Spoilage microorganisms cause food waste and loss of quality. While the foodborne pathogen outbreaks lead to thousands of hospitalizations and deaths. Essential oils (EOs), plant extracts, possess the required antimicrobial activities and thus their usage stands out as a feasible approach for controlling the undesirable bacterial growth in food systems. However, EOs are highly volatile and lose their activity upon exposure to environmental conditions. Recent research on species that are both uncommon and unfamiliar to the public has shown that wildlife value theory may not be applicable to the management of all species. This stands in contrast to studies on charismatic megafauna like deer, bear and elk, in which wildlife value orientations could be used to predict the acceptability of various management actions. Nutrient abatement programs, originally enacted to counter eutrophication have successfully suppressed nutrient loads and relative primary production in marine and freshwater systems. Recently, the additive impacts of invasive filter feeders have further reduced offshore primary production in several aquatic systems throughout the world. It has been hypothesized that the biological activity of these invasive species may sequester nutrients within the nearshore benthic environment, creating steep gradients in primary production between nearshore and offshore habitats. Diverse mechanics of the F-actin cytoskeleton mediate essential behaviors of cells, including cell division, migration, and shape change. Force generation by motor proteins and the resultant morphological change of cytoskeletal networks govern cellular processes such as migration and division. Cell stiffening and softening under external mechanical stimuli regulate cell shape. In this thesis, interplay between various cytoskeletal components during these processes is investigated using an agent-based computational model to elucidate mechanical factors underlying these processes. Radiant frying is an alternative technique to traditional immersion frying. While enjoyed by consumers, immersion fried foods are typically high in fat. Radiant frying uses infrared energy to produce finished products with similar sensory characteristics to traditional frying, but with significantly less fat. This research compared the efficacy of radiant and immersion frying using hash brown patties as a model food matrix by measuring temperature profiles, heat flux profiles, moisture and oil contents, peak shear force, color, mottling, and crust thickness as well as executing semi-trained and untrained sensory evaluations. Rats initially fear humans which can lead to negative affect, poor welfare, and difficult handling. Also, modeling and measuring positive affect states in rats can pose an additional challenge. Heterospecific play, or “tickling,” is a handling habituation technique that mimics rat rough-and-tumble play that is being used to study positive affect. It can also be used to reduce fear of human, improve welfare, and elicit a positive affect state. However, current studies implementing the technique in laboratory rats use a wide variety of protocols to achieve differential results. Bone morphogenetic proteins (BMPs) drive a range of cellular processes especially in the early stages of embryonic development. This family of proteins acts as one of the most important extracellular signals in development pattern formation across the animal kingdom. Improper lawn fertilization with excess N or phosphorus has the potential to contaminate both surface and groundwater. Invasive Burmese pythons are established in the Everglades and are altering the ecology of southern Florida. Their distribution in Florida is expanding northward into more urbanized and fragmented habitats. An understanding of the suitability of habitat throughout southern Florida for Burmese pythons and their interaction with Florida’s landscapes through movement behavior is vital for predicting the python’s ability to persist in habitats outside of the Everglades. In this thesis, we use ecological modeling to predict habitat suitability and to investigate personality-dependent dispersal.

The dopamine transporter (DAT) is a monoamine transporter that regulates dopamine (DA) neurotransmission by clearing DA from the synapse. DAT is acted on by a number of psychostimulant drugs, which block reuptake and potentiate DA signaling. Several inhibitors of DAT, both amphetamines and cathinones, also cause reversal of transport. Previous research has shown the importance of a conserved salt bridge in another monoamine transporter, the serotonin transporter, in amphetamine-induced efflux. In our studies here, we engineered a mutant Drosophila melanogaster DAT (dDAT D475N) designed to interrupt this salt bridge and examine the effects on amphetamine- and cathinone-induced efflux in vitro. …

Non-typhoidal Salmonella is the leading cause of foodborne illness in the United States, resulting in about 20,000 hospitalizations and nearly 380 deaths annually. The meat processing industry has been especially plagued by Salmonella, from meat-inherent sources and more alarmingly, cross-contamination. For ready-to-eat (RTE) meat products specifically, this can cause significant problems in processing facilities ensuring safe product for consumption, resulting in foodborne illness.

Laboratory mice are nocturnal, spending most of their daylight hours asleep. But they live in the diurnal world of human investigators and husbandry staff, who primarily work during this rest period. In humans, lack of sleep or sleep that occurs outside the normal circadian sleep period (as in shift work) has adverse effects. These include increased risk of cardiovascular disease, cancer, metabolic disorder, mood disorders, type II diabetes, and obesity. Listeria monocytogenes (Lm) is a foodborne pathogen, found ubiquitously in nature, and has a high morbidity rate among immunocompromised individuals, the elderly, and especially pregnant women and their fetuses resulting in abortion, stillbirth, and neonatal infection. There are currently no preventative medical interventions against Lm infection. The seventeen species and two subspecies described in the genus Calyptra are known to be obligate fruit piercers, with some species being of economic importance. Males within the genus have not only been observed piercing their fruit hosts, but have also been documented to occasionally feed on mammalian blood. Pro-Vitamin A (proVA) carotenoids, which are converted into retinol (Vitamin A) in the human body, have been the subject of human nutrition studies and are a target for biofortification of staple crops. Historically, β-carotene has been the principal target for enhancing levels of proVA, yet there is recent interest in enhancing the proVA carotenoid β-cryptoxanthin. Hypoxia inducible factors (HIFs) are central mediators of cellular responses to fluctuations of oxygen, an environmental regulator of stem cell activity. Muscle satellite cells are myogenic stem cells whose quiescence, activation, self-renewal and differentiation are influenced by microenvironment oxygen levels. However, the in vivo roles of HIFs in quiescent satellite cells and activated satellite cells (myoblasts) are poorly understood. Expression analyses indicate that HIF1α and HIF2α are preferentially expressed in pre- and post-differentiation myoblasts, respectively. An experimental study on the effect of helical strakes on vortex induced vibrations and the lock-in phenomenon in High Mast Lighting Towers (HMLTs) is investigated. Two multi-sided tapered scaled models are clamped in place in a subsonic wind tunnel that is equipped with a hot-wire sensor and a traverse mechanism. Phytophthora blight is a serious threat to the Midwest vegetable industry, because the oomycete pathogen responsible for this disease, Phytophthora capsici, has a wide host range, can spread quickly in fields, and produces resilient oospores that can survive in soil for years. Phytophthora capsici has become resistant to commonly used fungicides and resistant crop varieties are rare. Aflatoxin contamination in feed and feed ingredients is of high worldwide prevalence, posing serious risks to the livestock industry. A series of studies were conducted to better understand the comprehensive impact of aflatoxicosis and its interrelationships with dietary modifications in broiler chicks and ducks. Results of these studies emphasize the many factors that the severity of aflatoxicosis depends on, including animal species, age, presence of mycotoxin adsorbent, and concentration of major dietary nutrients. Proxy variables from palaeolimnological studies of lakes in the Prairie Pothole Region of North America have been used to infer large oscillations during the late Holocene between longer periods of high-salinity–dry conditions and shorter periods of low-salinity–wet conditions producing a normative pattern marked by the absence of hydrological stability. Studies of the historical rise in lake level at Devils Lake have identified 1980 as a transition point between two such hydroclimatic modes. Siderophores are low-molecular weight species utilized by bacteria for the sequestration of iron, an essential nutrient. Siderophores and their cognate receptors are considered to be virulence factors, due to their prominent role in pathogenicity. The work presented here focuses on ferrioxamine (FOx) as an “immutable” ligand for pathogen detection. A number of bacterial strains expressing high-affinity FOx receptors were identified by a proteomic BLAST search, and screened against microarrays patterned with FOx conjugates for detection using label-free optical imaging. Mass spectrometry (MS) imaging is a powerful investigation technique for a wide range of biological applications such as molecular histology of tissue, whole body sections, and bacterial films , and biomedical applications such as cancer diagnosis. MS imaging visualizes the spatial distribution of molecular ions in a sample by repeatedly collecting mass spectra across its surface, resulting in complex, high-dimensional imaging datasets. This dissertation consists of two studies at the United States Military Academy. Both studies involve the use of Course-based Undergraduate Research Experiences (CUREs). These experiences give students the ability to engage in undergraduate research at an early point in their academic career by replacing traditional laboratory activities with semester-long research projects. Both studies show an implementation of this type of instruction from the Center for Authentic Science Practice in Education (CASPiE). Study 1 shows the specific method of implementation at the military academy and explores learning-based outcomes. Primarily the outcome of critical thinking is demonstrated.
