NLRP10 is a nucleotide-binding oligomerization domain-like receptor that functions as an intracellular pattern recognition receptor for microbial products. Here, we generated a Nlrp10−/− mouse to delineate the role of NLRP10 in the host immune response and found that Nlrp10−/− dendritic cells (DCs) elicited sub-optimal IFNγ production by antigenspecific CD4+ T cells compared to wild-type (WT) DCs. In response to T-cell encounter, CD40 ligation or Toll-like receptor 9 stimulation, Nlrp10−/− DCs produced low levels of IL-12, due to a substantial decrease in NF-κB activation. Defective IL-12 production was also evident in vivo and affected IFNγ production by CD4+ T cells. Upon …

The Next Step In Civil Asset Forfeiture Reform: Passing The Civil Asset Forfeiture Reform Act Of 2014, Daniel Reed Nov 2017

The Next Step In Civil Asset Forfeiture Reform: Passing The Civil Asset Forfeiture Reform Act Of 2014, Daniel Reed

Catholic University Law Review

Civil asset forfeiture is an operation of legal fiction that enables the government to seize property without an underlying conviction of the property owner. Federal authorities bring thousands of civil asset forfeiture cases annually, often against the property of owners who have not been charged with a crime. Such cases can result in unjust outcomes and denials of due process to property owners. To address this controversy, Representative Tim Walberg proposed several reforms to federal civil asset forfeiture laws known as the Civil Asset Forfeiture Reform Act of 2014 (CAFRA 2014).

After discussing the history of civil asset forfeiture, this …

The efficacy of B cell depletion therapies in diseases such as nephrotic syndrome and rheumatoid arthritis suggests a broader role in B cells in human disease than previously recognized. In some of these diseases, such as the minimal change disease subtype of nephrotic syndrome, pathogenic antibodies and immune complexes are not involved. We hypothesized that B cells, activated in the kidney, might produce cytokines capable of directly inducing cell injury and proteinuria. To directly test our hypothesis, we targeted a model antigen to the kidney glomerulus and showed that transfer of antigen-specific B cells could induce glomerular injury and proteinuria. …

Impacts Of Hunting And Life-History Stage On The Stress Physiology And Body Condition Of Fall And Wintering Mallards (Anas Platyrhynchos), Jerad Richard Henson Nov 2017

Impacts Of Hunting And Life-History Stage On The Stress Physiology And Body Condition Of Fall And Wintering Mallards (Anas Platyrhynchos), Jerad Richard Henson

Electronic Theses and Dissertations Archive

Waterfowl face a multitude of stressors across the fall and winter. These stressors include energetic demands associated with annual cycle stage, weather, habitat availability, and waterfowl hunting seasons. Stressful stimuli elicit a physiologic stress response culminating with the release of corticosterone (CORT). CORT aids in survival and recovery over the short-term, but if elevated over a long period it can lead to decrements in health. To avoid the potential harmful effects of prolonged elevations in CORT, some birds seasonally dampen their response to a predictable stressor. The aim of this study was to examine the changes in stress physiology and …

Gingival Perfusion And Tissue Biomarkers During Early Healing Of Postextraction Regenerative Procedures: A Prospective Case Series, Lamees Alssum, Timothy D. Eubank, Sashwati Roy, Barbaros S. Erdal, Vedat O. Yildiz, Dimitris N. Tatakis, Binnaz Leblebicioglu

Clinical and Translational Science Institute

Background—Post-extraction alveolar bone loss, mostly affecting the buccal plate, occurs despite regenerative procedures. To better understand possible determinants, this prospective case series assessed gingival blood perfusion and tissue molecular responses in relation to postextraction regenerative outcomes. Methods—Adults scheduled to receive bone grafting in maxillary, non-molar, single tooth extraction site were recruited. Clinical documentation included probing pocket depth (PD), keratinized tissue width (KT), tissue biotype (TB), plaque (P) and bleeding. Wound closure was clinically evaluated. Gingival blood perfusion was measured by Laser Doppler Flowmetry (LDF). Wound fluid (WF) and gingival biopsies were analyzed for protein levels and gene expression, respectively, of …

'A Prey To Their Teeth': Puritan Sermons And Ministerial Writings On Indians During King Philip's War, Gregory Michna Nov 2017

'A Prey To Their Teeth': Puritan Sermons And Ministerial Writings On Indians During King Philip's War, Gregory Michna

Faculty Publications - History & Political Science

King Philip’s War encouraged the construction of barbaric Native American typologies by puritan ministers through sermons and wartime histories. While select Algonquians in New England invested in a hybridized identity as a “Praying Indian” during the decades before the war, martial circumstances encouraged ministers to emphasize indigenous savagery, which outweighed earlier efforts to demonstrate the Christian piety of native converts. English texts outlining God’s providential wrath linked the contemporaneous war to Old Testament punishments meted out to the Hebrews. This typological reading of current events also superimposed the characteristics of heathen barbarians on Native Americans irrespective of their allegiance, offering …

In Vivo Bioluminescence Imaging Of Nuclear Factor Kappab Activation: A Valuable Model For Studying Inflammatory And Oxidative Stress In Live Mice, Hong Zhu, Zhenquan Jia, Michael A. Trush, Y R. Li Nov 2017

In Vivo Bioluminescence Imaging Of Nuclear Factor Kappab Activation: A Valuable Model For Studying Inflammatory And Oxidative Stress In Live Mice, Hong Zhu, Zhenquan Jia, Michael A. Trush, Y R. Li

Osteopathic Medicine, Jerry M. Wallace School of

The nuclear factor kappaB (NF-κB) is a redox-sensitive transcription factor that plays a critical role in inflammation among other biological functions. This ROS Protocol article describes an in vivo bioluminescence imaging assay for assessing NF-κB activation using the commercially available transgenic mice carrying NF-κB response element-luciferase reporter gene (NF-κB-RE-Luc). Using the highly sensitive Berthold NightOwl LB981 in vivo bioluminescence imaging system, we are able to visualize the NF-κB activation in live mice under basal conditions, suggesting constitutive activation of NF-κB as a part of its fundamental biology. Treatment of mice with lipopolysaccharides (LPS) results in a drastic increase in bioluminescence, …

Stones And Bones: Catholic Responses To The 1812 Collapse Of The Mission Church Of Capistrano, Karin Vélez Nov 2017

Stones And Bones: Catholic Responses To The 1812 Collapse Of The Mission Church Of Capistrano, Karin Vélez

Faculty Publications

This essay delves into the 1812 collapse of the Great Stone Church at California’s Mission of San Juan Capistrano and its aftermath to consider how early modern Catholics in the greater Iberian world approached the material remains of ruined churches that contained human victims. Questions explored include how Franciscan missionaries reported and reacted to the calamity, why the casualties were disproportionately Indian and female, and what survivors did with the physical remnants of broken churches. Churches that collapsed on worshippers in Arequipa, Cuzco, Lima and Lisbon prior to 1812 are mustered for comparison. Overall, a pattern emerges of Catholics separating …
