Drug-induced cardiotoxicity is a critical challenge in the development of new drugs. Since the advent of human pluripotent stem cell-derived cardiomyocytes (CMs), researchers have explored ways to utilize these cells for in vitro preclinical drug screening applications. One area of interest is microphysiological systems (i.e. organ-on-a-chip), which aims to create more complex in vitro models of human organ systems, thus improving drug response predictions. In this dissertation, we investigated novel analysis methods and model platforms for detecting drug-induced cardiotoxicity using human induced pluripotent stem cell (iPSC)-derived cardiovascular cells.

Photoacoustic computed tomography(PACT), also known as optoacoustic tomography (OAT), is an emerging imaging technique that has developed rapidly in recent years. The combination of the high optical contrast and the high acoustic resolution of this hybrid imaging technique makes it a promising candidate for human breast imaging, where conventional imaging techniques including X-ray mammography, B-mode ultrasound, and MRI suffer from low contrast, low specificity for certain breast types, and additional risks related to ionizing radiation. MicroRNAs (miRNAs) are short single-stranded RNAs that function as the guide sequence of the post-transcriptional regulatory process known as the RNA-induced silencing complex (RISC), which targets mRNA sequences for degradation through complementary binding to the guide miRNA. Changes in miRNA expression have been reported as correlated with numerous biological processes, including embryonic development, cellular differentiation, and disease manifestation. Over the course of human development, the brain undergoes dramatic physical changes to achieve its final, convoluted shape. However, the forces underlying every cinch, bulge, and fold remain poorly understood. This doctoral research focuses on the mechanical processes responsible for early (embryonic) and late (preterm) brain development.

First, we examine early brain development in the chicken embryo, which is similar to human at these stages. Research has primarily focused on molecular signals to describe morphogenesis, but mechanical analysis can also provide important insights. Both within the United States and internationally, hexavalent chromium (Cr(VI)) is a contaminant of concern in drinking water supplies. The U.S. Environmental Protection Agency is considering a Cr(VI)-specific standard. Thus improved technologies for Cr(VI) removal in drinking water are needed. Iron electrocoagulation for Cr(VI) removal was examined at conditions directly relevant to drinking water treatment, and humic acid (HA) affects the performance of electrocoagulation in multiple ways. The success of the chromium treatment or remediation also relies on the stability of the Cr(III)-containing solids with respect to reoxidation under groundwater conditions. Manganese (hydr)oxide (Mn (hydr)oxide) minerals are ubiquitous in aquatic and terrestrial environments. These minerals have high surface areas and are highly sorptive and redox active. From decades of accumulated knowledge about natural redox cycling, we have found that Mn (hydr)oxides play critical roles as electron donors and acceptors in elemental geochemical cycling and biological metabolisms in nature. The vitreous humor of the eye is a biological hydrogel principally composed of fibrillary collagen interspersed with semi-flexible polyelectrolyte, hyaluronic acid (HA). Certain pathological conditions necessitate its removal and replacement. Current vitreous substitutes, such as silicone oils and perfluorocarbons, are hydrophobic, do not resemble the properties of the vitreous, and have known complications. An ideal vitreous substitute should have properties of the natural vitreous, perform its functions, and be biocompatible in the eye. The general objective of this work was to develop experimental methods based on magnetic resonance elastography (MRE) to characterize fibrous soft materials. Mathematical models of tissue biomechanics capable of predicting injury, such as traumatic brain injury (TBI), are of great interest and potential. However, the accuracy of predictions from such models depends on accuracy of the underlying material parameters. This dissertation describes work toward three aims. Influenza A viruses (IAV) are a major human and environmental pathogen. IAV successfully infects a diverse host range and adaptation of new viral strains to humans may cause pandemic events with high morbidity and mortality. As a member of the Orthomyxoviridae family, IAV inherently possesses a segmented genome, which enables a process of segment transmission between viruses following cellular co-infection, a process termed reassortment. Early animal embryogenesis entails a dynamic combination of embryonic cleavages, axial patterning, and gastrulation movements to shape a basic body plan. The underlying molecular signaling responsible for regulating this process remains poorly understood. In this thesis work, I first review recent progress in understanding of gastrulation movements in various model organisms brought by advances in imaging techniques. Brain extracellular concentration of the peptide amyloid-β (Aβ) is a major contributor to Alzheimer’s disease (AD) pathogenesis. High Aβ levels in the extracellular space precipitate aggregation of the peptide into soluble and insoluble toxic species. This process begins decades before cognitive impairment and triggers the cascade of pathology that eventually leads to AD. Synaptic activity is key to the regulation of extracellular Aβ levels. Presynaptic activity drives the production of Aβ, while postsynaptic receptor activation exhibits more nuanced regulation. The peroxisome is a vital organelle conserved through the entire eukaryotic lineage. In all examined species, peroxisomes are responsible for such essential processes as fatty acid beta-oxidation and metabolism of reactive oxygen species (ROS). In plants, peroxisomes have taken on additional specialized roles, such as production of some plant hormones and vitamins. In this work, I have uncovered novel factors regulating peroxisome number in model species Arabidopsis thaliana, and novel mechanisms governing how peroxisomes respond to salt stress. Multidrug resistant (MDR) Gram-negative bacteria represent a global health crisis. Traditional antibiotics create selective pressure and breed resistance, making our current drugs less and less effective. Targeting of bacterial virulence factors has gained interest as an alternative strategy to potentially circumvent this problem. Virulent Acinetobacter baumannii synthesize and secrete pre-acinetobactin, a small molecule metal chelator that scavenges life-sustaining iron and is critical for establishing infection. The work described herein sheds light on the unusual properties of a non-enzymatic isomerization from pre-acinetobactin to acinetobactin and proposes a siderophore-swapping mechanism by which A. baumannii expands its pH window for virulence. Forest restoration in the Sierra Nevada is receiving increased attention due to climate change, increasing populations in the wildland-urban interface, and the historical mismanagement of some forests, evidenced in part by the increasing frequency and severity of wildfires in recent years. The Ross Sea, Antarctica is a deep bay of the Southern Ocean that exhibits seasonal sea ice and is adjacent to a permanent ice shelf overlying seawater. In 2008 and 2009, imagery of the seafloor under the McMurdo Ice Shelf and under the seasonal ice in the Ross Sea was collected via remotely operated vehicle (ROV) at depths to 300 m. Distinct differences in Antarctic benthic communities were observed over multiple environmental gradients. Approximately 12%-20% of pregnant women smoke at some point during pregnancy, and 10% of pregnant women are reported to have smoked during the last 3 months of pregnancy. Smoking during pregnancy leads to developmental health risks for the fetus and child, including increased mortality, low birth weight, and developmental delays. The direct molecular targets of nicotine are nicotinic acetylcholine receptors (nAChRs) due to the similarities in structure between nicotine and acetylcholine. Anthropogenic pressures such as habitat loss and hunting pressures have caused terrestrial mammalian carnivores to decline by 95–99% in many regions of the world. These species are especially susceptible to fragmented habitats and a diminishing food supply because of their slow reproductive rates and high metabolic demands. In biodiversity hotspots such as California, understanding the range of human impacts to species is critical. This project assessed carnivore species distribution and occupancy by placing remote camera traps in a protected Reserve, agricultural, and developed land use types. Foreign accent, or the deviation from non-native speech, has a direct impact on communication and may even result in undesirable consequences for the speaker. Instead of perceiving statements as more difficult to understand, native speakers often perceive them as less trustworthy. However, the pronunciation of adult second language (L2) learners is extremely difficult to change, and L2 native-like pronunciation is rarely achieved after early childhood. The latest research suggests that explicit instruction about phonological awareness can contribute to better spoken comprehensibility even in adult L2 learners. The intriguing properties of reduced graphene oxide (rGO) have paved the way for a number of potential biomedical applications such as drug delivery, tissue engineering, gene delivery and bio-sensing. Over the last decade, there have been escalating concerns regarding the possible toxic effects, behaviour and fate of rGO in living systems and environments. This paper reports on integrative chemical-biological interactions of rGO with lung cancer cells, i.e. A549 and SKMES-1, to determine its potential toxicological impacts on them, as a function of its concentration.
