Glycerol-3-Phosphate Mediates Rhizobia-Induced Systemic Signaling In Soybean, M. B. Shine, Qing-Ming Gao, R. V. Chowda-Reddy, Asheesh K. Singh, Pradeep Kachroo, Aardra Kachroo

Glycerol-3-phosphate (G3P) is a well-known mobile regulator of systemic acquired resistance (SAR), which provides broad spectrum systemic immunity in response to localized foliar pathogenic infections. We show that G3P-derived foliar immunity is also activated in response to genetically-regulated incompatible interactions with nitrogen-fixing bacteria. Using gene knock-down we show that G3P is essential for strain-specific exclusion of non-desirable root-nodulating bacteria and the associated foliar pathogen immunity in soybean. Grafting studies show that while recognition of rhizobium incompatibility is root driven, bacterial exclusion requires G3P biosynthesis in the shoot. The HIV-1 envelope glycoprotein (Env) is sparsely incorporated onto assembling virus particles on the host cell plasma membrane in order for the virus to balance infectivity and evade the immune response. Env becomes trapped in a nascent particle on encounter with the polymeric viral protein Gag, which forms a dense protein lattice on the inner leaflet of the plasma membrane. While Env incorporation efficiency is readily measured biochemically from released particles, very little is known about the spatiotemporal dynamics of Env trapping events. Hagfish defend themselves from fish predators by producing large volumes of gill-clogging slime when they are attacked. The slime consists of seawater and two major components that are ejected from the slime glands: mucus and threads. The threads are produced within specialized cells and packaged into intricately coiled bundles called skeins. Skeins are kept from unraveling via a protein adhesive that dissolves when the skeins are ejected from the slime glands. Previous work revealed that hagfish slime glands have high concentrations of methylamines including trimethylamine N-oxide (TMAO), trimethylglycine (betaine) and dimethylglycine (DMG); however, the function of these compounds in the …

The Qualitative Report

The purpose of this study was to understand the meaning of online co-teaching for PhD faculty and teaching assistants (TAs). Narrative pedagogy underpinned the inquiry, which was designed to advance the discourse on mentorship of PhD future faculty. A faculty member and TA authors kept concurrent weekly journals or after-the-fact written reflections. The authors analyzed data as a team using a five-phase interpretive phenomenological analysis process to interpret the meaning of co-teaching for faculty and TAs. Lines of inquiry, central concerns, exemplars, shared meanings, and paradigm cases supported the overall interpretation, “You Learn When You Teach.” Co-mentorship should be a …

Neurological disorders have multiple etiologies such as genetics and age. Stress related disorders are becoming more common and diagnosed more often. Mental health disorders are no longer being treated as purely psychological. Mechanisms into the underlying factors of these disorders continue to bring this area to new frontiers. Chapter 2 of this dissertation focuses on the efforts made to prove the association of the protein FKBP51 to the stress response. We synthesized and used a molecular tool to discover and prove these interactions.

Considered the pulse controlling the biological interactions and contributing to the ecological survival of organisms, natural products have intrigued, challenged, and stimulated the scientific and non-scientific minds for millennia. Drug resistance and the lack of suitable potent drugs against neglected tropical diseases further motivate the biological activity assessment of these secondary metabolites, when possible.

Bone quality in diabetic patients is compromised, leading to weaker bones and increased fracture risk. However, the mechanism by which this occurs in diabetic bone remains to be fully elucidated. We hypothesized that elevated glucose and glucose variation would affect the function of osteocytes, essential regulators of bone homeostasis and quality. To first test this hypothesis, we used the IDG-SW3 osteocyte-like cell line to examine the effects of glucose levels on osteocyte function and viability in vitro. Interest in studying anti-microbial peptides (AMPs) has increased because of their potential as a future applicable antibiotic drug. In my research under supervision of Professor Greenfield, we attempt to understand the interaction of AMPs against membrane of bacteria. We are studying the structure of two novel hybrid AMPs LM7-1 and LM7-2 that were designed previously in Professor Martin’s research group (Cell and Molecular Biology Department). The bandgap near-infrared photoluminescence from the semiconducting single-walled carbon nanotubes is photostable, tunable, and sensitive to the local environment. Over the last 15 years, significant progress has been made in applying carbon nanotube photoluminescence toward a range of in vitro and in vivo biomedical applications. My research at URI NanoBio Engineering Lab encompasses the design and implementation of implantable and wearable biosensors in addition to developing in vitro molecular probes, based on single-walled carbon nanotubes. Animals communicate with potential mates using species-specific signals, and pheromones are powerful sexual signals that modify conspecific behavior to facilitate mate location. Among the vertebrates, snakes are especially adept in mate searching via chemical trailing, which is particularly relevant given that many snake species are invasive outside their native ranges. Chemical signals used in mate choice are, thus, potentially valuable tools for management of invasive snake species. The Burmese python (Python bivittatus) is an invasive snake in the Florida Everglades where it is negatively impacting native fauna. CD8 T cell exhaustion is the progressive, step-wise loss of T cell function due to persistent antigen stimulation. Given the importance of this phenomenon in sustaining chronic infections and preventing clearance of tumors, as well as potentially influencing autoimmune disorders, novel biomarkers must be established to monitor CD8 T cell exhaustion and the factors that influence its development (e.g. presence of CD4 helper cytokines, like IL-21). In this study, a six-day in vitro model of CD8 T cell exhaustion was characterized to evaluate the reduced functional capacity of these cells. Enteric pathogens sense numerous signals specific to the anatomical location in the intestine and integrate them with the complex regulatory networks to temporally and spatially regulate their virulence genes. MarA, SoxS, Rob and RamA are homologous transcription factors that belong to AraC family of proteins in Salmonella enterica that primarily were thought to be involved in rendering antibiotic resistance to bacteria by up regulating efflux pumps and down regulating outer membrane porins. At small spatial scales, the composition of species assemblages depends on local conditions and the dispersal of propagules from surrounding regions. In turn, processes taking place at broad temporal and spatial scales, including changes in regional physiography and climate, shape the species pools in these source regions. Research Question: Can sleep consolidation reduce the effects of an interruption during encoding, leading to improved accuracy on a delayed recognition task?

Consolidation: During sleep, memories acquired earlier are processed at a deeper level and strengthened by creating associations with previously-stored information (Rasch & Born, 2008). This process helps better integrate new information into existing long-term memory storage systems. Research indicates that the consolidation process can also prevent the effects of interference during memory retrieval (Robertson, 2012).

Querying the Past: LGBTQ Maine Oral History Project Collection

Bobbi Keppel is an 87-year old bisexual activist. Her father was a civil rights activist and union organizer; in part because of this, she felt she was a born “disruptor.” As a child, Bobbi Keppel was ill and struggled with being a “sickly kid.” She later married and had two children. During her marriage, she came out as bisexual with the support of her husband. She is a contributor to the classic anthology “By Any Other Name: Bisexual People Speak Out” (edited by Loraine Hutchins and Lani Ka'ahumanu). The Opioid Crisis is a national crisis affecting public, social, and economic healthcare. Nearly 50,000 deaths were caused by opioid overdoses in 2017. Current treatments of opioid addictions include the use of methadone and buprenorphine. These medications have been known to reduce opioid dependency, lower tolerances, increase the opioid overdose threshold, and lower overdose mortality. An analysis was conducted on prominent research studies investigating the effectiveness, safety, side effects, and influence of Methadone and Buprenorphine. Small interfering RNA (siRNA) therapy is a promising approach for treatment of a wide range of cancers, including breast cancers that display variable phenotypic features. To explore the general utility of siRNA therapy to control aberrant expression of genes in breast cancer, we conducted a detailed analysis of siRNA delivery and silencing response in vitro in 6 separate breast cancer cell models (MDA-MB-231, MDA-MB-231-KRas-CRM, MCF-7, AU565, MDA-MB-435 and MDA-MB-468 cells). Using lipopolymers for siRNA complexation and delivery, we found a large variation in siRNA delivery efficiency depending on the specific lipopolymer used for siRNA complexation and delivery. To preserve wetland ecosystem function, federal and state agencies have developed assessment procedures to better manage remaining wetland areas. Currently, wetland assessments do not consider microorganisms when determining wetland quality. This is notable, because fungi are often the primary decomposers of organic material and thus important players in nutrient cycling. The objective of this study is to quantify how wetland quality, as measured using the Ohio Rapid Assessment Method (ORAM), relates to fungal community composition.
