Exploration Of The Binding Pocket Of Histone Deacetylases: The Design Of Potent And Isoform-Selective Inhibitors, Abdullahi̇ İbrahi̇m Uba, Kemal Yelekçi̇

Turkish Journal of Biology

Histone deacetylases (HDACs) are enzymes that act on histone proteins to remove the acetyl group and thereby regulate the chromatin state. HDACs act not only on histone protein but also nonhistone proteins that are key players in cellular processes such as the cell cycle, signal transduction, apoptosis, and more. "Classical" HDACs have been shown to be promising targets for anticancer drug design and development. However, the selectivity of HDAC inhibitors for HDAC isoforms remains the motivation of current research in this field. Here, we explored Class I HDACs and HDAC6 by sequence alignment and structural superimposition, catalytic channel extraction, and …

Screening Of Yeast Isolates From Flowers For Effective Ethanol Production, Asma Chaudhary, Shuichi Karita Jan 2017

Screening Of Yeast Isolates From Flowers For Effective Ethanol Production, Asma Chaudhary, Shuichi Karita

Turkish Journal of Biology

The use of alternative substrates to produce biofuel is a striking option nowadays. This study aimed to screen bioethanol-producing yeast strains. From different flowers, 65 yeasts were isolated. Twelve isolates assimilated glucose by liberation of CO2 anaerobically. Out of these, only 5 yeast isolates fermented glucose in medium consisting of 0.8 g/L Mg2+ ions to produce 2.05 $\pm$ 0.03% ethanol. The selected five strains were identified as members of the genera Metschnikowia or Meyerozyma based on molecular characterization. Selected yeast strains were used for conversion of rice into bioethanol following dilute acid hydrolysis and fermentation. Consistent ethanol production was 1.80 …

Overexpression Of The Ibor Gene From Sweet Potato (Ipomea Batatas `Hoang Long') In Maize Increases Total Carotenoid And Ss-Carotene Contents, Thi Luong Tran, Thi Huong Ho, Duc Thanh Nguyen

Turkish Journal of Biology

Nutritional quality of most maize varieties is very low due to the lack of lysine and tryptophan and extremely low provitamin A carotenoids including ß-carotene, Α-carotene, and ß-cryptoxanthin. In this study, we report the successful overexpression of the IbOr gene in H145 and H95 inbred maize lines under the control of maize seed-specific promoter globulin 1 (Glo1) for the purpose of improving ß-carotene in maize. The results showed that the total carotenoid and ß-carotene content of all analyzed transgenic maize plants were significantly higher than those of wild-type lines. For H145-IbOr transgenic maize, in the best line (H145-IbOr.10), the total …

Impacts Of Aplysina Red Band Syndrome On Secondary Metabolite Profiles And Antibacterial Activity Of The Caribbean Sponge Genus Aplysina, Mafie Claire Vickers Jan 2017

Impacts Of Aplysina Red Band Syndrome On Secondary Metabolite Profiles And Antibacterial Activity Of The Caribbean Sponge Genus Aplysina, Mafie Claire Vickers

Honors Theses

Coral reef ecosystems continue to decline due to abiotic and biotic factors, including disease. Although much of the existing scientific literature has focused on coral diseases, reports of sponge diseases have been increasing. This is a cause for concern, as sponges are increasingly dominant components of coral reef communities, where they are responsible for a wide range of ecological functions. A sponge disease of particular interest is Aplysina Red Band Syndrome (ARBS), which impacts the Caribbean's most abundant genus of sponges, Aplysina. Three morphotypes of branching sponges within this genus are affected, including Aplysina fulva, and the thin and thick …

Relationships Between The Second To Fourth Digit Ratio (2d:4d) And Game-Related Statistics In Semi-Professional Female Basketball Players, Makailah Janae Dyer

Theses and Dissertations

Digit ratio (2D:4D) is a negative correlate of sports performance, although this relationship may be weak in open-skill sports. This study aimed to quantify relationships between 2D:4D and game-related statistics in female basketball players, and to quantify differences in mean 2D:4Ds between starters and reserves. Sixty-four female basketball players were measured for height, mass, and 2D:4D, with game-related statistics collected end-season. Partial correlations (adjusted for age and BMI) quantified relationships between 2D:4Ds and game- related statistics. Unpaired t-tests quantified differences in mean 2D:4Ds between starters and reserves. This study found that 2D:4D was a substantial negative correlate of blocks, rebounds, …

Parents Of Adolescent Children With Mental Illness: A Qualitative Exploration Of The Lived Experience, Lindsay Yates

Theses and Dissertations

The current exploratory study aimed to understand the experience of parents whose children have severe and persistent mental illness. It was hypothesized that themes developed from parents’ responses to interview questions would indicate the presence of a) grief, b) trauma, c) subjective burden, and d) objective burden. Participants were 13 parents, comprised of conveniently sampled biological parents and legal guardians. Each parent was independently interviewed using a semi-structured protocol. Interviews were audio recorded and transcribed by the principal investigator. Transcripts were analyzed using consensual qualitative analysis. An in-depth analysis of each transcript and cross analysis of transcripts yielded seven domains: …

Atypical Teratoid Rhabdoid Tumor: Two Case Reports And An Analysis Of Adult Cases With Implications For Pathophysiology And Treatment, Christopher Dardis, Jared Yeo, Kelly Milton, Lynn S. Ashby, Kris A. Smith, Shwetal Mehta, Emad Youssef, Jenny Eschbacher, Kathy Tucker, Laughlin Dawes, Neil Lambie, Elizabeth Algar, Elizabeth Hovey

Neurology

We present the first quantitative analysis of atypical teratoid rhabdoid tumors (ATRT) in adults, including two patients from our own institutions. These are of interest as one occurred during pregnancy and one is a long-term survivor. Our review of pathological findings of 50 reported cases of adult ATRT leads us to propose a solely ectodermal origin for the tumor and that epithelial-mesenchymal transition (EMT) is a defining feature. Thus, the term ATRT may be misleading. Our review of clinical findings shows that ATRT tends to originate in mid-line structures adjacent to the CSF, leading to a high rate of leptomeningeal …

Extending Data Curation Service Models For Academic Library And Institutional Repositories, Jon Wheeler Jan 2017

Extending Data Curation Service Models For Academic Library And Institutional Repositories, Jon Wheeler

University Libraries & Learning Sciences Faculty and Staff Publications

Many academic libraries have implemented institutional repositories (IR), which offer some potential for publishing and archiving data but can fall short of usability expectations and requirements. As general purpose repositories, IR's may lack the metadata and service capabilities to dynamically represent data and to effectively capitalize on complex, domain oriented metadata schema such as the Ecological Metadata Language (EML) or the ISO 19115-2014 Geographic information metadata standards. For libraries interested in providing data publishing services, one alternative to the domain agnostic nature of IR content and metadata models is to maintain the IR for traditional document types and in addition …

Exploring Racial Ideology And Coping As Moderators Of The Association Between Experiencing Racial Microaggressions And Psychological Outcomes In Black College Student, Terrence Harper Ii Jan 2017

Exploring Racial Ideology And Coping As Moderators Of The Association Between Experiencing Racial Microaggressions And Psychological Outcomes In Black College Student, Terrence Harper Ii

Master's Theses and Doctoral Dissertations

Recently, researchers have investigated how racial identity status attitudes may influence the relationship between experiencing racism and affective responses (i.e., it has been suggested that racial identity may act as a buffer against experiencing negative affect as a consequence of exposure to racial discrimination). While this question has received increased attention in recent years, this literature has not extensively examined this association with more current and refined conceptualizations of subtle discrimination (i.e., microaggressions) and racial ideology. Additionally, the impact of racial coping styles on this association has not yet been examined. As such, the purpose of this study was to …

Insulin-Like Growth Factor Signaling Regulates Developmental Trajectory Associated With Diapause In Embryos Of The Annual Killifish Austrofundulus Limnaeus, S. Cody Woll, Jason E. Podrabsky

Biology Faculty Publications and Presentations

Annual killifishes exhibit a number of unique life history characters including the occurrence of embryonic diapause, unique cell movements associated with dispersion and subsequent reaggregation of the embryonic blastomeres, and a short postembryonic life span. Insulin-like growth factor (IGF) signaling is known to play a role in the regulation of metabolic dormancy in a number of animals but has not been explored in annual killifishes. The abundance of IGF proteins during development and the developmental effects of blocking IGF signaling by pharmacological inhibition of the insulin-like growth factor I receptor (IGF1R) were explored in embryos of the annual killifish Austrofundulus …

The Challenge Of Enhancement & Adaptability In Healthcare: An Ethical Framework For Organizations, Gary Edwards Jan 2017

The Challenge Of Enhancement & Adaptability In Healthcare: An Ethical Framework For Organizations, Gary Edwards

Electronic Theses and Dissertations

This work defends enhancements that aim at promoting adaptability and formulates a framework for how healthcare organizations can cope with these sorts of enhancements. It begins by defending explicit approaches to defining enhancements and sketches a tripartite conception of enhancements dependent on well-being, social, and perception approaches. After assessing several major arguments for and against enhancement, it defends an adaptability justification for enhancements. In light of this justification, the remaining sections explore the adaptability argument’s implications for healthcare, justify an organizational approach for dealing with these implications, and finally formulate an organizational ethics framework for coping with the adaptability argument’s …

Strategies To Prevent Security Breaches Caused By Mobile Devices, Tony Griffin Jan 2017

Strategies To Prevent Security Breaches Caused By Mobile Devices, Tony Griffin

Walden Dissertations and Doctoral Studies

Data breaches happen almost every day in the United States and, according to research, the majority of these breaches occur due to a lack of security with organizations' mobile devices. Although most of the security policies related to mobile devices currently in place may meet the guidelines required by law, they often fail to prevent a data breach caused by a mobile device. The main purpose of this qualitative single case study was to explore the strategies used by security managers to prevent data breaches caused by mobile devices. The study population consisted of security managers working for a government …

Practical Guide And Atlas For The Diagnosis Of Fungal Infections, Afia Zafar, Kauser Jabeen, Joveria Farooqi

Books

Laboratory diagnosis of fungal infections remains challenging in South East Asia as it is a neglected field in most diagnostic centres in the developing world. Initial microscopic examination of clinical specimens for the presence of fungal elements followed by growth and eventual identification of isolates up to genus and species levels are very basic and important services that must be provided by any clinical microbiology laboratory. These services have considerable impact on selection of appropriate antifungal therapy and ultimate reduction in morbidity and mortality. With the realisation of scarcity of this service and expertise in Pakistan, our group decided to …

Disorganized/Disoriented (D) attachment has seen widespread interest from policy makers, practitioners, and clinicians in recent years. However, some of this interest seems to have been based on some false assumptions that (1) attachment measures can be used as definitive assessments of the individual in forensic/child protection settings and that disorganized attachment (2) reliably indicates child maltreatment, (3) is a strong predictor of pathology, and (4) represents a fixed or static "trait" of the child, impervious to development or help. This paper summarizes the evidence showing that these four assumptions are false and misleading. The paper reviews what is known about …

Characterization Of Chlamydomonas Reinhardtii Core Histones By Top-Down Mass Spectrometry Reveals Unique Algae-Specific Variants And Post-Translational Modifications, Aliyya Khan, Carlo Eikani, Hana Khan, Anthony Iavarone, James Pesavento

School of Science Faculty Works

The unicellular microalga Chlamydomonas reinhardtii has played an instrumental role in the development of many new fields (bioproducts, biofuels, etc.) as well as the advancement of basic science (photosynthetic apparati, flagellar function, etc.). Chlamydomonas' versatility ultimately derives from the genes encoded in its genome and the way that the expression of these genes is regulated, which is largely influenced by a family of DNA binding proteins called histones. We characterize C. reinhardtii core histones, both variants and their post-translational modifications, by chromatographic separation, followed by top-down mass spectrometry (TDMS). Because TDMS has not been previously used to study Chlamydomonas proteins, …
