Disclosed are various embodiments for transferring molecules from a surface for mass spectrometry and other sample analysis methods, and the like. A laser is focused onto a tip of an atomic force microscope to remove and capture a quantity of molecules from the surface, so they can be transferred to a mass spectrometer or another instrument for analysis.

Novel electrospun nanofibers and nanofibrous membranes, methods of manufacturing the same, and methods of using the same are provided. The nanofibers include a cactus mucilage, such as mucilage from Opuntia ficus-indica. An organic polymer can be added to the cactus mucilage before electrospinning. The nanofibrous membranes can be used in water filtration.

Background: We investigated interactions between genetic and psychological factors in predicting shoulder impairment phenotypes. We hypothesized that pro-inflammatory genes would display stronger relationships compared with pain-related genes when combined with psychological factors for predicting phenotypic changes.
Subjects and methods: Altogether, 190 participants completed a 5-day experimental protocol. An experimental shoulder injury model was used to induce physical impairment, and a priori selected genetic (pain-related, pro-inflammatory) and psychological (anxiety, depressive symptoms, pain catastrophizing, fear of pain, kinesiophobia) factors were included as predictors of interest. Impairment phenotypes were injury-induced deficits in range of motion (ROM) and strength. After controlling for age, sex, …

Myocardial toxicity and drug resistance caused by drug efflux are major limitations of doxorubicin (Dox)-based chemotherapy. Dox structure modification could be used to develop conjugates with an improved biological profile, such as antiproliferative activity and higher cellular retention. Thus, Dox thiol conjugates, Dox thiol (Dox-SH), thiol-reactive Dox-SS-pyridine (SS = disulfide), and a Dox-SS-cell-penetrating cyclic peptide, Dox-SS-[C(WR)4K], were synthesized. Dox was reacted with Traut's reagent to generate Dox-SH. The thiol group was activated by the reaction with dithiodipyridine to afford the corresponding Dox-SS-Pyridine (Dox-SS-Pyr). Crassostrea virginica, the eastern oyster, forms reefs that provide critical services and benefits to the resiliency of the surrounding ecosystem. Changes in environmental conditions, including salinity and temperature, can dramatically alter the services oysters provide by affecting their population dynamics. Climate warming may further exacerbate the effects of salinity changes as precipitation events increase in frequency, intensity, and duration. Temperature and salinity independently and synergistically influence gene expression and physiology in marine organisms. A three-dimensional, high-resolution, Finite-Volume Coastal Ocean Model (FVCOM) was used to study the dynamics of Barataria Estuary located in the Southeastern Louisiana. Three numerical experiments with different discharge scenarios, including the actual discharge (average ~ 160 m3 s-1) from the Davis Pond Diversion (DPD) over three months from April to June 2010, no discharge (NO), and the proposed Mid-Barataria Diversion (MBD) with a constant discharge of 850 m3 s-1, were conducted to investigate the impacts of river diversions on salinity gradients and residence times in the estuary. Sugarcane, being a complex aneu-polyploid, poses unique challenges to fine mapping of quantitative trait loci (QTL) controlling agronomic traits of interest. Genome-wide association studies (GWAS) could be a better alternative to identify molecular markers associated with specific traits due to high linkage disequilibrium in sugarcane. In the first study, marker-trait associations (MTAs) were evaluated for three sucrose traits, Brix, total recoverable sugar (TRS), and percent sucrose on 48 elite and historic Louisiana breeding clones using 1,062 alleles. These sucrose traits were highly correlated (P-value < 0.0001) at >0.96. TASSEL 5.1 and JMP Genomics 8.0 were compared with eight models run in each program. …

The effects of herbivory and chemical applications as potential inducers of resistance to herbivores in rice, Oryza sativa L., were explored through a series of field, greenhouse, and laboratory experiments. Studies were conducted to investigate the preference and performance of several herbivore pests of rice with a focus on, Lissorhoptrus oryzophilus Kuschel, the rice water weevil (RWW), under conditions where rice plants had been stressed by previous herbivory or chemical applications. The effects of defoliation by Spodoptera frugiperda (J.E. Smith), the fall armyworm, on resistance and tolerance of rice to RWW under field conditions were examined. Herbivory by fall …

Ceria is an earth-abundant material that has been widely used in heterogeneous catalysis, environmental catalysis, and energy applications thanks for its ability to readily convert between different oxidation states. The objective of this study is to theoretically elucidate the reaction mechanisms for the conversion of model organic compounds on ceria, in order to gain insights for the design of cost-effective and selective ceria-based catalysts. Acetaldehyde, acetic acid, and para-nitrophenyl phosphate monoester were selected as the model compounds to probe ceria surfaces. Avian and mammalian predation of Ashy Storm-Petrels (Oceanodroma homochroa) were documented during monthly nest monitoring in 1995–1998 and 2005–2015 and once-per-year monitoring in 1999–2004 at 5 colonies at Santa Cruz Island, California. Throughout the study period, predation by Barn Owls (Tyto alba) was documented at the sea cave colonies, and at Orizaba Rock, and likely contributed to reduced breeding success at Bat Cave in 1995–1997. Recent predation by Common Ravens (Corvus corax) caused reduced breeding success each year in 2012–2015 at Orizaba Rock (35%–48%) and in 2013 and 2015 at Bat Cave (49%–52%). Raven …

The western United States remains well below historical wildfire activity, yet misconceptions abound in the public and news media that the area burning by wildfire each year in the American West is unprecedented. We submit that short‐term records of wildfire and a disproportionate focus on recent fire trends within high‐profile science stoke these misconceptions. Furthermore, we highlight serious risks to long‐term water security (encompassing water supply, storage, and quality) that have only recently been recognized and are underestimated as the result of skewed perspectives of wildfire. Cellular actin assembly is controlled at the barbed ends of actin filaments, where capping protein (CP) limits polymerization. Twinfilin is a conserved in vivo binding partner of CP, yet the significance of this interaction has remained a mystery. Here, we discover that the C-terminal tail of Twinfilin harbors a CP-interacting (CPI) motif, identifying it as a novel CPI-motif protein. Twinfilin and the CPI-motif protein CARMIL have overlapping binding sites on CP. Further, Twinfilin binds competitively with CARMIL to CP, protecting CP from barbed-end displacement by CARMIL. Twinfilin also accelerates dissociation of the CP inhibitor V-1, restoring CP to an active …

Historical ecological research provides valuable insights for contemporary conservation management. Gaps in historical records, however, can limit the utility of that research. Future conservationists may therefore find themselves disadvantaged by the current societal trend of underinvestment in systematic collection of museum specimens and natural history information. To reduce that risk, we asked what managers and scientists could do today to better document the past and present conditions of Santa Cruz Island, California, as a means to improve both contemporary and future conservation. Marine crabs inhabit shallow coastal/estuarine habitats particularly sensitive to climate change, and yet we know very little about the diversity of their responses to environmental change. We report the effects of a rarely studied, but increasingly prevalent, combination of environmental factors, that of near-future pCO2 (~1000 µatm) and a physiologically relevant 20% reduction in salinity. We focused on two crab species with differing abilities to cope with natural salinity change, and revealed via physiological and molecular studies that salinity had an overriding effect on ion exchange in the osmoregulating shore crab, Carcinus maenas. The Rh blood group system (ISBT004) is the second most important blood group after ABO and is the most polymorphic one, with 55 antigens encoded by 2 genes, RHD and RHCE This research uses next-generation sequencing (NGS) to sequence the complete RHD gene by amplifying the whole gene using overlapping long-range polymerase chain reaction (LR-PCR) amplicons. The aim was to study different RHD alleles present in the population to establish reference RHD allele sequences by using the analysis of intronic single-nucleotide polymorphisms (SNPs) and their correlation to a specific Rh haplotype. OAPA A depth estimation algorithm based on vergence vision using a mechanical joint attached to two cameras is proposed. A Gaussian pyramid template-matching approach is used to align the view of the slave camera to the fixation point of the master camera. The master camera uses an object detection algorithm to find the target’s centroid and centers it relative to the image coordinates. Then, vergence movement of the slave camera is performed using a pyramid normalized cross-correlation algorithm. Simple geometric triangulation is employed to compute the depth of that target. This Article expands a previously published article, which introduced a novel problem to the centuries-old debate on the retributivist justification of punishment. The first article applied the problem of conjoined twins, where one commits a crime and the other is innocent, to pure retributivism. The conjoined twins problem showed that pure retributivism, which holds absolute duties to punish all who are guilty and none who are innocent, fails as a complete theory of punishment.
