Therapeutic proteins are becoming one of the fastest growing classes of drugs on the market. These large biologically-synthesized macromolecules are typically administered intravenously and are dependent upon cellular receptors and other active uptake mechanisms for delivery into tissues and cells. There is significant interest in developing cell-based assays to assess kinetics and mechanisms of endocytosis and transcytosis of large therapeutic proteins across various cell layers. An integrated vectorcardiogram (VCG) device having 3 pairs of electrodes placed in three orthogonal directions with the capability to continuously measure the electrical activity of the heart. All of the electrode pairs may be contained within a single miniaturized housing thereby eliminating the need for cumbersome wires required with current 12-lead ECG systems, while providing the same amount and quality of information as the 12-lead ECG system. The integrated VCG device also includes a communications function to allow the data collected by the electrodes to be transmitted wirelessly to a remote device, such as a pacemaker, to control its function …

The South American Cactus Moth, Cactoblastis cactorum (Berg), an invasive insect herbivore, has similar feeding habits and life history as a native Florida cactus moth, Melitara prodenialis Walker; however, regional Opuntia species only defend against M. prodenialis. Extracts of C. cactorum host plants and M. prodenialis host plants were compared to methyl jasmonate (MeJA)-treated Opuntia humifusa and MeJA-treated O. humifusa hairy root cultures using high performance thin layer chromatography. MeJA and C. cactorum treatments led to a reduction in tissue concentrations of terpenoid or phenolic compounds; however, significant increases in seven putative terpenoid compounds were observed in hairy root culture …

Pancreatic adenocarcinoma is one of the most leading causes of cancer-related deaths worldwide. Although recent advances provide various treatment options, pancreatic adenocarcinoma has poor prognosis due to its late diagnosis and ineffective therapeutic multimodality. Gemcitabine is the effective first-line drug in pancreatic adenocarcinoma treatment. However, gemcitabine chemoresistance of pancreatic adenocarcinoma cells has been a major obstacle for limiting its treatment effect. Our study found that p70S6K1 plays an important role in gemcitabine chemoresistence. MiR-145 is a tumor suppressor which directly targets p70S6K1 for inhibiting its expression in pancreatic adenocarcinoma, providing new therapeutic scheme. Regulation of order, such as orientation and conformation, drives the function of most molecular assemblies in living cells but remains difficult to measure accurately through space and time. We built an instantaneous fluorescence polarization microscope, which simultaneously images position and orientation of fluorophores in living cells with single-molecule sensitivity and a time resolution of 100 ms. We developed image acquisition and analysis methods to track single particles that interact with higher-order assemblies of molecules. We tracked the fluctuations in position and orientation of molecules from the level of an ensemble of fluorophores down to single fluorophores. We tested our system …

BACKGROUND:
ClinicalTrials.gov (CT) is an increasingly important resource for systematic reviewers attempting to identify published and unpublished clinical studies. In addition to clinical studies, however, some searches of the CT database also return systematic reviews (SRs). When I inquired about the SRs appearing in the results, the NLM Help Desk responded that “We do not recommend that systematic reviews be entered in ClinicalTrials.gov, since we only want the results of a clinical trial entered once. However, we will not refuse them if they are entered.” I wanted to find out how many SRs are included, describe their characteristics, and suggest …

The cognitive and social dexterity of humans is attributed to the neocortex, an area of the brain well accepted to elevate us beyond the mental processing of all other species. The cognitive ability of the cerebellum however, remains one of the most misunderstood. Autism spectrum disorders (ASDs), with a prevalence of 1 in 68 children, lack clear empirical explanations for abnormal neuropathology and potential neuronal disconnections influenced by the cerebellum. The current evaluation takes a comprehensive look at how the cerebellum, pre-frontal cortex, and basal ganglia are interconnected and influenced by each other as relevant to ASDs. With a better …

Hyperspectral Reflectance Signature Protocol For Predicting Subsurface Bottom Reflectance In Water: In-Situ And Analytical Methods, Charles R. Bostater, Tyler A. Rotkiske, Taylor Scott Oney

In-situ measurement of bottom reflectance signatures and bottom features in water are used to test an analytical based irradiance model protocol. Comparisons between predicted and measured bottom reflectance signatures are obtained using measured hyperspectral remote sensing reflectance signatures, water depth and water column constituent concentrations. Analytical solutions and algorithms are used to generate synthetic signatures of different bottom types. The analytical methodology used to simulated bottom reflectance contains offset and bias that can be corrected using spectral window based corrections. Example results are demonstrated for application to coral species, submerged aquatic vegetation and a sand bottom type. Spectral windows are …

Hyperspectral measurements of the water surface of urban coastal waters are presented. Oblique bidirectional reflectance factor imagery was acquired made in a turbid coastal sub estuary of the Indian River Lagoon, Florida and along coastal surf zone waters of the nearby Atlantic Ocean. Imagery was also collected using a pushbroom hyperspectral imager mounted on a fixed platform with a calibrated circular mechatronic rotation stage. Oblique imagery of the shoreline and subsurface features clearly shows subsurface bottom features and rip current features within the surf zone water column. Neurons in vertebrate central nervous systems initiate and conduct sodium action potentials in distinct subcellular compartments that differ architecturally and electrically. Here, we report several unanticipated passive and active properties of the cerebellar granule cell’s unmyelinated axon. Whereas spike initiation at the axon initial segment relies on sodium channel (Nav)-associated fibroblast growth factor homologous factor (FHF) proteins to delay Nav inactivation, distal axonal Navs show little FHF association or FHF requirement for high-frequency transmission, velocity and waveforms of conducting action potentials. In addition, leak conductance density along the distal axon is estimated as o1% that of somatodendritic membrane. The faster …
