Neuroticism And Stressful Life Events: Probing Mechanisms Underlying Vulnerability To Stress-Related Depression, Erin Bondy

Elevated neuroticism (N) potentiates the depressogenic effects of stressful life events (SLEs). We first replicate this association using longitudinal data (N=971 older adults) from the St. Louis Personality and Aging Network (SPAN) study. Here, SLEs prospectively predicted future depressive symptoms, especially among those reporting elevated N, even after accounting for prior depressive symptoms and previous SLE exposure (NxSLE interaction: p=0.016, ΔR2=0.003). These findings were further replicated in cross-sectional analyses of the Duke Neurogenetics Study (DNS), a young adult college sample with neuroimaging data (n=1,343: NxSLE interaction: p=0.019, ΔR2=0.003). OBJECTIVE: Rapid developments in understanding the molecular mechanisms underlying cognitive deficits in neurodevelopmental disorders have increased expectations for targeted, mechanism-based treatments. However, translation from preclinical models to human clinical trials has proven challenging. Poor reproducibility of cognitive endpoints may provide one explanation for this finding. We examined the suitability of cognitive outcomes for clinical trials in children with neurofibromatosis type 1 (NF1) by examining test-retest reliability of the measures and the application of data reduction techniques to improve reproducibility.

Mastery of jargon terms is an important part of student learning in biology and other science, technology, engineering, and mathematics domains. In two experiments, we investigated whether prelecture quizzes enhance memory for jargon terms, and whether that enhanced familiarity can facilitate learning of related concepts that are encountered during subsequent lectures and readings. Undergraduate students enrolled in neuroanatomy and physiology courses completed 10-minute low-stakes quizzes with feedback on jargon terms either online (experiment 1) or using in-class clickers (experiment 2). Helminth infection and dietary intake can affect the intestinal microbiota, as well as the immune system. Here we analyzed the relationship between fecal microbiota and blood profiles of indigenous Malaysians, referred to locally as Orang Asli, in comparison to urban participants from the capital city of Malaysia, Kuala Lumpur. We found that helminth infections had a larger effect on gut microbial composition than did dietary intake or blood profiles. Trichuris trichiura infection intensity also had the strongest association with blood transcriptional profiles. By characterizing paired longitudinal samples collected before and after deworming treatment, we determined that changes in serum zinc …

Impedance spectroscopy (IS) is an important tool for cell detection and characterization in medical and food safety applications. In this thesis, the Cal Poly Biofluidics Lab’s impedance spectroscopy system was re-evaluated and optimized for single-cell impedance spectroscopy. To evaluate the IS system, an impedance spectroscopy bioMEMS chip was fabricated in the Cal Poly Microfabcrication lab, software was developed to run IS experiments, and studies were run to validate the system. We evaluated the accuracy of a computerized particle analyzer (CPA) for high-throughput counting of Palmer amaranth (Amaranthus palmeri S. Watson) seeds and subsequently used the CPA to verify the accuracy of two subsampling methods for estimation of Palmer amaranth seed production. To determine accuracy of the CPA, 55 hand-counted samples, ranging from 500 to 5000 Palmer amaranth seeds, were drawn from field samples and counted with the CPA. The relationship between hand and CPA seed counts was described by a linear model (R-2 = 0.99) with a slope of 0.987 and a y-intercept of 3.49. Thus, very little discrepancy exists …

This dissertation explores a novel method of solving low-thrust spacecraft targeting problems using reinforcement learning. A reinforcement learning algorithm based on Deep Deterministic Policy Gradients was developed to solve low-thrust trajectory optimization problems. The algorithm consists of two neural networks, an actor network and a critic network. The actor approximates a thrust magnitude given the current spacecraft state expressed as a set of orbital elements. The critic network evaluates the action taken by the actor based on the state and action taken. The purpose of this study is to describe and understand what race means to first-generation African Caribbean immigrants. Specifically, the study seeks to understand these immigrants’ experience of being Black in America. African Caribbean immigrants migrate to America with a concept of race that is very different from that of the United States. However, upon arriving in America, they encounter the American construct of race that not only diverges significantly from that with which they are familiar, but find that they are being racialized in the American sense. Insulin is the most common treatment for hyperglycemia, such as that caused by type 1 or type 2 diabetes mellitus. Insulin causes cellular uptake and storage of glucose to maintain homeostasis and also plays important roles in other systems; an important example is regulation of potassium. In the periphery, insulin administration has been shown to increase the cellular uptake of potassium via Na+/K+ ATPase, leading to hypokalemia. Research in our lab and others has shown that insulin is a key regulator of cognitive function and local metabolism within the hippocampus. In recent years, Natural Language Processing in the field of machine learning has seen some major improvements. Data scientists have shown that neural networks are capable of breaking down semantics of sentences, translating languages, and answering complex questions with fast recall. While impressive, these feats all hinge on having access to a massive amount of clean text, or data sets with almost perfect grammar and spelling. The dissertation focuses on the design and synthesis of smart materials for the detection of molecules pertinent to environmental protection and healthcare. The use of computational simulations is pivotal toward advancing molecular design for targeted applications. Research was conducted to investigate the use of simulations to develop novel sensors with dual modes of signal transduction. The molecular properties were determined using computational modelling, and then used to elucidate the binding mechanism of the corresponding sensor complexes. Function modeling plays an essential role in academy design studies, yet a lack of acceptance in industry. A possible reason is that the designer must have well understand of the controlled vocabularies and grammars to utilize this method. The research presented in this dissertation is to fill this gap so that designers can use function-based design method without relevant knowledge. In graph-based function models, the function verbs and flow nouns are usually chosen from predefined vocabularies. In this case study, the researcher investigated the barriers to school administrators’ implementation of restorative practices as a management approach to student behavior. Based on the claim that restorative practices is a promising alternative to exclusionary disciplinary placements for changing behavior, the researcher used an emergent explanatory mixed methods design to better understand administrators’ resistance to change despite overwhelming evidence that exclusionary disciplinary practices are the primary contributors to national trends in disciplinary disproportionality and institutional discrimination. The researcher identified and analyzed barriers to create recommendations for addressing administrators’ resistance through organizational change strategies using systems theory.

Inoculation of detached strawberry leaves with Colletotrichum species may provide an accurate, rapid, nondestructive method of identifying anthracnose-resistant germplasm. The purpose of this study was to statistically compare two methods (visual and image analysis) of evaluating disease severity of strawberry germplasm screened for anthracnose resistance. Detached leaves of 77 susceptible and resistant strawberry clones were inoculated with one Colletotrichum gloeosporioides (Penz.) Penz. & Sacc. and two C. fragariae A. N. Brooks isolates. Taxonomic identification can be difficult when two or more species appear morphologically similar. DNA barcoding based on the sequence of the mitochondrial cytochrome c oxidase 1 gene (COI) is now widely used in identifying animal species. High‐resolution melting analysis (HRM) provides an alternative method for detecting sequence variations among amplicons without having to perform DNA sequencing. The purpose of this study was to determine whether HRM of the COI barcode can be used to distinguish animal species. Patellar instability is a debilitating condition, and trauma due to the dislocations may severely damage the articular cartilage of the patellofemoral joint. The lack of healing capacity of cartilage makes treatment of such lesions challenging. In young patients, preservation of native bone and cartilage is a priority, and therefore arthroplasty procedures should be avoided even in relatively larger lesions. Fresh osteochondral allografting has been described for these difficult and complex cases, with recent studies showing promising results. OBJECTIVE: To describe the findings of histopathology and genotyping studies in affected brain tissue from an individual with phacomatosis pigmentovascularis (PPV).
METHODS: A retrospective chart review of a 2-year 10-month-old male with a clinical diagnosis of PPV cesiomarmorata (or type V) was performed. Clinical features, brain imaging and histopathology findings, and genotyping studies in his affected brain tissue are summarized.

Amino acid availability in Gram-positive bacteria is monitored by T-box riboswitches. T-boxes directly bind tRNAs, assess their aminoacylation state, and regulate the transcription or translation of downstream genes to maintain nutritional homeostasis. Here, we report cocrystal and cryo-EM structures of Geobacillus kaustophilus and Bacillus subtilis T-box-tRNA complexes, detailing their multivalent, exquisitely selective interactions. The T-box forms a U-shaped molecular vise that clamps the tRNA, captures its 3' end using an elaborate 'discriminator' structure, and interrogates its aminoacylation state using a steric filter fashioned from a wobble base pair. Mindfulness practice, a present-centered approach to experience, has been shown to effectively enhance various dimensions of the human experience. Recent literature has explored how the practice of mindfulness meditation has provided benefits to practitioners seeking to enhance relationship dynamics. There are multiple examples of mindfulness-based relationship enhancement programs aiming to enhance the quality of intimate relationships in adults. This study utilizes a repeated-measures design to address whether a neutral mindfulness-based program, not geared at enhancing relationships, can significantly alter the quality of intimate relationships. Unsustainable agricultural practices are putting a strain on freshwater supplies in many parts of the world. More efficient irrigation techniques are going to be critical to feed a growing population. Data from the 2016 Arkansas Irrigation Survey was used to assess the adoption of three Irrigation Water Management (IWM) practices: multiple inlet rice irrigation, tailwater recovery, and storage reservoirs. Results indicate that these practices do not always lead to reported pumping time reductions, a proxy for water use savings. Statement of Significance: Cardiovascular disease is the leading cause of morbidity and mortality in long-term cancer survivors. Chemotherapy with doxorubicin and other anthracyclines is the most common cause of cardiac toxicity in treated cancer patients. These agents are used to manage both hematologic and solid-tumor malignancies. Cardiac manifestations often develop months to years after initial exposure, causing cardiomyopathy and heart failure. Osteopathic medicine approaches disease in a holistic manner attempting to prevent major complications earlier in the disease course. Preventing or reducing chemotherapy-induced cardiotoxicity would greatly improve the outlook for these patients and their overall wellbeing.

Methods to engineer nanomaterials and devices with uniquely tailored properties are highly sought after in fields such as manufacturing, medicine, energy, and the environment. The macromolecule deoxyribonucleic acid (DNA) enables programmable self-assembly of nanostructures with near arbitrary shape and size and with unprecedented precision and accuracy. Additionally, DNA can be chemically modified to attach molecules and nanoparticles, providing a means to organize active materials into devices with unique or enhanced properties. An opportunistic predation by Great Blue Heron (Ardea herodias) on Eastern cottontail rabbit (Sylvilagus floridanus) was observed in south Texas on 31 Oct 2019. The Great Blue Heron had already captured the Eastern cottontail rabbit at the first observation but the maneuvering process, killing, and ingestion were recorded photographically, which make this observation unique even though this might constitute the second report on Great Blue Heron eating Eastern cottontail rabbits.

Many recent theoretical and experimental techniques have been developed to probe the structurefunction relationships of complex biomolecules. The roles of RNAs are dependent upon various intricate structural motifs and interactions, including hairpins, pseudoknots, long range territory contacts, bulges and internal loops, that are not easily captured by these methods. Imaging of biologically significant molecules using plasmons of Metal Nanoparticles (MNPs) is gaining attention in the research community. Localized Surface Plasmon Resonance (LSPR) is the coherent oscillation of conduction electrons of MNPs. The biologically significant molecule is labeled with the fluorophore molecule to get the image. This approach is widely used in clinical practices, however, low intensity light emission from the labeled molecule makes it difficult to image the biologically significant material.
