Nutrient abatement programs, originally enacted to counter eutrophication have successfully suppressed nutrient loads and relative primary production in marine and freshwater systems. Recently, the additive impacts of invasive filter feeders have further reduced offshore primary production in several aquatic systems throughout the world. It has been hypothesized that the biological activity of these invasive species may sequester nutrients within the nearshore benthic environment, creating steep gradients in primary production between nearshore and offshore habitats. Diverse mechanics of the F-actin cytoskeleton mediate essential behaviors of cells, including cell division, migration, and shape change. Force generation by motor proteins and the resultant morphological change of cytoskeletal networks govern cellular processes such as migration and division. Cell stiffening and softening under external mechanical stimuli regulate cell shape. In this thesis, interplay between various cytoskeletal components during these processes is investigated using an agent-based computational model to elucidate mechanical factors underlying these processes. This thesis is composed of three independent studies.

Radiant frying is an alternative technique to traditional immersion frying. While enjoyed by consumers, immersion fried foods are typically high in fat. Radiant frying uses infrared energy to produce finished products with similar sensory characteristics to traditional frying, but with significantly less fat. This research compared the efficacy of radiant and immersion frying using hash brown patties as a model food matrix by measuring temperature profiles, heat flux profiles, moisture and oil contents, peak shear force, color, mottling, and crust thickness as well as executing semi-trained and untrained sensory evaluations. 3D Mesh segmentation is used in various applications such as object recognition, reconstruction, and analyzing structure of meshes. The method for 3D mesh segmentation based on sparse non-negative matrix factorization (NMF) was previously proposed. It represents a novel, and conceptually simpler, method than other comparable algorithms. However, this method still has potential to improve performance, results could have better consistency and uniqueness with faster computation time than the prior proposed algorithm. Snow and ice processes are a critical part of the Earth’s hydrological and climate cycles. These processes can serve as an important source of fresh water as well as a cause of flooding. Various missions have been proposed by NASA and ESA for the purpose of remote sensing of snow. Rats initially fear humans which can lead to negative affect, poor welfare, and difficult handling. Also, modeling and measuring positive affect states in rats can pose an additional challenge. Heterospecific play, or “tickling,” is a handling habituation technique that mimics rat rough-and-tumble play that is being used to study positive affect. It can also be used to reduce fear of human, improve welfare, and elicit a positive affect state. Advancements in computer vision are still not reliable enough for detecting video content including humans and their actions. Microtask crowdsourcing on task markets such as Amazon Mechnical Turk and Upwork can bring humans into the loop. However, engaging crowd workers to annotate non-public video footage risks revealing the identities of people in the video who may have a right to anonymity.

The fire-safety properties of these fuels, however, are unknown. Especially, the flammability limit of a fuel is a crucial parameter. There are various methods used by state agencies to process permit requests; the Indiana Department of Transportation employs a screening process that consists of the Tennessee Formula (allowable weight is determined and checked, for every axle group) in combination with the Gross Vehicle Weight limit. Invasive Burmese pythons are established in the Everglades and are altering the ecology of southern Florida. Their distribution in Florida is expanding northward into more urbanized and fragmented habitats. An understanding of the suitability of habitat throughout southern Florida for Burmese pythons and their interaction with Florida’s landscapes through movement behavior is vital for predicting the python’s ability to persist in habitats outside of the Everglades. In this thesis, we use ecological modeling to predict habitat suitability and to investigate personality-dependent dispersal.

Chip seals are among the most cost-effective surface treatments available for asphalt pavement preventive maintenance. Chip sealing typically consists of covering a pavement surface with asphalt emulsion into which aggregate chips are embedded. The asphalt emulsion cures through the evaporation of water, which helps to provide mechanical strength for the chip seal. Ultimately, the curing process enables the emulsion to adhere to the pavement while keeping the aggregate chips in place. This study investigated how brand equity was perceived on the Purdue University Residences’ Facebook page by applying a user experience method. From a review of previous literature, Website Experience Analysis was identified and performed to evaluate brand equity. This study addressed and explored various themes throughout the data. The results showed how page content and user interactions within a Facebook page influence participants’ perceptions of brand equity.

Aluminized ammonium perchlorate composite propellants (APCP) form large molten agglomerated particles that can result in poor combustion performance and increased two-phase flow losses. Quantifying agglomerate size distributions is important for assessing these losses for different types of aluminum fuels that can help improve rocket performance. It is highly desirable to measure particle sizes in-situ using non-intrusive optical methods, rather than conventional particle collection, which can have large uncertainties. The dopamine transporter (DAT) is a monoamine transporter that regulates dopamine (DA) neurotransmission by clearing DA from the synapse. DAT is acted on by a number of psychostimulant drugs, which block reuptake and potentiate DA signaling. Several inhibitors of DAT, both amphetamines and cathinones, also cause reversal of transport. Previous research has shown the importance of a conserved salt bridge in another monoamine transporter, the serotonin transporter, in amphetamine-induced efflux. In our studies here, we engineered a mutant Drosophila melanogaster DAT (dDAT D475N) designed to interrupt this salt bridge and examine the effects on amphetamine- and cathinone-induced efflux in vitro. …

Currently there is very little systematic work that quantifies the performance of energetic materials in terms of thrust or heat deposition applied to electronic circuits. A better understanding of the interactions between nano-scale energetic materials and electronic systems, as a function of stoichiometry is needed for enhanced defeat. Both of these needs are addressed in this research. Formulations of Al-CuO and Al-Bi2O3 nano-thermites were prepared at different equivalence ratios and selectively deposited onto silicon substrates and thrust and heat deposition of these materials was quantified. Liquid metal nanoparticles can be used to produce stretchable electronic devices. Understanding the mechanical properties of liquid metal nanoparticles is crucial to optimizing their use in various applications, especially printing of flexible, stretchable electronics. Smaller nanoparticles are desired for high-resolution printing and compatibility with existing scalable manufacturing methods; however, they contain less liquid metal and are more difficult to rupture than larger particles, making them less desirable for post-processing functionality.
