Heavy metals, such as arsenic, chromium, cadmium, nickel, mercury, and uranium are known to cause many human diseases and health complications after occupational or environmental exposure. Consequently, metals are environmental health concerns. This manuscript is an overview of the 9th Conference on Metal Toxicity and Carcinogenesis held in October 2016 in Lexington, Kentucky. Since 2000, this biennial meeting brings together experts in the field to discuss current and prospective research in an effort to advance research pertaining to metal toxicity and carcinogenesis. High doses of ionizing radiation induce acute damage to epithelial cells of the gastrointestinal (GI) tract, mediating toxicities restricting the therapeutic efficacy of radiation in cancer and morbidity and mortality in nuclear disasters. No approved prophylaxis or therapy exists for these toxicities, in part reflecting an incomplete understanding of mechanisms contributing to the acute radiation-induced GI syndrome (RIGS). Guanylate cyclase C (GUCY2C) and its hormones guanylin and uroguanylin have recently emerged as one paracrine axis defending intestinal mucosal integrity against mutational, chemical, and inflammatory injury. Here, we reveal a role for the GUCY2C paracrine axis in compensatory mechanisms opposing RIGS. …

The majority of pancreatic ductal adenocarcinomas (PDAC) rely on the mRNA stability factor HuR (ELAV-L1) to drive cancer growth and progression. Here, we show that CRISPR-Cas9–mediated silencing of the HuR locus increases the relative sensitivity of PDAC cells to PARP inhibitors (PARPi). PDAC cells treated with PARPi stimulated translocation of HuR from the nucleus to the cytoplasm, specifically promoting stabilization of a new target, poly (ADP-ribose) glycohydrolase (PARG) mRNA, by binding a unique sequence embedded in its 30 untranslated region. HuR-dependent upregulation of PARG expression facilitated DNA repair via hydrolysis of polyADP-ribose on related repair proteins. Accordingly, strategies to …

Evidence For The Early Onset Of The Ipswichian Thermal Optimum: Palaeoecology Of Last Interglacial Deposits At Whittlesey, Eastern England, H Langford, S Boreham, Rm Briant, Gr Coope, Et Al

School of Geography, Earth and Environmental Sciences

Fossiliferous deposits infilling a channel at Whittlesey, eastern England, are dated by amino acid racemization to the Last Interglacial, and pollen analysis indicates deposition in Ipswichian biozones Ip Ib and Ip IIb. Multidisciplinary palaeoenvironmental analyses of these deposits provide a rare insight into Ip Ib subzone conditions. Specifically, the Ip Ib deposits contain exotic thermophiles Naias minor, Belgrandia marginata, Bembidion elongatum, Pelochares versicolor, Caccobius schreberi, Onthophagus massai and Emys orbicularis, usually associated with Ip IIb. Combined palaeotemperature reconstructions based on beetle, ostracod and vertebrate assemblages of the Ip Ib deposits indicate that summers (mean July range +19 to +22°C) were …

Seeds From Fresh Conventional Tomatoes Germinate Faster Than Dried Or Organic Seeds, Christopher G. Brown, Kautz Evan, Cameron Kilpatrick, Mary Vu, Mallory Weaver

Conventionally-grown and organically-grown tomato seeds sourced from both fresh tomatoes and store-bought packages were germinated for seven days to evaluate the effect of seed source on germination rates. Seeds from fresh Roma tomatoes were prepared by allowing them to ferment in their own pulp for 24 hours, while commercially packaged dry seeds did not require any preparation. Once prepared, the seeds were spaced evenly on wet paper towels and stored in resealable plastic bags in groups of 10 for a total sample size of 100 seeds in each of four treatments. Photorhabdus temperata strain Hm is an entomopathogenic bacterium that forms a symbiotic association with Heterorhabditis nematodes. Here, we report a 5.0-Mbp draft genome sequence for P. temperata strain Hm with a G+C content of 44.1% and containing 4,226 candidate protein-encoding genes.

Exploring Biological Heterogeneity And Its Consequences At Tissue And Cellular Scales Through Mathematical And Computational Modeling, Romica Kerketta

This dissertation explores the effects of heterogeneity across different biological scales in cancer as well as normal cells. At the tissue scale, we investigated the variability present in the tumor microenvironment and its effect on patient chemotherapeutic outcomes using a mathematical model of drug transport. We found that parameters such as tumor blood perfusion and radius of blood vessel had an impact on the tumor cytotoxicity. This indicated that the physical microenvironment of the tumor is an important regulator of the tumor response to chemotherapy. Animal studies demonstrate that hyperlipidemia and renal lipid accumulation contribute to the pathogenesis of diabetic nephropathy (DN). We previously demonstrated that renal lipoproteins colocalize with biglycan, a renal proteoglycan. The purpose of this study was to determine whether prevention of renal lipid (apoB) accumulation attenuates DN. Biglycan-deficient and biglycan wild-type Ldlr−/− mice were made diabetic via streptozotocin and fed a high cholesterol diet. As biglycan deficiency is associated with elevated transforming growth factor-β (TGF-β), in some experiments mice were injected with either the TGF-β-neutralizing antibody, 1D11, or with 13C4, an irrelevant control antibody. The command and control (C2) of shared space resources are vulnerable to logical credential forgery and impersonation attacks among standardized and interoperable wireless radio frequency (RF) networks. Threats could come from trusted operators (insiders) or from external sources (outsiders). An attacker may gain unauthorized network access and illegally cross into C2 boundaries when conventional network authentication fails. This research proposes an integrated trust management system that uses both application-layer and physical-layer trust markers to authenticate users and their communication sources. The purpose of this research was to characterize jet fuel combustion emissions (JFCE) in an occupational setting. Prior research demonstrated that aircraft emit hazardous species, especially at engine start-up and ground idle. Complaints of eye, nose, and throat irritation from occupational exposures near aircraft exist. In this study JFCE were tested during an aeromedical evacuation engines running patient onload (ERO) on a C-130 Hercules at the 179th Airlift Wing, Mansfield-Lahm Air National Guard. Ultrafine particles, VOC, formaldehyde, carbon monoxide (CO), sulfuric acid, and metals were sampled simultaneously in approximate crew and patient breathing zones. To date, gene-based rare variant testing approaches have focused on aggregating information across sets of variants to maximize statistical power in identifying genes showing significant association with diseases. Beyond identifying genes that are associated with diseases, the identification of causal variant(s) in those genes and estimation of their effect is crucial for planning replication studies and characterizing the genetic architecture of the locus. However, we illustrate that straightforward single-marker association statistics can suffer from substantial bias introduced by conditioning on gene-based test significance, due to the phenomenon often referred to as “winner's curse.” We illustrate the ramifications of this bias …

Languages generally use musical pitch variation of the voice as part of their sound systems (Maddieson, 2011)—pitch variations that can be somewhat reminiscent of music. Music ability and/or training may influence language processing (e.g., Bidelman et al, 2011; Delogue et al, 2010). In particular, studies have concluded that there may be a relationship between absolute pitch and tone language acquisition (e.g., Lee, Lee, and Shi, 2011; Tillmann et al., 2011; Pfordresher and Brown, 2009). A botnet is a program designed to perform a specific task using multiple computers connected in a network. In this paper we will focus on botnets being used to distribute malicious programs. In the real world, botnets have been shown to exhibit more aggressive and sophisticated behaviour than traditional malware. Botnets are used to infect computer networks and hence their success depends on the properties of the networks. A modification of inertia (called MiHsC or quantized inertia) has been proposed that assumes that inertia is caused by Unruh radiation, and that this radiation is made inhomogeneous in space by either Rindler horizons caused by acceleration or the distant Hubble horizon. The former predicts the standard inertial mass, and the latter predicts galaxy rotation without dark matter and cosmic acceleration without dark energy.
