Kimmel Cancer Center Faculty Papers
Maintaining thiol homeostasis is an imperative for cancer cell survival in the nutrient-deprived microenvironment of solid tumors. Despite this metabolic vulnerability, a selective approach has yet to be developed to disrupt thiol homeostasis in solid tumors for therapeutic purposes. In this study, we report the identification of 2-mercaptopropionyl glycine disulfide (TTL-315) as a novel antimetabolite that blocks cell survival in a manner conditional on glucose deprivation. In the presence of glucose, TTL-315 lacks cytotoxic effects in normal cells where it is detoxified by reduction to 2-mercaptopropionyl glycine, a compound with known clinical pharmacologic and safety profiles. In several rodent models …

RNA processing and turnover play important roles in the maturation, metabolism and quality control of a large variety of RNAs thereby contributing to gene expression and cellular health. The TRAMP complex, composed of Air2p, Trf4p and Mtr4p, stimulates nuclear exosome-dependent RNA processing and degradation in Saccharomyces cerevisiae. The Mtr4 protein structure is composed of a helicase core and a novel so-called arch domain, which protrudes from the core. The helicase core contains highly conserved helicase domains RecA-1 and 2, and two structural domains of unclear functions, winged helix domain (WH) and ratchet domain. How the structural domains (arch, WH …

Background: Plant defense traits require resources and energy that plants may otherwise use for growth and reproduction. In order to most efficiently protect plant tissues from herbivory, one widely accepted assumption of the optimal defense hypothesis states that plants protect tissues most relevant to fitness. Reproductive organs directly determining plant fitness, including flowers and immature fruit, as well as young, productive leaf tissue thus should be particularly well-defended. To test this hypothesis, we quantified the cyanogenic potential (HCNp)—a direct, chemical defense—systemically expressed in vegetative and reproductive organs in lima bean (Phaseolus lunatus), and we tested susceptibility of these …

Toward A Future Adult Learning Community: If Socrates Had A Pc, Michael C. Pickett
Bollinger-Rosado Teaching & Learning Effectiveness Symposium Proceedings
The integration of educational technologies in adult learning environments is viewed from three distinct perspectives. The three perspectives are the structuralist, the means-end, and finally, a view of technology as an extension of oneself based in the situatedness of an experiential-type learning environment.
The various constructs of adult learning are reviewed and a framework for future adult learning communities is proposed that identifies the learner and educator provided components needed for an effective adult learning experience.

The Contribution Of Cancer Associated Fibroblasts In Breast Cancer Progression, Sanket H. Shah
Open Access Dissertations
Cancer Associated Fibroblasts (CAFs) are an integral component of the TME, and the majority of breast tumor stroma is comprised of CAFs. Multiple juxtacrine and paracrine interactions occur between cancer cells and CAFs that direct tumor progression. These interactions occur via soluble agents, including cytokines, hormones, growth factors, and secreted microRNAs. Some of these soluble agents induce MAPK activation in tumor cells. Our lab has established that MAPK activation in tumor cells is one of the key mechanisms involved in repression of ER. MAPK activation in tumor cells leads to alteration in miRNA expression. We previously identified a microRNA signature …
