Theses and Dissertations
Two vanadium(V) complexes were prepared for different projects. First the ONS tridentate ligand 14 was prepared according to literature procedure. This ligand was chosen to make the ONS dioxovanadium complex 15 for conjugation to a protein and to eventually elicit catalytic antibodies for the hydrolysis of the chemical weapon VX. Complex 15 was prepared in 65% overall yield and a full characterization was performed including X-ray diffraction. Initial screening performed at the Naval Research Laboratory indicated an IC50 of 84 nM for inhibition of a bacterial cholinesterase. It was also found to be 50% as effective as the control antiobiotic …
The Corrosion And Biofouling Characteristics Of Sealed Vs. Perforated Offshore Monopile Interiors: Experiment Design Comparing Corrosion And Environment Inside Steel Pipe, Monica M. Maher
Theses and Dissertations
This research addresses the need to improve on the existing methods for the corrosion control of the monopile interiors used to support wind powered turbines by incorporating a design that also enhances marine habitats and fisheries. Retrofitting monopile interiors with cathodic protection has been attempted on existing windfarms to mitigate corrosion, however, this can cause new problems including water acidification and hydrogen sulfide formation. Such chemistry changes can lead to unique localized corrosion concerns. This study investigated internal corrosion, chemistry and biofouling inside partially submerged hollow steel pipes. Watertight pipes with stagnant water inside were compared to pipes with holes …

Investigating Possible Scurvy In The Postclassic Maya Of Tipu, Emmalea Gomberg
Investigating Possible Scurvy In The Postclassic Maya Of Tipu, Emmalea Gomberg
Master's Theses
Although residing in a tropical environment, numerous researchers have posited the possibility of scurvy in the Maya in recent years. The historic population of Tipu in western Belize dating to AD 1541-1638 is one of the largest and best preserved Maya series, and was therefore evaluated for possible presence of scurvy since the diet appears typical of the pre-contact Petén populations. The temporal bones and greater wings of the sphenoid were analyzed for scorbutic activity in 143 individuals, along with periodontal disease and antemortem tooth loss to determine if there was a co-occurrence with scurvy.
Overall, 27% of the sample …
Pseudomonas/Brachypodium As A Model System For Studying Rhizosphere Plant Microbe Interactions Under Water Stress, Janiece Mcwilliams
Pseudomonas/Brachypodium As A Model System For Studying Rhizosphere Plant Microbe Interactions Under Water Stress, Janiece Mcwilliams
Master's Theses
In contrast to well-studied mechanisms of drought tolerance in plants, the interactions between plants and their microbiome during water stress are still poorly understood. This missing knowledge is crucial for the exploitation of beneficial microbial communities to improve the sustainability of agriculture under changing climatic conditions. The research described here bridged this gap by focusing on the molecular interactions between beneficial rhizobacterium Pseudomonas synxantha 2-79 and annual grass Brachypodium distachyon Bd21. 2-79 exemplifies a group of rhizobacteria associated with dryland wheat, while B. distachyon originates from the Middle East and has emerged as a model for valuable biomass, food, forage, …

Rethinking Service: Visual Arts At The Land-Grant University And In Extension Programming, Meghan L.E. Kirkwood
Journal of Extension
In 2014, land-grant institutions observed the centenary of the Smith-Lever Act, which established the Cooperative Extension program. Traditionally, Cooperative Extension has engaged in and disseminated findings from research in areas such as agricultural sciences and public policy, but rarely the visual arts, despite the prevalence of visual arts departments at land-grant institutions. Faculty in the Department of Visual Arts at North Dakota State University (NDSU) desired to know whether there was potential for collaboration with NDSU Extension. This article addresses findings from the resulting survey-based study and the implications of those findings.
Ensuring Food Safety As Demand For Improved Food System Efficiency Increases, Abigail Snyder, Kate Shumaker, Nicole Nelsen
Ensuring Food Safety As Demand For Improved Food System Efficiency Increases, Abigail Snyder, Kate Shumaker, Nicole Nelsen
Journal of Extension
Food security and safety issues have been central to Extension programming since its inception, but emerging concerns over the issues of food waste and system inefficiency have brought new challenges to the forefront. Social change ideas related to reducing food waste include use of secondary quality produce, date label elimination, repurposing of food scraps, and donation and gleaning of food. Implementation of these ideas often intersects with issues of food safety. Therefore, it is incumbent on Extension professionals nationwide to consider how best to use content and expertise to address modern food security, food waste, and food safety challenges.

Varying Dataset Resolution Alters Predictive Accuracy Of Spatially Explicit Ensemble Models For Avian Species Distribution, Claire M. Curry, Jeremy D. Ross, Andrea Contina, Eli S. Bridge
School of Integrative Biological & Chemical Sciences Faculty Publications
Species distribution models can be made more accurate by use of new “Spatiotemporal Exploratory Models” (STEMs), a type of spatially explicit ensemble model (SEEM) developed at the continental scale that averages regional models pixel by pixel. Although SEEMs can generate more accurate predictions of species distributions, they are computationally expensive. We compared the accuracies of each model for 11 grassland bird species and examined whether they improve accuracy at a statewide scale for fine and coarse predictor resolutions. We used a combination of survey data and citizen science data for 11 grassland bird species in Oklahoma to test a spatially …

A Survey And Use-Wear Analysis Of Wickliffe Thick Pottery In The Southeastern United States, Anthony P. Farace
Theses
The Wickliffe Thick pottery type, an unusual vessel with a globular body, thick wall, and funnel-like opening at the bottom, has been assumed to be related to salt production and/or juice pressing. The following project presents the results of a use-wear analysis in order to understand Wickliffe Thick’s possible uses demonstrating that past conclusions likely need revision. A systematic, macroscopic analysis of ceramic sherds from more than 20 Mississippian sites throughout Kentucky, Missouri, and Illinois are included in the study. Use-wear on the samples occur in a low frequency. Although other factors such as a white efflorescence, and Wickliffe Thick’s …

Venezuela: A Uniquely Senian Insight Into A Human Rights Crisis, Andrea I. Scheer
Brooklyn Journal of International Law
For over twenty decades, Venezuelan political leaders have blatantly disregarded their citizens’ human rights, leading to the downfall of Venezuela’s economy and democratic institutions, including severe food and medicine shortages, as well as staggering inflation rates. As a result, Venezuela provides a unique affirmation of the Capabilities Approach introduced by Professor Amartya Sen, which focuses not only on the freedoms that individuals possess, but also on what individuals are capable of doing as possessors of these freedoms. This Note seeks to use Sen’s Capabilities Approach to understand the nature and scope of Venezuela’s multidimensional crisis, arguing that a Senian approach …
Involvement Of Gdap1 In The Regulation Of Mtorc1 Activity In A Drosophila Mliv Model, Kristen Clemons
Involvement Of Gdap1 In The Regulation Of Mtorc1 Activity In A Drosophila Mliv Model, Kristen Clemons
Dissertations and Theses (Open Access)
The master regulator of metabolism and growth, mechanistic target of rapamycin complex 1 (mTORC1), is responsible for maintaining metabolic homeostasis by sensing nutrient and energy levels within the cell to promote or inhibit translation and autophagy accordingly. In the childhood neurodevelopmental disorder Mucolipidosis type IV (MLIV), mTORC1 activity is decreased. The underlying mechanism for reduced mTORC1 signaling in MLIV is poorly understood. The gene encoding ganglioside-induced differentiation associated protein 1 (GDAP1) is transcriptionally upregulated in MLIV. This project investigated the involvement of GDAP1 in MLIV disease pathology. Using the UAS/GAL4 system in an established MLIV Drosophila model, we …
A Compressed Sensing Approach To Detect Immobilized Nanoparticles Using Superparamagnetic Relaxometry, Sara Thrower
A Compressed Sensing Approach To Detect Immobilized Nanoparticles Using Superparamagnetic Relaxometry, Sara Thrower
Dissertations and Theses (Open Access)
Superparamagnetic relaxometry (SPMR) is an emerging technology that leverages the unique properties of biologically targeted superparamagnetic iron oxide nanoparticles to detect cancer. The use of ultra-sensitive sensors enables SPMR to detect tumors ten times smaller than current imaging methods. Reconstructing the distribution of cancer-bound nanoparticles from SPMR measurements is challenging because the inverse problem is ill posed. Current methods of source reconstruction rely on prior knowledge of the number of clusters of bound nanoparticles and their approximate locations, which is not known in clinical applications. In this work, we present a novel reconstruction algorithm based on compressed sensing methods that …
Behavioral Insights Into Nociceptor Function: A Systematic Approach To Understanding Postsurgical And Neuropathic Pain Mechanisms In Rats, Max Odem
Dissertations and Theses (Open Access)
Postsurgical and neuropathic pain are each clinically common, and often associated with ongoing pain. Ongoing pain has been linked to ongoing activity (OA) in human C-fiber nociceptors. Preclinical studies using rodent neuropathic models have concentrated on allodynia driven by OA generated in non-nociceptive Aβ fibers, but little attention has been paid to postsurgical pain in sham controls or to C-fiber nociceptor OA promoting ongoing pain.
Operant assays that reveal negative motivational and cognitive aspects of voluntary pain-related behavior may be particularly sensitive to pain-related alterations. In the mechanical conflict (MC) test, rodents can freely choose to escape from a brightly …

A Role For Epac1 And Epac2 In Nociceptor Hyperexcitability And Chronic Pain After Spinal Cord Injury, Samantha Berkey
Dissertations and Theses (Open Access)
Chronic pain is a major complaint of those living with spinal cord injury (SCI), affecting 65-80% of the SCI population, but the treatment options remain limited or non-existent. The cAMP sensor EPAC has previously been shown to play a key role in chronic inflammatory and neuropathic pain, though the contribution from each of its two main isoforms, EPAC1 and EPAC2, is unclear. Here I test the hypothesis that both EPAC1 and EPAC2 play a key role in the maintenance of persistent nociceptor hyperexcitability and chronic pain after SCI.
Using both a T9 SCI mouse model and a T10 SCI rat …
Adenylyl Cyclase Type 9: Regulation And Cardiac Function, Tanya A. Baldwin, Tanya A. Baldwin
Adenylyl Cyclase Type 9: Regulation And Cardiac Function, Tanya A. Baldwin, Tanya A. Baldwin
Dissertations and Theses (Open Access)
Abnormalities in cardiac stress signaling underlie a number of cardiovascular diseases (e.g. arrhythmias and heart failure). Cardiac stress signaling pathways normally integrate signals from the sympathetic nervous system to promote efficient contraction and relaxation under stress. Sympathetic control through β-adrenergic stimulation is propagated by adenylyl cyclase (AC). AC synthesizes cyclic AMP (cAMP), an important second messenger that initiates signaling pathways to modulate physiological and pathophysiological functions of the heart, including the activation of PKA and subsequent phosphorylation of ion channels, contractile machinery, and stress response proteins that enhance cardiac function. Alterations of cAMP signaling occur in the failing heart and …

IL-1α Blockade Reduces Immune Suppression in the Early Tumor Micro-Environment
Brenda Melendez, B.S.
Advisory Professor: Gregory Lizee, PhD.
Immunotherapy against melanoma has shown great promise in the clinic for treating advanced-stage patients. However, a major barrier against effective T cell mediated cytotoxicity is immunosuppression in the tumor micro-environment. It has been described that tumors secrete pro-inflammatory cytokines capable of modulating immune responses that favors the growth of tumor cells. Specifically, IL-1 plays a critical role in myeloid cell recruitment and activation, which can in turn inhibit T cell activity in vivo. Moreover, IL-1 is also known to up-regulate immune inhibitory …
Heme Peroxidase Hpx-2 Protects Caenorhabditis Elegans From Pathogens, Yi Liu
Heme Peroxidase Hpx-2 Protects Caenorhabditis Elegans From Pathogens, Yi Liu
Dissertations and Theses (Open Access)
Heme-containing peroxidases are important components of innate immunity. Many of them functionally associate with NADPH oxidase (NOX)/dual oxidase (DUOX) enzymes by using the hydrogen peroxide they generate in downstream reactions. In Caenorhabditis elegans, a ShkT-containing peroxidase, SKPO-1, and the dual oxidase, BLI-3, contribute to resistance against the human pathogen Enterococcus faecalis, likely functioning in conjunction with each other. However, the exact mechanism(s) by which peroxidases and NOX/DUOXs contribute to pathogen resistance remain unclear. Here, we demonstrated that another peroxidase, HPX-2 (Heme-PeroXidase 2), is required for resistance against some, but not all pathogens. Tissue specific …
A Flexible Service-Oriented Approach To Address Hydroinformatic Challenges In Large-Scale Hydrologic Predictions, Michael Antonio Souffront Alcantara
A Flexible Service-Oriented Approach To Address Hydroinformatic Challenges In Large-Scale Hydrologic Predictions, Michael Antonio Souffront Alcantara
Theses and Dissertations
Water security is defined as a combination of water for achieving our goals as a society, and an acceptable level of water-related risks. Hydrologic modeling can be used to predict streamflow and aid in the decision-making process with the goal of attaining water security. Developed countries usually have their own hydrologic models; however, developing countries often lack hydrologic models due to factors such as the maintenance, computational costs, and technical capacity needed to run models. A global streamflow prediction system (GSPS) would help decrease vulnerabilities in developing countries and fill gaps in areas where no local models exist by providing …
Characterization Of Sip68 For Its Role In Plant Stress Signaling, Saroj Chandra Lohani
Characterization Of Sip68 For Its Role In Plant Stress Signaling, Saroj Chandra Lohani
Electronic Theses and Dissertations
Glucosyltransferases catalyze the transfer of glucose molecules from an active donor to acceptor molecules and are involved in many plant processes. SIP68, a tobacco glucosyltransferase protein, is a SABP2-interacting protein. It was identified in a yeast two-hybrid screen using SABP2 as bait and tobacco proteins as prey. SABP2, converts methyl salicylate to salicylic acid (SA) as a part of the signal transduction pathways in SA-mediated defense signaling. Subcellular localization is a crucial aspect of protein functional analysis to assess its biological function. The recombinant SIP68 tagged with eGFP was expressed transiently in Nicotiana benthamiana and observed under confocal microscopy. Fluorescent …

Sip-428, A Sir2 Deacetylase Enzyme And Its Role In Biotic Stress Signaling Pathway, Bal Krishna Chand Thakuri
Electronic Theses and Dissertations
SABP2 (Salicylic Acid Binding Protein 2) plays a vital role in the salicylic acid signaling pathway of plants both regarding basal resistance and systemic acquired resistance against pathogen infection. SIP-428 (SABP2 Interacting Protein-428) is a Silent information regulator 2 (SIR2) like deacetylase enzyme that physically interacts with SABP2 in a yeast two-hybrid interaction and confirmed independently by a GST pull-down assay. We demonstrated that SIP- 428 is an NAD+ dependent SIR2 deacetylase enzyme. Transgenic tobacco plants silenced in SIP- 428 expression via RNAi showed enhanced basal resistance to microbial pathogens. Moreover, these SIP-428-silenced lines also exhibited a robust induction of …
Dreadd Targeting Projections From The Nucleus Accumbens To The Ventral Pallidum With Nicotine Self-Administration, Amanda Smith
Dreadd Targeting Projections From The Nucleus Accumbens To The Ventral Pallidum With Nicotine Self-Administration, Amanda Smith
Electronic Theses and Dissertations
Projections from the Nucleus Accumbens (NAc) to the Ventral Pallidum (VP) play a critical role in motivation and reward. Rewards and reward-associated cues are thought to alter this pathway by suppressing GABA release to the VP, however, the role of the NAc to VP pathway has never been investigated with regard to nicotine self-administration. We hypothesized that increasing GABAergic signaling from the NAc to the VP would decrease the reinforcing effects of nicotine. To increase GABA release, CRE dependent DREADD was expressed in the NAc of male rats. Administration of low dose clozapine activated the DREADD receptor and showed a …

Island Identities: Analysis Of The Vernacular Building Patterns On South Carolina Barrier Islands, Claire Elizabeth Bushemi
All Theses
Long a retreat for the wealthy and once Union staging areas during the Civil War, the barrier islands of South Carolina represent themselves as a unique beach experience. In the early years of the eighteenth century, builders on the islands utilized ocean winds to neutralize the effects of summer sun to make buildings ideal for recreation. The islands were then marketed as having unique identities and have developed connotations of individuality that endure today. This thesis explores the qualities that distinguish and identify these unique environments by examining and analyzing the physical fabric of a cross section of seven of …

Analysis Of Frozen Desserts Using Low-Temperature Scanning Electron Microscopy (Lt-Sem), Kinsey Elizabeth Macdonald
All Theses
Commercial vanilla ice cream and other frozen desserts from the United States were analyzed for ice crystal length using low-temperature scanning electron microscopy (LT-SEM). Average ice crystal length was determined using multiple micrographs of each sample/product. Out of the products tested, 11 out of 15 samples had an average ice crystal length above the consumer sensory threshold limit of 55 µm. Products containing stabilizers tended to have smaller average ice crystal lengths than products without stabilizers. With a few exceptions, lower fat products tended to have larger ice crystals because there was less fat to stabilize the ice crystals. Four …
