Protein adsorption on solid surfaces is a common yet complicated phenomenon that is not fully understood. Self-assembled monolayers have been utilized in many studies, as well-defined model systems for studying protein-surface interactions in the atomic level. Various strategies, including the use of single component SAMs[1, 2], combinations of long and short alkanethiolates with methyl- and hydroxyl- terminal groups[3, 4], and using mixtures of alkanethiolates with similar chain length and varying terminal functional group [5] have been used to effectively control the surface wettability and determine the effect of surface composition and wettability on protein adsorption. Cationic polymers have shown potential as gene delivery vectors due to their ability to condense DNA and protect it from cellular and restriction nucleases. Dendrimers are hyperbranched macromolecules with precisely defined molecular weights and highly symmetric branches stemming from a central core. The nanosize, tunable surface chemistries and ease of surface functionalization has made dendrimers an attractive alternative to conventional linear polymers for DNA delivery applications. Current chemotherapeutics exhibit debilitating side effects as a result of their toxicity to healthy tissues. Reducing these side effects by developing chemotherapeutics with selectivity for cancer cells is an active area of research. Phototherapy is one promising modality for selective treatment, where drug molecules are “turned on” when irradiated with light, reducing damage to healthy tissues by spatially restricting the areas exposed to irradiation. A second approach to improve selectivity is to exploit the differences in cancerous versus healthy cells, such as increased metabolism and/or upregulation of cell surface receptors. Lignin is one of the most abundant and underutilized biopolymers on earth. Primarily composed on three monolignol units (sinapyl, coniferyl, and p-coumaryl alcohol), lignin is formed through a radical pathway resulting in an assortment of linkages, of which the β-O-4 linkage is the most prevalent (up to 60% in some hardwood species). In planta, lignin plays an important role in water transport and in protecting plants from chemical and biological attack. Traditional attempts to depolymerize lignin have focused on the cleavage of β-O-4 linkages via thermal or reductive routes. However these pathways lead to low-value, unstable product mixtures. Colorimetric methods combined with color-changing chemical probes are widely used as simple yet effective tools for identifying and quantifying a wide variety of molecules in solution. For nucleic acids (DNA and RNA), perhaps the most commonly used colorimetric probe is potassium permanganate, which can be used to identify single-stranded pyrimidines (thymine and cytosine) in polymers. Unfortunately, permanganate is not an effective probe for identifying purines (adenine and guanine), especially in the presence of the more reactive pyrimidines. A Qualitative Exploration Of Masculinity Among Community College Males, Marianne Fontes

The purposes of this grounded theory study were to explore how traditional-aged community college males experience masculinity and to examine how they engage with community college support systems. Eleven diverse participants engaged in three interviews each in which they were asked to reflect on their definitions of masculinity as well as their activities on and off campus. Open, axial, and selective coding methods were employed, and a new theory emerged. The new theory suggests that the participants' hesitation to make concrete educational decisions is related to their resistance to manhood. Filth flies are a significant pest at animal production facilities, leading to economic losses that include reductions in animal weight and milk production. Pesticides and biological control organisms are two important components of Integrated Pest Management (IPM) programs against filth flies. Biological control against filth flies frequently involves augmentative releases of pupal parasitoids. This thesis explores computational methods for automatically detecting and quantifying synapses in complex 3D neuronal images. The resulting approach is a novel combination of traditional image processing, machine learning algorithms, and multi-channel comparison methods designed to overcome the unique challenges posed by these images. The methods investigated combine the strengths of each of these components in order to produce an overall method that is capable of fully detecting the synapses in large 3D confocal neuron images with minimal interaction. Human annotation of 3D neuron images remains prohibitively difficult and subjective, and computational analysis tools are highly desirable in the expanding …

Use of chemical signals for sexual communication is found in a large proportion of insects. Pheromones may also act as honest signals of the sender's health and resources. Most research has focused on female-produced pheromones used for attracting mates over large distances or on the role pheromones play in species and mate recognition. Little work has been done exploring male produced pheromones, and how pheromones may be involved in mate assessment. The purpose of the following thesis was to investigate how transwomen (otherwise known locally as waria) of Indonesia, navigate their gender identities within their specific locales in central Java. Particularly, I focus how my respondents do this within Yogyakarta, which I define as a cultural hub where local custom and law known as adat predominates and two other field sites located in the periphery of Yogyakarta. For the past decade, next-generation sequencing (NGS) has been used to undertake genomics-scale projects in molecular biology. This method of sequencing involves randomly fragmenting a sample of nucleic acid and randomly generating millions of short reads. The large number of reads moves the assembly and other analyses to be performed by computer algorithms. As well as sequencing full chromosomes, full RNA extracts can be sequenced to determine levels of gene expression and exon boundaries. This thesis examines a definition of reliability that can be applied to targeted nanoconjugate chemotherapy in cancer and also simulates a stochastic differential equation model to attempt to predict the estimated time until remission when using targeted nanoconjugate chemotherapy utilizing a recursive algorithm to attempt to find the dose of nanoconjugates that will result in remission by an expected time T. Two general assumptions have often been made about folivorous primates: first, that feeding competition is low or absent because leaves are a superabundant and evenly dispersed resource, and as a result, that feeding competition does not constrain the size of folivorous groups. However, many folivorous primates maintain smaller group sizes. In this study, I aimed to examine this 'folivore paradox' by testing the hypothesis that mantled howler monkeys (Alouatta palliata) deplete localized patches of young leaves and therefore experience feeding competition. A butanol fermentation pathway was engineered in Escherichia coli through cloning and chromosomal integration of the butanol genes (thiL, CA_C2873, hbd, crt, bcd, etfA, etfB, adhe1, adhe2, bdhAB) from Clostridium acetobutylicum ATCC824. Initially, a minimal set of butanol pathway genes (thiL, hbd, crt, bcd, etfAB, adhe2) was transcriptionally fused with a pflBp6 promoter and were integrated into the chromosome of E. coli EG10, a strain lacking endogenous competing fermentation pathways, resulting in strain EG60. Anaerobic fermentations showed that EG60 was able to produce 100 muM butanol, suggesting that the butanol pathway genes were effectively expressed for butanol production. Although the …

Oxygen isotope measurements (δ¹⁸O) provide valuable insight into climate conditions. These data are advantageous as paleoclimate proxies, but records are sparse in high latitude waters, where calcareous microfossils are poorly preserved in marine sediments. Over glacial timescales, high latitude regions likely have the widest variability in terms of the δ¹⁸O values of marine waters; therefore, the paucity of data in these regions greatly hinders our ability to reconstruct global climatic and environmental change. There is an immediate need for new antibiotics. Antimicrobial resistance is rising at an alarming rate. In addition, certain select agents are a risk for bioterrorism that necessitate the discovery of new antibiotics. The methylerythritol phosphate (MEP) pathway is a metabolic pathway that produces the isoprenoid precursors, isopentyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP). Pathogenic organisms continue to develop resistance to modern-day antibiotics, while there has been a significant decline in the development of antibiotics over the last 30 years. One particular pathway of interest in the development of new antibiotics is the biosynthesis of isoprenoids. These compounds are essential chemical building blocks in pathogenic organisms such as Plasmodium falciparum, Mycobacterium tuberculosis, and Burkholderia pseudomallei. Due to its capability to ferment biomass-derived pentose and hexose, Escherichia coli is considered a potential biocatalyst for production of bioethanol. Gene deletions and promoter replacements were previously employed to engineer a non-transgenic homoethanol pathway in E. coli [glucose (or 1.2 xylose) => (glycolysis) => 2 pyruvate + 2 NADH => (pyruvate dehydrogenase, aceEF-lpd operon) => 2 acetyl-CoA + 4 NADH => (alcohol dehydrogenase, adhE) => 2 acetaldehyde + 2 NADH => (alcohol dehydrogenase) => 2 ethanol]. This thesis is constructed to delineate mechanisms that influence bystander behavior in the presence of bullying phenomena. Purpose for this study is derived from the shallow empirical history of differentiation between passive outsiders and intervening defenders, particularly when concerned with student populations within the United States. This study included child participants in grades four and five from the Midwestern region. White and bur oak savanna was once the dominant vegetation type in the Midwestern United States prior to European settlement. Within the oak savanna biome were pockets of oak-hickory closed forested sites, usually occurring in mesic areas and on the east side of rivers. Restoration efforts to restore the land back to savanna and closed canopy oak dominated forests are met with varying success. The objective of this thesis was to investigate the culture conditions necessary to promote high cell densities, lipid productivities, and harvesting yields from the locally endemic microalgae Monoraphidium sp. Dek19 in a wastewater environment. The alga was inoculated in final effluent acquired from a local wastewater treatment facility in DeKalb, Illinois. The alga was cultivated in 1L Erlenmeyer flasks at 10°C, 70 μmol m⁻² s⁻¹ light, 14:10 light:dark cycle. Culture agitation was found to be an important condition for algal growth and nutrient removal. Very few student-athletes will go on to become a professional in their sport; therefore, the vast majority will need to pursue a non-sport career once their collegiate athletic eligibility has ended. Research indicates that a strong athletic identity and identity foreclosure coincide with lower levels of career maturity, which contributes to the struggle that student-athletes often face with their post-sport transition. The majority of student-athletes compete in nonrevenue-producing collegiate sports (e.g., cross country and soccer), but this subgroup has not been adequately examined. This thesis examines American gender concepts of masculinity and femininity depicted in six Modern Age superhero comic books. Specifically, it examines how the American gender concepts of hegemonic masculinity, hypersexualized masculinity, emphasized femininity, and hypersexualized femininity are depicted in three Marvel and three DC comic books. This examination found that despite gender differences defined by biological essentialism that legitimates gender inequality, female and male superheroes are much more alike than different through what I term "visionary gender". Habitual bipedal striding locomotion is a distinctive feature of hominins. The human fossil record, however, shows diversity in lower limb anatomy suggesting functional-mechanical differences in locomotion across these fossil taxa. Different forms of bipedality appear in the skeleton of australopithecines such as Australopithecus africanus, Australopithecus afarensis, and Australopithecus sediba. This study seeks to test the hypothesis that australopithecine lower limbs exhibit a mechanically different form of bipedality relative to modern humans, a character that is evident in lower limb flexibility; it is likely that leg flexibility allowed australopithecines to climb trees as well as walk bipedally.
