According to the Center for Disease Control’s (CDC) report to the Congress, there are 2.2 million emergency department visits; 80,000 hospitalizations; and 50,000 deaths each year due to traumatic brain injury. Adults 65 years and older account substantially for the majority of the hospitalization and deaths. Over 70% of the traumatic brain injuries of the older adults are classified as mild to moderate; however, even with these milder injuries, older adults present with a significantly higher morbidity and mortality compared to all other age groups (LeBlanc et al., 2006). Key variables such as trauma exposure (TE) and internalizing symptoms (e.g., posttraumatic stress disorder [PTSD]) have been shown to correlate with non-medical use of prescription drugs (NMUPD); however, the temporal associations between these phenotypes remain poorly understood. Moreover, there is a paucity of research that incorporates the influence of genetic factors in the etiology of NMUPD. Although it has been demonstrated that drug use disorders are moderately heritable, research aimed at identifying the specific genes conferring risk is virtually non-existent for NMUPD. Adverse childhood experiences (ACEs) are linked to increased mental health problems in children, but their association with mental health services utilization is not well known. This secondary analysis used 2016 National Survey of Children’s Health data from two samples: children aged 6-17-years-old with a mental or behavioral condition in need of treatment or counseling (N = 5,723); and a subsample of children who experienced at least one ACE (n = 3,812). Multiple logistic regression and latent class analysis (LCA) were performed to examine the association between ACEs and mental health services utilization. Background. DNA methylation (DNAm) is a removable chemical modification to the DNA sequence intimately associated with genomic stability, cellular identity, and gene expression. DNAm patterning reflects joint contributions from genetic, environmental, and behavioral factors. As such, differences in DNAm patterns may explain interindividual variability in risk liability for complex traits like major depression (MD). Hundreds of significant DNAm loci have been identified using cross-sectional association studies. This dissertation builds on that foundational work to explore novel statistical approaches for longitudinal DNAm analyses. Methods. 2.5 million people sustain a traumatic brain injury (TBI) annually in the United States. Although there is potential for functional recovery following TBI, there is no definitive treatment to improve recovery after TBI. Our lab has shown that TBI enhances an endogenous neurogenic response in the subventricular zone and hippocampus. TBI-induced neural stem cells (NSCs) can integrate into regions such as the hippocampus and olfactory bulb. Although the mechanism behind TBI-enhanced neurogenesis remains unknown, the Notch signaling pathway has been implicated as a regulator in the maintenance and survival of NSCs.

In this study, Naegleria fowleri, a pathogenic amoeba and the causative agent of Primary Amebic Meningoencephalitis (PAM), was utilized to determine the presence or absence of classically conserved Golgi molecules featured in the expression of a Golgi apparatus. Previous studies concluded no Golgi expression via light microscopy and transmission electron microscopy, but a recent report on Naegleria gruberi indicated the presence of dispersed Golgi tubules. Empathy, the ability to feel and/or understand another’s emotional state, plays a significant role in interpersonal interactions, mitigating hostility and enhancing affiliation and helping. However, empathy also biases interpersonal reactions. For example, at the group level empathy can become amplified towards members of their ingroup and blunted towards individuals in outgroups, a term called parochial empathy. Currently, no validated measures of parochial empathy at the dispositional level exist, and development of such a scale would be important to understanding the role of group-based emotions in prejudice and discrimination against outgroups. Psychostimulants elicit their psychological and behavioral effects through interactions with the monoamine transporters. These compounds are classified as either substrates or reuptake inhibitors, depending on their mechanism of action, and have varying potencies at the three human monoamine transporters, hDAT, hSERT, and hNET. Substrates and reuptake inhibitors also have distinct electrophysiological signatures: reuptake inhibitors cause hyperpolarization in HEK293 cells stably expressing the monoamine transporters, whereas substrate transport mediates a depolarizing current. Recent studies have shown that substrate-mediated depolarization is sufficient to activate L-type calcium channels, a characteristic which has been used to develop a novel calcium assay which uses calcium …

Over the past few decades, there has been increased research focus on carbon cycling within aquatic systems, especially with the changing global climate. Inland waters play a major role in the global carbon cycle, but the fundamental features remain poorly understood, particularly the large lakes of the world. Background:Non-medical use of prescription opioids (NMUPO) has become a clear threat to public health. Young adults (aged 18 to 25) have a high risk of NMUPO. My prior work on Chinese undergraduates indicates a high prevalence of lifetime NMUPO (49.2%). Health behavior theories propose that outcome expectancies are robust psychosocial determinants of substance use. Literature has identified the influence of outcome expectancies on alcohol and drug use. Psychedelics are a subset of hallucinogenic drugs that exert their characteristic effects through agonist activity at the serotonin receptor 2A (5-HT2A). In this study, I aimed to characterize the modulatory role of the metabotropic glutamate subtype 2 receptor (mGluR2) in the 5-HT2A-specific rodent model of hallucinogenic action, head-twitch response (HTR). Secondly, I aimed to explore if 5-HT2A agonist-induced deficits in prepulse inhibition (PPI) of the startle response, an additional model of hallucinogenic action, could be produced in mice. Over 200,000 cases of breast cancer are diagnosed every year. Nearly 20% of these patients supplement their diets with some form of vitamin D. This high frequency of vitamin D supplement use may be due in part to research suggesting that cancer patients with higher serum vitamin D3 levels have better prognoses than patients with low serum vitamin D3. However, double-blind clinical trials on the efficacy of vitamin D3 supplementation in breast cancer have been inconclusive. Prolactin (PRL) and its receptor (PRLr) have been implicated in the development and progression of human breast cancer. PRL activates its receptor and induces activation of proximal Janus kinase 2 (Jak2) for signal transduction. Here, we sought to determine the role of PRLr-associated peptidyl-prolyl isomerase, cyclophilin A (CypA), in modulating structure/function relationships of the PRLr. It was demonstrated that CypA mediated PRL-induced conformational change of the CFP- and -YFP tagged forms of the PRLr cytoplasmic-tail, whereas CypA inhibition by NIM811 (N-methyl-4-isoleucine cyclosporin) or knockdown blocked the conformational change of the PRLr assessed by Fluorescence Resonance Energy Transfer (FRET) signal or …

The objective of this thesis is to formulate and solve the sensor placement problem for damage localization in a sensor network. A Bayesian estimation problem is formulated with the time-of-flight (ToF) measurements. In this model, ToF of lamb waves, which are generated and received by piezoelectric sensors, is the total time for each wave to be transmitted, reflected by the target, and received by the sensor. The ToF of the scattered lamb wave has characteristic information about the target location. After the rebellion over the killing of Michael Brown, the US Justice Department reported that over-policing for the sake of monetary extraction was taking place in Ferguson, MO, with non-White and people in poverty being disproportionately targeted at the hands of the police. Breast cancer mortality is mainly due to distant recurrence of the disease arising from dormant tumor cells established by cancer therapies. Patients who initially respond to cancer therapies often succumb to distant recurrence of the disease. Latina/o emerging adults living in southern states may be especially at risk of being disproportionately affected by HIV. In 2015, Latina/os aged 20 to 24 accounted for 14.7% of all 20-24-year old’s living with HIV in southern states, despite the fact that 20 to 24-year-olds make up 3.7% of the region’s overall population of individuals living with HIV (CDC, 2016; U.S. Census Bureau, 2017). Purpose: This study examined the impact of an acute bout of exercise on LXRα mRNA expression in isolated PBMCs from aerobically fit middle-aged males, and its associations with cellular inflammation. Furthermore, association between cellular age and LXRα were investigated. Methods: LXRα mRNA, plasma free fatty acids, IL-6, and MCP-1 responses were quantified following acute moderate intensity exercise and throughout recovery. To examine LXRα’s associations with cellular inflammation, PBMCs were stimulated with palmitate. Finally, telomere length was quantified as an indicator of cellular age. Results: LXRα mRNA expression was reduced at 90 minutes of recovery and did not influence systemic inflammation. …

The purpose of this inquiry is to identify the barriers and facilitators that can advance an inpatient rehabilitation facility to become a high reliability organization and advance interdisciplinary teamwork.

One of the most important factors in accessing dental care is having dental insurance. For people with low incomes, Medicaid is the main source of health insurance. Medi-Cal is California’s Medicaid program. Adult dental services were mostly eliminated in Medi-Cal in 2009 due to the economic downturn and partially restored in 2014. The objective of this study is to evaluate the effect of change in Medi-Cal dental coverage, specifically the partial restoration of adult dental coverage in 2014, on dental care utilization among Medi-Cal beneficiaries. Widely effective treatment for alcohol use disorder is not yet available, because the exact biological mechanisms that underlie this disorder are not completely understood. One way to gain a better understanding of these mechanisms is to examine the genetic frameworks that contribute to the risk for developing this disorder. This dissertation examines genetic association data in combination with gene expression networks in the brain to identify functional groups of genes associated with alcohol consumption and dependence.

A Longitudinal Study Of The Stress-Buffering Effects Of Romantic Relationships On Alcohol Outcomes In College Students Exposed To Trauma, Rebecca L. Smith

This study examined interplay between interpersonal trauma (IPT), relationship status, relationship satisfaction (SAT), and partner substance use (PSU), and whether these relationship dimensions moderated associations between IPT and alcohol outcomes. Data came from a longitudinal study of college students (N=9,911; 61%female; 49%White). Precollege IPT increased the likelihood of being in a relationship, while college-onset IPT decreased the likelihood. IPT predicted lower SAT and higher PSU. Individuals with precollege IPT consumed more alcohol than those without IPT, but this was mitigated for those in relationships. The differential mobility classifier (DMC) is one of the core components in electrical mobility particle sizers for sizing sub-micrometer particles. Designing the DMC requires knowledge of the geometrical and constructional imperfection (or tolerance). Studying the effects of geometrical imperfection on the performance of the DMC is necessary to provide manufacturing tolerance and it helps to predict the performance of geometrically imperfect classifiers, as well as providing a calibration curve for the DMC. Childhood sexual abuse (CSA) is a stressful life experience with lasting/far-reaching health and psychopathological consequences. Our laboratory recently identified a significantly increased frequency of acquired chromosomal anomalies (assessed using the cytokinesis-blocked micronucleus assay) in adult female twins exposed to CSA when compared to their unexposed co-twin. Abstract
Although great strides have been made over the decades in development and optimization of anti-cancer therapies, even highly effective drugs often fail to completely eliminate tumors. Residual tumor cells can enter into a state of dormancy for prolonged periods of time but eventually are able to regain proliferative capacity and reemerge as chemotherapy-resistant disease. Because recurrent disease is a leading contributor to patient’s mortality, it is paramount to identify strategies for effectively destroying residual tumor cells.

An exo-atmospheric nuclear detonation releases up to 80 percent of its’ energy as X-rays. Satellite’s solar cells and their protective coatings are vulnerable to low energy X-ray radiation. Cold X-rays (~1-1.5 keV) are absorbed close to the surface of materials causing the blow-off and rapid formation of Warm Dense Plasmas (WDPs), particularly in a gap between the unshielded active elements of solar cells. College students have an increased risk for cannabis use, trauma exposure, and posttraumatic stress disorder (PTSD). Cannabis use disorder (CUD) and PTSD comorbidity is high, and given the negative consequences of the comorbidity (e.g., poor academic outcomes), there is a need to understand comorbid CUD-PTSD etiology. Two primary etiologic models exist: self-medication (i.e., PTSD à CUD) and high-risk (i.e., CUD à PTSD) hypotheses.
