PURPOSE

To characterize outer retina structure in best vitelliform macular dystrophy (BVMD) and to determine the effect of macular lesions on overlying and adjacent photoreceptors.

METHODS

Five individuals with BVMD were followed prospectively with spectral domain optical coherence tomography and confocal and nonconfocal split-detector adaptive optics scanning light ophthalmoscopy (AOSLO). The AOSLO cone photoreceptor mosaic images were obtained within and around retinal lesions. Cone density was measured inside and outside lesions. In 2 subjects, densities were compared with published measurements acquired ∼2.5 years before. One subject was imaged 3 times over a 5-month period.

RESULTS

The AOSLO imaging demonstrated that …

Executive Function, Disability, And Agency, Kevin Timpe Oct 2016

Executive Function, Disability, And Agency, Kevin Timpe

University Faculty Publications and Creative Works

This paper considers how a number of particular disabilities can impact agency primarily by affecting what psychologists refer to as ‘executive function.’ Some disabilities, I argue, could decrease agency even without fully undermining it. I see this argument as contributing to the growing literature that sees agency as coming in degrees. The first section gives a broad outline of a fairly standard approach to agency. The second section relates that framework to the existing literature, which suggests that agency comes in degrees. The third section considers the psychological literature on executive function with a particular focus on how aspects of …

Reasonable Accommodations And The Ada Amendments’ Overlooked Potential, Jeannette Cox Oct 2016

Reasonable Accommodations And The Ada Amendments’ Overlooked Potential, Jeannette Cox

School of Law Faculty Publications

There is “a dearth of precedent” outlining the scope of the Americans with Disabilities Act’s reasonable accommodations provision. The “little precedent” available “remains severely underdeveloped,” “in a state of chaos,” and leaves “many issues unresolved.” Circuit splits abound. For example, courts widely differ in their perspectives about whether the ADA requires employers to permit employees with disabilities to work from home. Similarly, in circumstances in which an employee with a disability can no longer do his or her current job, courts differ on the question of whether the ADA requires the employer to prefer the employee with a disability for …

Development And Implementation Of High Fidelity Human Models For The Investigation Of Blast And Non-Penetrating Projectile Impact, Candice F. Cooper

Mechanical Engineering ETDs

Military operations abroad have highlighted the effects of several types of physical traumas including traumatic brain injury and behind armor blunt trauma. While previous approaches toward understanding and mitigating trauma caused by blast or blunt impact relied upon physical testing of animal subjects, post mortem human subjects, or human tissue surrogates, recent advances in computational capability have spurred a growing area of research in computational investigations into wound injury and its mitigation. The development of high-fidelity human torso and head-neck-torso models are presented here. These models are employed in blast and non-penetrating projectile impact simulations in order to demonstrate the …

A Francisella-Like Endosymbiont In The Gulf Coast Tick Evolved From A Mammalian Pathogen, Jonathan G. Gerhart, Abraham S. Moses, Rahul Raghavan

Biology Faculty Publications and Presentations

Ticks (order Ixodida) vector pathogenic bacteria that cause diseases in humans and other mammals. They also contain bacteria that are closely related to pathogens but function as endosymbionts that provide nutrients that are missing from mammalian blood—their sole food source. For instance, mammalian pathogens such as Coxiella burnetii and Francisella tularensis, as well as Coxiella-like and Francisella-like endosymbionts (CLEs and FLEs, respectively) occur in ticks worldwide. However, it is not clear whether the pathogens evolved from symbionts or symbionts from pathogens. Recent studies have indicated that C. burnetii likely originated from a tick-associated ancestor, but the origins …

Proline (Pro) accumulation is one of the most prominent changes in plant metabolism during drought and low water potential; however, the regulation and function of Pro metabolism remain unclear. We used a combination of forward genetic screening based on a Proline Dehydrogenase1 (PDH1) promoter-luciferase reporter (PDH1pro:LUC2) and RNA sequencing of the Pro synthesis mutant p5cs1-4 to identify multiple loci affecting Pro accumulation in Arabidopsis (Arabidopsis thaliana). Two mutants having high PDH1pro:LUC2 expression and increased Pro accumulation at low water potential were found to be alleles of Cytochrome P450, Family 86, Subfamily A, Polypeptide2 (CYP86A2) and Long Chain Acyl Synthetase2 (LACS2), …

Physiological Reports published by Wiley Periodicals, Inc. on behalf of the American Physiological Society and The Physiological Society. A volume-regulated anion channel (VRAC) has been electrophysiologically characterized in innumerable mammalian cell types. VRAC is activated by cell swelling and mediates the volume regulatory efflux of Cl− and small organic solutes from cells. Two groups recently identified the mammalian leucine-rich repeat containing protein LRRC8A as an essential VRAC component. LRRC8A must be coexpressed with at least one of the other four members of this gene family, LRRC8B-E, to reconstitute VRAC activity in LRRC8−/− cells. LRRC8 genes likely arose with the origin …

Patients with tricho-dento-osseous (TDO) syndrome, an ectodermal dysplasia caused by mutations in the homeodomain transcription factor DLX3, exhibit enamel hypoplasia and hypomineralization. Here we used a conditional knockout mouse model to investigate the developmental and molecular consequences of Dlx3 deletion in the dental epithelium in vivo. Dlx3 deletion in the dental epithelium resulted in the formation of chalky hypomineralized enamel in all teeth. Interestingly, transcriptomic analysis revealed that major enamel matrix proteins and proteases known to be involved in enamel secretion and maturation were not affected significantly by Dlx3 deletion in the enamel organ. In contrast, expression of several ion …

The research in this study investigated two core focal points: the possible shift in a teacher’s transformational practices after participation in professional development on Essential Questions (McTighe & Wiggins, 2013) and whether the implemented transformational practices led to a possible difference in student questioning skills in grades two through eight on the Measure of Questioning Skills, student achievement in STAR Math, and STAR Reading in …

Characterization Of Histological Changes In The Microvasculature Of Rat Skeletal Muscle After Spinal Cord Injury, Sally Lin Oct 2016

Characterization Of Histological Changes In The Microvasculature Of Rat Skeletal Muscle After Spinal Cord Injury, Sally Lin

Master's Theses (2009 -)

The purpose of this study was to determine whether there are histological changes in the microvasculature of rat skeletal muscle following chronic spinal cord injury both above and below the level of injury. This study is important because microvascular structure likely impacts muscle performance and cardiovascular health. To the best of our knowledge, this is the only study to investigate microvascular structure within rat skeletal muscle after spinal cord injury. We hypothesized structural remodeling would occur in both the myofibers and microvasculature, which would then manifest in differences in myofiber cross sectional area and microvascular diameter, wall thickness, wall to …

Optimization Of The Bowtie Gap Geometry For A Maximum Electric Field Enhancement, Tsenguun Byambadorj Oct 2016

Optimization Of The Bowtie Gap Geometry For A Maximum Electric Field Enhancement, Tsenguun Byambadorj

Master's Theses (2009 -)

Optimization of the geometry of a metallic bowtie gap at radio frequency is presented in this thesis. Since the design and fabrication of a plasmonic device (nanogap) at nanoscale is challenging, the results of this study can be used to estimate the best design parameters for nanogap structure. The geometry of the bowtie gap including gap size, tip width, metal thickness, and tip angle are investigated at macroscale to find the maximum electric field enhancement across the gap. This thesis focuses on the simulation portion of a work that consists of experimental and simulation platforms. The simulation platform is created …

Plasticized Polymer Coatings For Sh-Saw Sensors For High Sensitivity And Long-Term Monitoring Of Btex Analytes In Liquid Phase, Pintu Adhikari Oct 2016

Plasticized Polymer Coatings For Sh-Saw Sensors For High Sensitivity And Long-Term Monitoring Of Btex Analytes In Liquid Phase, Pintu Adhikari

Master's Theses (2009 -)

BTEX compounds (benzene, toluene, ethylbenzene, and xylene) are constituents of crude oil and hazardous to human health. Among them, benzene has the lowest maximum contaminant level for drinking water because of its carcinogenicity. Spills or leakage from underground storage tanks or hazardous waste sites can contaminate nearby groundwater with these volatile organic compounds. Therefore, it is very important to detect the presence of BTEX contamination as early as possible in order to start the remediation process and maintain a healthy environment. To develop an in-situ continuous monitoring sensor system, shear horizontal surface acoustic wave (SH-SAW) sensor devices are being investigated …

Baby Dust To All! Identity Construction In Two-Week Wait Online Discussion Forums, Danielle R. Derose

Master's Theses (2009 -)

Women with a self-identified infertility status sometimes choose to address this status by seeking medical intervention. There are a variety of methods available when attempting to conceive using medical treatments, with the choice heavily dependent on the health of each partner. A common first approach by reproductive endocrinologists is that of intrauterine insemination, or IUI. Women undergoing IUI invest significant time and money into the process and often must undergo procedures or take medication that can be enormously distressing. Once the IUI is complete, the woman must wait an emotional two weeks before she finds out if she is pregnant …

Climate Change Impacts On Freshwater Wetland Hydrology And Vegetation Cover Cycling Along A Regional Aridity Gradient, Philip A. Fay, Glenn R. Guntenspergen, Jennifer H. Olker, W. Carter Johnson

Natural Resource Management Faculty Publications

Global mean temperature may increase up to 6°C by the end of this century and together with precipitation change may steepen regional aridity gradients. The hydrology, productivity, and ecosystem services from freshwater wetlands depend on their future water balance. We simulated the hydrology and vegetation dynamics of wetland complexes in the North American Prairie Pothole Region with the WETLANDSCAPE model. Simulations for 63 precipitation × temperature combinations spanning 6°C warming and −20% to +20% annual precipitation change at 19 locations along a mid-continental aridity gradient showed that aridity explained up to 99% of the variation in wetland stage and hydroperiod …

Mechanism Of Rifampicin Inactivation In Nocardia Farcinica, Heba Abdelwahab, Julia S.Martin Del Campo, Yumin Dai, Camelia Adly, Sohby El-Sohaimy, Pablo Sobrado

Chemistry Faculty Research & Creative Works

A novel mechanism of rifampicin (Rif) resistance has recently been reported in Nocardia farcinica. This new mechanism involves the activity of rifampicin monooxygenase (RifMO), a flavin-dependent monooxygenase that catalyzes the hydroxylation of Rif, which is the first step in the degradation pathway. Recombinant RifMO was overexpressed and purified for biochemical analysis. Kinetic characterization revealed that Rif binding is necessary for effective FAD reduction. RifMO exhibits only a 3-fold coenzyme preference for NADPH over NADH. RifMO catalyzes the incorporation of a single oxygen atom forming an unstable intermediate that eventually is converted to 20-N-hydroxy-4-oxo-Rif. Stable C4a-hydroperoxyflavin was not detected by rapid …

Microbiome Sharing Between Reef-Building Corals And Associated Epibiotic Gastropods In French Polynesia, Emily R. Schmeltzer Oct 2016

Microbiome Sharing Between Reef-Building Corals And Associated Epibiotic Gastropods In French Polynesia, Emily R. Schmeltzer

Capstone Projects and Master's Theses

Every macroorganism has a unique association of microbes, also known as a microbiome. This is a complex association in reef-building corals, and research has shown that these microbes are a functional extension of the coral animal itself. Together these components of host, symbiotic zooxanthellae, protists, bacteria, algae, archaea, viruses, and fungi, form what is now known as the coral holobiont. Recent studies have shown that the microbes found in the surface mucus layer of corals play a key role in holobiont ecology. Microbial interactions in the coral surface mucus layer are especially important when considering the increasing prevalence of coral …

Evolutionary Interpretations Of Mycobacteriophage Biodiversity And Host-Range Through The Analysis Of Codon Usage Bias, Laura A. Esposito, Swati Gupta, Fraida Streiter, Ashley Prasad, John J. Dennehy

Publications and Research

In an genomics course sponsored by the Howard Hughes Medical Institute (HHMI), undergraduate students have isolated and sequenced the genomes of more than 1,150 mycobacteriophages, creating the largest database of sequenced bacteriophages able to infect a single host, Mycobacterium smegmatis, a soil bacterium. Genomic analysis indicates that these mycobacteriophages can be grouped into 26 clusters based on genetic similarity. These clusters span a continuum of genetic diversity, with extensive genomic mosaicism among phages in different clusters. However, little is known regarding the primary hosts of these mycobacteriophages in their natural habitats, nor of their broader host ranges. As such, it …

A preliminary genome sequence has been assembled for the Southern Ocean salp, Salpa thompsoni (Urochordata, Thaliacea). Despite the ecological importance of this species in Antarctic pelagic food webs and its potential role as an indicator of changing Southern Ocean ecosystems in response to climate change, no genomic resources are available for S. thompsoni or any closely related urochordate species. Using a multiple-platform, multiple-individual approach, we have produced a 318,767,936-bp genome sequence, covering >50% of the estimated 602 Mb (±173 Mb) genome size for S. thompsoni. Using a nonredundant set of predicted proteins, >50% (16,823) of sequences showed significant homology …
