Diviva Localization During Sepiva Depletion In Mycobacterium Smegmatis, Madeline Carlson May 2019

Diviva Localization During Sepiva Depletion In Mycobacterium Smegmatis, Madeline Carlson

2019 Spring Honors Capstone Projects - Archive

Mycobacterium is a genus of bacteria best known for Mycobacterium tuberculosis(Mtb), which causes tuberculosis. Proteins that are distinctive to mycobacteria can be studied in closely related species of Mtb in order to find potential new tuberculosis drugs based on unique mycobacterial processes. In this study, the model organism Mycobacterium smegmatis (Msmeg) was used to study the protein, SepIVA. SepIVA is a protein found in Mycobacteria, which is thought to be similar to a group of proteins that function in assisting with cell wall synthesis during cell division and cell elongation. To study the possible functions of SepIVA, the localization of …

Young Germ Cell Depleted Ovaries In Post-Reproductive Mice And Its Effects On Immune Function, Mckenna Walters May 2019

Young Germ Cell Depleted Ovaries In Post-Reproductive Mice And Its Effects On Immune Function, Mckenna Walters

Undergraduate Honors Capstone Projects

It has previously been shown that young, cycling ovarian transplantation in aged female mice increased the general health and life span in regard to their post-reproductive health. It has further been hypothesized that this enhancement of health is directly influenced by the ovarian somatic cells. To address this hypothesis, transplants of young germ cell depleted and germ cell containing ovaries were performed on female mice. The purpose of this study is to continue to discern the reproductive influence on aging health, specifically in the area of immunological well-being. Control group mice were separated by age and treatment mice were subsequently …

Development Of A Lectin-Fc Fusion Protein With Antiviral And Anti-Cancer Activity., Matthew William Dent

Electronic Theses and Dissertations

This thesis describes the development of a novel lectin-Fc fusion protein and its antiviral and anti-cancer activity. The molecule, Avaren-Fc (AvFc), is a fusion of a variant of the actinomycete lectin actinohivin (Avaren) and the Fc region of human IgG1, and is selective for the terminal α1,2-mannose residues found at the ends of high-mannose-type glycans that can be found on the surface of certain heavily glycosylated viruses and cancer cells. Here, AvFc was found to be able to neutralize simian immunodeficiency virus as well as Hepatitis C virus with nanomolar IC50 values. Furthermore, AvFc recognizes a number of cell …

Development Of Mouse Models For Respiratory And Neurological Disease Caused By Enterovirus D68 And Evaluation Of Antiviral Therapies, Brett L. Hurst

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Enterovirus D68 (EV-D68) is a virus that normally causes disease in children. While this virus typically causes a respiratory infection, in 2014, a large outbreak of the virus was associated with patients that had paralysis of the arms or legs. Even though the virus was discovered in 1962, little was known about the life cycle of the virus or its ability to cause disease. An animal model of disease was needed to understand how the virus causes disease and to develop antiviral compounds to target the virus life cycle.

We adapted the virus by serial-passage in lung tissues from mice …

Male Reproductive Infection And Sexual Transmission Of Zika Virus In An Immunocompromised Mouse Model, Chad S. Clancy May 2019

Male Reproductive Infection And Sexual Transmission Of Zika Virus In An Immunocompromised Mouse Model, Chad S. Clancy

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Zika virus (ZIKV) is a sexually transmitted viral infection most frequently transmitted by mosquitoes. The source of infectious virions in the male reproductive tract has yet to be elucidated. The goals of the studies included developing and characterizing two mouse models for reproductive transmission studies and demonstration of sexual transmission of virus via artificial insemination. The mouse strains used in the study lacked receptors to interferon molecules, key signaling proteins of the host immune response. Inflammation severity was assessed during acute disease, 5-11 days after infection using a novel histopathology grading system. ZIKV proteins and genome were initially detected in …

School of Medicine Publications

The association between tuberculosis (TB) and diabetes mellitus (DM) had a common place in the literature up to the first half of the 20th century, but virtually disappeared with the discovery of insulin to treat DM and antibiotics to cure TB. In the late 1990s the literature began to re-emerge with the worldwide increase in type 2 DM, particularly in TB-endemic countries. Today, type 2 DM is the most prevalent comorbidity among TB patients and the World Health Organization considers it a threat to TB control. We summarize the literature on TB and DM up to the 1960s. Then we …

Experimental Immunology And The Potential For Osteopathological Reconstructions : Pursuing An Experimental Foundation For The Skeletal Inflammatory Index., Megan Elizabeth Duncanson May 2019

Experimental Immunology And The Potential For Osteopathological Reconstructions : Pursuing An Experimental Foundation For The Skeletal Inflammatory Index., Megan Elizabeth Duncanson

Electronic Theses and Dissertations

Individuals who mount a strong inflammatory response may produce a shift in the systemic levels of inflammatory mediators, which may lead to a potential hyper-inflammatory phenotype (HIP). Systemic inflammation can increase severity in local inflammatory processes detected in bone lesions. This thesis investigated in vitro if human immune cells pre-treated with inflammatory inducers would affect the inflammatory response against Porphyromonas gingivalis or Staphylococcus aureus (both associated with osteological lesions). We exposed human peripheral blood mononuclear cells to bacterial lysates, or pro-inflammatory cytokines. Sequentially, we exposed the same culture to either P. gingivalis or S. aureus. The final expression of …

Background:

Catheter Associated Urinary Tract Infections (CAUTI) are considered a preventable hospital-acquired infection [2]. Centers for Medicaid and Medicare no longer reimburse hospitals for preventable CAUTIs [2]. A concerted effort to decrease our indwelling urinary catheter (IUC) rate has led to a decrease in the number of infections related to these catheters at Maine Medical Center (MMC) in Portland, Maine. Starting in 2012 as a result of The Joint Commission National Patient Safety Goal, the inter-professional CAUTI committee focus has been on decreasing utilization of IUC[1]. After an initial large decrease in utilization, the rates have flattened. The effort over …

Tuberculous Peritonitis: An Unusual Diagnosis For Vague Abdominal Pain, Zachary Ciochetto, Michel Hanna May 2019

Tuberculous Peritonitis: An Unusual Diagnosis For Vague Abdominal Pain, Zachary Ciochetto, Michel Hanna

MERF 2019 - Case Reports

Mycobacterium Tuberculosis (MTB) continues to affect millions of people worldwide and remains a significant cause of mortality in those who contract the disease and do not receive proper treatment. This phenomena also explains why this disease continues to be transmitted when they are not properly isolated. Predominantly the focus is on pulmonary TB but that is only one of many complications of the disease. Miliary TB occurs when the infection hematogenously disseminates throughout the body and causes further complications. When the infection seeds other organ sites it causes unique complications and one of the more worrisome is peritonitis. We report …

Idea, Form, Reality – The Implications Of Natural Ventilation Strategies On Public Buildings In Temperate Maritime Climates, Michael P.G. Haslam

Masters

Natural ventilation is one of the prime form givers in environmentally responsive architecture – in its detail and its architecturally charged form of stacks and voids. It facilitates our understanding of a response to natural systems – not just in how it responds visually to warm and cool air but also in the adaptive approach to thermal comfort. The purpose of this research is to investigate the impacts and implications of natural ventilation strategies for different typologies of public and commercial buildings. This is with the aim of attempting to define a vocabulary of ventilation techniques and details that are …

Epidemiology Of Tuberculosis In Internationally Displaced Children Resettling In Harris County, Gabriella S. Lamb May 2019

Epidemiology Of Tuberculosis In Internationally Displaced Children Resettling In Harris County, Gabriella S. Lamb

Dissertations and Theses (Open Access)

Background More than 300,000 refugees arrived in the United States (U.S.) from 2010-2015, and Texas accepts the 2nd highest number of refugees. Texas also accepts large numbers of asylees, parolees, and special immigrant visa holders. Additionally, a large proportion of trafficked persons in the U.S. live in or pass through Texas. Foreign-born children are disproportionately affected by tuberculosis (TB) and account for two-thirds of U.S. childhood TB cases. Children are at greater risk for progression from TB infection to disease and experience greater morbidity and mortality from TB disease. This makes screening for and treatment of TB infection in children …

Trends In Advanced Hiv Disease And Long-Term Outcomes Of Hivinfected Children Treated With Antiretroviral Therapy In Eastern And Southern Africa: 2003-2017, Peter James Elyanu

Dissertations and Theses (Open Access)

Despite the over 10-years’ experience providing pediatric antiretroviral therapy(ART) and implementing different WHO guidelines, data on updated trends (after 2010) in advanced disease at ART start, and on long-term survival of children receiving ART are scarce. Without these data, the impact of recent WHO guidelines on early treatment initiation and long-term survival remains unclear. First, we examined trends in advanced disease at ART start, then described 10-year survival and investigated risk factors of early mortality in children treated with ART and lastly examined the association between rapid ART initiation and outcomes (mortality and loss to follow-up) of HIV infected. We …

The HPV vaccine protects against diseases and cancers caused by the …

Approximate Dynamic Programming: Health Care Applications, Amir Ali Nasrollahzadeh May 2019

Approximate Dynamic Programming: Health Care Applications, Amir Ali Nasrollahzadeh

All Dissertations

This dissertation considers different approximate solutions to Markov decision problems formulated within the dynamic programming framework in two health care applications. Dynamic formulations are appropriate for problems which require optimization over time and a variety of settings for different scenarios and policies. This is similar to the situation in a lot of health care applications for which because of the curses of dimensionality, exact solutions do not always exist. Thus, approximate analysis to find near optimal solutions are motivated. To check the quality of approximation, additional evidence such as boundaries, consistency analysis, or asymptotic behavior evaluation are required. Emergency vehicle …

Access Before Evidence And The Price Of The Fda's New Drug Authorities, Erika Lietzan May 2019

Access Before Evidence And The Price Of The Fda's New Drug Authorities, Erika Lietzan

Faculty Publications

Sometimes drug innovation seems to happen in reverse. Patients enjoy a treatment for years even though the treatment has not been approved by the FDA or proven safe and effective to the FDA's standards. (Sometimes this happens because the FDA has declined to take enforcement action.) The agency encourages companies to perform the work necessary to satisfy the United States "gold standard" for new drug approval, however, by promising exclusivity in the marketplace. When a company does this work, at considerable expense, the results are predictable. The new drug is expensive, and patients and payers (and sometimes policymakers) are outraged. …

Frozen Pre-Embryo Practice In Missouri, Mary M. Beck, L. "Joanna" Beck Wilkinson May 2019

Frozen Pre-Embryo Practice In Missouri, Mary M. Beck, L. "Joanna" Beck Wilkinson

Faculty Publications

McQueen v. Gadberry was an Eastern District dissolution dispute over frozen pre-implantation embryos formed from McQueen’s eggs and Gadberry’s sperm. The St. Louis County trial court found the pre-embryos to be marital property of a special character and awarded them jointly to each of the former spouses. The appellate court affirmed. McQueen is an important decision because the finding that embryos are marital property was an issue of first impression that affects many Missouri families. Infertility is a common problem, couples frequently utilize assisted reproductive technologies (ART) to form families, and the extra frozen pre-implantation embryos (hereinafter pre-embryos) have fueled …

PCOM Capstone Projects

Childhood vaccination has been one of public health’s greatest achievements in its history. Just over the past 20 years, immunizations averted more than 21 million hospitalizations and 732,000 deaths in the US (Skinner, 2017). However, healthcare professionals, public health advocates, and researchers are still witnessing an unceasing tension between the anti-vaccination movement and pro-vaccination organizations. Since the invention of the first vaccines, individuals from various religious sects, cultural backgrounds, and ethnic groups have been questioning the efficacy of vaccines, the possible health risks and side effects that may be introduced, and the morality behind the act of vaccinating. With questionable …

The Potential Synergistic Effect F Egcg-S And Antibiotics On The Eskape Bacteria Group, Summer Elsayed

Theses, Dissertations and Culminating Projects

The primary etiology of nosocomial infections derives from the following pathogenic ESKAPE organisms: Enterococcus faecium/faecalis, Staphyloccoccus aureus, Klebsiella pneumoniae, Acinetobacter baumanni, Pseudomonas aeruginosa and Enterobacter species. These opportunistic organisms are multidrug resistant and have the ability to form biofilm, which pose a great threat to clinical medicine. Evidence has shown catechins found in green tea (Camellia sinensis), have antibacterial activity. In this study, a modified polyphenolic catechin compound Epigallocatechin-3-gallate stearate (EGCG-S) was used to research its potential to inhibit ESKAPE growth and biofilm formation. In addition, synergistic ability of EGCG-S with antibiotics was also studied. Disk diffusion and colony forming …
