Uncoupling The Folding And Binding Of An Intrinsically Disordered Protein, Anusha Poosapati, Emily Gregory, Wade M. Borcherds, Lucia B. Chemes, Gary Daughdrill Aug 2018

Uncoupling The Folding And Binding Of An Intrinsically Disordered Protein, Anusha Poosapati, Emily Gregory, Wade M. Borcherds, Lucia B. Chemes, Gary Daughdrill

Molecular Biosciences Faculty Publications

The relationship between helical stability and binding affinity was examined for the intrinsically disordered transactivation domain of the myeloblastosis oncoprotein, c-Myb, and its ordered binding partner, KIX. A series of c-Myb mutants was designed to either increase or decrease helical stability without changing the binding interface with KIX. This included a complimentary series of A, G, P, and V mutants at three non-interacting sites. We were able to use the glycine mutants as a reference state and show a strong correlation between binding affinity and helical stability. The intrinsic helicity of c-Myb is 21%, and helicity values of the …

Quantifying The Relations Among Neurophysiological Responses, Dimensional Psychopathology, And Personality Traits., Takakuni Suzuki Aug 2018

Quantifying The Relations Among Neurophysiological Responses, Dimensional Psychopathology, And Personality Traits., Takakuni Suzuki

Open Access Dissertations

The Research Domain Criteria (RDoC) model, one of the transdiagnostic dimensional models of psychopathology proposed by the National Institute of Mental Health, emphasizes the integration of basic biological science into clinical research. The event-related negativity (ERN), a deflection in electroencephalogram after making an error, is a promising neurophysiological indicator for such endeavor. However, RDoC model posits that ERN is related to three of the five broad dimensions without any indication of how ERN relates differently across the dimensions. Recent research indicate that ERNs elicited from different tasks do not elicit the exact same ERNs. Therefore, constructing theoretically guided tasks and …

Microbial Changes In Gene Expression Level In Response To Environmental Conditions In The Delaware Bay, Margi I. Patel Aug 2018

Microbial Changes In Gene Expression Level In Response To Environmental Conditions In The Delaware Bay, Margi I. Patel

All Theses

Bacteria dominate in abundance, diversity and potentially metabolic activity in many environments. Our current knowledge on the influence of specific individual taxa on these processes is largely lacking. To bridge these gaps, I chose three near complete metagenome-assembled genomes (MAGs) from the Delaware Bay, phylogenetically associated with the Roseobacter clade, to compare the functional potential of the MAGs to their closest relatives. I also characterized the relative activity of one MAG by using normalized gene expression levels and differential gene expression. The normalized number of transcripts per sample revealed whether or not specific genes/pathways were being expressed at the time …

Temporal And Ecological Community Dynamics Of Water-Cooling Tower Associated Legionella Spp., Emlyn Hammer Aug 2018

Temporal And Ecological Community Dynamics Of Water-Cooling Tower Associated Legionella Spp., Emlyn Hammer

All Theses

Man-made water systems can have complex microbial communities. Legionella spp. are ubiquitous in these environments and, with favorable conditions, can lead to outbreaks of Legionnaires disease. Included in these water systems are free living amoebas (FLA) and potential other eukaryotes that may act as a reservoir for Legionella survival and replication. Associations with these hosts can play a role in Legionnaires' disease outbreaks. A water cooling tower in Aikens, SC associated with L. pneumophila was measured every month for environmental parameters (temperature, bromine (Br), chlorine (Cl), pH, and dissolved oxygen). Water and sediment samples were collected every week for six …

Biocementation Of Simulant Martian Regolith, Jason Gleaton

All Theses

Here on Earth biocementation has been extensively studied and holds promise in producing a more energy efficient and environmentally benign alternative construction materials compared to the standard method of production. This research is focused on developing bioprocesses to produce bioconcrete columns using potentially Mars-compatible microalgae and simulated Martian regolith. A marine microalga, Thraustochytrium striatum was tested to make Martian regolith-based columns in the presence of CaCl2/urea.

Three different biogrouting methods were investigated including simultaneous, sequential and batch circulation of microalga cell biomass and CaCl2/urea in the columns. The need of post-biogrouting column soaking was studied to …

Using Snake Genomes To Illuminate The Patterns And Mechanisms Of Rapid Adaptation, Daren Carter Card Aug 2018

Using Snake Genomes To Illuminate The Patterns And Mechanisms Of Rapid Adaptation, Daren Carter Card

Biology Dissertations - Archive

One of the most important and interesting goals in evolutionary biology is to understand the mechanisms generating biodiversity and adaptive novelty. Ever-evolving genomic techniques have served as a catalyst for this work, enabling rapid increases in our knowledge of diverse taxa. By leveraging a combination of phylogeographic, population genetic, and comparative genomic methods, I established two snake systems with unique attributes that showed promise for increasing our understanding of important evolutionary questions related to local adaptation and convergence. Using sampling from several island populations of Boa imperator with similar adaptive phenotypes (e.g., reduced body size and craniofacial morphological shifts), I …

A New Approach For Performance Evaluation Of Bridge Infrastructure Using Terrestrial Lidar And Advanced Mathematical Modeling, Ali Shafikhani Aug 2018

A New Approach For Performance Evaluation Of Bridge Infrastructure Using Terrestrial Lidar And Advanced Mathematical Modeling, Ali Shafikhani

Civil Engineering Dissertations - Archive

High plastic expansive clays when subjected to different climatic conditions undergo large ground movements causing distress to infrastructures including bridges, pavements, buildings, retaining structures, and others. Performance assessment of these structures built on problematic soils such as expansive clays is important to reduce maintenance and extending the design life of infrastructure. Rapid developments in remote sensing technologies with precise evaluation have influenced the monitoring techniques for assessing the health condition of civil infrastructure projects. While these technologies have considerably aided in performance evaluation, cogent procedures for evaluating the ground movements are still required that integrates technologies, climatic factors, soil behavior …

Chronic Cadmium Exposure Alters Erα Dependency And Drug Sensitivity Of Breast Cancer Cells, Mathew Bloomfield Aug 2018

Chronic Cadmium Exposure Alters Erα Dependency And Drug Sensitivity Of Breast Cancer Cells, Mathew Bloomfield

Dissertations, Masters Theses, Capstones, and Culminating Projects

The global prevalence of breast cancer in women illustrates the importance of identifying factors that contribute to disease onset and progression. Endogenous and environmental agents that interact with estrogen receptor alpha (ERα) have been shown to play a role in breast cancer etiology. Evidence from epidemiological studies and animal models has suggested that cadmium, a heavy metal that can activate ERα, contributes to the development and progression of breast cancer. Additionally, our lab showed that chronic cadmium exposure altered the expression of several ERα-responsive genes and increased the malignancy of MCF7 breast cancer cells. Although these studies support cadmium’s function …

Best Practices In A Clinical Development Project Management Office (Pmo) To Achieve A Reference Standard In The Pharmaceutical Industry, Russell Guinto Aug 2018

Best Practices In A Clinical Development Project Management Office (Pmo) To Achieve A Reference Standard In The Pharmaceutical Industry, Russell Guinto

Dissertations, Masters Theses, Capstones, and Culminating Projects

Medications are developed by the pharmaceutical industry starting with the discovery phase, proceeds to preclinical trials, moves into clinical trials (progressing from Phase I to Phase III), and if the data are positive, may lead to Food and Drug Administration (FDA) approval. Once approved, post-marketing surveillance for safety is required as long as the drug is marketed to consumers. This phase may also include clinical trial Phase IV studies if additional safety testing is required. This process usually takes between ten to fifteen years, with clinical development taking seven to ten years of that time (1). Clinical development can …

Detecting A Probiotic Product Within The Gut Of Broiler Chickens, Anneka Pisula Aug 2018

Detecting A Probiotic Product Within The Gut Of Broiler Chickens, Anneka Pisula

Master's Theses

As of January 2017, the U.S. poultry industry banned the use of antibiotics and now relies on alternatives such as probiotics to help protect animal health. Although probiotic use is not a new concept in the poultry industry, identifying the best combination of bacterial strains to generate an effective probiotic formula requires further investigation. This study aimed to detect a probiotic product of four bacterial strains (Pedioccoccus acidilactici, Pediococcus pentosaceus, Lactobacillus plantarum, and Bacillus subtilis) in a feeding trial with broiler chickens. Birds given the probiotic were predicted to show an improved growth performance with …

The Roles Of Tid1, Ndj1, And Spo16 In Distributive Segregation During Saccharomyces Cerevisiae Meiosis, Ethan Atticus Shaw Aug 2018

The Roles Of Tid1, Ndj1, And Spo16 In Distributive Segregation During Saccharomyces Cerevisiae Meiosis, Ethan Atticus Shaw

Master's Theses

Meiosis is a specialized form of cell division in sexually reproducing eukaryotes. Crossovers are physical connections formed between homologous chromosomes during meiosis; these connections help ensure normal segregation of homologous chromosomes at meiosis I. However, the yeast Saccharomyces cerevisiae and other eukaryotes can still segregate homologs properly even in the absence of some crossovers. This is due to a backup mechanism known as distributive segregation, which correctly segregates non-crossover chromosomes at a higher rate than if segregation were completely random. To study distributive segregation, we have generated diploid yeast with one homeologous chromosome pair consisting of a Saccharomyces cerevisiae chromosome …

Antimicrobial Efficacy And Safety Of A Novel Gas Plasma-Activated Catheter Lock Solution, Sudhir Bhatt, Poonam Mehta, Chen Chen, Dayle A. Daines, Leonard A. Mermel, Hai-Lan Chen, Michael G. Kong

Bioelectrics Publications

Antimicrobial lock solutions are important for prevention of microbial colonization and infection of long-term central venous catheters. We investigated the efficacy and safety of a novel antibiotic-free lock solution formed from gas plasma-activated disinfectant (PAD). Using a luminal biofilm model, viable cells of methicillin-resistant Staphylococcus aureus, Staphylococcus epidermidis, Pseudomonas aeruginosa, and Candida albicans in mature biofilms were reduced by 6 to 8 orders of magnitude with a PAD lock for 60 min. Subsequent 24-h incubation of PAD-treated samples resulted in no detectable regrowth of viable bacteria or fungi. As a comparison, the use of a minocycline-EDTA-ethanol lock solution for 60 …

Bee Assemblages Data, Bernadette Maria Mach, Daniel A. Potter Aug 2018

Bee Assemblages Data, Bernadette Maria Mach, Daniel A. Potter

Entomology Research Data

Urban and suburban landscapes can be refuges for biodiversity of bees and other pollinators. Public awareness of declining pollinator populations has increased interest in growing plants that provide floral resources for bees. Various publications and websites list “bee-friendly” plants, but such lists are rarely based on empirical data, nor do they emphasize flowering trees and shrubs, which are a major component of urban landscapes. We quantified bee visitation to 72 species of flowering woody landscape plants across 373 urban and suburban sites in Kentucky and southern Ohio, USA, sampling and identifying the bee assemblages associated with 45 of the most …
