Abstract: Mind as Spirit in the World of Culture. Philosophy of Culture, "Naturalistic" Transcendental Philosophy and the Question of the Space of Culture. In this essay I present Cassirer's philosophy of culture as a special application of Kant's transcendental philosophy. I then compare and contrast it to another tradition, namely the Pittsburgh School, especially McDowell. The starting point here is the Sellarsian concept of the 'Space of reasons', which McDowell expands through his Aristotelian-inspired concept of 'Second nature'. Cryo-electron microscopy (cryo-EM) is a structure determination method for large molecular complexes. As more and more atomic structures are determined using this technique, it is becoming possible to perform statistical characterization of side-chain conformations. Two data sets were involved to characterize block lengths for each of the 18 types of amino acids. One set contains 9131 structures resolved using X-ray crystallography from density maps with better than or equal to 1.5 Å resolutions, and the other contains 237 protein structures derived from cryo-EM density maps with 2-4 Å resolutions. We evaluated transgenic lines of sugarcane modified with the barley chitinase class-II gene to create resistance against the red rot causative agent Colletotrichum falcatum Went. Local sugarcane cultivar SP93 was transformed with a 690-bp coding sequence of the chitinase-II gene under the influence of a polyubiquitin promoter. Transgenic sugarcane lines (T$_{0}$) overexpressing the chitinase gene were obtained through a particle bombardment method with 13.3% transformation efficiency. Four transgenic sugarcane lines, SCT-03, SCT-05, SCT-15, and SCT-20, were tested for resistance against red rot by in vitro antifungal assays. Breast cancer is the most common cancer among women and the molecular pathways that play main roles in breast cancer regulation are still not completely understood. MicroRNAs (miRNAs) and transcription factors (TFs) are important regulators of gene expression. It is important to unravel the relation of TFs, miRNAs, and their targets within regulatory networks to clarify the processes that cause breast cancer and the progression of it. Lipophilic Antioxidants In Edible Weeds From Agricultural Areas, Helena Sircelj, Maja Mikulic Petkovsek, Robert Veberic, Metka Hudina, Ana Slatnar

The present paper reports the contents of individual lipophilic antioxidants in fourteen species of edible common agricultural weeds, typical of agricultural areas such as fields and orchards. Young edible green aboveground parts of weeds were analyzed for their chlorophyll, carotenoid, and tocopherol qualitative profiles and contents. To the best of our knowledge, this is the first report on the complete lipophilic antioxidant composition of the edible weeds examined in this study. The results revealed that all examined leafy plant species are good sources of lipophilic antioxidants, the richest source being Urtica dioica (255.64 mg 100 $g^{-1}$ fwt), followed by Cardamine …

Insect pests are among the major constraints rendering drastic decreases in crop yield. The expression of stacked insecticidal genes in crops can lead to resistance durability and can delay the development of resistance in target insect pests. The present study was designed to introduce an insect resistance trait in locally cultivated Turkish tobacco cultivars (Basma and Nail) with pyramided insecticidal genes. Glyphosate is a widely used herbicide that inhibits the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS)-encoding aroA gene in the shikimate pathway. The discovery and cloning of the aroA gene with high resistance is central to breeding a transgenic glyphosate-resistant plant. A novel aroAPantoea gene from Pantoea G-1 was previously isolated and cloned. The aroAPantoea enzyme was defined as a new class I EPSPS with glyphosate resistance. The aroAPantoea gene was introduced into tobacco through Agrobacterium-mediated transformation. The transgenic tobacco plants were confirmed by PCR, RT-PCR, and Southern blot. Escherichia coli is an extensively used host for the production of recombinant proteins, making its N-terminal methionine aminopeptidase (MAP) an attractive candidate for studies on posttranslational protein processing. The present study describes the recombinant production and properties of MAP from the DH5α strain of E. coli. The soluble and active enzyme was produced in E. coli BL21 (DE3) RIL - codon plus cells under a T7 promoter system and purified by anion-exchange chromatography. Inhibiting ceramidase activity in cancer cells has been identified as a promising target for cancer therapy in recent studies. Thus, we examined the possible role of ceranib-2, a novel ceramidase inhibitor, on growth and apoptotic mechanisms of the human normal glia cell line (HNA), human glioma cell lines (T-98G and U-87MG), and a rat glioma cell line (C6). Effects of drought stress on growth, predawn xylem water potential (ψpd), osmotic solutes (soluble sugar and proline), and stomatal conductance were assessed in two oak (Quercus cerris L. and Quercus robur L.) seedlings. Seedlings of both species were subjected to three drought treatments with the following irrigation intervals: well-watered (control: irrigation every 2-3 days), moderate drought stress (irrigation every 15 days), and severe drought stress (irrigation every 30 days). Drought-stressed seedlings of Q. cerris and Q. robur had more negative predawn xylem water potential than their well-watered seedlings. A carboxylesterase isolated from Aeromonas caviae MTCC 7725 was immobilized by entrapping it in chitosan coated calcium alginate beads. This was characterized by scanning electron microscopy (SEM), energy dispersive X-ray spectroscopy (EDX), and Fourier transform infrared spectroscopy (FTIR). The activity of the native and immobilized enzyme was measured at various temperatures, pH levels, and organic solvents. The optimum temperature for activity of the native enzyme was found to be 40 °C and this increased to 50 °C on immobilization. The immobilized enzyme showed enhanced stability and high residual activity in various organic solvents as compared to the free enzyme. An …

A number of differentially expressed proteins (DEPs) were identified in comparative two-dimensional gel electrophoresis analysis of dry and 24-h water-imbibed seeds of maize F1 hybrid DHM 117 (BML 6 x BML 7) and its parental inbreds. Of the DEPs, 53.4% (86/161) in dry seeds and 58% (127/219) in water-imbibed seeds exhibited a nonadditive pattern in the F1 hybrid as compared to parental inbreds. The mammalian centrosome/cilium complex is composed of the centrosome, the primary cilium, and the centriolar satellites,which together function in key cellular processes including signaling. Defective assembly, maintenance, and function of the centrosome/cilium complex cause the human genetic diseases known as ciliopathies, which are characterized by a multitude of developmental syndromes including retinal degeneration and kidney cysts. Pepper fruit (Capsicum annuumL.) is one of the best sources of carotenoids, carbohydrates, flavonoids, phenols, and antioxidant activity important in the human diet. The levels of bioactive compounds are variable and may be affected by cultivar, maturity stage, and boron concentration in the soil solution. A greenhouse experiment was conducted with the pepper cultivars Solario, Osho, Odysseo, and Arlequin treated with 5 boron concentrations (0.5, 1, 2.5, 5, and 10 mg B L-1) for 70 days. Antibiotic resistance is one of the most important problems of our era and hence the discovery of new effective therapeutics is urgent. At this point, studying the allosteric communication pathways in the bacterial ribosome and revealing allosteric sites/residues is critical for designing new inhibitors or repurposing readily approved drugs for this enormous machine. Isoflavones prevent the incidence of cancer, reduce cardiovascular problems, and decrease menopausal symptoms. In soybean (Glycine soja Sieb. & Zucc.) seeds, the content is greatly affected by growing conditions. The variations in soy-isoflavone content were investigated by growing 346 Korean wild soybean accessions from natural habitats under the same agroclimatic conditions. High-performance liquid chromatography was used to measure the profiles of six soy-isoflavones, i.e. glycosides (daidzin, glycitin, genistin) and malonyl glycosides (malonyl-daidzin, malonyl-glycitin, and malonyl-genistin). The soy-isoflavone content of the wild soybeans ranged from 0.5 to 5.51 mg/g with an average of 2.78 mg/g. The highest mean total soy-isoflavone was …

This 2-year field study investigated the effects of different wheel traffic passes, manure amounts, and manure application methods on soil temperature, soil moisture, CO2 emission, and soil O2 content. To achieve this purpose, three different wheel traffic applications (no traffic, one pass, and two passes) were used. In the experiments, two different methods of manure applications (surface and subsurface) and three different farmyard manure amounts were used with a control plot (N0), 40 Mg ha-1 (N40), and 80 Mg ha-1 (N80). The genetic potential of accessions from Solanum section Lycopersicon (S. lycopersicum L., S. lycopersicum var. cerasiforme, S. pimpinellifolium L., and S. habrochaites Knaap & Spooner) for breeding tomato taste has been studied in three environments with clonal replicates. The environment clearly affected the accumulation and level of variation of sugars and acids and derived variables through a direct effect. It seems that photosynthetically active radiation would exert a major effect on sugar accumulation while in the case of organic acids the effect of temperature might be more important. The effect of altitude, shooting period, and tea grade on the catechins and caffeine contents of Turkish black tea was investigated. The experimental factors significantly affected all of the parameters analyzed. Epigallocatechin gallate was the major catechin of black tea (0.719-1.007 g 100 g-1 dm) followed by epigallocatechin (0.627-0.791 g 100 g-1 dm). Additionally, caffeine (1.810-2.175 g 100 g-1 dm), theaflavin (3.201-6.000 μmol g-1), and thearubigin (8.577-12.635 g 100 g-1 dm) contents were determined. Efficiency in fruit trees is connected with flower quantity and quality. Flower abortions occur largely depending on locations and years in apricot varieties that are selective in terms of climate requirements. In this study carried out with varieties having a wide range of chilling requirements in İzmir and Malatya, flower and leaf bud drop rates differed according to varieties and was found low in Malatya. During the flower bud and bloom period, Tokaloğlu, Precoce de Tyrinthe, and Iğdır varieties did not show anomalies in Malatya while Precoce de Tyrinthe had a low rate of abnormality in İzmir. In this study, the effect of field dodder (Cuscuta campestris Yunck.) on the leaf and tuber yield of sugar beet fields in Kahramanmaraş Province of Turkey was investigated. Several therapeutic products based on mesenchymal stem cells (MSCs) have been translated into clinical applications. MSCs should undergo in vitro culture before a sufficient quantity can be achieved. Hence, both expansion kinetics and the biological characteristics of derived cells from primary culture are pertinent to their applications. In the present study, MSCs were isolated from rat bone marrow and adipose tissue (designated as bMSCs and aMSCs, respectively) and cells were comparatively analyzed regarding cell morphology, proliferation, colony formation, differentiation potential, and immunophenotype following the long-term subculture. There is no consensus on the properties of an ideal dressing for treating wounds. The aim of this study was to investigate the efficacy of dressings using topically administered bitter melon extract with olive oil, pure olive oil, nitrofurazone, and saline in the healing of ischemic wounds. A sample group of 48 rats was used in the trial. Their wounds were treated with bitter melon extract, pure olive oil, nitrofurazone, and saline.
