Modern antiretroviral therapies have provided HIV-1 infected patients longer lifespans and better quality of life. However, several neurological complications are now being seen in these patients due to HIV-1 associated injury of neurons by infected microglia and astrocytes. In addition, these effects can be further exacerbated with opiate use and abuse. One possible mechanism for such potentiation effects of opiates is the interaction of the mu opioid receptor (MOR) with the chemokine receptor CCR5 (CCR5), a known HIV-1 co-receptor, to form MOR-CCR5 heterodimer. Cytochrome p450 (CYP450) enzymes are predominantly involved in Phase I metabolism of xenobiotics. In this study, the CYP450 isoforms involved in xanthotoxol metabolism were identified using recombinant CYP450s. In addition, the inhibitory effects of xanthotoxol on eight CYP450 isoforms and its pharmacokinetic parameters were determined using human liver microsomes. CYP1A2, one of CYP450s, played a key role in the metabolism of xanthotoxol compared to other CYP450s. Xanthotoxol showed stronger inhibition on CYP3A4 and CYP1A2 compared to other isoenzymes with the IC50 of 7.43 μM for CYP3A4 and 27.82 μM for CYP1A2. Metabolic syndrome (MetS) encompasses metabolic abnormalities that substantially increase risk for chronic illnesses. MetS and stress are closely related; the pathophysiology of MetS involves dysregulated stress response in both the physiological and psychological domains. In an effort to further clarify the relationship between metabolic abnormalities and autonomic dysregulation, we used ambulatory impedance cardiography to examine indicators of cardiac autonomic control (CAC) in a sample of 50 adult primary care patients with and without MetS. Indices of sympathetic and parasympathetic influences on cardiovascular functioning were assessed in the context of psychological stressors and compared across experimental groups and examined in relation …

However we define comics, it is safe to claim that in general they consist of two main components: images and language. With some exceptions, the vast majority of comics include linguistic elements: speech balloons, thought balloons, narrative boxes, sound effects, and ambient language (language used in the background, as on store fronts, t-shirts, restaurant menus, and the like). Comics scholarship examines the language used in comics to say something about narrative, character development, even the nature of comics themselves. We describe a method for simply characterizing the size and shape of a nanopore during solution-based fabrication and surface modification, using only low-overhead approaches native to conventional nanopore measurements. Solution-based nanopore fabrication methods are democratizing nanopore science by supplanting the traditional use of charged-particle microscopes for fabrication, but nanopore profiling has customarily depended on microscopic examination. Our approach exploits the dependence of nanopore conductance in solution on nanopore size, shape, and surface chemistry in order to characterize nanopores. Measurements of the changing nanopore conductance during formation by etching or deposition can be analyzed using our method to characterize the nascent …

Part I of this article delineates the harms to children from marriage bans, harms that the Obergefell Court relied upon to recognize the fundamental right to marry for same-sex couples. Part II explains how Obergefell missed an opportunity to advance the constitutional rights of children by failing to invoke well-established equal protection law. This discussion briefly catalogues the omitted child-centered cases that warranted a more robust analysis of children's rights. This series of cases begins with the Brown v. Board of Education decision, and then turns to a number of post-Brown equal protection cases that explicitly prohibited state practices that …

Attitudes And Experiences With Older Adults: A Case For Service Learning For Undergraduates, Hardeep Obhi, Erin Woodhead

The current study examined whether relationship quality with older adults currently and in childhood, as well as experience with older adults, was associated with biases toward older adults and interest in working with older adults as a possible career area. The authors sampled undergraduate students (N = 753, M = 18.97 years, SD = 2.11 years) from a Northern California university. In hierarchical regression analyses, higher perceived quality of relationships with older adult family members, higher perceived social support, and lower perceived conflict from relationships with older adults was significantly associated with positive attitudes toward older adults. OBJECTIVE: Small intestinal neuroendocrine tumors (SI-NETs) are often detected after they have become metastatic. Using a novel protein array, we identified pathways important in SI-NET metastasis development in surgically resected patients. METHODS: Paired primary tumors and liver metastases from 25 patients undergoing surgical resection for metastatic SI-NETs were harvested. Extracted proteins were separated by sodium dodecyl sulfate gel and multiplex immunoblots were performed with 136 antibodies. Significant Analysis of Microarray was used to select for differentially expressed proteins. A tissue microarray was constructed from 27 archived specimens and stained by immunohistochemistry. Background Irreversible inhibition of Bruton's tyrosine kinase (BTK) by ibrutinib represents an important therapeutic advance for the treatment of chronic lymphocytic leukemia (CLL). However, ibrutinib also irreversibly inhibits alternative kinase targets, which potentially compromises its therapeutic index. Acalabrutinib (ACP-196) is a more selective, irreversible BTK inhibitor that is specifically designed to improve on the safety and efficacy of first-generation BTK inhibitors. Methods In this uncontrolled, phase 1-2, multicenter study, we administered oral acalabrutinib to 61 patients who had relapsed CLL to assess the safety, efficacy, pharmacokinetics, and pharmacodynamics of acalabrutinib. Sleep quality and memory are both relevant topics in today’s society, especially among college students.
Purpose: To determine if there is a correlation between sleep quality and short-term memory including objective and subjective measures.
Methods: This study consisted of 25 participants, 6 males and 19 females, ages ranging from 19 to 22 (20.8±0.8 years), who avoided stimulants, caffeine or other sleep altering drugs for at least eight hours. Virginia’s Warbler (Oreothlypis virginiae) reaches the northeastern limit of its breeding range in the southern Black Hills of South Dakota, where it is considered a species of conservation concern. This population occurs in pine–juniper–shrub habitat in several canyons in extreme western Custer County. Additional such habitat occurs in the southern Black Hills south and east of the documented breeding populations, but these areas have received few systematic surveys, and whether Virginia’s Warbler occupies them was uncertain. With an increasing global cancer burden, new methods of cancer prevention are desired to improve overall survival. In particular, pancreatic cancer (PC) is in desperate need of new strategies to improve the dismal 8% overall 5-year survival rate. Studies have shown aberrant activation of the Hedgehog pathway to be associated with very poor prognosis in multiple cancer types, including pancreatic. Prevention strategies utilizing dietary agents have been shown to regulate multiple cancer hallmark pathways, but little is known regarding their effect on Hedgehog. Graves’ disease (GD) is an organ-specific autoimmune disease, and thyrotropin (TSH) receptor (TSHR) is a major autoantigen in this condition. Since the extracellular domain of human TSHR (TSHR-ECD) is shed into the circulation, TSHR-ECD is a preferentially immunogenic portion of TSHR. Both genetic factors and environmental factors contribute to development of GD. Inheritance of human leukocyte antigen (HLA) genes, especially HLA-DR3, is associated with GD. TSHR-ECD protein is endocytosed into antigen-presenting cells (APCs), and processed to TSHR-ECD peptides. Wide crosses have been used for decades as a method for transferring novel genetic material and traits in plant breeding. Historically, many products of wide crosses require tedious and inefficient surgical embryo rescue prior to embryo abortion to recover single plantlets. We have utilized transgenic switchgrass (Panicum virgatum L. cv Alamo) as a pollen donor in conjunction with antibiotic or herbicide selection for recovery of intra-and interspecific F1 crosses by using developing ovules from the female parent and selecting for embryogenic cultures derived from the in situ immature embryo. Fluorescent nanocrystals, specifically quantum dots, have been a useful tool for many biomedical applications. For successful use in biological systems, quantum dots should be highly fluorescent and small/monodisperse in size. While commonly used cadmium-based quantum dots possess these qualities, they are potentially toxic due to the possible release of Cd2+ ions through nanoparticle degradation. Indium-based quantum dots, specifically InP/ZnS, have recently been explored as a viable alternative to cadmium-based quantum dots due to their relatively similar fluorescence characteristics and size. Many of the Supreme Court’s important holdings concerning campaign finance law are not pure matters of constitutional interpretation. Rather, they are “contingent” constitutional determinations: the Court’s conclusions rest in substantial part on legislative facts about the world that the Court finds, intuits, or assumes to be true. While earlier commentators have recognized the need to improve legislative factfinding by the Supreme Court, other aspects of its treatment of legislative facts—particularly in the realm of campaign finance—require reform as well.
Stare decisis purportedly insulates the Court’s purely legal holdings and interpretations from future challenge. Factually contingent constitutional rulings should, in contrast, …

Psychological distress, including depression and anxiety, has been associated with increased risk for sexual revictimization in youth who have experienced child sexual abuse. The present study utilized assessment information from treatment seeking youth with histories of sexual abuse to explore specific risk indicators for revictimization—risk taking, social problems, maladaptive cognitions, and posttraumatic stress—that may be indicated by self-reported distress. The relationship between initial levels of distress and change in symptoms over a 12-week course of treatment was also explored. Participants were 101 youth referred to a child-focused therapeutic group for victims of sexual abuse, 65 youth referred to an adolescent-focused …

Apoptotic Induction Via Membrane/Dna Damage And Metabolic Inactivation By Synthetic Food Colorants In Allium Cepa Root Meristem, Prajitha Vazhangat, John Ernest Thoppil

The present study investigates the cytotoxic and genotoxic effects of two frequently employed synthetic food colorants, lemon yellow and orange red, using an Allium cepa assay. Uptake of food colorants by root cells at different concentrations and varied exposures significantly altered the cytogenetic system, membrane integrity, mitochondrial function, root growth, and cell division. The food colorants induced significant DNA damage, micronucleus formation, and other severe chromosomal aberrations at genetic level and inhibited cell division as well as root growth. Severe DNA damage, membrane damage, and metabolic inactivation observed in the result are indicative of apoptotic cell death. These results confirm …

Public understanding of science and public engagement around science policy issues takes place within a complex system involving public opinion, values, identities, social groups, media, and social, economic, and political structures and influences. This paper offers a theory of agency that bridges the psychological and sociological and thereby offers a theoretical framework for understanding the system underlying public understanding of science, support for science, and the implications of public engagement. Agency theory integrates elements of well-established social science theories into an understanding of human agency. Strategies for improved public engagement are discussed.

Phycocyanin is an abundant protein-pigment complex in many photosynthetic microbes and the feedstock for several emerging products in the food and pharmaceutical industries. The purpose of this project was to research the effectiveness of sonication technology to improve the efficiency and yields of phycocyanin production from the cyanobacterium Spirulina platensis. Sonication is the application of sound energy to agitate and break particles or cells. When used to augment the current methods of phycocyanin production, it has the potential benefit of reducing cell extraction volumes and increasing the yield of phycocyanin. Insects with complex life cycles provide an ideal system for investigating the relationship between sex-skewing bacterial symbionts and the secondary loss of sexual reproduction. This research utilized the insect family Adelgidae, a group notorious for having peculiar and complex life cycles. In this family, some species have lost their sexual generation entirely, trapping them in asexual reproduction. Additionally, these insects are known to harbor multiple symbionts, both obligate and facultative. The array of facultative symbionts that have been detected includes the infamous sex-skewing symbiont Wolbachia. Many insects live on nutrient-poor diets; to compensate, they have evolved obligate associations with microorganisms that are transmitted directly between generations. The bacterial associates of some Hemiptera insects have been identified and understood, but several remain unknown. Our objective is to identify the diversity of bacterial endosymbionts in Issidae using modern microscopy and molecular methods, and compare their identities to known symbionts from related insects. To do so, we will extract DNA from insect samples, amplify bacterial symbiont 16S genes, sub-clone PCR products, sequence the products, and determine their phylogenetic relationships to other insect symbionts and free-living bacteria. Tobacco mosaic virus (TMV), named for the plant it was first discovered in, can infect hundreds of different species. It has no known invertebrate vectors, but TMV is spread mechanically through contact with other plants, clothes, tools, contaminated soil and seed borne. While TMV is known to be exceptionally stable, it has been reported that treatment of tools with powdered milk can significantly reduce the infection rate of the virus. While there is a seed treatment available for conventional agriculture, there is no seed treatment available for organic production.
