Kristin Allison ’15, a native of Old Fort, N.C., applied to Gardner-Webb University on a whim. She was invited to interview for a scholarship and was impressed with the campus and the people. “As soon as I stepped foot on campus, I fell in love,” she recalled. “It truly felt like home, and the staff, students, and interviewers were incredibly kind and welcoming. A few weeks later, I received word that I had been offered a full scholarship to attend Gardner-Webb.”

This thesis comprises of three computer aided drug design studies utilizing molecular docking and molecular dynamic simulations: (i) a lead optimization study virtually screening an initial library of ~120000 lead compounds targeting fungal leucyl tRNA synthetase, (ii) an exploratory study to understand the binding pathway of BRACO19 to a parallel telomeric DNA G-quadruplex by MD simulations and compare with experimentally solved X-ray crystal structure (iii) a comparative study to understand the lack of selectivity of BRACO19 to various topologies of human telomeric DNA G-quadruplex over DNA duplex.

Purpose: To describe a novel biochemical marker in dried blood spots suitable to improve the specificity of newborn screening for Pompe disease.
Methods: The new marker is a ratio calculated between the creatine/creatinine (Cre/Crn) ratio as the numerator and the activity of acid α-glucosidase (GAA) as the denominator. Using Collaborative Laboratory Integrated Reports (CLIR), the new marker was incorporated in a dual scatter plot that can achieve almost complete segregation between Pompe disease and false-positive cases.
Results: The (Cre/Crn)/GAA ratio was measured in residual dried blood spots of five Pompe cases and was found to be elevated (range 4.41–13.26; 99%ile …

This research evaluates two different Biological Nitrogen Removal (BNR) systems for enhanced nitrogen removal in decentralized wastewater treatment. The first study evaluated the performance of Hybrid Adsorption and Biological Treatment Systems (HABiTS) at the pilot scale with and without stage 1 effluent recirculation. HABiTS is a system developed at the bench scale in our laboratory and was designed for enhanced BNR under transient loading conditions. It consists of two stages; an ion exchange (IX) onto clinoptilolite media coupled with biological nitrification in the aerobic nitrification stage 1 and a Tire-Sulfur Adsorption Denitrification (T-SHAD) system in the anoxic denitrification stage 2. …

Transcription-Driven Dna Methylation Setting On The Mouse Peg3 Locus, Corey L. Bretz, Joomyeong Kim

© 2017 Taylor & Francis Group, LLC. The imprinting of the mouse Peg3 domain is controlled through the Peg3-DMR, which obtains its maternal-specific DNA methylation during oogenesis. In the current study, we deleted an oocyte-specific alternative promoter, termed U1, which is localized 20 kb upstream of the Peg3-DMR. Deletion of this alternative promoter resulted in complete removal of the maternal-specific DNA methylation on the Peg3-DMR. Consequently, the imprinted genes in the Peg3 domain become biallelic in the mutants with maternal transmission of the deletion. Waterfowl face a multitude of stressors across the fall and winter. These stressors include energetic demands associated with annual cycle stage, weather, habitat availability, and waterfowl hunting seasons. Stressful stimuli elicit a physiologic stress response culminating with the release of corticosterone (CORT). CORT aids in survival and recovery over the short-term, but if elevated over a long period it can lead to decrements in health. To avoid the potential harmful effects of prolonged elevations in CORT, some birds seasonally dampen their response to a predictable stressor. The mechanisms by which immune checkpoint blockade modulates tumor evolution during therapy are unclear. We assessed genomic changes in tumors from 68 patients with advanced melanoma, who progressed on ipilimumab or were ipilimumab-naive, before and after nivolumab initiation (CA209-038 study). Tumors were analyzed by whole-exome, transcriptome, and/or T cell receptor (TCR) sequencing. In responding patients, mutation and neoantigen load were reduced from baseline, and analysis of intratumoral heterogeneity during therapy demonstrated differential clonal evolution within tumors and putative selection against neoantigenic mutations on-therapy. Transcriptome analyses before and during nivolumab therapy revealed increases in distinct immune cell subsets, activation of specific …

The Landscape Of Mtdna Modifications In Cancer: A Tale Of Two Cities, Kate L. Hertweck, Santanu Dasgupta
Biology Faculty Publications and Presentations
Mitochondria from normal and cancerous cells represent a tale of two cities, wherein both execute similar processes but with different cellular and molecular effects. Given the number of reviews currently available which describe the functional implications of mitochondrial mutations in cancer, this article focuses on documenting current knowledge in the abundance and distribution of somatic mitochondrial mutations, followed by elucidation of processes which affect the fate of mutations in cancer cells. The conclusion includes an overview of translational implications for mtDNA mutations, as well as recommendations for future research uniting mitochondrial variants and tumorigenesis.

We performed a set of experiments using a direct imaging method to investigate the diffusion process of iron oxide, Fe2O3, nanoparticles. We studied concentration fluctuations that move against the concentration gradient and induce disturbances in the interface between the iron oxide suspension and water in the sample cell. Using this imaging method in combination with the differential dynamic algorithm for image processing, we are able to extract information about the power, size, and lifetime of the fluctuations. We performed this experiment both in the presence and in the absence of a 4.2 mT magnetic field. We …

Glyphosate, the main ingredient in Roundup®, is the most widely used herbicide in the world. It is also used in combination with another effective herbicide, 2,4-D, in the formulation called Enlist Duo®. The EPA approved the use of Enlist Duo® on certain crops including those genetically modified to be resistant to both herbicides. The predicted significant increase in the use of these herbicides raised concerns from the general public because both compounds have been classified as possibly or probably carcinogenic. Phidianidines are a class of compound that has been extracted from the sea mollusk Phidianis militaris. These compounds have been shown to exhibit a variety of useful properties such as antihistamine effects, anti-cancer activity, agonism of the µ-opioid receptor and neuroprotection. The biological activities are thought to be caused by the 1,2,4- oxadiazole ring found within the molecule. The goal of this project is to synthesize analogues of phidianidine that contain a 1,2,3-triazole instead of the 1,2,4 oxadiazole ring using a method that will allow for other regions of the molecule to be changed. Zebrafish are a highly-valued model organism used for developmental biology research. Zebrafish can be used for genetic manipulation and hence, many mutant and transgenic lines exist. It is impractical to maintain lines of adult zebrafish, due to resource constraints and the need to continuously produce new generations. Therefore, a practical way to preserve zebrafish lines is to freeze sperm and retrieve lines using in vitro fertilization of fresh eggs. The goal of this study was to investigate the effects of sports-related head injury on balance, attention, and memory. Reliable differences have been found using measures that directly tap into brain functioning, such as the auditory oddball task combined with EEG recording. We hypothesized that athletes reporting a diagnosed concussion or participation in high-risk sports would have compromised balance and neurocognitive functioning compared to athletes in low risk sports. Forty-five undergraduate participants were identified as either concussed, non-concussed in high-risk sports, or non-concussed in low-risk sports using a survey of athletic history, head trauma and demographics. Moringa oleifera is a tree native to a number of Asian, African, and Central American countries and has been used in traditional medicine for an assortment of medicinal uses for centuries. Due to bioactive compounds within Moringa leaves, it is believed that Moringa leaf extract may possess cancer-selective antiproliferative properties. Previous research has been conducted in regards to this topic, but poor experimental design due to lack of necessary controls limits the legitimacy of anticancer claims. Mastery of pragmatics, which refers to the language used in different social interactions, is key to effective communication in daily life. However, studies have found that mothers of children with an autism spectrum disorder (ASD) or Fragile X Syndrome (FXS) are at an increased risk for impaired pragmatic language skills. Though many measures of pragmatic language exist for children, very few are available for these mothers. Two commonly used measures for the mothers include the Pragmatic Rating Scale (PRS) and the Pragmatic Language Subscale of the Broad Autism Phenotype Questionnaire (BAPQ). Multigeneration Effects Of Chronic Exposure To Polycyclic Aromatic Hydrocarbons In Gulf Killifish (Fundulus Grandis), Andrea Yammine

The goal of this study was to examine potential adaptation within two generations of Gulf killifish exposed to polycyclic aromatic hydrocarbons (PAH) while also investigating the use of mucus as an effective and non-lethal alternative to determine concentrations of 11-ketotestosterone (11-KT) and vitellogenin (VTG) of F. grandis. Multiple biomarkers were used to assess adaptation within these fish, such as lysozyme (enzyme critical in innate immunity), and 11-ketotestosterone and vitellogenin (endocrine hormones often affected by PAHs).
Mucal concentrations of 11-KT and VTG were determined and evaluated for effectiveness through comparative assays with blood plasma. This method was then applied to …

Easily-calculated metrics describing potential impacts of dams are often used as preliminary investigations, or in analyses describing hydrologic impact over large scales and involving many dams. However, common dam impact metrics overwhelmingly reflect effects of medium to large reservoirs. Indicators applied to describe a dam’s capacity to alter downstream river flows typically consider reservoir storage as a sole mechanism of hydrologic impact. Herein, metrics of hydraulic size are proposed to encompass potential impacts related to both storage and diversion of water. Hydrologic effect due to storage and diversion computed across a sample of diversion and non-diversion dams exemplifies that hydrologic …
