Liberation Of Recalcitrant Cell Wall Sugars From Oak Barrels Into Bourbon Whiskey During Aging, Jarrad Wade Gollihue, Mitchell D. Richmond, Harlen Wheatley, Victoria G. Pook, Meera Nair, Isabelle A. Kagan, Seth Debolt

Horticulture Faculty Publications

Oak barrels have been used by humans for thousands of years to store and transport valuable materials. Early settlers of the United States in Kentucky began charring the interior of new white oak barrels prior to aging distillate to create the distinctively flavored spirit we know as bourbon whiskey. Despite the unique flavor and cultural significance of "America's Spirit", little is known about the wood-distillate interaction that shapes bourbon whiskey. Here, we employed an inverse method to measure the loss of specific wood polysaccharides in the oak cask during aging for up to ten years. We found that the structural …

Markey Cancer Center Faculty Publications

Collagen prolyl 4-hydroxylase (P4H) expression and collagen hydroxylation in cancer cells are necessary for breast cancer progression. Here, we show that P4H alpha 1 subunit (P4HA1) protein expression is induced in triple-negative breast cancer (TNBC) and HER2 positive breast cancer. By modulating alpha ketoglutarate (α-KG) and succinate levels P4HA1 expression reduces proline hydroxylation on hypoxia-inducible factor (HIF) 1α, enhancing its stability in cancer cells. Activation of the P4HA/HIF-1 axis enhances cancer cell stemness, accompanied by decreased oxidative phosphorylation and reactive oxygen species (ROS) levels. Inhibition of P4HA1 sensitizes TNBC to the chemotherapeutic agent docetaxel and doxorubicin in xenografts and patient-derived …

Metabolic Reprogramming Of Murine Cardiomyocytes During Autophagy Requires The Extracellular Nutrient Sensor Decorin., Maria A. Gubbiotti, Erin L. Seifert, Ulrich Rodeck, Jan B. Hoek, Renato V. Iozzo

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

The extracellular matrix is a master regulator of tissue homeostasis in health and disease. Here we examined how the small, leucine-rich, extracellular matrix proteoglycan decorin regulates cardiomyocyte metabolism during fasting in vivo. First, we validated in Dcn-/- mice that decorin plays an essential role in autophagy induced by fasting. High-Throughput metabolomics analyses of cardiac tissue in Dcn-/- mice subjected to fasting revealed striking differences in the hexosamine biosynthetic pathway resulting in aberrant cardiac O-β-N-Acetylglycosylation as compared with WT mice. Functionally, Dcn-/- mice maintained cardiac function at a level comparable with nonfasted animals whereas fasted WT mice showed …

Numerical Analysis Of Leakage Through Defective Geomembrane Liners In Embankment Dams, Sarper Demirdogen Oct 2018

Numerical Analysis Of Leakage Through Defective Geomembrane Liners In Embankment Dams, Sarper Demirdogen

USF Tampa Graduate Theses and Dissertations

Placing a geomembrane liner in the core of a dam is an alternative construction technique to traditional clay core types. This study aims to assess the performance of such internal geomembrane sealing systems in an earthen dam. Two-dimensional (2D) numerical analysis was performed to evaluate leakage through defective seams within an earthen dam. Five possible applications of internal geomembrane systems were initially modeled to locate the zero-pressure lines in an earthen dam. Then, another application where the geomembrane is placed on the upstream face was modeled to compare the upstream and internal geomembrane systems. The results of this study show …

From The River To The Gulf: An Investigation Of Biogeochemical Cycling In Wetland Soils And Coastal Shelf Sediments, Katie Bowes Oct 2018

From The River To The Gulf: An Investigation Of Biogeochemical Cycling In Wetland Soils And Coastal Shelf Sediments, Katie Bowes

LSU Master's Theses

Louisiana is home to 40% of the coastal wetlands in the lower 48 states, yet accounts for 80% of the coastal wetland loss in this region. This loss is attributed to decreased sediment supply, levee alteration, sea level rise, channelization, and subsidence. The levee system in Louisiana disconnected coastal wetlands from the main stem of the Mississippi River (MSR), reducing the amount of land-building sediment that reaches coastal wetlands. This disconnection also allows a greater percentage of river discharge, including agricultural contaminants and runoff, to flow into the northern Gulf of Mexico (NGOM).

The 2012 Coastal Master Plan proposed eight …

Diffusion Through The Ex Vivo Vitreal Body – Bovine, Porcine, And Ovine Models Are Poor Surrogates For The Human Vitreous, Sara Shafaie, Victoria Hutter, Marc B. Brown, Michael T. Cook, David Y.S. Chau

Pharmacology and Animal Models in Research Collection

The human vitreous humour is a complex gel structure whose composition and physical properties can vary considerably from person to person and also change with age. To date, the viscoelastic properties of the human vitreous gel has not been thoroughly investigated and despite many years of intensive research, an ideal vitreous substitute remains a challenge. Understanding the physical structure and properties of the vitreous is of fundamental and therapeutic interest, providing a clear insight into diffusion and transport of administered ophthalmic drug molecules into the vitreous. A number of mammalian surrogates, mainly bovine, porcine and ovine vitreous humours have been …

Integrating Multiple Genetic Detection Methods To Estimate Population Density Of Social And Territorial Carnivores, Sean M. Murphy, Ben C. Augustine, Jennifer R. Adams, Lisette P. Waits, John J. Cox Oct 2018

Integrating Multiple Genetic Detection Methods To Estimate Population Density Of Social And Territorial Carnivores, Sean M. Murphy, Ben C. Augustine, Jennifer R. Adams, Lisette P. Waits, John J. Cox

Forestry and Natural Resources Faculty Publications

Spatial capture–recapture models can produce unbiased estimates of population density, but sparse detection data often plague studies of social and territorial carnivores. Integrating multiple types of detection data can improve estimation of the spatial scale parameter (σ), activity center locations, and density. Noninvasive genetic sampling is effective for detecting carnivores, but social structure and territoriality could cause differential detectability among population cohorts for different detection methods. Using three observation models, we evaluated the integration of genetic detection data from noninvasive hair and scat sampling of the social and territorial coyote (Canis latrans). Although precision of estimated density was …

Micronutrient Availability In Alternative Foods During Agricultural Catastrophes, David Denkenberger, Joshua M. Pearce

Civil and Architectural Engineering Faculty Research

Several catastrophes could block the sun, including asteroid/comet impact, super volcanic eruption, and nuclear war with the burning of cities (nuclear winter). Previous work has analyzed alternate food supplies (e.g., mushrooms growing on dead trees, bacteria growing on natural gas). This was shown to be technically capable of feeding everyone with macronutrients (protein, carbohydrates, and lipids) and minerals, although economics and politics remain uncertain. The present work analyzes vitamin availability in such alternative food scenarios. The vitamin content of various alternate foods is compared to the US recommended daily allowance (RDA) as well as the average requirement defined by the …

A Pokébalancing Act: The Management Of Japanese Cultural “Odor ” In Pokémon, Nicholas Raes Oct 2018

A Pokébalancing Act: The Management Of Japanese Cultural “Odor ” In Pokémon, Nicholas Raes

Theses and Dissertations

In this project, I analyzed two seasons of the Pokémon anime to discover patterns of cultural “odor” management employed by the show’s producers and editors. In the first season of Pokémon, the producers of the anime included many symbolic representations of Japanese cultural artifacts within the visual features of the show. However, between the Japanese and US versions of the season, most instances of Japanese linguistic text were either “scrubbed” from scenes entirely or replaced with English text. These techniques struck a careful balance between including and excluding Japanese specificity within the first season, subtly expressing to non-Japanese audiences the …

Enteric septicemia of catfish, columnaris disease and streptococcosis, caused by Edwardsiella ictaluri, Flavobacterium columnare and Streptococcus iniae, respectively, are the most common bacterial diseases of economic significance to the pond-raised channel catfish Ictalurus punctatus industry. Certain management practices are used by catfish farmers to prevent large financial losses from these diseases such as the use of commercial antibiotics. In order to discover environmentally benign alternatives, using a rapid bioassay, we evaluated a crude extract from the roots of muscadine Vitis rotundifolia against these fish pathogenic bacteria and determined that the extract was most active against F. columnare. Subsequently, several isolated …

The Introduced Argentine Ant (Linepithema Humile) On The California Channel Islands: Distribution And Patterns Of Spread, Christina L. Boser, Korie Merrill, Robert N. Fisher, Ida Naughton, David A. Holway

Western North American Naturalist

The Argentine ant (Linepithema humile) is a widespread and abundant introduced species that disrupts ecosystems throughout its introduced range. This invader was inadvertently introduced to Santa Catalina, San Clemente, Santa Cruz, and San Nicolas Islands at various points during the past century but currently appears to be absent from the remaining Channel Islands. Multiple spatially disjunct infestations on each invaded island individually range in size from <500 m2 to >427 ha and encompass a variety of habitats, including large areas that are entirely dominated by native perennial vegetation. The existence of multiple infestations on individual islands suggests that inadvertent …

Whereas VHL inactivation is a primary event in clear cell renal cell carcinoma (ccRCC), the precise mechanism(s) of how this interacts with the secondary mutations in tumor suppressor genes, including PBRM1, KDM5C/JARID1C, SETD2, and/or BAP1, remains unclear. Gene expression analyses reveal that VHL, PBRM1, or KDM5C share a common regulation of interferon response expression signature. Loss of HIF2α, PBRM1, or KDM5C in VHL-/-cells reduces the expression of interferon stimulated gene factor 3 (ISGF3), a transcription factor that regulates the interferon signature. Moreover, loss of SETD2 or BAP1 also reduces the ISGF3 level. Finally, ISGF3 is strongly tumor-suppressive in a xenograft …

The Role Of Forests In Securing Hydropower Needs Of Cambodia, Mohit Kaura Oct 2018

The Role Of Forests In Securing Hydropower Needs Of Cambodia, Mohit Kaura

USF Tampa Graduate Theses and Dissertations

Hydropower relies on protection of watersheds to regulate water and sediment yields. Deforestation accelerates the rate of soil erosion, thereby increasing the amount of river sediments heading to the dam's reservoir, decreasing the longevity of the dam. In Cambodia in particular, recent deforestation rates are among the largest on the planet, and forests are expected to disappear within the lifespan of proposed dams. The cost of protecting and restoring forested watersheds can be considered as an annual investment towards sustainable reservoir management and hydropower generation. A modeling framework is developed to estimate the sediment accumulation in reservoirs from deforestation-driven soil …
