Sequence Capture And Next-Generation Sequencing Of Ultraconserved Elements In A Large-Genome Salamander, Catherine E. Newman, Christopher C. Austin

© 2016 John Wiley & Sons Ltd Amidst the rapid advancement in next-generation sequencing (NGS) technology over the last few years, salamanders have been left behind. Salamanders have enormous genomes—up to 40 times the size of the human genome—and this poses challenges to generating NGS data sets of quality and quantity similar to those of other vertebrates. However, optimization of laboratory protocols is time-consuming and often cost prohibitive, and continued omission of salamanders from novel phylogeographic research is detrimental to species facing decline. An important result of Arkhipov-Bezrukavnikov-Ginzburg relates constructible sheaves on the affine Grassmannian to coherent sheaves on the dual Springer resolution. In this paper, we prove a positive-characteristic analogue of this statement, using the framework of 'mixed modular sheaves' recently developed by the first author and Riche. As an application, we deduce a relationship between parity sheaves on the affine Grassmannian and Bezrukavnikov's 'exotic t-structure' on the Springer resolution.

Comparison of plate convergence with the timing and magnitude of upper crustal shortening in collisional orogens indicates both shortening deficits (200–1700 km) and significant (10–40%) plate deceleration during collision, the cause(s) for which remains debated. The Greater Caucasus Mountains, which result from postcollisional Cenozoic closure of a relict Mesozoic back-arc basin on the northern margin of the Arabia-Eurasia collision zone, help reconcile these debates. Here we use U-Pb detrital zircon provenance data and the regional geology of the Caucasus to investigate the width of the now-consumed Mesozoic back-arc basin and its closure history. …

Does Leaf Shedding Protect Stems From Cavitation During Seasonal Droughts? A Test Of The Hydraulic Fuse Hypothesis, Brett T. Wolfe, John S. Sperry, Thomas A. Kursar

During droughts, leaves are predicted to act as ‘hydraulic fuses’ by shedding when plants reach critically low water potential (Ψplant), thereby slowing water loss, stabilizing Ψplant and protecting against cavitation-induced loss of stem hydraulic conductivity (Ks). We tested these predictions among trees in seasonally dry tropical forests, where leaf shedding is common, yet variable, among species. We tracked leaf phenology, Ψplant and Ks in saplings of six tree species distributed across two forests. Species differed in their timing and extent of leaf shedding, yet converged in shedding leaves as they approached the Ψplant value associated with a 50% loss of …

A recent study reported considerable sediment trapping by three large channel bars downstream 18–28 km of the Mississippi–Atchafalaya River diversion (commonly known as the Old River Control Structure, ORCS) during the 2011 Mississippi River flood. In this study, we analyzed 3-decadal morphological changes of the 10-km river channel and the three bars to elucidate the long-term effects of river engineering including diversion, revetment and dike constructions. Satellite images captured between 1985 and 2015 in approximate 5-year intervals were selected to estimate the change of channel morphology and bar surface area. Development of devices through design, prototyping, testing, and fabrication is especially necessary for enhancement of research and eventual application in cryobiology. The advent of 3-dimensional printing offers unique opportunities for this process, given that the materials involved are suitable for use in cryogenic temperatures. We report herein that 3-D printing with polylactic acid (PLA) thermoplastic is ideally suited for cryobiology device development. Devices that are designed and standardized in open-source fashion can be electronically distributed and created locally on increasingly affordable 3-D printers, and can accelerate cryobiology findings and improve reproducibility of results.

Effective conservation measures for endangered species require basic knowledge of habitat use and critical environmental variables influencing the species' occurrence. However, setting priorities may be difficult when multiple endangered species inhabit the same area and differ in habitat use. This study characterized the physical and chemical environment at 30 sites along a 2.5 km stretch of stream associated with the Baños del Azufre hydrogen sulphide spring complex, home to the only population of the widemouth gambusia (Gambusia eurystoma) in existence. Also present in the spring is Poecilia sulphuraria, a narrowly endemic and endangered species known from only a few populations. …

Background: Salinity is one of the many abiotic stresses limiting rice production worldwide. Several studies were conducted to identify quantitative trait loci (QTLs) for traits associated to salinity tolerance. However, due to large confidence interval for the position of QTLs, utility of reported QTLs and the associated markers has been limited in rice breeding programs. The main objective of this study is to construct a high-density rice genetic map for identification QTLs and candidate genes for salinity tolerance at seedling stage. We use an ab initio B3LYP approximation to perform a quantitative analysis of the electronic and structural properties of hydrogen terminated (100), (110), and (111) diamond surfaces. We also study the charge transfer produced by the adsorption of a number of molecular species, including NO2, NO, and O3. Our results shed light on the understanding of hydrogenated diamond surface conductivity and demonstrate that combination of surface termination and molecular species is key to control the hole doping on diamond. West Nile virus (WNV) is a neurotropic ssRNA flavivirus that can cause encephalitis, meningitis, and death in humans and mice. Human TLR7 and TLR8 and mouse TLR7 recognize viral ssRNA motifs and induce antiviral immunity. However, the role of mouse TLR8 in antiviral immunity is poorly understood. In this article, we report that TLR8-deficient (Tlr8−/−) mice were resistant to WNV infection compared with wild-type controls. Efficient WNV clearance and moderate susceptibility to WNV-mediated neuronal death in Tlr8−/− mice were attributed to overexpression of Tlr7and IFN-stimulated gene-56 expression, whereas reduced expression of the proapoptotic gene coding Bcl2-associated X protein was observed. …

In response to the challenge of maximising the effectiveness of one-shot information literacy (IL) sessions, library faculty at Lehman College experimented with the flipped classroom model. This research paper reports the results of a multi-semester quantitative study of the flipped classroom in business management and education one-shot sessions. Researchers explored two research questions: Do students in a flipped session demonstrate greater knowledge before their session than students in a control session? and Do flipped and control students demonstrate significant, positive improvement in knowledge after their session? The researchers used pre- and post-tests to evaluate two crucial aspects of the flipped …

Flipped teaching is a pedagogical model in which the roles of the instructor and the students in a flipped context are redefined. Within this unique pedagogical context, researchers suggest that, in order to maximize the learning process for students, assessment should follow a student-centered approach (Talbert, 2015; Honeycutt & Garrett, 2014).Utilising assessment as a learning tool through layering and scaffolding in the flipped context engages students in the learning process, encourages continuous assessment of student learning, creates opportunities for implementing critical thinking, helps students gain a deeper understanding of concepts, allows formative feedback and eventually yields improved outcomes. This article offers an impressionistic look back over the past five decades, from 1968 to 2016, in Catholic theology in the United States. At the heart of this story are Christology, the world of grace, and their relationship. This memoir unfolds in three parts: “Running on Empty, 1968–1980”; “Jesus and the World of Grace, 1980–2016”; “Can Liberal Catholics Come Back?” It identifies the most neuralgic question left to us from this period: How is Christ related to the world of grace?

Smart grid is an intelligent cyber physical system (CPS). The CPS generates a massive amount of data for efficient grid operation. In this paper, a big data-driven, cloud-based information and communication technology (ICT) framework for smart grid CPS is proposed. The proposed ICT framework deploys hybrid cloud servers to enhance scalability and reliability of smart grid communication infrastructure. Because the data in the ICT framework contains much privacy of customers and important data for automated controlling, the security of data transmission must be ensured. Some individuals and groups seem to adopt emerging ideas and innovations more readily than others. Since any emerging innovation or technology inherently comes with a higher degree of uncertainty and risk, the group or individual must deal with the anxiety created by innovative behavior. Individuals who are less anxious and risk averse may tend to adopt innovations more easily than others. The landscape throughout the range of the Eastern Box turtle (Terrapene carolina carolina) has been altered significantly since the late 1880s by habitat fragmentation. In this study, the spatial ecology and seasonal movement patterns of the Eastern Box turtle in contrasting habitat types are investigated. Eastern Box turtles had home ranges that averaged (mean = 3.77 ± 2.82 ha, minimum convex polygon) in size among individuals. There was no significant difference between mean annual home range size and movement patterns between males and females; however, home range size for male turtles was larger during the summer versus spring activity season …

Chloroplast Dna Phylogenetics Of The North American Chestnuts And Chinquapins (Castanea Mill., Fagaceae), Matthew T. Perkins

Evolutionary relationships and genetic structure of the North American Castanea were investigated using chloroplast DNA sequence data. Six plastome loci were PCR-amplified and sequenced in 77 accessions representing the three currently recognized North American Castanea species. Diagnostic morphological character states and a unique haplotype were shared among C. pumila and a plant tentatively identified as C. dentata in one sympatric site, suggesting past hybridization and chloroplast capture. Surprisingly, the cpDNA phylogeny did not agree with previous taxonomic treatments. The inability to distinguish between deep coalescence and interspecific hybridization as the causes of haplotype sharing makes phylogenetic reconstruction of the North …
