The staphylococcal accessory regulator (sarA) plays an important role in Staphylococcus aureus infections including osteomyelitis, and the msaABCR operon has been implicated as an important factor in modulating expression of sarA. Thus, we investigated the contribution of msaABCR to sarA-associated phenotypes in the S. aureus clinical isolates LAC and UAMS-1. Mutation of msaABCR resulted in reduced production of SarA and a reduced capacity to form a biofilm in both strains. Biofilm formation was enhanced in a LAC msa mutant by restoring the production of SarA, but this was not true in a UAMS-1 msa mutant. Similarly, …

Dynamical systems have been of interest to biologists and mathematicians alike. Many processes in biology lend themselves to dynamical study. Movement, change, and response to stimuli are dynamical characteristics that define what is 'alive'. A scientific relationship between these two fields is therefore natural. In this thesis, I describe how my PhD research variously related to biological, mathematical, and computational problems in cell biology. In chapter 1 I introduce some of the current problems in the field. In chapter 2, my mathematical model of firefly luciferase in vivo shows the importance of dynamical models to understand systems. Tsetse flies (Diptera: Glossinidae) house a taxonomically diverse microbiota that includes environmentally acquired bacteria, maternally transmitted symbiotic bacteria, and pathogenic African trypanosomes. Sodalis glossinidius, which is a facultative symbiont that resides intra and extracellularly within multiple tsetse tissues, has been implicated as a mediator of trypanosome infection establishment in the fly’s gut. Tsetse’s gut-associated population of Sodalis are subjected to marked temperature fluctuations each time their ectothermic fly host imbibes vertebrate blood. The molecular mechanisms that Sodalis employs to deal with this heat stress are unknown. In this study, we examined the thermal tolerance and heat shock response of …

Similarity analysis is the subject of several studies that primarily collect, process, and analyze movement data of various types, including animals, humans, vehicles, hurricanes, and eye tracking. The movement studies often focus on characteristics of a mobile entity (moving object) over time and space. Such studies usually are interested in tracking the changes to the moving object. Therefore, the size of the object becomes irrelevant. Each mobile object is considered a moving point through time and space. Water‐borne hormone measurement is a noninvasive method suitable for amphibians of all sizes that are otherwise difficult to sample. For this method, containment‐water is assayed for hormones released by the animal. Originally developed in fish, the method has expanded to amphibians, but requires additional species‐specific validations. We wanted to determine physiological relevance of water‐borne corticosterone in spotted salamanders (Ambystoma maculatum) by comparing concentrations to those taken using established corticosterone sampling methods, such as plasma. Using a mixture of field and laboratory studies, we compared water‐borne corticosterone levels to other traditional methods of sampling corticosterone for spotted salamander larvae, metamorphs, and …

Pre-Commitment To Moral Values, Nicholas R. Herzog, Justin F. Landy, Emma E. Levine

When faced with reoccurring tradeoffs between moral values, people can address them by considering the specifics of each case or by setting policies that predetermine how they will address similar cases. Previous research on moral judgment has often focused on isolated tradeoffs, and therefore, it is unclear which decision strategies are preferred in contexts with reoccurring tradeoffs. Across our studies, participants judged people who precommitted to always prioritizing one value more positively than people who adjusted their priorities based on the specifics of each case. Our findings have important implications for understanding public perceptions of complex policies.

The Social Consequences Of Absolute Moral Proclamations, Elizabeth Huppert, Nicholas R. Herzog, Justin F. Landy, Emma E. Levine

Across six studies (N = 3348), we find that people prefer targets who make absolute proclamations (i.e. "It is never okay for people to lie" ) over targets who make ambiguous proclamations ("It is sometimes okay for people to lie" ), even when both targets tell equivalent lies. Preferences for absolutism stem from the belief that moral proclamations send a true signal about moral character--they are not cheap talk. Therefore, absolute proclamations signal moral character, despite also signaling hypocrisy. This research sheds light on the consequences of absolute proclamations and identifies circumstances in which hypocrisy is preferred over consistency.

There are limited data on the lesion volume changes following spinal cord injury (SCI). In this study, a meta-analysis was performed to evaluate the volume size changes of the injured spinal cord over time among animal studies in traumatic SCI. Following the Preferred Reporting Items for Systematic Reviews and Meta-Analyses (PRISMA) guidelines, we conducted a comprehensive electronic search of English literature of PubMed and EMBASE databases from 1946 to 2015 concerning the time-dependent changes in the volume of the spinal cord following mechanical traumatic SCI. New York is experiencing dramatic population growth among horse-and-buggy driving populations. Farming is recognized as essential to the maintenance of these cultures. Nationally, these groups, particularly in larger settlements, struggle to stay active in farming. However, Old Order Mennonites run contrary to trends within the plain sects as well as agriculture at large. With over 570 households, a central New York settlement of Groffdale Conference Mennonites grew by 2,700% over the last three decades. They have one of the highest percentages of their population in agriculture, operating 99% of the dairy farms in the area, yet with a herd size …

Lighting and marking recommendations for animal-drawn buggies and wagons were first established in 2001 through an American Society of Agricultural and Biological Engineers (ASABE) Engineering Practice, EP576.1. Many Anabaptist communities who primarily rely on animal-drawn vehicles utilize this practice for marking their buggies and wagons; however they do not utilize the practice for their low-profile vehicles, such as pony carts. Visibility for pony carts on public roads is important to protect the operators, typically women and children. Two 24 h BioBlitzes were conducted in September, 2018, and April, 2019, at Grassy Pond Recreational Area in Lowndes County, Georgia. Teams of scientists, students, and public participants surveyed plants, insects, amphibians and reptiles, birds, mammals, fungi, lichens, and aquatic microorganisms. Both events included educational programs with informational walks and activities targeted to youth. In the fall, 654 species and 55 additional genera were recorded. In the spring, there were 581 species and 54 additional genera recorded. Two fungi and 38 lichens were first reports for Lowndes County; one fungus was a first for Georgia. One lichen may be a …

The Church of Jesus Christ of Latter-day Saints has had a presence in Tonga since July of 1891, when Mormon missionaries, Brigham Smoot and Alva Butler arrived in Nuku’alofa on the island of Tongatapu.1 Although the Tongan mission was organized as early as 1916, the Church did not began to grow at a steady rate in Tonga until the 1920s. The first stake in Tonga was created in 1968, and the Nuku’alofa Tonga Temple was dedicated in 1983. However, where did Tongan Latter-day Saints attend the temple between from the 1920s until 1983?

Good People Don't Need Medication: How Moral Character Beliefs Affect Medical Decision-Making, Sydney Elizabeth Scott, Justin F. Landy

How do people make decisions? Prior research focuses on how people's cost-benefit assessments affect which medical treatments they choose. We propose that people also worry about what these health decisions signal about who they are. Across four studies, we find that medication is thought to be the "easy way out", signaling a lack of willpower and character. These moral beliefs lower the appeal of medications. Manipulating these beliefs--by framing medication as a signal of superior willpower or by highlighting the idea that treatment choice is just a preference--increases preferences for medication.

The author was the director or co-director of a specialty Deaf psychiatric inpatient unit for 17 of its 23 years. In Part 1 of this article, the author reflected on the lessons learned about deaf psychiatric patients from this experience. These lessons include recognition of the wide continuum of communication skills and deficits of the deaf persons served and the significant numbers of deaf patients who have poor communication skills in any language. The author also supported the hypothesis that many deaf psychiatric patients have a particular disorder involving language dysfluency, related mainly to language deprivation and an array of …
