Current antifungal therapies have limited effectiveness in treating invasive fungal infections. Furthermore, the development of new antifungal is currently unable to keep pace with the urgent demand for safe and effective new drugs. Auranofin, an FDA-approved drug for the treatment of rheumatoid arthritis, inhibits growth of a diverse array of clinical isolates of fungi and represents a new antifungal agent with a previously unexploited mechanism of action. In addition to auranofin's potent antifungal activity against planktonic fungi, this drug significantly reduces the metabolic activity of Candida cells encased in a biofilm. Unbiased chemogenomic profiling, using heterozygous S. cerevisiae deletion strains, …

The Anchor: 2017

The Anchor began in 1887 and was first issued weekly in 1914. Covering national and campus news alike, Hope College’s student-run newspaper has grown over the years to encompass over two-dozen editors, reporters, and staff. For much of The Anchor's history, the latest issue was distributed across campus each Wednesday throughout the academic school year (with few exceptions). In recent years The Anchor moved to monthly print issues and a more frequently updated website. The Anchor is now published in print twice per academic semester. Occasionally, the volume and/or issue numbering is irregular.

Increased expression of NOX4 protein is associated with cancer progression and metastasis but the role of NOX4 in cell proliferation and invasion is not fully understood. We generated NOX4 knockout HeLa cell lines using the CRISPR-Cas9 gene editing system to explore the cellular functions of NOX4. After transfection of CRISPR-Cas9 construct, we performed T7 endonuclease 1 assays and DNA sequencing to generate and identify insertion and deletion of the NOX4 locus. We confirmed the knockout of NOX4 by Western blotting. NOX4 knockout cell lines showed reduced cell proliferation with an increase of sub-G1 cell population and the decrease of S/G2/M …

The hippocampal formation is a brain structure integrally involved in episodic memory, spatial navigation, cognition and stress responsiveness. Structural abnormalities in hippocampal volume and shape are found in several common neuropsychiatric disorders. To identify the genetic underpinnings of hippocampal structure here we perform a genome-wide association study (GWAS) of 33,536 individuals and discover six independent loci significantly associated with hippocampal volume, four of them novel. Of the novel loci, three lie within genes (ASTN2, DPP4 and MAST4) and one is found 200 kb upstream of SHH. A hippocampal subfield analysis shows that a locus within the MSRB3 gene shows evidence …

Patent Law’S Reproducibility Paradox, Jacob S. Sherkow Jan 2017

Patent Law’S Reproducibility Paradox, Jacob S. Sherkow

Duke Law Journal

Clinical research faces a reproducibility crisis. Many recent clinical and preclinical studies appear to be irreproducible—their results cannot be verified by outside researchers. This is problematic for not only scientific reasons but also legal ones: patents grounded in irreproducible research appear to fail their constitutional bargain of property rights in exchange for working disclosures of inventions. The culprit is likely patent law’s doctrine of enablement. Although the doctrine requires patents to enable others to make and use their claimed inventions, current difficulties in applying the doctrine hamper or even actively dissuade reproducible data in patents. This Article assesses the difficulties …

Biblical Foundations Of Modern Social Work Values And Practice Competencies, Laura Racovita-Szilagyi, Ph.D., M.S.W, Mioara Diaconu, Ph.D., M.S.A., M.S.W.

The Journal of Biblical Foundations of Faith and Learning

Modern social work values and practice competencies establish the foundation from which social work educators teach future practitioners much needed knowledge and skills for competent practice. The profession’s two major organizations provide the platform upon which this paper was crafted. The National Association of Social Workers (NASW) provides the profession’s Code of Ethics, which presents the values and ethical principles guiding social work practice; the Council on Social Work Education (CSWE) provides the competency-based framework for social work education. Through the ages, the Bible has presented guidelines regarding: working with the poor and other vulnerable populations in their social context, …

Gene Therapy For Inherited Blood Diseases, From Viral Vectors To Gene Editing, Fulvio Mavilio Jan 2017

Gene Therapy For Inherited Blood Diseases, From Viral Vectors To Gene Editing, Fulvio Mavilio

Scale-up and Manufacturing of Cell-based Therapies V

Twenty-five years ago, genetically modified bone marrow cells were administered for the first time to a child suffering from adenosine deaminase (ADA) deficiency, a rare disorder of the immune system. Since then, gene therapy has struggled to find its place in clinical medicine, amid a rollercoaster of successes and setbacks and hype and skepticism with little precedent in modern times. Recently, a series of authoritative clinical studies proved that transplantation of genetically modified hematopoietic stem cells can cure severe diseases like immunodeficiencies, hemoglobinopathies and metabolic diseases, contributing to transforming gene therapy into one of the hottest area of investment for …

A Mixed Methods Study Of Collaborating Teacher Leadership In Professional Development Schools (Pdss), Nedd James Johnson Jan 2017

A Mixed Methods Study Of Collaborating Teacher Leadership In Professional Development Schools (Pdss), Nedd James Johnson

Theses and Dissertations

Professional Development Schools (PDSs) provide teachers with multiple new opportunities for leadership. As teachers are encouraged to assume new roles outside of the classroom and beyond the school, it is important to examine the concept of teacher leadership in practice--specifically, how it is perceived by those most closely associated with it. This study identifies a shared understanding of teacher leadership from collaborating teachers, using a sequential explanatory mixed methods design in PDSs within a large, diverse southern New Jersey school district-university partnership. The two-phase study examines perceptions about teacher leaders through data collected from the Teacher Leadership Inventory (TLI) (Angelle …

Can Clinical Staging Of Primary Amoebic Meningoencephalitis Be Of Any Therapeutic Benefit, Abdul Mannan Baig Dr., Armaghan-E-Rehman Mansoor

Department of Biological & Biomedical Sciences

Naegleria fowleri, is a free-living amoeba (FLA) known to infect humans and cause a fatal disease called primary amoebic meningoencephalitis (PAM). All of the patients are commonly admitted to the emergency room (ER). Often, treatment in the ER is delayed due to the rarity of disease leading to a delay in the diagnosis and late arrival of patients to the ER. The attempts to reduce raised intracranial pressure and subsequent herniation of the brain stem are challenging and become the cause of death in the affected patients during their stay in ER. Use of drugs like mannitol to reduce raised …

Automatic Analysis Of Surface Electromyography Of Reflexes For Central Nervous System Inhibition During Pregnancy, Neda Parvin Jan 2017

Automatic Analysis Of Surface Electromyography Of Reflexes For Central Nervous System Inhibition During Pregnancy, Neda Parvin

Theses and Dissertations

Eclampsia is a life-threatening neurological complication (seizures or coma) of pregnancy, and represents progression of pre-eclampsia (high blood pressure and protein in the urine isolated to pregnancy). Even though the exact mechanism of pre-eclampsia and its neurological manifestations have yet to be definitively established, it is known that there is a loss of inhibitory impulses between the cerebral cortex and the spinal cord resulting in exaggerated reflexes. In this work, we establish normative measures for deep tendon reflexes (DTR) during pregnancy. We quantified the surface electromyogram (EMG) at the knee and ankle of DTRs in 279 subjects. The signals were …

Dangerous Fictions In Shakespeare's Richard Ii, Terence D. Wride Jan 2017

Dangerous Fictions In Shakespeare's Richard Ii, Terence D. Wride

Student Works

In Shakespeare's Richard II, Shakespeare deposes monarchy by exposing the dangerous fictions of The King’s Two Bodies, Carl Schmitt's definition of sovereignty as expounded in Political Theology, and the English tradition of the divine right of kings and royal prerogative. By examining Ernst Kantorowicz's explication of the king's body politic and body natural as found his book The King's Two Bodies, I argue that Shakespeare critiques the popular political theology of his time by exposing the negative political repercussions of an ill-defined body politic. What past scholars have overlooked and failed to do is provide a concrete definition of …

Tears In Heaven: Religiously And Culturally Sensitive Laws For Preventing The Next Pandemic, Eloisa C. Rodriguez-Dod, Aileen Maria Marty, Elena Maria Marty-Nelson

Catholic University Law Review

This Article argues that laws created to curtail the spread of deadly contagious diseases need to be drafted and implemented in ways that maximize acceptance of an affected communities’ cultural and religious beliefs. When laws are put in place that are inconsistent with community mores, the overall goal of stopping an epidemic is threatened. Communities often distrust government and other relief organizations who mandate rules and regulations that impinge their religious and cultural beliefs; thus, these regulations geared at helping communities can paradoxically undermine the goal of preventing the spread of infectious disease.

This Article focuses on the need for …

Faculty Publications

Background

Existing literature suggests that endocrine measures, including the steroid hormones of cortisol and Dehydroepiandrosterone (DHEA), as well as the DHEA to cortisol ratio in the human hair can be used as promising biomarkers of chronic stress among humans. However, data are limited regarding the validity of these measures as biomarkers of chronic stress among people living with HIV (PLWH), whose endocrine system or hypothalamic pituitary adrenal (HPA) axis may be affected by HIV infection and/or antiretroviral therapy (ART) medications.

Method

Using hair sample data and self-reported survey from 60 PLWH in China, we examined the validity of three endocrine …

Background

The link between DNA methylation, obesity, and adiposity-related diseases in the general population remains uncertain.

Methods and Findings

We conducted an association study of body mass index (BMI) and differential methylation for over 400,000 CpGs assayed by microarray in whole-blood-derived DNA from 3,743 participants in the Framingham Heart Study and the Lothian Birth Cohorts, with independent replication in three external cohorts of 4,055 participants. We examined variations in whole blood gene expression and conducted Mendelian randomization analyses to investigate the functional and clinical relevance of the findings. We identified novel and previously reported BMI-related differential methylation at 83 CpGs …

Familial Lung Cancer: A Brief History From The Earliest Work To The Most Recent Studies, Anthony Musolf, Claire Simpson, Mariza De Andrade, Diptasri Mandal, Colette Gaba, Ping Yang, Yafang Li

Dartmouth Scholarship

Lung cancer is the deadliest cancer in the United States, killing roughly one of four cancer patients in 2016. While it is well-established that lung cancer is caused primarily by environmental effects (particularly tobacco smoking), there is evidence for genetic susceptibility. Lung cancer has been shown to aggregate in families, and segregation analyses have hypothesized a major susceptibility locus for the disease. Genetic association studies have provided strong evidence for common risk variants of small-to-moderate effect. Rare and highly penetrant alleles have been identified by linkage studies, including on 6q23–25. Though not common, some germline mutations have also been identified …
