Trans-activation response (TAR) DNA-binding protein 43 (TDP-43) is a natively dimeric 414-residue protein that is encoded by the human TARDBP gene that has important implications in the pathogenesis of the neurodegenerative disorders ALS, FTD, and CTE. TDP-43 has been found hyperphosphorylated and ubiquitinated in the aggregates of the affected neurons of these diseases. The discovery of the presence of TDP-43 positive inclusions in brain matter of patients with CTE has made repetitive brain injury a possible environmental stimulus for aggregation in TDP-43 proteinopathies. Mucin type O-glycosylation is a common modification of proteins in higher organisms found on cell surfaces and secreted proteins. O-glycosylation is important in a variety of biological processes from development, the control of serum phosphate levels, and playing role in vascular and heart diseases and cancer. A family of 20 transferases (GalNAc-Ts) initiate mucin type O-glycosylation in humans by adding the sugar GalNAc onto Ser or Thr residues on targeted proteins. BACKGROUND: Periodontitis is an inflammatory disease caused by poly-microbial infection that leads to destruction of connective tissue and alveolar bone. It is well documented that bacteria-derived virulent factors that can act on Toll-like receptors (TLRs), represented by Lipopolysaccharides (LPS), are engaged in the initiation of inflammatory responses. However, while LPS is also produced by the bacteria colonized in the healthy periodontal tissue, inflammation is not induced by LPS in those periodontal healthy subjects, suggesting the requirement of additional factor to upregulate the LPS-mediated inflammatory response in periodontal tissue. Atropine Sulfate (AS) auto-injector (AtroPen®) is being used as an effective and safe antidote for the treatment of organophosphate (OP) pesticides or nerve gas poisoning. The use of AtroPen® is associated with several drawbacks including: bulky size, availability, affordability, invasiveness, and administration errors. Previously, AS fast disintegrating sublingual tablets (FDSTs) were developed and the feasibility of AS sublingual permeability were demonstrated. However, AS permeability was delayed due to the negative impact of higher doses of AS on FDST’s physical characteristics. Disclosed are methods, compositions, reagents, systems, and kits to prepare nitroxide-functionalized brush-arm star polymer organic radical contrast agent (BASP-ORCA) as well as compositions and uses thereof. Various embodiments show that BASP-ORCA display unprecedented per-nitrox-ide and per-molecule transverse relaxivities for organic radical contrast agents, exceptional stability, high water solubility, low in vitro and in vivo toxicity, and long blood compartment half-life. These materials have the potential to be adopted for tumor imaging using clinical high-field 1H MRI techniques.

Bacterial antibiotic resistance modulation by small signaling molecules is an emerging mechanism that has been increasingly reported in recent years. Several studies indicate that indole, an interkingdom signaling molecule, increases bacterial antibiotic resistance. However, the mechanism through which indole reduces antibiotic resistance is largely unknown. In this study, we demonstrated a novel mechanism for indole-mediated reversal of intrinsic antibiotic resistance in Lysobacter. This reversal was facilitated by a novel BtuD-associated dual-function importer that can transfer both vitamin B12 and antibiotics. Can we imagine a Blue Humanities that takes the non-relation as a starting point for ecological thought? I believe we can. Following Shakespeare and Deleuze, this essay engages in a thought experiment that, if it is not too absurd, might, like the ship of fools of medieval times, unmoor the Blue Humanities from its current safe harbor by putting the thought of ‘our’ world under erasure. The United Methodist Church (UMC) as an institution has strict language against lesbian and gay people and against the ordination of the lesbian and gay people. Prior research has focused on how denominations and congregations discuss issues around sexuality, how attitudes have shifted around issues of sexuality, and how to provide spiritual care for the LGBTQ+ community. This qualitative study interviewed lesbian and gay ordained elders in the UMC to learn about their experiences of serving as clergy people in the UMC even though there are prohibitive statements in church documents. EPR imaging at low frequency is a powerful tool to obtain important biological information in vivo in a non-invasive way. Properties of nitroxide and trityl radical imaging reagents have been studied. Developments in rapid scan imaging techniques are reported that improve efficiency of experiments and user-friendliness of software.

Relaxation and signal-to-noise ratio (S/N) in pulse experiments on trityl radicals were measured at frequencies between 400 MHz and 1.5 GHz. Relaxation time increases as the frequency increases and the radical concentration decreases. Oftentimes, primary care providers lack the knowledge, confidence, and time to monitor chronic pain patients closely resulting in possible inappropriate prescribing of opioids. Purpose: The project aim was to improve primary care provider knowledge and confidence in managing chronic pain patients in the primary care setting using a step model that was developed based on the Centers for Disease Control (CDC) guidelines as well as online education modules produced by the CDC. Bradyrhizobium japonicum is a Gram-negative soil bacterium commonly known for its agriculturally significant mutualistic relationship with soybean. In this symbiosis, the bacteria and plant undergo complex molecular signaling characterized by sent and received signals resulting in the formation of infection threads and root nodules. This research aimed to compare two related bacterial outer membrane proteins, FegA and FhuA, associated with the molecular signaling between the bacteria and plant. Previous work has led to the hypothesis that the N-terminal domain (NTD) of FegA in B. japonicum is needed for a functional symbiosis to occur. Anorexia Nervosa (AN) is a psychiatric disorder characterized by significantly low body weight and over-evaluation of weight and shape in self-identity. The complex etiology of Anorexia Nervosa renders many current treatments ineffective, thereby prolonging the course of the illness. Among the many psychological and behavioral features of AN, cognitive features like obsessiveness, rigidity, and perfectionism are often described. Patients hold firm beliefs about an ideal body weight and find it difficult to alter their thinking patterns. In this dissertation work, a semi-quantitative assay using a lipidomics approach and an absolute quantitative assay using liquid chromatography-mass spectrometry techniques were developed (LC-MS). In the lipidomics approach, ultra-high pressure liquid chromatography in tandem with quadrupole time of flight (UHPLC-QTOF) mass spectrometry was used to profile, compare and quantitate the human plasma lipids in Alzheimer’s disease subjects (AD) and normal cognitive controls (NCCs). This study explores language policies in “almost-bilingual” classrooms, in which most but not all students share a home language. Teachers who are bilingual face a dilemma in these settings. Should they draw on shared linguistic expertise to benefit the majority while excluding a few, or should they forego significant benefits for most in the interest of equity? This qualitative study examines the classroom language policies and practices of one English-as-a second-language (ESL) teacher at a majority-Latino high school.
