Computational Analysis Of Broad Complex Zinc-Finger Transcription Factors, Barbara Mascareno-Shaw
Computational Analysis Of Broad Complex Zinc-Finger Transcription Factors, Barbara Mascareno-Shaw
The Pegasus Review: UCF Undergraduate Research Journal
Studies of transcription factors have provided important contributions toward understanding the cell morphology and developmental regulation of fruit flies based on sequence analysis of amino acid substitutions. In this report we describe the construction of four protein-DNA models based on two experimentally determined X-ray crystal structures (2DRP and 1A1H) using computational simulations of Z1, Z2, Z3, and Z4 zinc-finger DNA binding proteins. Once these models were constructed, 3 mutants were generated for the Z1/Z3 proteins. We evaluated the construction of the models and mutants against the templates using molecular mechanics (MM3) calculations in an attempt to determine the effect of …

Microtubule-Actin Crosslinking Factor 1 And Plakins As Therapeutic Drug Targets, Quincy A. Quick
Microtubule-Actin Crosslinking Factor 1 And Plakins As Therapeutic Drug Targets, Quincy A. Quick
Biology Faculty Research
Plakins are a family of seven cytoskeletal cross-linker proteins (microtubule-actin crosslinking factor 1 (MACF), bullous pemphigoid antigen (BPAG1) desmoplakin, envoplakin, periplakin, plectin, epiplakin) that network the three major filaments that comprise the cytoskeleton. Plakins have been found to be involved in disorders and diseases of the skin, heart, nervous system, and cancer that are attributed to autoimmune responses and genetic alterations of these macromolecules. Despite their role and involvement across a spectrum of several diseases, there are no current drugs or pharmacological agents that specifically target the members of this protein family. On the contrary, microtubules have traditionally been targeted …

Kruppel-like factor 4 (Klf4) is a zinc-finger-containing protein that plays a critical role in diverse cellular physiology. While most of these functions attribute to its role as a transcription factor, it is postulated that Klf4 may play a role other than transcriptional regulation. Here we demonstrate that Klf4 loss in neural progenitor cells (NPCs) leads to increased neurogenesis and reduced self-renewal in mice. In addition, Klf4 interacts with RNA-binding protein Staufen1 (Stau1) and RNA helicase Ddx5/17. They function together as a complex to maintain NPC self-renewal. We report that Klf4 promotes Stau1 recruitment to the 3′-untranslated region of neurogenesis-associated mRNAs, …

”It Tells All Of Our History!” The Boi-Bumbá Festival Of Manaus, Marnie K. Watson
College of Humanities and Public Affairs
This article examines the discourse on modernity and nostalgia in Manaus, the capital of the Brazilian state of Amazonas, with a focus on the boi-bumbá festival, a folkloric dramatic dance that tells the story of the death and resurrection of a rancher’s beloved bull. Although most of the population lives in urban areas and engages in wage labor, the region is still popularly imagined as a “place that time forgot”: a natural area largely free of modern human intervention. This paper discusses how the boi-bumbá of Amazonas rose to stratospheric popularity in the context of urbanization, industrialization, political change, and …
Rare Parthenogenic Reproduction In A Common Reef Coral, Porites Astreoides, Alicia A. Vollmer
Rare Parthenogenic Reproduction In A Common Reef Coral, Porites Astreoides, Alicia A. Vollmer
HCNSO Student Theses and Dissertations
Multiple stressors have caused a decline in coral populations. Broadcast spawning corals once dominated the Florida Reef Tract (FRT), but since their decline, smaller brooding corals, soft corals, and macroalgae are replacing them. Brooding corals are more resilient to current threats in part because they are reproductive throughout much of the year and their larvae are competent to settle after release. Despite the ubiquity of brooders on Florida reefs, much of their reproductive strategy remains unknown. This study aimed to examine paternity as a function of colony size and density in Porites astreoides, a common brooding coral in the …

The Río Chiriqui basin is one of 52 major watershed areas, or cuencas, in the Republic of Panama. It occurs in western Panama, discharges into the Pacific Ocean, and includes portions of Volcán Barú on its northern extent. The Río Caldera occupies the northernmost subbasin of this basin. Two of its tributaries, Quebrada Grande and Quebrada Jaramillo, occur in close proximity and on opposite sides, and have different recent geologic histories and current land use patterns. During the course of investigating the caddisfly fauna of these two drainages, three new species of the microcaddisfly genus Neotrichia Morton (Trichoptera: Hydroptilidae) were …

An early, overlooked description of the genus Atelodesmis (Coleoptera, Cerambycidae, Lamiinae) is attributed to Chevrolat (in Duponchel and Chevrolat 1841), with A. mannerheimii Duponchel and Chevrolat, 1841 as its type species, and the genus redescribed. Atelodesmis Buquet, 1857, is a junior synonym and primary homonym of Atelodesmis Chevrolat. Atelodesmis hirticornis Buquet, 1857 and A. vestita Buquet, 1857 are synonymized with A. mannerheimii. Fallaxdesmis is described as a new genus with Atelodesmis unicolor Buquet, 1857 as type species. Atelodesmis piperita Bates, 1855 is transferred to Eupogonius and newly recorded for the state of Oaxaca, Mexico. A new species for Mexico …

© 2018 The Author(s). Background: Formation of nucleosomes along eukaryotic DNA has an impact on transcription. Major transcriptional changes occur in response to low external phosphate (Pi) in plants, but the involvement of chromatin-level mechanisms in Pi starvation responses have not been investigated. Results: We mapped nucleosomes along with transcriptional changes after 24-h of Pi starvation in rice (Oryza sativa) by deep sequencing of micrococcal nuclease digested chromatin and ribosome-depleted RNA. We demonstrated that nucleosome patterns at rice genes were affected by both cis- and trans-determinants, including GC content and transcription. Also, categorizing rice genes by nucleosome patterns across the …

We documented the first record of the non-native species Piaractus brachypomus (Characiformes: Serrasalmidae) in Lago Petén-Itzá, Petén, Guatemala. One brined specimen was donated by a local fisherman in San Benito, El Petén. The captured specimen was reportedly collected in the southern basin of the lake. We used the mitochondrial gene cytochrome oxidase I (COI) to validate identity of the specimen. We hypothesize that the local aquarium trade is the most likely source of introduction of the non-native Pacu in Lago Petén-Itzá.

Analyses of long-term ecosystem dynamics offer insights into the conditions that have led to stability vs. rapid change in the past and the importance of disturbance in regulating community composition. In this study, we (1) used lithology, pollen, and charcoal data from Mallín Casanova (47°S) to reconstruct the wetland, vegetation, and fire history of west-central Patagonia; and (2) compared the records with independent paleoenvironmental and archeological information to assess the effects of past climate and human activity on ecosystem dynamics. Pollen data indicate that Nothofagus-Pilgerodendronforests were established by 9,000 cal yr BP. Although the biodiversity of the understory increased …
An Examination Of Factors That Affect The Utilization Of Mental Health Services By Adolescents, Nichole A. Murray
An Examination Of Factors That Affect The Utilization Of Mental Health Services By Adolescents, Nichole A. Murray
USF Tampa Graduate Theses and Dissertations
Mental disorders among adolescents are on the rise and are among the most common chronic conditions, constituting a national epidemic. Prevalence rates indicate that 20–40% of the adolescent population in the U.S. suffers from emotional or behavioral difficulties significant enough to lead to functional impairment. Further, estimates suggest that more than 20% of the adolescent population has a diagnosable mental disorder, which significantly impacts their functioning substantially at school, at home and in their communities. Despite national agendas to address the mental health needs of adolescents, studies suggest that approximately 50 percent of adolescents with mental health needs do not …

Optimal Wavelength Selection For Optical Spectroscopy Of Hemoglobin And Water Within A Simulated Light-Scattering Tissue, Mikael Marois, Stephen L. Jacques, Keith D. Paulsen
Dartmouth Scholarship
An algorithm that selects optimal wavelengths for spectral fitting of diffuse light reflectance spectra using a nonnegative least squares method is presented. Oxyhemoglobin, deoxyhemoglobin, and water are considered representative absorbers, but the approach is not constrained or limited by absorber selection provided native basis spectra are available. The method removes wavelengths iteratively from a scattering-modulated absorption matrix by maximizing the product of its singular values and offers considerable improvements over previously published wavelength selection schemes. Resulting wavelength selections are valid for a broad range of optical properties and yield lower RMS errors than other wavelength combinations. The method is easily …

Background: Breast cancer is intrinsically heterogeneous and is commonly classified into four main subtypes associated with distinct biological features and clinical outcomes. However, currently available data resources and methods are limited in identifying molecular subtyping on protein-coding genes, and little is known about the roles of long non-coding RNAs (lncRNAs), which occupies 98% of the whole genome. lncRNAs may also play important roles in subgrouping cancer patients and are associated with clinical phenotypes. Methods: The purpose of this project was to identify lncRNA gene signatures that are associated with breast cancer subtypes and clinical outcomes. We identified lncRNA gene signatures …

Identification Of Individuals For Directorship Roles: Evaluation Of A University’S Succession Management, Nana Yaw Oppong, Nancy Oduro-Asabere
The Qualitative Report
Identification of potential individuals for leadership roles is a critical aspect of a succession management programme, as other aspects of the programme depend on an effective identification. This study evaluates how the University of Cape Coast (UCC) in Ghana identifies potential non-academic senior members for directorship roles. We collected qualitative data through in-depth interviewing of nine directors at the university. We analysed the data using constant comparison analysis by developing three themes, under each of which we presented similar categories of data. We found that the criteria for identification of potential directors include seniority; both internal and external sources; and …

Evaluation Of Experimental Populations And Glandular-Haired Varieties Of Alfalfa For Alfalfa Blotch Leafminer (Diptera: Agromyzidae) Feeding Injury, R. C. Venette, W. D. Hutchison, E. C. Burkness, C. C. Sheaffer
The Great Lakes Entomologist
Following the spread of the alfalfa blotch leafminer, Agromyza frontella (Rondani) (Diptera: Agromyzidae), into Minnesota and Wisconsin U.S.A. during 1994-1997, three field trials were conducted in Minnesota to assess the potential for leafminer resistance among several sources of alfalfa (Medicago sativa), germplasm. In 1998, 86 entries were evaluated, most of which were experimental populations. In addition, six commercial varieties of alfalfa were evaluated. Of the six varieties, four had been bred for various levels of glandular-hair expression, specifically for resistance to the potato leafhopper, Empoasca fabae (Harris) (Homoptera: Cicadellidae). In two of three trials, we found no significant differences …
