Intratumoral Sting Activation Sensitizes Poorly Immunogenic Cancers To Checkpoint Blockade Immunotherapy, Casey Ager

Unprecedented durable tumor regression can be achieved in patients with checkpoint blockade immunotherapy, yet these responses are rare and are not observed in all tumor types. Castration resistant prostate cancer (CRPC) and pancreatic adenocarcinoma (PDAC) are two common lethal cancers that remain refractory to checkpoint blockade, requiring development of new therapeutic approaches to promote responsivity in patients with these diseases. Both CRPC and PDAC are characterized by an immunosuppressive tumor milieu rich in cells of the myeloid lineage, which regulate T cell activity in many ways independent of the coinhibitory receptor axis. Technological innovations have produced large multi-modal datasets that range in multiplatform genomic data, pathway data, proteomic data, imaging data and clinical data. Integrative analysis of such data sets have potentiality in revealing important biological and clinical insights into complex diseases like cancer. This dissertation focuses on Bayesian methodology establishment in integrative analysis of radiogenomics and pathway driver detection applied in cancer applications. Over one billion of the world’s population are smokers, with increasing tobacco use in low-and middle-income countries. However, information about the methodology of studies on tobacco control is limited. We conducted a literature search to examine and evaluate the methodological designs of published tobacco research in Sub-Saharan Africa (SSA) over the past 50 years. The first phase was completed in 2015 using PubMed, Embase, CINAHL, and Cochrane Central Register of Controlled Trials. An additional search was completed in February 2017 using PubMed. Tardive dyskinesia (TD) is a side effect of antipsychotic medications used to treat schizophrenia (SCZ) and other mental health disorders. No study has previously used pyrosequencing to quantify DNA methylation levels of the DLGAP2 gene; while the quantitative methylation levels among CpG sites within a gene may be correlated. To deal with the correlated measures among three CpG sites within the DLGAP2 gene, this study analyzed DNA methylation levels of the DLGAP2 gene using a linear mixed model (LMM) in a Chinese sample consisting of 35 SCZ patients with TD, 35 SCZ without TD (NTD) and 34 healthy controls (HCs) …

Introduction: Penile transplantation, in its infancy, has the potential to reestablish functional outcomes for men with penile loss and disfigurement. However, significant bioethical considerations are pertinent, and systematic discussions are necessary to safely progress implementation. Aim: To determine the attitude of health practitioners toward the penile transplant and identify the key aspects of concern pertinent to the operation and clinical care. Methods: Health care professionals from the United States responded to either email invitation, web link, or social media post on Facebook to complete a questionnaire investigating perceptions and attitudes toward penile transplantation. PURPOSE: To examine the influence of three distinct training phases on isometric mid-thigh pull (IMTP) measures in well-trained weightlifters. METHODS: Pre- and post-block IMTP data from 11 collegiate weightlifters was used for analysis. The mean of the best two attempts from each athlete for measures of PF and RFD from 0-50ms, 0-100ms, 0-150ms, 0-200ms, and 0-250ms were used for comparison. In total, results from five timepoints for each of the 11 athletes were examined in order to compare the effects of the three training phases. RESULTS: A repeated measures ANOVA revealed no statistically significant (p ≥ 0.05) effects of training …

African Wild Dogs (Lycaon pictus) are an endangered species of canid from Sub-Saharan Africa. They are very social communal hunters, and are capable of chasing down prey for long stretches of time. Wild dogs benefit a savannah ecosystem by regulating the populations of their prey so that it does not become unhealthy and overgrown. Like many organisms, wild dogs are also in competition with other predators for resources, namely lions and hyenas, who are capable of stealing their kills and occasionally injuring and killing them. Women’s entry into the legal academy in significant numbers—first as students, then as faculty—was a 1970s and 1980s phenomenon. The photocatalytic degradation of simazine (SIM) by Zinc oxide/graphene oxide (ZnO/GO) composites, under visible light irradiation, was investigated to determine various reaction parameters, of initial pH, ZnO loading on GO, and catalyst mass. A pH of 2 was determined to be the optimal reaction pH for all ZnO/GO catalysts. A loading mass of 40mg for the 20 & 30mmol composites; whereas a 10mg mass was determined to be most effective for the 10mmol composite. The reaction followed second order kinetics for the degradation process, the reaction dependence was determined to be on the concentration of SIM in solution. All ZnO/GO …

The Gulf of Mexico provides substantial resources along its 1,680-mile coastline, spanning 5 U.S. states and providing millions of pounds of seafood and hundreds of millions in revenue annually. Continued success of the fishery relies on the ability of a large, complex, multiagency network to find the best solutions to balance the needs of humans, as well as ensure long-term sustainability. Data from the Hispanic Association of Colleges and Universities asserts that increasing the number of Hispanics that graduate is vital for our country’s future. The purpose of this research is to utilize publicly available data to estimate the current rate of occurrence of undiagnosed High Blood Pressure (HBP) in young adults (18 to 24 years old) and the out-of-pocket healthcare expenses associated with complications of HBP, to provide feasible alternatives to mitigate the effects on a region's economy. The Sylmar earthquake of 1971 caused significant damage to slender, non-ductile reinforced concrete (RC) shear wall buildings in California. A later survey by the Concrete Coalition in 2011, under the guidance of EERI members, indicated that there are over 3000 vulnerable concrete buildings in California [8]. This led to City of Los Angeles (LA) Ordinance 193893 enacted in 2015, which requires mandatory upgrades to these concrete buildings by 2035. Traditionally post-earthquake structural engineering reconnaissance consists of a team of experts who are deployed to the field to record and capture earthquake damage data, which is later uploaded into online repositories. Despite many advances to these data archives in recent years, the entries in online repositories often have limited metadata which make it difficult and time consuming to extract specific damage evidence that can be used for meaningful analysis. This report outlines the author’s contributions to overcoming these challenges via the development of a neural network that automatically filters and classifies post-earthquake civil infrastructure damage data after a seismic event. …

IN THE CURRENT ERA of the #MeToo movement and prominent sexual assault disclosures, these types of crimes are finally receiving the attention they deserve. Our society is acknowledging what research has been telling us for years about the high incidence of sexual violence in the United States. According to a recent Centers for Disease Control and Prevention (CDC) report, more than 36% of women have experienced contact sexual violence in their lifetime and 19% have experienced attempted or completed rape. Investigating The Association Between Fantasy Proneness And Emotional Distress: The Mediating Role Of Cognitive Coping Strategies, Am Bacon, Jj Charlesford

Fantasy proneness (FP; the tendency to immersion in imagination) is linked to psychopathology and suggested to be a maladaptive coping strategy. However, some evidence suggests it can be a positive trait. We examined whether the FP-emotional distress relationship is mediated by coping strategy. Participants completed the Depression, Anxiety and Stress Scale, the Cognitive Emotion Regulation Questionnaire measure of coping strategies, the Creative Experiences Questionnaire FP scale (Study 1; N=248) and the newly developed Fantasy Questionnaire which measures fantasy across two subscales, creative and imaginative (Study 2; N=208). In Study 1, FP presented positive associations with emotional distress, with maladaptive coping …

Using data from the World Bank and the World Values Survey, I use a linear regression model to assess how attitudes toward education, jobs, and over-all equal rights impact women's account ownership.

Environmental health issues are complex and require interdisciplinary and community engagement approaches to better understand them and inform policy. As one example, lead exposure has a number of dangerous neurological effects, including developmental delays and learning deficits. Potential lead exposure through drinking water and paint are areas of concern. In a 2018 document, the Environmental Protection Agency (EPA) estimated that up to 20% of lead exposure comes from drinking water alone. PURPOSE OF REVIEW: A wide variety of triggers prompt attacks in episodic migraine. Although experimental triggers such as glyceryl trinitrate reliably produce migraine, natural triggers are much less predictable and vary in importance between individuals. This review describes the most common triggers in episodic migraine and provides strategies for managing them in clinical practice.
RECENT FINDINGS: Multiple migraine attack triggers have been established based on patient surveys, diary studies, and clinical trials. Stress, menstrual cycle changes, weather changes, sleep disturbances, alcohol, and other foods are among the most common factors mentioned. Focal electrical stimulation of the brain incites a cascade of neural activity that propagates from the stimulated region to both nearby and remote areas, offering the potential to control the activity of brain networks. Understanding how exogenous electrical signals perturb such networks in humans is key to its clinical translation. To investigate this, we applied electrical stimulation to subregions of the medial temporal lobe in 26 neurosurgical patients fitted with indwelling electrodes. Networks of low-frequency (5-13 Hz) spectral coherence predicted stimulation-evoked increases in theta (5-8 Hz) power, particularly when stimulation was applied in or adjacent to white matter. In amyotrophic lateral sclerosis (ALS), mitochondrial dysfunction and oxidative stress form a vicious cycle that promotes neurodegeneration and muscle wasting. Irisin, a muscle-origin protein derived from the extracellular domain of the fibronectin domain-containing 5 protein (FNDC5), has been shown to modulate mitochondria welfare through paracrine action. Here, we test the hypothesis that irisin contributes to cardioprotection after myocardial infarction by preserving mitochondrial function in cardiomyocytes. Animal model studies show that intravenous administration of exogenous irisin produces dose-dependent protection against ischemia/reperfusion (I/R)-induced injury to the heart as reflected by the improvement of left ventricular ejection fraction and the reduction in serum level of cTnI (n = 15, P < 0.05). I/R-induced apoptosis of cardiomyocytes is reduced after irisin treatment. The irisin-mediated protection has, at least in part, an effect on mitochondrial function because administration of irisin increases irisin staining in the mitochondria of the infarct area. Irisin also reduces I/R-induced oxidative stress as determined by mitochondrial membrane potential evaluation and superoxide FLASH event recording (n = 4, P < 0.05). The interaction between irisin and superoxide dismutase2 (SOD2) plays a key role in the protective process because irisin treatment increases SOD activity (n = 10, P < 0.05) and restores the mitochondria localization of SOD2 in cardiomyocytes (n = 5, P < 0.05). These results demonstrate that irisin plays a protective role against I/R injury to the heart. Targeting the action of irisin in mitochondria presents a novel therapeutic intervention for myocardial infarction.
