This fairy tale, with elements of Chicken Little, is set on a farm. Delivering isotopic tracers for metabolic studies in rodents without overt stress is challenging. Current methods achieve low label enrichment in proteins and lipids. Here, we report noninvasive introduction of 13C6-glucose via a stress-free, ad libitum liquid diet. Using NMR and ion chromatography-mass spectrometry, we quantify extensive 13C enrichment in products of glycolysis, the Krebs cycle, the pentose phosphate pathway, nucleobases, UDP-sugars, glycogen, lipids, and proteins in mouse tissues during 12 to 48 h of 13C6-glucose feeding. Applying this approach to patient-derived lung tumor xenografts (PDTX), we show that the liver supplies glucose-derived Gln …

As academic education and research increasingly take advantage of geospatial data and methodologies, we see a corresponding exponential growth in the number of available geospatial resources in the form of GIS datasets and scanned historical maps. However, users can experience difficulty finding these resources due to the unconnected multitude of platforms and clearinghouses that host them. Additionally, the resources are not always well described with web semantic metadata that facilitates discovery. In response to this challenge, The Big Ten Academic Alliance Geospatial Data Project began in 2015 to provide discoverability, facilitate access, and connect scholars to geospatial resources. Social Anxiety Disorder (SAD) and Alcohol Use Disorder (AUD) are highly prevalent and frequently co-occur. The results of population studies suggest that SAD tends to precede AUD, and the results of laboratory studies suggest that alcohol use facilitates social behaviors in socially anxious individuals. Therefore, we posited that, in a modern context, a tendency to consume alcohol may be positively selected for among socially anxious individuals by its effect on the likelihood of finding a partner and reproducing. Metabolic homeostasis is coordinately controlled by diverse inputs. Understanding these regulatory networks is vital to combating metabolic disorders. The nematode Caenorhabditis elegans has emerged as a powerful, genetically tractable model system for the discovery of lipid regulatory mechanisms. Here we introduce DBL-1, the C. elegans homolog of bone morphogenetic protein 2/4 (BMP2/4), as a significant regulator of lipid homeostasis. We used neutral lipid staining and a lipid droplet marker to demonstrate that both increases and decreases in DBL-1/BMP signaling result in reduced lipid stores and lipid droplet count. Niche Cadherins Control The Quiescence-To-Activation Transition In Muscle Stem Cells., Aviva J. Goel, Marysia-Kolbe Rieder, Hans-Henning Arnold, Glenn L. Radice, Robert S. Krauss

Many adult stem cells display prolonged quiescence, promoted by cues from their niche. Upon tissue damage, a coordinated transition to the activated state is required because non-physiological breaks in quiescence often lead to stem cell depletion and impaired regeneration. Here, we identify cadherin-mediated adhesion and signaling between muscle stem cells (satellite cells [SCs]) and their myofiber niche as a mechanism that orchestrates the quiescence-to-activation transition. Conditional removal of N-cadherin and M-cadherin in mice leads to a break in SC quiescence, with long-term expansion of a regeneration-proficient SC pool. Kobayashi Takiji's (1903-1933) Kanikōsen (The Crab Cannery Ship, 1929), the outstanding work from the proletarian literary movement, experienced an influx of new adaptations into various mediums during the years that preceded and followed the "Kanikōsen boom" of 2008. This thesis focuses on two manga adaptations that provide readers with starkly different takes on the original story. Using theories by Scott McCloud and Azuma Hiroki, I first attempt to draw parallels between the form of manga and that of the novel. Over the past forty years, the Food and Drug Administration (FDA) has successfully restricted consumers' access to home-testing applications based on the notion that it should protect individuals from their own reactions to test results. In the 1970s, the FDA briefly denied women access to home pregnancy tests that were identical to those used in laboratories. In the late 1980s and early 1990s, it relied on concerns about consumer responses to HIV status results to justify a categorical ban on applications for HIV home-testing technology. This work aimed at understanding the interactions of 1) three physiologically relevant transition metal ions copper(II), cobalt(II) and manganese(II) with five tri- or tetra-dentate metal chelators (ligand or L) and 2) the resulting ML complexes with two known metalloenzyme inhibitors, 8-hydroxyquinoline (8-HQ) and acetohydroxamic acid (AHA). The ultimate goal of the work was to identify ligands that can form enzyme active site M2+ structural mimetics capable of interacting with metalloenzyme active site inhibitors. The precise timing of action potentials generated in the nervous system is crucial for generating adequate behavior. Once generated, action potentials travel along axons towards the neurons or muscles they innervate. Axons are also responsible for preserving the temporal fidelity of the generated action potentials. One challenge axons face is that they can be of considerable length, and exposed to changes in internal and external conditions. Temperature fluctuations, for example, affect the ion channels that generate and propagate action potentials causing changes in action potential speed.
