Salicylate hydroxylase (NahG) is a flavin-dependent monooxygenase that catalyzes the decarboxylative hydroxylation of salicylate into catechol in the naphthalene degradation pathway in Pseudomonas putida G7. We explored the mechanism of action of this enzyme in detail using a combination of structural and biophysical methods. NahG shares many structural and mechanistic features with other versatile flavin-dependent monooxygenases, with potential biocatalytic applications. The crystal structure at 2.0 Å resolution for the apo form of NahG adds a new snapshot preceding the FAD binding in flavin-dependent monooxygenases. The kcat/Km for the salicylate reaction catalyzed by the holo form is …

The salmonid fisheries of the Columbia River Basin (CRB) have enormous socioeconomic, cultural, and ecological importance to numerous diverse stakeholders (eg state, federal, tribal, nonprofit), and there are a wide array of opinions and perspectives on how these fisheries should be managed. Although we appreciate Duda et al.'s commentary, it offers only one perspective of many in this context. The objective of our paper (Hand et al. 2018) was to provide justification for “the importance of social–ecological perspectives when communicating conservation values and goals, and the role of independent science in guiding management policy and practice for …

When Do Psychosocial Explanations Of Psychiatric Problems Increase Stigma? Self-Report And Implicit Evidence, Ryan Thibodeau, Gabrielle L. Amberger
When Do Psychosocial Explanations Of Psychiatric Problems Increase Stigma? Self-Report And Implicit Evidence, Ryan Thibodeau, Gabrielle L. Amberger
Psychology Faculty/Staff Publications
Background and objectives
Biomedical explanations of psychiatric problems, compared to psychosocial explanations, may amplify psychiatric stigma. One limitation of existing research is the measurement of almost exclusively self-reported stigma. This study evaluated the stigma-related effects of biomedical versus psychosocial explanations of schizophrenia using conventional self-report and two other measurement approaches that may tap more deeply held attitudes.
Methods
One hundred three undergraduates listened to a vignette describing a man with (1) schizophrenia of biomedical origin, (2) schizophrenia of psychosocial origin, or (3) diabetes. They then completed an Implicit Association Test, conventional self-report stigma measures, and projected other measures that captured …

Mfd Protects Against Oxidative Stress In Bacillus Subtilis Independently Of Its Canonical Function In Dna Repair, Holly Anne Martin, Katelyne E. Porter, Carmen Vallin, Tatiana Ermi, Natalie Contreras, Mario Pedraza-Reyes, Eduardo Robleto
Life Sciences Faculty Research
Background: Previous reports showed that mutagenesis in nutrient-limiting conditions is dependent on Mfd in Bacillus subtilis. Mfd initiates one type of transcription-coupled repair (TCR); this type of repair is known to target bulky lesions, like those associated with UV exposure. Interestingly, the roles of Mfd in repair of oxidative-promoted DNA damage and regulation of transcription differ. Here, we used a genetic approach to test whether Mfd protected B. subtilis from exposure to two different oxidants. Results: Wild-type cells survived tert-butyl hydroperoxide (t-BHP) exposure significantly better than Mfd-deficient cells. This protective effect was independent of UvrA, a component of the canonical …

Many granting agencies allow reviewers to know the identity of a proposal’s Principal Investigator (PI), which opens the possibility that reviewers discriminate on the basis of PI race and gender. We investigated this experimentally with 48 NIH R01 grant proposals, representing a broad spectrum of NIH-funded science. We modified PI names to create separate White male, White female, Black male, and Black female versions of each proposal, and 412 scientists each submitted initial reviews for three proposals. We find little to no race or gender bias in initial R01 evaluations, and additionally find that any bias that might have been …
