Interrogation Of Possible Imaging Conditions For Radiation Sensitive Metal Organic Frameworks In Transmission Electron Microscopes, Wei-Yu Pan
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Although transmission electron microscopy (TEM) is extremely powerful at the characterization of materials on the nano- and atomic scales, the advantages introduce accompanying drawbacks. That is, high energy electrons offer the structural information while imparting radiation damage to the samples. Metal organic frameworks are a new class of materials that have potential applications in gas storage and catalysis. A direct imaging of their lattice structures is highly desired. However, these samples are extremely radiation sensitive. In order to characterize their structures, low-dose TEM need to be performed. To date, there is a lack of a systematic of radiation study of …

Synthesis Of Binary Nanocrystalline Aluminum Alloys Through High Energy Ball Milling, Lacey Lynn Schwab
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Nanocrystalline metals have useful mechanical properties such as high strength, improved wear resistance, and longer fatigue life; however, they are relatively unstable – grain boundary doping is a viable method towards stabilization [1-3]. After comparing the work of Murdoch and Schuh, that used a thermodynamic model to estimate grain boundary segregation enthalpy to the experimental work of Umbrajkar et al. | it was realized that mechanical alloying is a processing route for grain boundary stabilization [10,11]. Whether or not chemical mixing has occurred is a good indicator of a powder system’s potential for grain boundary doping. 99Al1Si, 99Al1Mg, 99Al1Zr, and …

Fabrication Of A Novel Biosensor For Macromolecules Detection Through Molecular Imprinting Technique, Yingjie Yu
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
There is an increasing need for precise molecular detection as a diagnostic tool for early identification of diseases, pathogens, and abnormal protein levels in the body. Typical chemical analytical methods are generally costly, unstable, and time-consuming. Molecular imprinting (MI) technique, based on the “lock and key model†, could be a simple method to overcome those shortcomings. In this study, a self-assembled monolayer (SAM) was employed as a platform to fabricate MI biosensor for detection of macromolecules. I demonstrated that, when the monolayer was formed on a rough surface, this method was in fact templating molecules in three dimensions, and hence …

Engineering Interface And Analysis In Orthopedic Surgery: Resection Of Bone Tumors And Reconstruction Using Biomimetic Implants, Carlos Gabriel Helguero Alcivar
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
As healthcare grows more advanced, the next step is an individualized approach to patient care. Skeletal tumor resection provides complex challenges for surgeons. The goal of providing an accurate wide resection in sarcomas from preoperative plans while trying to preserve as much functionality remains a difficult task. We propose to leverage new devel- opments in 3-D printing, computer aided design (CAD), and finite element analysis (FEA) to design and manufacture personalized cutting guides in order to increase the accuracy of childhood bone cancer resections, minimize post-resection fracture risk, and maximize al- lograft osteointegration. On the other side, after proper resection, …

A Medical Decision Model For Tinnitus Treatment Assessment, Hong Don Ihn
A Medical Decision Model For Tinnitus Treatment Assessment, Hong Don Ihn
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Tinnitus means objective somatosounds, and subjective auditory or sensorineural perception of noise without external physical sounds. There are several tinnitus treatments, but it is difficult to ascertain how otorhinolaryngologists rank the alternatives of treatments due to the insufficiency of available evidence and standards. A hierarchical decision model (HDM), a popular multi-criteria decision analysis (MCDA) method, can help to implement a comprehensive assessment of tinnitus treatments and rational decision-making to select the appropriate tinnitus treatments. The HDM considers a holistic approach using multiple dimensions, criteria and expert judgments to acquire the relative ranking of candidate tinnitus treatments. The four dimensions selected …
The Stream Of Consciousness In Richard Foreman's Lava, Jungyoon Han
The Stream Of Consciousness In Richard Foreman's Lava, Jungyoon Han
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Richard Foreman’s Lava (1989) is chosen to be studied in terms of stream of consciousness as one of the representative plays of Ontological-Hysteric Theatre. Given the nature of stream of consciousness, continuation of experiences, this play is analyzed through its manifestation process itself. The stream of consciousness is found to consist of three main features; Fragmentation, Mismatch, and Rituality. These features are not only belong to Richard Foreman’s Lava but also have been descended and regarded as the main characteristics of avant-garde theaters. This study relates the manifestation of stream of consciousness in Lava with the avant-gardism. It pursues to …
Phic31 Integrase As A More Efficient Method Of Transgenesis In Zebrafish, Andrew Scortea
Phic31 Integrase As A More Efficient Method Of Transgenesis In Zebrafish, Andrew Scortea
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Zebrafish transgenesis is conventionally performed via the tol2 transposable element system as it is the most straightforward, providing quick and easy transgenesis. Despite being commonly used in the zebrafish community, tol2 mediated transgenesis has issues that cause workflow slowdowns and delays. The biggest of these issues are the random nonspecific integrations which can occur multiple times within a genome. In order to generate a stable transgenic line with a single insertion, one must perform several generations of outcrosses and subsequent screening. Thus, while tol2 transgenesis is effective at generating founder animals, a considerable amount of time and labor is required …
Transcriptional Regulation Of Mmp-9 And Mmp-14 By P53 And Its Mutant R280k, Yingjiao Xue
Transcriptional Regulation Of Mmp-9 And Mmp-14 By P53 And Its Mutant R280k, Yingjiao Xue
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Breast cancer is the most common cancer in women and metastasis is the primary cause for mortality. Matrix metalloproteinases (MMPs) play important roles in cancer cell migration and invasion. Tumor suppressor gene TP53 is the most frequently mutated gene in cancer, and p53 mutation occurs in invasive breast cancer with higher frequency than in non-invasive breast cancer. However, the nature of relationship between p53 and MMPs remains inconclusive. Here we show that wild type p53 could repress transcriptional activity of MMP-9 and MMP-14, while the DNA-contact mutant p53 R280K could upregulate the transcription of MMP-9 and MMP-14. This regulation might …
Aging-Related Elevation Of Sphingoid Bases Shortens Yeast Chronological Life Span By Compromising Mitochondrial Function, Jae Kyo Yi
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Aging is the biological process of growing older in a deleterious sense, senescence. However, what we have gained only comprises a partial and incomplete understanding of the fundamental molecular mechanisms. Increasing evidence suggests that dysregulation of the metabolism of sphingolipids including sphingoid bases (SBs, long-chain bases) might play an important role in the aging. We know neither how SBs are regulated during yeast aging nor how they, in turn, regulate it. Herein, I demonstrate that the yeast alkaline ceramidases (YPC1 and YDC1) and SB kinases (LCB4 and LCB5) cooperate in regulating SBs during the aging process and that SBs shorten …
Using Quantitative Silac Proteomics To Identify Mek1 Substrates During Yeast Meiosis, Raymond Theodore Suhandynata
Using Quantitative Silac Proteomics To Identify Mek1 Substrates During Yeast Meiosis, Raymond Theodore Suhandynata
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
The exchange of reciprocal information between homologous chromosomes in conjunction with sister chromatid cohesion ensures proper segregation of homologous chromosomes during Meiosis I. Improper segregation of homologous chromosomes during meiosis leads to chromosomal imbalance, which in humans can lead to genetic disorders such as Down syndrome (Trisomy 21) and Turner syndrome (monosomy of the X chromosome). Meiotic recombination begins with the initiation of double-strand breaks (DSBs) and the biased repair of these breaks by the homologous chromosome. This inter-homolog (IH) bias is upheld by the meiotic recombination checkpoint kinase Mek1. Mek1 is a meiosis specific serine/threonine kinase that prevents cells …
Evaluation Of Contrast Agents Used In Computed Tomography, Michael Joseph Bonvento
Evaluation Of Contrast Agents Used In Computed Tomography, Michael Joseph Bonvento
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Contrast agents have been routinely employed in radiographic applications for more than 80 years and in computed tomography (CT) for more than 50 years. These agents have high atomic number increasing the photoelectric interaction probability thereby reducing photon intensity projected through the contrast containing structure and improving available image contrast. A wide variety of contrast agents have been examined over the years, however, iodine and barium containing agents make up the vast majority of applications. Other agents are or may become available which may find utility due to reduced toxicity, better characteristics for imaging or dose reduction. This is particularly …

Diffuse Correlation And Optical Spectroscopies For The Monitoring Of Spinal Cord Hemodynamics, Angela Sofia Kogler
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Spinal cord ischemia, caused by a prolonged loss of blood flow to the spinal cord, can occur as a result of direct trauma to the spinal cord or as a complication of major vascular, spine and spinal cord surgery, and surgeries for the stabilization of spinal cord trauma, often resulting in partial or complete paresis/paralysis. Current electrophysiological methods employed to detect spinal cord ischemia, namely somatosensory and motor evoked potentials, are indirect, temporally insensitive, nonspecific, and cumbersome. There is no technology currently available that can directly and immediately detect the onset of spinal cord ischemia, nor provide immediate feedback on …
Oxidized Graphene Nanoparticles As A Delivery System For The Bioactive Sphingolipid Ceramide, Cassandra Suhrland
Oxidized Graphene Nanoparticles As A Delivery System For The Bioactive Sphingolipid Ceramide, Cassandra Suhrland
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
In recent years there has been a growing appreciation for the active functions certain lipids have in cellular metabolism. These "bioactive lipids" have been found to be an important part of many cell signaling processes. In particular, the bioactive sphingolipid ceramide has emerged as an important player in cancer biology. Dysregulation of ceramide metabolism has been shown to impart resistance to drug and radiation treatment in certain types of cancer, with low levels of specific ceramide species conferring resistance to apoptosis. It has also been shown that restoring levels of these ceramides in cancer cells through exogenous delivery also restores …

Carbon Nanotubes As Localized Microwave Hyperthermia Agents, Shawn Xie
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Carbon nanotubes (CNTs) in recent years have been cited to provide great potential for biomedical applications such as drug delivery, imaging and hyperthermia. In particular, it has been shown that they are able to interact with a broad array of electromagnetic radiation, including microwaves. The latter is of great interest for cancer imaging and therapy due to its non-ionizing nature and unique balance of spatial resolution and penetration depth, as well as ability to thermally ablate tumor tissue. Combined with microwaves, CNTs can serve as a platform with high selectivity of microwave absorption, enabling selective microwave thermal therapy, and effective …

Synthetic, Medicinal, And Computational Studies On Novel Anticancer, Antitubercular, And Anti-Nociceptive Agents, Longfei Wei
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Multidrug resistance is a major obstacle to effective cancer chemotherapy. New-generation taxoids SB-T-1214 and SB-T-1216 have shown two to three orders of magnitude higher potency than those of paclitaxel and docetaxel against multidrug-resistant cancer cell lines. A structure-activity relationship (SAR) study has been conducted in order to test which moiety of the new-generation taxoids account for the high potency against multidrug resistant cancer cell lines. In order to selectively deliver new-generation taxoids into tumor cells, tumor-targeted drug delivery systems with a smart methyl-branched disulfide linker have been developed in the Ojima laboratory. The metabolic stability of this disulfide linker has …

Don’T Look Now: The Child In Horror Cinema And Media | Don’T Look Now: The Child In Horror Cinema And Media, Hans Staats
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
This dissertation examines the definition of the monstrous child in horror cinema and media. Spanning from 1955 to 2009 and beyond, the question of what counts as a monstrous child illuminates a perennial cinematic preoccupation with monstrosity and the relation between the normal and the pathological. Films like The Night of the Hunter (Charles Laughton, 1955), The Bad Seed (Mervyn LeRoy, 1956), Village of the Damned (Wolf Rilla, 1960), The Innocents (Jack Clayton, 1961), Don’t Look Now (Nicolas Roeg, 1973), Profondo rosso/Deep Red (Dario Argento, 1975), Carrie (Brian De Palma, 1976), Halloween (John Carpenter, 1978), Child’s Play (Tom Holland, 1988), …

Othering The Mother: Traumatic Effects Of Motherhood On The Formation Of Identity In Toni Morrison’S Beloved | Othering The Mother: Traumatic Effects Of Motherhood On The Formation Of Identity In Toni Morrison’S Beloved, Gabriella Elizabeth Greco
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
As a text based heavily on tropes from both American Gothic literature and the slave narrative genre, Toni Morrison’s novel Beloved serves as a complex and delicate commentary on racial ambiguities and the psychologically haunting effects of African American trauma. These ambiguities simultaneously serve as reasons for and consequences of the various traumas that strain the interpersonal relationships between the female characters. Aside from the most blatant and gruesome trauma of the novel—the infanticide that Sethe commits as an act of mercy for her unnamed child—many of the recurring traumatic memories evoke the challenges of motherhood during slavery. | As …

From Text To Screen: John Green And The Rise Of Digital Literacy, Jessica Pond
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Social media is rapidly becoming more prevalent in modern society. Businesses, politicians, and educators are using social media as an increasingly important tool of communication and information. In my thesis, I examine the implications of the growing importance of social media on young adult authors and the way that literacy is perceived. While social media is often seen as existing purely for entertainment, many educational professionals are bringing aspects of social media into classrooms as instructive and informational tools. Through close examination of the works of John Green and his activities on social media, this thesis addresses the ways in …

Digitizing The Novel, 1987-2010, Bradley Reina
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
The novel is digital, it was digital, and it will be digital. Most authors have written on word processors and most publishers have made books with some form of desktop publishing software since the early 1990s. The first novels for digital display were written and published in the late 1980s. From a literary perspective, the question is whether such digital-born literature translates into palpable changes in the novel form, why, and how. Previous theories of the meeting of digital technology and literature have all too often presented a predetermined fate for this pairing—an essentialist vision of literature, technology, or both. …

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
With the concern for human rights always present to this project, I explore in the initial chapters the precariousness of the first-person singular subjective “I†in both testimonial and fictional texts and its bearing on human rights. In practice, first-person narratives, whether as life-writing or fictional, are textual spaces in which writers’ concerns that deal with human conditions are frequently articulated. By examining the first-person narrative function in selected texts of Gertrude Stein, Virginia Woolf, James Baldwin, and Samuel Beckett, this dissertation shows how conceptual imaginings of the concept of the human and human embodiment are unsettled. These exemplar modernist …

Jane Austen From Page To Screen: How Current Society And " Pride And Prejudice" Have Shaped " Northanger Abbey," " Persuasion," And " Mansfield Park" On Film, Stephanie Ann Reese
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
While Jane Austen’s novels were published over two hundred years ago, her stories continue to captivate audiences through print and the screen. There seems to be a growing tradition of adapting certain novels, such as Pride and Prejudice, over other ones like, Northanger Abbey, Persuasion, and Mansfield Park. In this paper I explore how the former three novels differ from Pride and Prejudice from their heroines and heroes to the romantic relationships between the two protagonists. Then I take into consideration the large impact of Colin Firth’s Mr. Darcy in influencing Austen’s other heroes such as Captain Wentworth, Henry Tilney …
Thermal And Near-Infrared Analyses For Understanding Martian Surface Mineralogy Through Orbital Measurements, Laboratory Studies And Statistical Models, Cong Pan
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Understanding the geologic evolution of Mars requires a detailed characterization of Martian surface compositions and their geologic settings. Because much of the Martian surface is covered with regolith or unconsolidated materials, in this work I focused on key locations in which to characterize Martian crustal materials as well as assess the degree of influence from physical sorting processes on measured surface compositions from orbit. In Chapter 2, I identified and characterized 26 crater central peaks with distinctive spectral signatures from surrounding plains with thermal infrared and visible/near-infrared data, based on a global survey of Martian impact craters between 10-200 km …

The Answer Was Cooperative: How Anthropologists In Peru Redefined The "National Problem," 1948-1975, Ying-Ying Chu
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
The scholarly literature has often depicted the agricultural cooperative model implemented during the Peruvian Agrarian Reform in 1969 as an imposed policy. This dissertation challenges the state-centric view by exploring intellectual and policy debates, which were strongly influenced by grassroots developments, between 1948 and 1975. It shows how the cooperative model evolved from a radical project of the left into a well-accepted strategy of rural development and flourished in Peru during the post-World War II era. This study highlights the critical role played by a new generation of anthropologists and other social scientists, who mediated between the government, international aid …

Words That Make The Man: Major League Baseball, The Media, And American Manhood, 1945-1962, Elizabeth O'Connell Gennari
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Baseball and the media have been linked throughout the American national pastime’s history. Its rise as a professional game coincided with the Circulation Wars between Joseph Pulitzer and William Randolph Hearst, who used sports as one means of selling newspapers. The expansion of radio programming linked listeners across the nation, and made Babe Ruth one of the first national celebrities. Baseball likewise helped fill airspace in the early days of television, and allowed viewers unable (or unwilling) to attend a ballpark the opportunity to see players like Mickey Mantle and Willie Mays perform. This dissertation examines the role of the …

An Investigation Of Pelagophyte Ecology, Yoonja Kang
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Pelagophytes are important picoplankton in the open ocean and in coastal waters where Aureococcus anophagefferens and Aureoumbra lagunensis form ecosystem-disruptive brown tides. Despite decades of research, factors facilitating brown tides are not completely understood. This dissertation is a comparative study of pelagophytes assessing processes facilitating brown tides and transcriptional similarities among open ocean and coastal pelagophytes. During my dissertation, top-down (e.g. grazing) and bottom-up (e.g. nutrients) processes shaping phytoplankton communities were examined during brown tides in the Indian River Lagoon, Florida, USA. While moderate levels of ammonium promoted Aureoumbra growth over picoprokaryotes, grazing pressure on Aureoumbra was near absent permitting …

Ancient Greek Women: Weavers, Painters And Patrons, Mikaela Batista
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Linda Nochlin and the Guerrilla Girls, amongst others, have advocated for the equal representation of women artists. I have found that this contemporary concern of gender inequality can be traced back to Classical Antiquity. The patriarchal structure that women lived in assisted in the construction of female roles within society. Through these roles we can examine their contributions to the art world of Ancient Greece as weavers, painters and patrons. The analysis of their artistic significance led to their historiography by writers such as Pliny the Elder and Giovanni Boccaccio. Their written works, created in Ancient Rome and 14th century …
Increasing College Students’ Interest And Engagement In Stem: A Comparison Of Strategies For Challenging Stem Stereotypes | Increasing College Students’ Interest And Engagement In Stem: A Comparison Of Strategies For Challenging Stem Stereotypes, Jiyun Elizabeth Shin
Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)
Increasing science, technology, engineering, and mathematics (STEM) graduates has become an important part of the education agenda in the U.S. in recent years. Stereotypes about STEM (i.e. | belief that STEM abilities are innate, and that European American men are best suited for STEM) have been identified as one of the critical factors that may contribute to low recruitment and retention of STEM students. Drawing from the literatures on biological essentialism and role models, this study compared different strategies for challenging STEM stereotypes among undergraduate students in STEM and non-STEM fields. STEM stereotypes were challenged directly with research articles that …
