Synthesis And Bioevaluation Of Methionine Depletion Agents, Houssine Ikhlef Jan 2019

Synthesis And Bioevaluation Of Methionine Depletion Agents, Houssine Ikhlef

Electronic Theses and Dissertations

The native polyamines (putrescine, spermidine, and spermine) are low molecular weight amines that exist as polycations at physiological pH. These polycations interact with DNA, RNA, and influence many cellular processes. Intracellular polyamine levels are maintained via a balance of polyamine biosynthesis, catabolism, and transport. Ornithine, an amino acid obtained from L-arginine, is decarboxylated by ornithine decarboxylase (ODC) to form putrescine. The biosynthesis of the higher polyamines, spermidine and spermine, requires the addition of an amino-propyl group donated by decarboxylated S-adenosylmethionine (dc-SAM), which itself is derived from the amino acid L-methionine (Met). Tumor cells are heavily reliant on methionine because it …

Lifetime History Of Concussions And Behavioral Measures Of Anhedonia, John O'Donnell Jan 2019

Lifetime History Of Concussions And Behavioral Measures Of Anhedonia, John O'Donnell

Electronic Theses and Dissertations

Concussions are the most common neuropsychological problem in the United States and are associated with sequelae such as cognitive complaints and depression-related symptoms. Recent research suggests that head trauma is associated with anhedonia and that concussions have the potential to damage axons and postsynaptic connections in neural circuits that play a role in reward processing. Anhedonia may be better understood as an overarching construct with multiple subtypes including motivational, decisional, and consummatory. The current study examines the relationship between lifetime concussion history and subtypes of anhedonia using behavioral measures of reward processing: the Effort Expenditure for Rewards Task (EEfRT), Probabilistic …

Do Global Cities Make Green Cities? How Global Governance Impacts Transportation In Bogotá And Medellín, Eleanor Jackson Jan 2019

Do Global Cities Make Green Cities? How Global Governance Impacts Transportation In Bogotá And Medellín, Eleanor Jackson

Honors Theses

This thesis examines how global and local governance has combined to deliver effective and sustainable public transportation in cities by comparing Bogotá’s bus rapid transit (BRT) system, TransMilenio, with Medellín’s mass transit system, STIMVA, often referred to as Metro de Medellín. After considering the rationales used to justify local and global authority over climate change, this analysis problematizes the supposed benefits of empowering global and local actors by highlighting the conflicts of interest that plague the elites who mediate the global and the local. In analyzing the global and local interactions, this work draws from extensive literature to highlight three …

Human Action Recognition In Videos Using Transfer Learning, Kaiqiang Huang, Sarah Jane Delany, Susan Mckeever

Session 1: Active Vision, Tracking, Motion Analysis

A variety of systems focus on detecting the actions and activities performed by humans, such as video surveillance and health monitoring systems. However, published labelled human action datasets for training supervised machine learning models are limited in number and expensive to produce. The use of transfer learning for the task of action recognition can help to address this issue by transferring or re-using the knowledge of existing trained models, in combination with minimal training data from the new target domain. Our focus in this paper is an investigation of video feature representations and machine learning algorithms for transfer learning for …

Atp Induced Molecular Disassembly Of Cytolethal Distending Toxin's B/C Heterodimer, George Huhn

Electronic Theses and Dissertations

Cytolethal distending toxin (CDT) is a virulence factor produced by many Gram-negative bacteria, including Haemophilus ducreyi. This fastidious pathogen is the causative agent of genital cancroid. CDT is a heterotrimeric toxin with an AB2 structure consisting of a cell-binding "B" domain (CdtA + CdtC) and a catalytic "A" domain (CdtB) that has DNase activity. This toxin assembles in the bacterial periplasm that lacks ATP and is secreted into the extracellular environment. After cell binding, CDT is internalized by endocytosis and travels through the endosomes and Golgi before arriving in the endoplasmic reticulum (ER). CdtA is lost from the holotoxin before …

In this dissertation we present some results relating to three topics on phase retrieval. Chapters 1 and 2 contain the relevant background materials. In chapter 3, we introduce the notion of exact phase-retrievable frames as a way of measuring a frame's redundancy with respect to its phase retrieval property. We show that, in the d-dimensional real Hilbert space case, exact phase-retrievable frames can be of …

It Takes Two To Tango: The Toxin-Chaperone Relationship, Alisha Kellner Jan 2019

It Takes Two To Tango: The Toxin-Chaperone Relationship, Alisha Kellner

Electronic Theses and Dissertations

Cholera toxin (CT) enters the cell via receptor-mediated endocytosis and travels in a retrograde fashion to the endoplasmic reticulum (ER). The catalytic A1 subunit (CTA1) is then displaced from the rest of the holotoxin, unfolds, and is exported to the cytosol where it regains an active conformation for the ADP-ribosylation of its G-protein target. We have shown that the cytosolic chaperones Hsp90 and Hsc70 are required for CTA1 translocation to the cytosol. We have also shown that both are able to independently bind and refold CTA1. Using libraries of CTA1-derived peptides, we have identified a single Hsc70 binding site, YYIYVI …
