Gcn5 Impacts Fgf Signaling At Multiple Levels And Activates C-Myc Target Genes During Early Differentiation Of Embryoid Bodies, Li Wang Aug 2017

Gcn5 Impacts Fgf Signaling At Multiple Levels And Activates C-Myc Target Genes During Early Differentiation Of Embryoid Bodies, Li Wang

Dissertations and Theses (Open Access)

Precise control of gene expression during development is orchestrated by transcription factors, signaling pathways and co-regulators, with complex cross-regulatory events often occurring. Growing evidence has identified chromatin modifiers as important regulators for development as well, yet how particular chromatin modifying enzymes affect specific developmental processes remains largely unclear. Embryonic stem cells (ESCs) are self-renewing, pluripotent, and have the abilities to generate almost all cell types in adult tissues. The dual capacity of ESCs to self-renew and differentiate offers unlimited potential for studying gene regulation events at specific developmental stages in vitro that parallel developmental events during embryogenesis in vivo. …

Purpose: Pazopanib, a multireceptor tyrosine kinase inhibitor targeting primarily VEGFRs1–3, is approved for advanced soft tissue sarcoma (STS) and renal cell cancer. Downstream of VEGFR, trametinib is an FDA-approved MEK inhibitor used for melanoma. We hypothesized that vertical pathway inhibition using trametinib would synergize with pazopanib in advanced STS. Experimental Design: In an open-label, multicenter, investigator-initiated National Comprehensive Cancer Network (NCCN)-sponsored trial, patients with metastatic or advanced STS received pazopanib 800 mg and 2 mg of trametinib continuously for 28-day cycles. The primary endpoint was 4-month progression-free survival (PFS). Secondary endpoints were overall survival, response rate, and disease control rate. …

Frequency Sensitive Mechanism In Low-Intensity Ultrasound Enhanced Bioeffects, April D. Miller, Abdoulkadri Chama, Tobias M. Louw, Anuradha Subramanian, Hendrik J. Viljoen

Department of Chemical and Biomolecular Engineering: Faculty Publications

This study presents two novel theoretical models to elucidate frequency sensitive nuclear mechanisms in low-intensity ultrasound enhanced bioeffects. In contrast to the typical 1.5 MHz pulsed ultrasound regime, our group previously experimentally confirmed that ultrasound stimulation of anchored chondrocytes at resonant frequency maximized gene expression of load inducible genes which are regulatory markers for cellular response to external stimuli. However, ERK phosphorylation displayed no frequency dependency, suggesting that the biochemical mechanisms involved in enhanced gene expression is downstream of ERK phosphorylation. To elucidate such underlying mechanisms, this study presents a theoretical model of an anchored cell, representing an in vitro …

Impulsive Personality Dimensions Are Associated With Altered Behavioral Performance And Neural Responses In The Monetary Incentive Delay Task, Ruolei Gu, Yang Jiang, Seth Kiser, Chelsea L. Black, Lucas S. Broster, Yue-Jia Luo, Thomas H. Kelly

Behavioral Science Faculty Publications

Individual differences in dimensions of impulsivity personality including disinhibition and sensation seeking modulate approach responses to reinforcing stimuli, such as drugs and money. The current study examined the effects of monetary incentive on both behavioral performance and electrophysiological activity among individuals varying in disinhibition and sensation seeking. The monetary incentive delay (MID) task was completed under electroencephalogram (EEG) recording. Behavioral data showed that higher disinhibition and sensation-seeking were associated with lower performance accuracy. Event-related potential (ERP) data showed that high reinforcement cues elicited a larger late positive component (LPC) than other conditions among high disinhibition participants, indicating its strong emotional …

Msh1-Mediated Non-Genetic Phenotypic Variation In Plants: Prospects For Epigenetic Breeding In Sorghum Bicolor, Dikungwa Ketumile Aug 2017

Msh1-Mediated Non-Genetic Phenotypic Variation In Plants: Prospects For Epigenetic Breeding In Sorghum Bicolor, Dikungwa Ketumile

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Sorghum is the fifth most important cereal crop grown in the world, and is the primary staple food for many people in developing countries. Like other major crops, its improvement is constrained by depletion of genetic diversity within breeding germplasm. Investigated in this study is the unique creation of non-genetic phenotypic variation through altering MutS Homolog1 (MSH1), a plant-specific gene, and the potential it presents for sorghum improvement. Suppression of MSH1 results in non-genetic developmental reprogramming. The derived MSH1 memory lines, when used in crossing, result in heritable phenotypic variation that enhances plant vigor and agronomic performance. A …

Analysis Of Cereal Aphid Feeding Behavior And Transcriptional Responses Underlying Switchgrass–Aphid Interactions, Kyle G. Koch

Department of Entomology: Dissertations, Theses, and Student Research

Switchgrass, Panicum virgatum L., is a perennial warm-season grass that is a model species for the development of bioenergy crops. However, the sustainability of switchgrass as a bioenergy feedstock will require efforts directed at improved biomass yield under a variety of stress factors. The objectives of this research were to: (1) elucidate greenbug and yellow sugarcane aphid feeding behavior on resistant and susceptible switchgrasses using electronic penetration graphs (EPG), (2) define transcriptional changes before and during insect feeding through RNA-seq to identify candidate resistance genes in a hybrid switchgrass cultivar, and (3) utilize methods in RNA sequencing of insects to …

Beringian Biogeography: Patterns Of Intercontinental Dispersal And The Structuring Of A Holarctic Tapeworm Genus, Genevieve M. Haas Aug 2017

Beringian Biogeography: Patterns Of Intercontinental Dispersal And The Structuring Of A Holarctic Tapeworm Genus, Genevieve M. Haas

All NMU Master's Theses

The opening and closing of the Bering Land Bridge due to Pleistocene climate fluctuations facilitated the exchange of taxa between the Palearctic and Nearctic. While many studies have worked toward elucidating the role of Beringia in assembling northern faunas, relatively little work has focused on parasites. Here I examine the number and direction of transberingian colonization events within the Holarctic tapeworm genus, Arostrilepis Mas-Coma & Tenora, 1997. I performed maximum likelihood and multi-locus coalescent phylogenetic reconstructions using mitochondrial and nuclear DNA sequences. Biogeographic ancestral range estimations were conducted on the resulting species phylogeny. My systematic reconstructions reveal as many as …

Post-Activation Potentiation: Decay Or Fatigue Delay, Ryan L. Meidinger Aug 2017

Post-Activation Potentiation: Decay Or Fatigue Delay, Ryan L. Meidinger

All NMU Master's Theses

Post-activation potentiation has been shown to improve jumping performance and other ballistic activities. The improvements in performance have been attributed to four main mechanisms, but the most important mechanism to the current study is the improvement in neural activity that leads to greater levels of potentiation. Post-activation potentiation has been shown to be stimulated by a maximal activity, called a conditioning contraction, and can be used as a warm up. In studies that have not shown the effects of post-activation potentiation, the proposed reason is fatigue, but the interaction of post-activation potentiation and fatigue have not been thoroughly tested. The …

Basigin-2 Mediated Activation Of Erk1/2 Signaling In Human Glioblastoma Multiforme Cells, Erik R. Peterson Aug 2017

Basigin-2 Mediated Activation Of Erk1/2 Signaling In Human Glioblastoma Multiforme Cells, Erik R. Peterson

All NMU Master's Theses

Glioblastoma multiforme (GBM) is the most common malignant form of human brain cancer. GBM tumor cells overexpress the protein Basigin (Bsg) at the cell surface where it contributes to malignancy via stimulation of matrix metalloproteinase (MMP) expression in surrounding normal tissues, resulting in the degradation of the extracellular matrix (ECM) surrounding tumors, promoting remodeling of the tumor borders, stimulating growth. In work by Belton et al. (2008), human uterine endometrial cells treated with a recombinant form of human basigin possessing the extracellular domain of the Bsg protein (rBsg-ECD) showed activation of the Mitogen-Activated Protein Kinase (MAPK) signaling pathway proteins, ERK1/2. …

Go For A Walk! Monitoring Walking For Depressed Mood, Michael E. Shrake Aug 2017

Go For A Walk! Monitoring Walking For Depressed Mood, Michael E. Shrake

All NMU Master's Theses

Literature suggests that exercise has benefits for health and mood. For people suffering from depression however, even summoning motivation to go for a walk can seem impossible, no matter the alleged benefits. My study builds on current articles suggesting that wearable technology like activity-tracking bracelets can have significant effects on motivation and physical health goals. The current study followed 11 individuals who met the minimum threshold for depression as measured by the Patient Health Questionnaire-9 (PHQ-9) for a period of 28 days and tracked their walking using a Jawbone UP activity bracelet. The results showed that daily activity differed significantly …

Comparison Of Aquaponics And Hydroponics On Basil (Ocimum Basilicum) Morphometrics And Essential Oil Composition, Lauren E. Wilson, Nathan C. Duncan, D. Andrew Crain

RURALS: Review of Undergraduate Research in Agricultural and Life Sciences

Soilless farming, though in practice for over two millennia, is becoming more prevalent in modern food production as it not only saves water and space but also provides an effective option for indoor urban farming. There are currently two major models of soilless farming—hydroponics and aquaponics. While both systems are effective methods, very few studies compare the two systems. The objective of this study was to compare the water quality, basil (Ocimum basilicum) productivity, and basil essential oil profiles from plants grown in a hydroponic and newly established aquaponic system. Basil plants from two age groups (young plants …

From Efficacy To Effectiveness To Broad Implementation: Evolution Of The Body Project, Carolyn Becker, Eric Stice

Psychology Faculty Research

Objective: At the turn of the millennium, eating disorders (EDs) prevention was largely nonexistent. No program had reduced future onset of EDs in even a single trial, and most had not reduced ED symptoms. Sixteen years later, the ED prevention field has translated basic risk factor research into interventions, with demonstrated efficacy and effectiveness in reducing ED risk factors and symptoms, as well as future ED onset in some trials. This article reviews the aforementioned progress focusing on a model intervention (i.e., the Body Project [BP]).

Method: The article is a qualitative review of the existing BP literature.

Results: Although …

Resolving Spatial And Temporal Variability In Dissolved Organic Matter Characteristics Within Combined Agricultural And Stormwater Conveyances, Bryce A. Mihalevich Aug 2017

Resolving Spatial And Temporal Variability In Dissolved Organic Matter Characteristics Within Combined Agricultural And Stormwater Conveyances, Bryce A. Mihalevich

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

In many urban areas, stormwater runoff can threaten the ecological health of streams and downstream water bodies. Due to the increased impervious nature of urban landscapes, runoff is more “flashy” and as a result, high concentrations of pollutants can be transported in shorter periods of time than in more natural environments. One pollutant of concern is dissolved organic matter (DOM). DOM is important within aquatic ecosystems, but excess amounts can cause depletion in dissolved oxygen concentrations and can negatively affect aquatic organisms. This study investigated changes in DOM quantity and sources within the Northwest Field Canal (NWFC), an urban water …

The Role Of Disease And Ectoparasites In The Ecology Of Nestling Golden Eagles, Benjamin Michael Dudek Aug 2017

The Role Of Disease And Ectoparasites In The Ecology Of Nestling Golden Eagles, Benjamin Michael Dudek

Boise State University Theses and Dissertations

Climate and anthropogenic land use changes can alter biological communities and affect disease infection rates and parasite species distribution and abundance. Management to mitigate the threats of emerging infectious diseases and parasite species requires identifying and understanding factors that influence individual susceptibility within populations. Golden eagles (Aquila chrysaetos) in southwestern Idaho face several current and emerging threats, including a landscape-mediated diet shift that has increased the potential for disease infection, and warming temperatures that may increase the distribution and abundance of hematophagous ectoparasites. We examined prevalence of Trichomonas gallinae infection in golden eagle nestlings across western North America …

Landscape-Scale Manipulation Of The Acoustic Environment Alters The Distribution Of Breeding Birds And Arthropods, Elizeth Cinto-Mejia Aug 2017

Landscape-Scale Manipulation Of The Acoustic Environment Alters The Distribution Of Breeding Birds And Arthropods, Elizeth Cinto-Mejia

Boise State University Theses and Dissertations

Oil and gas development has rapidly increased across the world over the last several decades. Anthropogenic noise, an invisible pollutant that alters animal distribution and behavior, could be responsible for documented wildlife population declines near loud compressor stations in energy extraction fields. We experimentally played back compressor noise, creating a “phantom natural gas field” in a large-scale experiment, and tested the effects of noise on songbird distributions during the breeding season and on arthropod distributions. Further, to begin to understand the influence of noise produced by different types of extraction infrastructure, we examined the effects of sound intensity and bandwidth, …

Unknown image type identification is the problem of identifying unknown types of images from the set of already provided images that are considered to be known, where the known and unknown sets represent different content types. Solving this problem has a lot of security applications such as suspicious object detection during baggage scanning at airport customs, border protection via remote sensing, cancer detection, weather and disaster monitoring, etc. In this thesis, we focus on identification of unknown landscape images. This application has a huge relevance to the context of a smart nation where it can be applied to major national …

Using Silkworms As A Host To Spin Spider Silk-Like Fibers, Xiaoli Zhang Aug 2017

Using Silkworms As A Host To Spin Spider Silk-Like Fibers, Xiaoli Zhang

All Graduate Theses and Dissertations, Spring 1920 to Summer 2023

Using silkworms as the potential host to spin spider silk-like fibers is an area of intense research world-wide. The conventional methods used to create transgenic silkworms hosting spider silk-like gene limits the incorporation of spider silk-like protein and do not improve the mechanical performance of the composite silkworm/spider silk fibers. In this dissertation, synthetic spider ampullate genes were incorporated into the precise site of the fibroin heavy chain or light chain using the latest genome editing technology CRISPR/cas9 guided non-homologous end joining as opposed to conventional random integration using transposon-based piggyBac system. These protocols, with extensive applicability to other silkworm …

Cell-Free Synthesis Of Proteins With Unnatural Amino Acids: Exploring Fitness Landscapes, Engineering Membrane Proteins And Expanding The Genetic Code, Song Min Schinn

Theses and Dissertations

Unnatural amino acids (uAA) expand the structural and functional possibilities of proteins. Numerous previous studies have demonstrated uAA as a powerful tool for protein engineering, but challenges also remain. Three notable such challenges include: (1) the fitness of uAA-incorporated proteins are difficult to predict and time-consuming to screen with conventional methods, (2) uAA incorporation in difficult-to-express proteins (e.g. membrane proteins such as G-protein coupled receptors) remain challenging, and (3) the incorporation of multiple types of uAA are still limited. In response, we pose cell-free protein synthesis (CFPS), a rapid and versatile in vitro expression system, as a platform to explore …

Electrophilic Catalysis Using Heterobimetallic Complexes, Whitney Kaye Walker Aug 2017

Electrophilic Catalysis Using Heterobimetallic Complexes, Whitney Kaye Walker

Theses and Dissertations

Conventional ligand design in transition metal catalysis capitalizes on the ability of phosphorous, nitrogen, carbon, oxygen, and sulfur-based donors to modify the steric and electronic properties of a reactive metal center. Heterobimetallic transition metal complexes that contain a dative metal-metal bond provide a unique approach to ligand design where the reactivity of the metal center can be modified by metal-metal electronic communication. Our laboratory is interested in using the unique properties of heterobimetallic complexes to address significant limitations in current transition metal catalysis. My PhD work has focused on the ability of early/late transition metal heterobimetallic complexes to facilitate catalysis …

Object Language/On Defining Sculpture, Thaddeus Barak Moore Celia-Zoellner

UNLV Theses, Dissertations, Professional Papers, and Capstones

Object Language

In the current era we in the Western, developed world, have almost universal free and uninhibited access to almost every piece of information in existence. Increasingly, regardless of the source, material presented to us as fact has become increasingly suspect. Together, these two things mean this endless stream of data is useless. The question is how to combat this decline, how to reverse the process of a meaningless, constant data-dump. The answer lies in the language used to communicate information. Language is the means by which we communicate complex ideas and knowledge from person to person. Language is …

Automated Quantification Of White Blood Cells In Light Microscopic Images Of Injured Skeletal Muscle, Yang Jiao Aug 2017

Automated Quantification Of White Blood Cells In Light Microscopic Images Of Injured Skeletal Muscle, Yang Jiao

UNLV Theses, Dissertations, Professional Papers, and Capstones

Muscle regeneration process tracking and analysis aim to monitor the injured muscle tissue section over time and analyze the muscle healing procedure. In this procedure, as one of the most diverse cell types observed, white blood cells (WBCs) exhibit dynamic cellular response and undergo multiple protein expression changes. The characteristics, amount, location, and distribution compose the action of cells which may change over time. Their actions and relationships over the whole healing procedure can be analyzed by processing the microscopic images taken at different time points after injury. The previous studies of muscle regeneration usually employ manual approach or basic …

Development And Psychometric Evaluation Of The Unfinished Business In Bereavement Scale, Kara Klingspon Aug 2017

Development And Psychometric Evaluation Of The Unfinished Business In Bereavement Scale, Kara Klingspon

UNLV Theses, Dissertations, Professional Papers, and Capstones

Bereavement is one of life's greatest challenges, but most grievers recover within approximately six months after the loss. Prolonged Grief Disorder or Complicated Grief describes the 10-20% who continue to struggle with chronic and severe symptoms such as yearning and/or longing for the deceased. Those with prolonged grief are at elevated risk for a number of detrimental physical and mental health outcomes. Unfinished business, which refers to a subjective perception that something was left undone, unsaid, or unresolved with the deceased, is one marker indicating greater risk for such symptomology. Although a common target for intervention, no empirically validated tool …
