User-Based Photometer Analysis Of Effluent From Advanced Nitrogen-Removal Onsite Wastewater Treatment Systems, Bianca N. Ross, George W. Loomis, Kevin P. Hoyt, José A. Amador

Natural Resources Science Faculty Publications

Advanced nitrogen-removal onsite wastewater treatment systems (OWTS) are used to reduce total nitrogen (N) levels in domestic wastewater. Maintaining system performance requires regular monitoring and in situ rapid tests can provide an inexpensive option for assessing treatment performance. We used a portable photometer to measure ammonium and nitrate concentrations in final effluent from 46 advanced N-removal OWTS, sampling each site at least three times in 2017. To assess photometer accuracy, we compared measurements made using the photometer with those determined by standard laboratory methods using linear regression analysis and a two-tailed t test to compare regression parameters to those for …

Comparing Fishers' And Scientists' Estimates Of Size-At-Maturity And Maximum Body Size As Indicators Of Overfishing, Elizabeth Layli Mclean, Graham E. Forrester Jan 2018

Comparing Fishers' And Scientists' Estimates Of Size-At-Maturity And Maximum Body Size As Indicators Of Overfishing, Elizabeth Layli Mclean, Graham E. Forrester

Natural Resources Science Faculty Publications

We tested whether fishers’ local ecological knowledge (LEK) of two fish life-history parameters, size at maturity (SAM) at maximum body size (MS), was comparable to scientific estimates (SEK) of the same parameters, and whether LEK influenced fishers’ perceptions of sustainability. Local ecological knowledge was documented for 82 fishers from a small-scale fishery in Samaná Bay, Dominican Republic, whereas SEK was compiled from the scientific literature. Size at maturity estimates derived from LEK and SEK overlapped for most of the 15 commonly harvested species (10 of 15). In contrast, fishers’ maximum size estimates were usually lower than (eight species), or overlapped …

HSV-1 causes a life-long infection in its host and has evolved multiple strategies to facilitate infection and evade the immune response. This virus has been found to enter cells by both endocytosis and fusion. The way the virus exploits endocytosis is not fully understood. Recent studies have uncovered roles of Rab GTPases, key regulators in intracellular membrane trafficking pathways, in distinct steps of the HSV-1 life cycle (Raza et al., 2018). This study will focus on analyzing the levels of the late endosomal regulator Rab7 expression in macrophages infected with HSV-1. Revealing the effect of virus on the levels of …

Faculty & Staff Scholarship

Forward genetics remains a powerful method for revealing the genes underpinning organismal form and function, and for revealing how these genes are tied together in gene networks. In maize, forward genetics has been tremendously successful, but the size and complexity of the maize genome made identifying mutant genes an often arduous process with traditional methods. The next generation sequencing revolution has allowed for the gene cloning process to be significantly accelerated in many organisms, even when genomes are large and complex. Here, we describe a bulked-segregant analysis sequencing (BSA-Seq) protocol for cloning mutant genes in maize. Our simple strategy can …

Assessment Of A Mouse Xenograft Model Of Primary Colorectal Cancer With Special Reference To Perfluorooctane Sulfonate, Jeffrey H. Wimsatt, Caitlin Montgomery, Laurel S. Thomas, Charity Savard, Rachel Tallman, Kim Innes, Nezar Jrebi

Faculty & Staff Scholarship

Colorectal cancer ranks third among the most commonly diagnosed cancers in the United States. Current therapies have a range of side effects, and the development of a reliable animal model to speed the discovery of safe effective preventative therapies would be of great value. A cross-sectional study in a large Appalachian population recently showed an association between low circulating levels of perfluorooctane sulfonate (PFOS) and a reduced prevalence of colorectal cancer. A study using APCmin(C57BL/6J-ApcMin/J) mice prone to familial adenomatous polyposis found PFOS was protective when exposure occurred during tumor development. To test the possible benefit of PFOS on spontaneous …

Genetic Assessment Of Inbred Chicken Lines Indicates Genomic Signatures Of Resistance To Marek's Disease, Lingyang Xu, Yanghua He, George E. Liu, Huanmin Zhang, Hans H. Cheng, Robert L. Taylor Jr, Jiuzhou Song

Faculty & Staff Scholarship

Background: Marek’s disease (MD) is a highly contagious pathogenic and oncogenic disease primarily affecting chickens. However, the mechanisms of genetic resistance for MD are complex and not fully understood. MD-resistant line 63 and MD-susceptible line 72 are two highly inbred progenitor lines of White Leghorn. Recombinant Congenic Strains (RCS) were developed from these two lines, which show varied susceptibility to MD.

Results: We investigated genetic structure and genomic signatures across the genome, including the line 63 and line 72, six RCSs, and two reciprocally crossed flocks between the lines 63 and 72 (F1 63 × 72 and F1 72 × …

Evaluating The Effectiveness Of Soil-Native Bacteria In Precipitating Calcite To Stabilize Expansive Soils, Bhaskar C. S. Chittoori, Malcolm Burbank, Md Touhidul Islam

Civil Engineering Faculty Publications and Presentations

The use of chemical additives to stabilize expansive soils is a common practice. However, the environmental concerns associated with the greenhouse gas generation during the production of these chemicals have launched engineers in search of sustainable stabilization alternatives. Microbial induced calcite precipitation (MICP) is a bio-cementation technique that could be a potential solution to this problem. Typically, MICP is achieved via bio-augmentation; however, bio-stimulation was argued to be a more realistic alternative due to its field implementation potential. Hence, in this research study, two expansive soils with varying plasticity characteristics were examined to understand the potential of MICP in treating …

Bioinformatic Workflows For Generating Complete Plastid Genome Sequences—An Example From Cabomba (Cabombaceae) In The Context Of The Phylogenomic Analysis Of The Water-Lily Clade, Michael Gruenstaeudl, Nico Gerschler, Thomas Borsch

Biological Sciences Faculty Publications

The sequencing and comparison of plastid genomes are becoming a standard method in plant genomics, and many researchers are using this approach to infer plant phylogenetic relationships. Due to the widespread availability of next-generation sequencing, plastid genome sequences are being generated at breakneck pace. This trend towards massive sequencing of plastid genomes highlights the need for standardized bioinformatic workflows. In particular, documentation and dissemination of the details of genome assembly, annotation, alignment and phylogenetic tree inference are needed, as these processes are highly sensitive to the choice of software and the precise settings used. Here, we present the procedure and …

Plant Sciences and Entomology Faculty Publications

Invasive Phragmites australis is widespread in North America and despite decades of management and large annual expenditures (>5 million US$) using physical and chemical means, local populations and the species range are expanding. Allowing continued expansion does not only threaten native wetland biota but also an endemic North American subspecies Phragmites australis americanus. We used extensive multi-pronged investigations in Europe and North America to evaluate host specificity and impact of two European stem mining noctuid moths, Archanara geminipuncta and A. neurica. Both moth species are specific to the genus Phragmites and both show a very strong, but …

Finding Multiple Roots Of Systems Of Nonlinear Equations By A Hybrid Harmony Search-Based Multistart Method, Gisela C. V. Ramadas, Edite M. G. P. Fernandes, Ana Maria A. C. Rocha

Applied Mathematics & Information Sciences

A multistart (MS) clustering technique to compute multiple roots of a system of nonlinear equations through the global optimization of an appropriate merit function is presented. The search procedure that is invoked to converge to a root, starting from a randomly generated point inside the search space, is a new variant of the harmony search (HS) metaheuristic. The HS draws its inspiration from an artistic process, the improvisation process of musicians seeking a wonderful harmony. The new hybrid HS algorithm is based on an improvisation operator that mimics the best harmony and uses the idea of a differential variation, borrowed …

An Inverse Problem For Delay Differential Equations: Parameter Estimation, Nonlinearity, Sensitivity, Fathalla A. Rihan, Abdulla A. Azamov, Hebatallah J. Al-Sakaji

Applied Mathematics & Information Sciences

This article presents the theoretical framework to solve inverse problems for Delay Differential Equations (DDEs). Given a parameterized DDE and experimental data, we estimate the parameters appearing in the model, using least squares approach. Some issues associated with the inverse problem, such as nonlinearity and discontinuities which make the problem more ill-posed, are studied. Sensitivity and robustness of the models to small perturbations in the parameters, using variational approach, are also investigated. The sensitivity functions may provide guidance for the modelers to determine the most informative data for a specific parameter, and select the best fit model. The consistency of …

Existence Results For Impulsive Fractional Neutral Integro-Differential Equations With Nonlocal Conditions, A. Sivasankari, A. Leelamani, Dumitru Baleanu

Applied Mathematics & Information Sciences

We present a new prospective of the existence of a mild solution for impulsive fractional neutral integro-differential equations with nonlocal conditions in Banach spaces. In light of ideas for Banach contraction principle and Krasnoselskii-Schaefer’s fixed point theorem, we build up the existence results with resolvent operator and η -norm. At last, a case is given to represent the acquired outcomes .

Department of Educational Psychology: Faculty Publications

Genomic variation underlying major depressive disorder (MDD) likely involves the interaction and regulation of multiple genes in a network. Data-driven co-expression network module inference has the potential to account for variation within regulatory networks, reduce the dimensionality of RNA-Seq data, and detect significant geneexpression modules associated with depression severity. We performed an RNA-Seq gene co-expression network analysis of mRNA data obtained from the peripheral blood mononuclear cells of unmedicated MDD (n = 78) and healthy control (n = 79) subjects. Across the combined MDD and HC groups, we assigned genes into modules using hierarchical clustering with a dynamic …
