There has been a dearth of information that explains group beliefs about chronic diseases. The overall aim of this study was to use both qualitative and quantitative methods to understand consensus in illness beliefs among diabetic and hypertensive patients. The cultural consensus and common-sense model of self-regulation were used to provide a theoretical framework for the findings. Older diabetic and hypertensive adults recruited from the community took part in a semi-structured interview and answered questionnaires. Results indicated that all participants shared a single cultural belief regarding these chronic illnesses. Contrary to expectations, there were no differences in cohesive beliefs between …

Current bacterial chemotherapy faces an overwhelming reduction in efficacy due to an increase in bacterial resistance; resulting in a higher clinical demand for novel therapeutics and the investigation for their respective targets. Vitamin pathways exogenous to the human body represent an attractive target for drug development since the enzymatic machinery is solely and ubiquitously to many infectious agents. HMP kinase [E.C 2.7.1.49] is a key enzyme in the thiamine metabolism. It catalyzes two subsequent phosphorylations of 4-amino-5-hydroxymethyl-2-methylpyrimidine (HMP), making it indispensable for microbial survival. Disinfection by-products (DBPs) are a significant concern in drinking water quality. Haloacetic acids, which are generated from disinfection processes, are one of the major groups in disinfection by-products. Because they induce potential harmful effects for human health, the US government has already established regulations to control the levels of haloacetic acids in drinking water. In order to control the presence of haloacetic acids in drinking water and monitor other potential harmful compounds, an appropriate analytical method is needed. Globally, the human population continues to grow and move into urban areas causing a range of effects on local ecosystems. Elevated temperatures, altered hydrology, and higher concentrations of atmospheric pollutants in urban environments create novel conditions for resident organisms. My research focused on one particular aspect of human alteration to urban environments – nitrogen (N) deposition – and its effects on native plants across an urban to rural gradient in the southern Great Plains, North Texas (NTX), USA. Despite recent advances unraveling mechanisms of host–pathogen interactions in innate immunity, the participation of purinergic signaling in infection-driven inflammation remains an emerging research field with many unanswered questions. As one of the most-studied oral pathogens, Porphyromonas gingivalis is considered as a keystone pathogen with a central role in development of periodontal disease. This pathogen needs to evade immune-mediated defense mechanisms and tolerate inflammation in order to survive in the host. Sexually transmitted infections affect hundreds of millions of worldwide. Both human immunodeficiency virus (HIV-1 and -2) and herpes simplex virus-2 (HSV-2) remain incurable, urging the development of new prevention strategies. While current prophylactic technologies are dependent on strict user adherence to achieve efficacy, there is a dearth of delivery vehicles that provide discreet and convenient administration, combined with prolonged-delivery of active agents. Transgender populations demonstrate significant sexual diversity, with over three-fourths (77%) of transgender individuals identifying as sexual minorities (Grant et al., 2011); however, a majority of research on sex, sexuality, and sexual practice has focused exclusively on cisgender and heterosexual individuals and, perhaps more importantly, addressed these issues a heteronormative and cisnormative perspective (Nichols, 2014). Global augmentation of protein O-GlcNAcylation occurs in response to a myriad of stressors and confers a survival advantage at the cellular level. This protective phenomenon has been demonstrated to mediate cardioprotection through various in vitro and in vivo studies during ischemia-reperfusion, myocardial infarction, and oxidative stress; however, relatively little is known of the regulation of protein O-GlcNAcylation. Protein O-GlcNAcylation is regulated by two antagonistic enzymes, namely, O-GlcNAc transferase (OGT) and O-GlcNAcase (OGA). Ablation of cardiomyocyte OGT, the enzyme that catalyzes the addition of O-GlcNAc to proteins, exacerbates cardiac dysfunction during infarct-induced heart failure (HF). This dissertation will explore the potential of a semi-empirical Hamiltonian, developed by the research group at the University of Louisville, in predicting the existence of new families of low-dimensional boron nanostructures based on icosahedral α-B12 clusters, and in tuning the band gaps of h-BN sheets with graphene domains and holey graphene. This semi-empirical Hamiltonian models electron-electron and electron-ion interactions using environment-dependent (ED) functions, and ion-ion interactions via usual pairwise terms. Peptoanaerobacter stomatis is a newly appreciated taxon, present in high numbers in oral biofilms from periodontal patients. However, little is known about this organism’s potential virulence and its interaction with the host immune response. Therefore, to better understand the role of P. stomatis in periodontitis, we studied its interactions with neutrophils. Human neutrophils were challenged with P. stomatis strain CM2 or S. aureus AF488 (positive control) for 30 min. Bacterial internalization was examined by ImageStreamX imaging flow cytometry. Critical Limb ischemia (CLI) is a serious manifestation of peripheral artery disease. Advanced CLI patients are poor candidates for vascular surgeries. Numerous studies have shown that hypoxia inducible factor 1α (HIF1-α) plays an important role in recovery from murine hindlimb ischemia (HLI). HIF1-α is regulated by oxygen dependent prolyl hydroxylases (PHDs). Previous studies have shown that inhibition of PHDs by metal quenchers and viral delivery of HIF1-α improves blood flow to the ischemic limb. The goal of this project was to examine the role of endothelin-1 (ET-1) as a mediator in the pathway between air pollution exposure and the development of vascular injury. A human cohort and male mice (C57BL/6 and ecSOD-Tg) were used to evaluate changes in the ET-1 system in response to exposures of fine particulate matter (PM2.5). Human ET-1 levels were significantly associated with environmental factors and markers of vascular change, but were decreased with increased PM2.5. No association was seen between ET-1 and endothelial progenitor cells (EPCs) except for EPC-4, possibly indicating a regulatory relationship with this specific population. In …

Invasive species pose a threat to aquatic and terrestrial ecosystems by decreasing biodiversity (Didham et al. 2005). Amur bush honeysuckle (Lonicera maackii) has typically reduced native plant diversity and altered animal communities by influencing animal abundance and activity (Collier and Vankat 2002). This study was intended to determine whether honeysuckle density or other characteristics of forested stands influence avian diversity, whether impacts are seasonally dependent, and whether correlations exist between attributes of forested stands and honeysuckle density. Cisplatin is a nephrotoxic chemotherapeutic that causes acute kidney injury (AKI) in 30% of patients. Although recovery can occur after one episode of cisplatin-induced AKI, studies have indicated multiple episodes may lead to the development of chronic kidney disease (CKD), an irreversible disease with no current treatments. The standard mouse model of cisplatin-induced AKI consists of one, high dose of cisplatin (> 20 mg/kg) that is lethal to the animal three days later. This model doesn’t accurately reflect the repeated dosing regimen patients receive, and doesn’t allow for long-term outcome studies of pathologies associated with CKD. We have developed a …

Our understanding of worry in children and adolescents has been increasingly enhanced over the past decade through the downward extension of adult cognitive models. Although our knowledge about the cognitive factors that place children at risk for worry has grown, little is known about these processes within African American youth. The epidermal growth factor receptor (EGFR) is a receptor tyrosine kinase (RTK) that is an integral component of proliferative signaling. When activated by a ligand at the plasma membrane, EGFR undergoes clathrin-mediated endocytosis. This spatial regulation of the receptor is an important regulator of receptor expression as it mediates its degradation. Endocytosis also has implications on EGFR downstream signaling, though the details are not fully understood. Ras is the most commonly activated oncogene in human cancer. Activated Ras drives cell growth and proliferation by activating multiple mitogenic signaling pathways. However, Ras also has the paradoxical ability to promote anti-growth, pro-apoptotic, and pro-senescent signaling. The signaling pathways of many of these biological effectors remain poorly defined. One group of proteins capable of promoting Ras-induced apoptosis and cell cycle arrest is the RASSF family of tumor suppressors. Background: The numerous virulence factors of Porphyromonas gingivalis are essential for the organism to invade host tissues while escaping immune responses [1, 2]. Some of these factors, like major proteases referred to as gingipains, are secreted by a unique secretion system called PerioGate or Type IX Secretion System (T9SS). T9SS consists of a number of membrane and periplasmic proteins, which provide transportation only for proteins that possess a conserved C-terminal domain (CTD). Objective: This project sought to understand the gene regulatory networks that drive parotid salivary gland acinar cells to terminally differentiate, and drive expression of terminal differentiation genes in dedifferentiated ParC5 cells. Methodology: Laser capture microdissection was used to isolate acinar cells at multiple time points during differentiation. This important step allowed us to measure gene expression in a single and important cell type. Persistent infection (PI) with Bovine Viral Diarrhea Virus (BVDV) is a major source of economic loss for the livestock industry. Persistent infection occurs when a dam is exposed to the virus during 42 to 125 days of gestation and the virus is passed to the fetus without an adaptive immune response. These PI calves serve as continuous sources of infection to others. Supplementation of zilpaterol hydrochloride (ZH; Zilmax®) to cattle has been suggested to have a negative impact on well-being of cattle. The objective of this study was to evaluate the hematological and serum biochemical profiles in feedlot heifers supplemented with ZH in a minimal stress environment.
