Snail1, a key transcription factor of epithelial–mesenchymal transition (EMT), is subjected to ubiquitination and degradation, but the mechanism by which Snail1 is stabilized in tumours remains unclear. We identify Dub3 as a bona fide Snail1 deubiquitinase, which interacts with and stabilizes Snail1. Dub3 is overexpressed in breast cancer; knockdown of Dub3 resulted in Snail1 destabilization, suppressed EMT and decreased tumour cell migration, invasion, and metastasis. These effects are rescued by ectopic Snail1 expression. IL-6 also stabilizes Snail1 by inducing Dub3 expression, the specific inhibitor WP1130 binds to Dub3 and inhibits the Dub3-mediating Snail1 stabilization in vitroand in vivo. …

Effectiveness Of Intuitive Eating Intervention Through A Text Messaging Program Among College Students, Tessa J. Loughran Feb 2017

Effectiveness Of Intuitive Eating Intervention Through A Text Messaging Program Among College Students, Tessa J. Loughran

Theses and Dissertations

A critical time period to prevent overweight may be the college age years. College students experience newfound food independence, possibly resulting in the formation of poor eating habits, such as overeating, or binge eating, placing them at high risk for obesity. Additionally, college students with high perceived stress levels are more likely to experience emotional eating, which may lead to weight gain. Intuitive Eating (IE) practice focuses on listening to the hunger and satiety signals, in hopes that it will prevent overeating, resulting in weight loss. Recently, texting intervention has been seen to be a successful intervention platform for behavior …

Background. A Nugent score > 7 has been defined as the gold standard for the diagnosis for bacterial vaginosis (BV), though it is resource intensive and impractical as point of care testing. We sought to determine if colorimetric assessment of vaginal pH can accurately predict the occurrence of BV. Methods. We performed a planned subanalysis of 1,216 pregnant women between 13 0/7 and 19 6/7 weeks who underwent vaginal examination as part of a randomized controlled trial. Using a standardized technique, specimens were obtained for colorimetric assessment and two separate slides for Gram staining. These slides were subsequently evaluated by two …

The Roots And Repercussions Of Moroccan Soft Wheat Consumption, Production, And Import Patterns, Anna Murphy Feb 2017

The Roots And Repercussions Of Moroccan Soft Wheat Consumption, Production, And Import Patterns, Anna Murphy

University Honors Theses

Following the conclusion of World War II, Moroccans' khobz—bread—consumption experienced one fundamental and detrimental change: the homogenization of recipes from an assortment of diverse ingredients, such as barley, winter wheat, durum, and seeds, to almost solely soft wheat. The homogenizing trend towards soft wheat is an outcome of Western influence, trade policies, international markets, economic growth and integration. Specifically, the United States' development initiatives—namely Green Revolution technology transfers and food aid—rendered detrimental effects on Moroccan wheat patterns of consumption, production, and import, which were further perpetuated and extended by Moroccan governance post-independence. Analyzing the ramifications of soft wheat patterns …

A Mendelian Randomization Study Of The Effect Of Calcium On Coronary Artery Disease, Myocardial Infarction And Their Risk Factors, Lin Xu, Shi Lin Lin, C. Mary Schooling

Publications and Research

Meta-analyses of randomized controlled trials (RCTs) suggest calcium could have adverse effects on cardiovascular disease, although these findings are controversial. To clarify, we assessed whether people with genetically higher calcium had a higher risk of coronary artery disease (CAD), myocardial infarction (MI) and their risk factors. We used a two-sample Mendelian randomization study. We identified genetic variants (single nucleotide polymorphisms (SNPs)) that independently contributed to serum calcium at genome-wide significance which we applied to large extensively genotyped studies of CAD, MI, diabetes, lipids, glycaemic traits and adiposity to obtain unconfounded estimates, with body mass index (BMI) as a control outcome. …

Representative Bureaucracy, Street-Level Bureaucrats And Bureaucratic Discretion In Federal Disaster Assistance, Jason David Rivera Feb 2017

Representative Bureaucracy, Street-Level Bureaucrats And Bureaucratic Discretion In Federal Disaster Assistance, Jason David Rivera

Journal of Public Management & Social Policy

This paper explores the role of representative bureaucracy and bureaucratic discretion in the allocation of federal disaster assistance to Hurricane Sandy survivors. Through the analysis of focus groups and key informant interviews, this study finds that FEMA home inspectors are not diverse in reference to race and gender; however, inspectors are found to be diverse in reference to place of origin. Although the role that race and gender play in the allocation of resources is found to be unclear, the region from which inspectors come from is found to be influential in the allocation of aid to survivors. As such, …

Participant Observation: Enhancing The Impact Measurement In Community Based Participatory Research, Brent Hammer, Fay Fletcher, Alicia Hibbert Feb 2017

Participant Observation: Enhancing The Impact Measurement In Community Based Participatory Research, Brent Hammer, Fay Fletcher, Alicia Hibbert

The Qualitative Report

Participant observation parallels the principles of community based participatory research (CBPR), recognizing that each community should be understood in its own context. Using fieldnotes from the Métis Settlements Life Skills Journey (MSLSJ) program, the authors explore the benefits and challenges of using participant observation in CBPR program evaluation. Participant observation was incorporated in 2014 and 2015 as researchers sought a complementary perspective and context to determine the impact of the program. The authors explore relationships with a large number of stakeholders (children, facilitators, community members, and project staff) and discuss ensuring the participant observer’s perspective is not privileged above others.

International Journal of Exercise Science: Conference Proceedings

Dual-energy x-ray absorptiometry (DXA) is viewed as a superior method of body composition assessment, but whole-body DXA scans are impacted by variation in pre-assessment activities, such as eating and drinking. DXA software now allows for estimation of visceral adipose tissue (VAT), which has been implicated in a number of diseases. It is unknown to what extent food and fluid intake affect VAT estimates. PURPOSE: determine the effects of acute high-carbohydrate (HC) and very low-carbohydrate (VLC) diets on DXA estimates of VAT. METHODS: Male and female adults completed two one-day dietary conditions in random order: a VLC diet (1 – …

Mitochondrial Dysfunction In Diaphragm Muscle Precedes The Cachectic Phenotype In Llc Tumor-Bearing Mice., Conner A. Benson, David E. Lee, Jacob L. Brown, Megan E. Rosa-Caldwell, Tyrone A. Washington, Nicholas P. Greene, Michael P. Wiggs

International Journal of Exercise Science: Conference Proceedings

The defining feature of cancer cachexia is extensive weight loss and skeletal muscle atrophy. It is clinically important because cachexia reduces patient survival, results in functional impairment, and is estimated to be directly responsible for 20-40% of cancer deaths. Unfortunately, no clinical therapy exists and therefore, it is important to understand the molecular mechanisms responsible for rapid muscle wasting. Compared to limb muscles, the diaphragm is relatively understudied in cancer cachexia, but is likely to be adversely affected because cachexia is a systemic disease. Wasting of the primary inspiratory muscle may result in difficulty breathing and inability to adjust minute …

Effects Of Hypohydration On Muscular Performance In Females: An Ongoing Study, Joshua J. Gann, Thomas L. Andre, Paul S. Hwang, Brooke L. Roemer, Darryn S. Willoughby

International Journal of Exercise Science: Conference Proceedings

Dehydration (~3%) has been shown to negatively affect anaerobic performance. A majority of this research has been conducted using male participants. Like males, females have been shown to underestimate sweat loss, which could lead to insufficient rehydration and thus hypohydration. Additionally, due to differences in thermoregulation, it is possible that females respond to hypohydration differently than males. To date, no research has examined the effects of previous night dehydration on muscular performance in females. PURPOSE: The purpose of this ongoing study was to determine the effects of previous night dehydration on muscular strength, muscular endurance, lower body power, and …

Exercise Does Not Stimulate Citrate Synthase Activity In Cachectic Muscle, Sarah Senger, Kira Pamerleau, Derek Wallace, Paul Rivas, Nicholas Musi, A. Pratrap Kumar, Darpan I. Patel

International Journal of Exercise Science: Conference Proceedings

Cachexia causes metabolic alterations in skeletal muscle mitochondria stimulated by inflammatory imbalance towards pro-inflammatory signaling. Previous work by our group has demonstrated that exercise significantly increases muscle mass in cases of advanced tumors in transgenic mice. Furthermore, we have discovered that the natural product Nexrutine® has anti-inflammatory properties which can be beneficial in protecting mitochondria. PURPOSE: To compare the effects of exercise and Nexrutine® on mitochondrial density in skeletal muscle taken from transgenic adenocarcinoma of the mouse prostate (TRAMP) models. METHODS: This project is a continuation of a larger study investigating the effects of exercise and Nexrutine® on the attenuation …

Effects Of Dose Timing On Fluid Excretion During Sodium-Aided Hyperhydration Protocols, Shelby L. Greene, Elizaveta Roslanova, David Morris Feb 2017

Effects Of Dose Timing On Fluid Excretion During Sodium-Aided Hyperhydration Protocols, Shelby L. Greene, Elizaveta Roslanova, David Morris

International Journal of Exercise Science: Conference Proceedings

Co-consumption of sodium and water has been shown to be superior in promoting hyperhydration compared to consumption of an equal amount of water alone. Most sodium-aided hyperhydration studies have provided subjects with a bolus of fluid followed by a urine collection period. However the effect of providing equal amounts of fluid in a single vs. multiple doses over time on fluid retention has not been systematically studied. PURPOSE: To compare the effects of different dosing strategies on urine excretion levels following the consumption of consistent amounts of sodium and water. METHODS: Urine excretion was measured during five separate …

The Valsalva Maneuver For Assessment Of Cardiovagal Baroreflex Sensitivity, Carmen Campalans, Carsten Flores-Hansen, Scott Matjeka, Clarissa Quezada, Donovan L. Fogt, William H. Cooke

International Journal of Exercise Science: Conference Proceedings

The cardiovagal baroreflex is fundamental to understanding normal autonomic neural function, but can be difficult to assess. PURPOSE: In this study, we highlight the utility of the Valsalva maneuver for the assessment of cardiovagal baroreflex sensitivity during both vagal inhibition and activation. METHODS: Thirteen supine male subjects (21 ± .7 yrs; 173 ± 2 cm; 78 ± 4 kg) were instrumented with an electrocardiogram and a finger photoplethysmograph (Finometer, to assess beat-by-beat arterial pressure). Subjects then exhaled against resistance to an expiratory pressure of 40 mmHg for 15 s followed by a 2-min recovery. Subjects performed three trials, and we …

Time-Course Of Diuresis During Sodium-Aided Hyperhydration Protocols With And Without Caffeine, Elizaveta Roslanova, David M. Morris Feb 2017

Time-Course Of Diuresis During Sodium-Aided Hyperhydration Protocols With And Without Caffeine, Elizaveta Roslanova, David M. Morris

International Journal of Exercise Science: Conference Proceedings

When used alone, both caffeine and sodium-aided hyperhydration (SAH) are ergogenic. When caffeine is used in conjunction with SAH, diuresis occurs, but hyperhydration is still achieved albeit at lower levels when compared to SAH alone. Caffeine has been shown to be ergogenic when taken as little as 5 min, or as much as 6 hr prior to the start of exercise. Thus, determining the temporal aspects of caffeine induced diuresis when used in conjunction with SAH may lead to superior pre-exercise nutritional strategies. PURPOSE: To investigate the time-course of caffeine-induced diuresis during a 90 min SAH protocol. METHODS Subjects …

Nexrutine® Mimics Exercise Preservation Of Muscle In Tramp Mice By Decreasing Proteolysis-Inducing Factor, Derek J. Wallace, Sarah Senger, Kira Pamerleau, Darpan Patel

International Journal of Exercise Science: Conference Proceedings

Cachexia is an adverse effect of prostate cancer (PCa) characterized by skeletal muscle loss which significantly impacts mortality and quality of life. Recently, proteolysis-inducing factor (PIF) mediated activation of the ubiquitin proteasome system has been identified as a potential target for treating cachexia. Oncologists encourage nutritional and physical intercession to improve outcomes, however, it is unclear how these interventions impact PIF activity. PURPOSE: To test the hypothesis that Nexrutine® (a natural bark extract of the Amur cork tree with anti-inflammatory capabilities) and exercise can independently attenuate muscle loss in a transgenic adenocarcinoma of mouse prostate (TRAMP) model by modulating …
