Exploring Mechanisms Linking Signal Transduction And Cancer Stem Cell Expansion, Kibeom Jang

Increasing evidence indicates that cancer stem cells (CSCs) comprise a distinct self-renewing sub-population that can generate cancerous progeny with reduced replicative potency. CSCs are important therapeutic targets: they may not only initiate tumors but also mediate recurrence and metastasis. Most anti-cancer drugs kill the bulk cancer population. CSCs either proliferate too slowly for targeting by cycle active drugs, or escape chemotherapy by drug expulsion or greater DNA repair, leading to recurrence. The work in this thesis was undertaken to identify novel mechanisms leading to CSCs expansion. My first project identified a novel mechanism whereby vascular endothelial growth factor A (VEGFA) …

The surfaces of Ni50Ti50 shape memory alloys (SMAs) were patterned by laser scribing. This method is more simplistic and efficient than traditional indentation techniques, and has also shown to be an effective method in patterning these materials. Different laser energy densities ranging from 5 mJ/pulse to 56 mJ/pulse were used to observe recovery on SMA surface. The temperature dependent heat profiles of the NiTi surfaces after laser scribing at 56 mJ/pulse show the partially-recovered indents, which indicate a "shape memory effect (SME)" Experimental data is in good agreement with theoretical simulation of laser induced shock wave propagation inside NiTi SMAs. …

Predators that feed on chemically-defended prey often experience non-lethal effects that result in learned avoidance of the prey species. Some predators are able to consume toxic prey without ill-effect. The Chinese mantid is able to consume cardenolide-containing monarch caterpillars without immediate adverse effects. Although they discard the caterpillars’ gut contents, mantids consume sequestered cardenolides. Although consumption of these cardenolides does not elicit an acute response, there may be long-term costs associated with cardenolide consumption. We tested the hypothesis that consumption of monarch caterpillars will adversely affect adult mantid biomass gain and reproductive condition. We reared mantids from egg to adult …

Pomona Faculty Publications and Research
A one-pot Pd-catalyzed conversion of aryl iodide to aryl sulfonyl fluorides using DABSO and Selectfluor has been developed generating aryl sulfonyl fluorides in good to excellent yields. The reaction results in the generation of electronically and sterically diverse sulfonyl fluorides. Additionally, sulfonyl fluorides can be converted to aryl sulfonamides and sulfonic esters using Cs2CO3 under mild conditions.

Hendra Virus Fusion Protein Transmembrane Domain Contributes To Pre-Fusion Protein Stability, Stacy Webb, Tamas Nagy, Hunter Moseley, Michael G. Fried, Rebecca Ellis Dutch
Molecular and Cellular Biochemistry Faculty Publications
Enveloped viruses utilize fusion (F) proteins studding the surface of the virus to facilitate membrane fusion with a target cell membrane. Fusion of the viral envelope with a cellular membrane is required for release of viral genomic material, so the virus can ultimately reproduce and spread. To drive fusion, the F protein undergoes an irreversible conformational change, transitioning from a metastable pre-fusion conformation to a more thermodynamically stable post-fusion structure. Understanding the elements that control stability of the pre-fusion state and triggering to the post-fusion conformation is important for understanding F protein function. Mutations in F protein transmembrane (TM) domains …

Medication Dosage Calculation: A Mathematical Intervention For Nursing Students, Theresa D. Cowan
Medication Dosage Calculation: A Mathematical Intervention For Nursing Students, Theresa D. Cowan
Dissertations
Nurse educators continue to express concern over the struggle that nursing students have with mathematical calculations necessary to compute medication dosage. This applied dissertation designed, implemented, and evaluated a structured program of mathematical remediation incorporating dimensional analysis. This program targeted nursing students with demonstrated deficiencies in computation skills. The degree of mathematical preparation that the student was exposed to prior to enrollment into nursing courses was considered worthy of review. The age-old adage of gender differences that were theorized to affect the mathematical ability of students was taken into regard as well. Best practices included a curriculum devised to address …

2003, April – Land Use History And Mapping In California’S Central Coast Region; Report No. Wi-2003-03; Newman, W. B, Watson, F.G.R,
Miscellaneous Monterey and San Luis Obispo County Documents and Reports
Land use change has a long history in the region, which once held the State Capitol at Monterey. The region been transformed several times, following the introduction of Europeans and their grain crops in the 1800s, the development of groundwater-based irrigation in the late 1920s, and the expansion of vineyards and urban areas in more recent years.
Land management must be aware of the history of the land, and of its current spatial state. The early chapters of this report review the major land use systems of the region and their history. The latter chapter presents a new remotely sensed …

This work was intended to develop self-assembly lipids for incorporating G-protein coupled receptors (GPCRs) in order to improve the success rate for nuclear magnetic resonance spectroscopy (NMR) structural elucidation. We hereby report the expression and purification of uniformly 15N-labeled human cannabinoid receptor-2 domain in insect cell media. The domain was refolded by screening several membrane mimetic environments. Different q ratios of isotropic bicelles were screened for solubilizing transmembrane helix 6, 7 and 8 (TMH67H8). As the concentration of dimyristoylphosphocholine (DMPC) was increased such that the q ratio was between 0.16 and 0.42, there was less crowding in the cross peaks …

Pathogenic gene fusions have been identified in several histologic types of salivary gland neoplasia, but not previously in acinic cell carcinoma (AcCC). To discover novel gene fusions, we performed whole-transcriptome sequencing surveys of three AcCC archival cases. In one specimen we identified a novel HTN3-MSANTD3 gene fusion, and in another a novel PRB3-ZNF217 gene fusion. The structure of both fusions was consistent with the promoter of the 5' partner (HTN3 or PRB3), both highly expressed salivary gland genes, driving overexpression of full-length MSANTD3 or ZNF217. By fluorescence in situ hybridization of an expanded AcCC case series, we observed MSANTD3 rearrangements …
Ecological Pleiotropy Suppresses The Dynamic Feedback Generated By A Rapidly Changing Trait, John Delong
Ecological Pleiotropy Suppresses The Dynamic Feedback Generated By A Rapidly Changing Trait, John Delong
School of Biological Sciences: Faculty Publications
Population dynamics may carry a signature of an ecology- evolution-ecology feedback, known as eco-evolutionary dynamics, when functionally important traits change. Given current theory, the absence of a feedback from a trait with strong links to species interactions should not occur. In a previous study with the Didinium-Paramecium predator-prey system, however, rapid and large-magnitude changes in predator cell volume occurred without any noticeable effect on the population dynamics. Here I resolve this theory-data conflict by showing that ecological pleiotropy—when a trait has more than one functional effect on an ecological process—suppresses shifts in dynamics that would arise, given the links between …

Background: Rift Valley fever (RVF) is a mosquito-borne viral zoonosis of ruminants and humans that causes outbreaks in Africa and the Arabian Peninsula with significant public health and economic consequences. Humans become infected through mosquito bites and contact with infected livestock. The virus is maintained between outbreaks through vertically infected eggs of the primary vectors of Aedes species which emerge following rains with extensive flooding. Infected female mosquitoes initiate transmission among nearby animals, which amplifies virus, thereby infecting more mosquitoes and moving the virus beyond the initial point of emergence. With each successive outbreak, RVF has been found to expand …

Is Biomass A Reliable Estimate Of Plant Fitness?, Brett Steven Younginger, Dagmara Sirova, Mitchell B. Cruzan, Daniel J. Ballhorn
Biology Faculty Publications and Presentations
The measurement of fitness is critical to biological research. Although the determination of fitness for some organisms may be relatively straightforward under controlled conditions, it is often a difficult or nearly impossible task in nature. Plants are no exception. The potential for long-distance pollen dispersal, likelihood of multiple reproductive events per inflorescence, varying degrees of reproductive growth in perennials, and asexual reproduction all confound accurate fitness measurements. For these reasons, biomass is frequently used as a proxy for plant fitness. However, the suitability of indirect fitness measurements such as plant size is rarely evaluated. This review outlines the important associations …
