแขวนลอยสูง นอกจากนี้ยังมีส่วนประกอบของสารลดแรงตึงผิว ทำให้น้ำเสียปนเปื้อนน้ำมันตัดมีลักษณะเป็นอิมัลชันที่มีความคงตัวสูงและยากในการบำบัดด้วยกระบวนการทางชีวภาพและกายภาพทั่วไป ในงานวิจัยนี้ได้ทำการศึกษาประสิทธิภาพและสภาวะที่เหมาะสมของการบำบัดน้ำเสียปนเปื้อนน้ำมันตัดความเข้มข้นต่ำที่ไม่สามารถบำบัดได้หมดจากกระบวนการขั้นต้นและน้ำเสียปนเปื้อนน้ำมันตัดความเข้มข้นสูงที่มีค่าใกล้เคียงกับความเข้มข้นของน้ำมันตัดที่เหลือจากการใช้งานในกระบวนการจริงด้วยกระบวนการเฟนตันและโฟโตเฟนตัน ในงานวิจัยนี้ศึกษาความเข้มข้นของน้ำมันตัดเริ่มต้น 0.1 1.0 4.0 9.0 และ 22.0 กรัมต่อลิตร โดยใช้เฟอร์รัสซัลเฟตเป็นตัวเร่งปฏิกิริยาและควบคุมพีเอชที่ 3.0 ± 0.2 จากการศึกษาประสิทธิภาพการบำบัดน้ำเสียปนเปื้อนน้ำมันตัดความเข้มข้น 0.1 และ 1.0 กรัมต่อลิตร โดยใช้เฟอร์รัสไอออน 50 มิลลิกรัมต่อลิตร และอัตราส่วนของเฟอร์รัสไอออนต่อสารละลายไฮโดรเจนเปอร์ออกไซด์ที่ 1: 2.5 ด้วยกระบวนการเฟนตัน พบว่าสามารถบำบัดซีโอดีได้ร้อยละ 88.97 ± 5.63 และ 91.98 ± 1.68 ตามลำดับ เมื่อทำการบำบัดด้วยกระบวนการโฟโตเฟนตันพบว่าอัตราส่วนของเฟอร์รัสไอออนต่อสารละลายไฮโดรเจนเปอร์ออกไซด์ที่ 1: 10 มีประสิทธิภาพในการบำบัดซีโอดีของน้ำเสียปนเปื้อนน้ำมันตัดทั้งสองความเข้มข้นได้สูงสุดร้อยละ 96.21 ± 5.23 และ 91.95 ± 1.52 ตามลำดับ เมื่อทำการบำบัดน้ำเสียปนเปื้อนน้ำมันตัดความเข้มข้นสูงที่ 4.0 9.0 และ 22.0 กรัมต่อลิตร โดยใช้ความเข้มข้นของเฟอร์รัสไอออน 75 150 และ 300 มิลลิกรัมต่อลิตร และสารละลายไฮโดรเจนเปอร์ออกไซด์ 375 300 และ 1500 มิลลิกรัมต่อลิตร ตามลำดับ พบว่าสามารถบำบัดซีโอดีได้มากกว่าร้อยละ 87 โดยประสิทธิภาพการบำบัดซีโอดีของกระบวนการเฟนตันและโฟโตเฟนตันไม่มีความแตกต่างกันอย่างมีนัยสำคัญ (ระดับความเชื่อมั่น 95%) เนื่องจากในการศึกษาครั้งนี้ใช้น้ำเสียสังเคราะห์ที่มีความขุ่นสูง ทำให้บดบังแสงอัลตราไวโอเลตระหว่างการบำบัด ส่งผลให้การเพิ่มแสงอัลตราไวโอเลตไม่มีผลต่อการบำบัดน้ำเสียปนเปื้อนน้ำมันตัด รวมถึงศึกษาการนำตะกอนจากกระบวนการเฟนตันและโฟโตเฟนตันกลับมาใช้ในการบำบัดน้ำเสียปนเปื้อนน้ำมันตัดใหม่ จากผลการศึกษาพบว่าสามารถนำตะกอนกลับมาใช้เป็นตัวเร่งปฏิกิริยาแทนการเติมเฟอร์รัสไอออนในระบบได้จำนวน 2 ครั้ง โดยตะกอนที่นำกลับมาใช้ซ้ำนั้นจะมีการสะสมของสารอินทรีย์และอนุภาคของน้ำมันตัดในตะกอนเพิ่มขึ้นจากการบำบัดครั้งก่อน ทำให้มีความเข้มข้นของสารอินทรีย์เพิ่มสูงขึ้นตามรอบการใช้ตะกอนซ้ำ ส่งผลให้มีประสิทธิภาพการบำบัดซีโอดีลดลงตามจำนวนรอบการบำบัด

This research investigated the effects of microplastic accumulation on the floc characteristics and mechanism of membrane fouling in a sequencing batch membrane bioreactor (SB-MBR). At first, the jar test experiment was performed for 120 min. The results indicate that spiking microplastics of 100-1,000 MPs/L resulted in an increase in floc size by 27.9±9.2% to 70.9±5.1%, while the control set (without microplastic) showed a greater increase in the floc size at 113.3±5.6% of the initial floc size. The novel knob-socket (KS) model provides a construct to interpret and analyze the direct contributions of amino acid residues to the stability in α-helical protein structures. Based on residue preferences derived from a set of protein structures, the KS construct characterizes intra- and inter-helical packing into regular patterns of simple motifs. The KS model was used in the de novo design of an α-helical homodimer, KSα1.1. There are many enzymes required for efficient and proper pyrimidine biosynthesis. The two that are most important and were discussed in this thesis are aspartate transcarbamoylase (ATCase) and dihydroorotase (DHOase). Both play an important role in not only pyrimidine biosynthesis production, but also mechanistic regulation of de novo synthesis. Anthrax is an infection caused by Bacillus anthracis. This chapter provides an overview of the social and psychological factors contributing to coping with and adjusting to disability by summarizing models; clinical implications, evidence-based practice, and measurement issues; and empirical research related to the process of coping with and adjustment to disability. While the models are useful for understanding a portion of the experience of people with disabilities and their coping and adjustment process, these reactions are not universal and not always experienced in an orderly sequence.
